Development of a Plasmid Vector(pSS4) for the Use in Animal Transgenesis (유전자 변환 동물 생산을 위한 Plasmid Vector(pSS4)의 개발)

  • Transgenic animals have become an important tool in the basic and applied sectors of genetic and biomedical sciences. In particular transgenes provide clear-cut markers in the spatial and temporal analysis of developing embryos for the understanding of developmental mechanisms. For the long-term use of plasmid vector in a particular purpose it would be necessary to develop one's own vector system which can be properly expressed in eukaryotic system. Plasmids were constructed from ori region of pUC19 and early region of SV40 through various steps. LacZ gene coding for $\beta$-galactosides was fused to early gene of SV40 in translational in-frame. Poly(A) tailing site of SV40 was inserted at the 3' lacZ so that initiation, elongation and terminatin be controlled by SV40 transcription (pSS4). Biological function of the constructed pSS4 was demonstrated via microinjection of the plasmid into fertilized loach eggs and subsequent detection of $\beta$-galactosidase in developing embryos. The result indicate that the newly constructed pSS4 is functional in a eukaryotic system in vivo. Thus pSS4 may be used as an efficient tool for the study of embryogenesis and a basic carrier for various genes for animal transgenesis.

Characterization of X-ray Detector for CCD-based Electronic Portal Imaging Device (CCD를 이용한 전자포탈영상장치의 엑스선 계측기 특성에 관한 연구)

  • 금속판/형과스크린 계측기와 CCD 카메라를 이용한 방사선영상장치가 현재 전자포탈영상에 널리 쓰이고 있다. 이 장치의 효율적인 영상획득을 위해 계측효율이 좋고, 공간분해능력이 뛰어난 금속판/ 형과스크린 계측기의 두께를 최적화할 필요가 있었다. 이 논문에서는 금속판과 형광스크린의 두께가 계측효율과 공간분해능에 미치는 영향이 연구되었다. 이 결과는 치료 엑스선 영상장치에 쓰일 수 있는 금속판/형과스크린 계측기의 최적화된 두께를 결정하는데 쓰일 수 있다. 몬테칼로 방법을 이용하여 계산한6 MV 선형가속기에서 발생되는 엑시선의 에너지 스펙트럼을 바탕으로, 여러 가지 두께의 금속판/형광스크린에 대하여 계측효율과 공간분해능을 계산하였고, 이를 실험을 통해 검증하였다. 계측효율은 입사된 엑스선의 에너지가 형광스크린에 흡수된 비율로 계산되며, 공간분해능은 흡수된 에너지의 공간 분포를 통해 계산되었다. 계측효율은 금속판의 두께에 의해, 공간분해능은 형광스크린의 두께에 의해 결정될 수 있음을 본 연구를 통해 확인할 수 있었고, 이로써 특정이용에 관련된 금속판/형광스크린의 두께에 대한 서로 보상 (trade-off) 관계에 있음을 계산과 측정결과를 통해 확인할 수 있었고, 이로써 특정이용에 관련된 금속판/형광스크린 계측기의 최적화된 두께를 산출할 수 있게 되었다. 계산을 바탕으로 CCD를 이용한 전자포탈영상장치의 시작품을 설계 및 제작하였고 팬텀을 이용하여 영상을 얻었다. 단일 프레임 영상은 노이즈가 많으나, 프레임 평균 방법을 이용하여 영상의 질을 향상시킬 수 있었다.

Damage state evaluation of experimental and simulated bolted joints using chaotic ultrasonic waves

  • Ultrasonic chaotic excitations combined with sensor prediction algorithms have shown the ability to identify incipient damage (loss of preload) in a bolted joint. In this study we examine a physical experiment on a single-bolt aluminum lap joint as well as a three-dimensional physics-based simulation designed to model the behavior of guided ultrasonic waves through a similarly configured joint. A multiple bolt frame structure is also experimentally examined. In the physical experiment each signal is imparted to the structure through a macro-fiber composite (MFC) patch on one side of the lap joint and sensed using an equivalent MFC patch on the opposite side of the joint. The model applies the waveform via direct nodal displacement and 'senses' the resulting displacement using an average of the nodal strain over an area equivalent to the MFC patch. A novel statistical classification feature is developed from information theory concepts of cross-prediction and interdependence. This damage detection algorithm is used to evaluate multiple damage levels and locations.

  • The segmentation of moving object in video sequence is a core technique of intelligent image processing system such as video surveillance, traffic monitoring and human tracking. A typical method to segment a moving region from the background is the background subtraction. The steps of background subtraction involve calculating a reference image, subtracting new frame from reference image and then thresholding the subtracted result. One of famous background modeling is Gaussian mixture model (GMM). Even though the method is known efficient and exact, GMM suffers from a problem that includes false pixels in ROI (region of interest), specifically shadow pixels. These false pixels cause fail of the post-processing tasks such as tracking and object recognition. This paper presents a method for removing false pixels included in ROT. First, we subdivide a ROI by using shape characteristics of detected objects. Then, a method is proposed to classify pixels from using histogram characteristic and comparing difference of energy that converts the color value of pixel into grayscale value, in order to estimate whether the pixels belong to moving object area or shadow area. The method is applied to real video sequence and the performance is verified.

  • A study to have read does an improvement of an error restoration technology based on the edge direction interpolation that a stop image cared for inside frame correction more than with an image restoration way of a transfer error or with an aim. A way proposed to is based on edge direction detection method of a block utilizing the edge direction which will adjust a part damaged a sweater to a remaining part here. The rest of error pixel used non linear Midian filter for process later data information by the final stage and did interpolation. The examination result shows a good recuperation tendency and low accounts time of a way proposed to realization possibility of a real time image processing.

Channel Error Detwction and Concealment Technqiues for the MPEG-2 Video Standard (MPEG-2 동영상 표준방식에 대한 채널 오차의 검출 및 은폐 기법)

  • In this paper, channel error characteristics are investigated to alleviate the channel error propagation problem of the digital TV transmission systems. First, error propagation problems, which are mainly caused by the inter-frame dependancy and variable length coding of the MPEG-2 baseline encoder, are intensively analyzed. Next, existing channel resilient schemes are systematically classified into two kinds of schemes; one for the encoder and the other for the decoder. By comparing the performance and implementation cost, the encoder side schemes, such as error localization, layered coding, error resilience bit stream generation techniques, are described in this paper. Also, in an effort to consider the parcticality of the real transmission situation, an efficient error detection scheme for a decoder system is proposed by employing a priori information of the bit stream syntas, checking the encoding conditions at the encoder stage, and exploiting the statistics of the image itself. Finally, subsequent error concealment technique based on the DCT coefficient recovery algorithm is adopted to evaluate the performance of the proposed error resilience technique. The computer simulation results show that the quality of the received image is significantly improved when the bit error rate is as high as 10$^{-5}$ .

Effects of local structural damage in a steel truss bridge on internal dynamic coupling and modal damping

  • Structural health monitoring of steel truss bridge based on changes in modal properties was investigated in this study. Vibration measurements with five sensors were conducted at an existing Warren truss bridge with partial fractures in diagonal members before and after an emergency repair work. Modal properties identified by the Eigensystem Realization Algorithm showed evidences of increases in modal damping due to the damage in diagonal member. In order to understand the dynamic behavior of the bridge and possible mechanism of those increases in modal damping, theoretical modal analysis was conducted with three dimensional frame models. It was found that vibrations of the main truss could be coupled internally with local vibrations of diagonal members and the degree of coupling could change with structural changes in diagonal members. Additional vibration measurements with fifteen sensors were then conducted so as to understand the consistency of those theoretical findings with the actual dynamic behavior. Modal properties experimentally identified showed that the damping change caused by the damage in diagonal member described above could have occurred in a diagonal-coupled mode. The results in this study imply that damages in diagonal members could be detected from changes in modal damping of diagonal-coupled modes.

A statistical framework with stiffness proportional damage sensitive features for structural health monitoring

  • A modal parameter based damage sensitive feature (DSF) is defined to mimic the relative change in any diagonal element of the stiffness matrix of a model of a structure. The damage assessment is performed in a statistical pattern recognition framework using empirical complementary cumulative distribution functions (ECCDFs) of the DSFs extracted from measured operational vibration response data. Methods are discussed to perform probabilistic structural health assessment with respect to the following questions: (a) "Is there a change in the current state of the structure compared to the baseline state?", (b) "Does the change indicate a localized stiffness reduction or increase?", with the latter representing a situation of retrofitting operations, and (c) "What is the severity of the change in a probabilistic sense?". To identify a range of normal structural variations due to environmental and operational conditions, lower and upper bound ECCDFs are used to define the baseline structural state. Such an approach attempts to decouple "non-damage" related variations from damage induced changes, and account for the unknown environmental/operational conditions of the current state. The damage assessment procedure is discussed using numerical simulations of ambient vibration testing of a bridge deck system, as well as shake table experimental data from a 4-story steel frame.

On the Interval Detection of Implosive Stop Sounds by Frame Energy Difference (프레임간 에너지 차를 이용한 음성신호의 종성 폐쇄음 구간 검출에 관한 연구)

  • Preprocessing in speech recognition system is useful, for it reduces some of the complicated procedures required for the final recognition. In this paper, we suggest a new preprocessing algorithm for detecting the intervals of implosive stop sounds. Implosive stop sounds follow vowels in Korean language, and its characteristic is included in the region of vowels. When an implosive stop is pronounced, the velum is quickly colsed, thus its energy decays abruptly and the closure lasts for about 50 to 150 msec. The enegy difference between adjacent frames is chosen as a parameter which represents well the above features.

  • Communication performance requirements are described as QoS parameters in multimedia communication Protocols including SCTP. One of the important QoS parameters is the reliability of the transfer. As a QoS parameter, the reliability defines error detection, report and correction mechanisms. Including SCTP, conventional error control mechanisms, however, do not consider the integrated viewpoint of multimedia data. In this paper, I have designed and proposed the multimedia oriented error control method based on multimedia characteristics. The proposed scheme should have the reduction effect of the communication resources such as frame buffer, procesing power and bandwidth as well as satisfy the requirements of users.
