Anti-oxidative effects of fractionated Pulsatilla koreana $N_{AKAI}$ extracts (백두옹 분획층의 항산화 효과)

  • Objectives : This study was designed to investigate the effects of anti-oxidation of fractionated Pulsatilla koreana $N_{AKAI}$ (PK) extracts. And we examined to determine that a certain fractionated extract has the best anti-oxidative effects between the fractionated PK extracts. Methods : Anti-oxidative effects of fractionated PK extracts was measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cell was measured. After these examination, we determined a fraction that has best anti-oxidative effects. Results : Fractionated PK extracts inhibited radicals effectively. Also in RAW 264.7 cell, intracellular oxidation has inhibited by PK extracts. In these tests, ethyl acetate (EA) fraction has the best anti-oxidative effects among PK extracts. Conclusions : This results demonstrate that PK extracts exhibit anti-oxidative effects. And EA fraction has the best inhibition effects among the six fractions of PK.

Fractionated Aged Black Garlic Extracts Enhance Growth of Anti-My-10 Hybridoma Cells and Production of IgG1 Antibody

  • Aged black garlic (ABG) was extracted with 20% ethanol and water (crude extracts) and fractionated into three categories (>10, 3-10, and <3 kDa). The effect of crude extract supplements on anti-My-10 hybridoma cell growth and IgG1 antibody production was investigated in suspension culture with a chemically defined protein-free medium. We observed that supplementation of ABG to the cell culture medium stimulated anti-My-10 hybridoma cell growth and production of IgG1 antibody, particularly with fractionated ABG of low molecular weight. The stimulation depended upon the concentration and the size of the fractionated ABG. We also found that the growth-promoting activity was not correlated with high antibody production. These results suggest that fractionated ABG is a novel and promising alternative as an animal cell culture supplement.

Broad Spectrum Anti-bacterial Activity of the Leaves, Stem and Root Barks of Myristica subabulata

  • A number of Myristica species are used in herbal medicine for a variety of ailments. The methanol extracts of the leaves and the stem and root barks of Myristica subabulata were fractionated into petrol, dichloromethane and finally ethyl acetate. These were then screened against a total of 31 micro-organisms comprising of 13 G +ve, 12 G ve, one protozoa and 5 moulds. A broad-spectrum anti-bacterial activity was observed. As compared to fractionated the un-fractionated extracts exhibited much better level of activity.

Anti-oxidative and Anti-inflammatory Effect of Melia Toosendan in Mouse Macrophage Cells (마우스 대식세포에서 천련자의 항산화 및 항염증 효과)

  • Objectives: Melia toosendan(MT) has been used as a traditional Korean herbal medicine, and today it is used as a medication for colic, side aches, heartache and other disorders of liver. The aim of this study was to determine whether fractionated extracts of MT inhibit free radical generation such as DPPH radical, superoxide radical and nitric oxide, production of nitrite, an index of NO, PGE2, iNOS, COX-2 and proinflammatory cytokines in lipopolysaccharide(LPS)-treated RAW 264.7 macrophages. Methods: MT extract prepared with methanol, and then fractionated with hexane, dichloromethane, ethylacetate, n-butanol and water. Inhibitory effect of MT onto free radical generation was determined by measuring DPPH, superoxide anions and nitric oxide scavenging activities in vitro. Cytotoxic activity of extracts on RAW 264.7 cells was measured using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt(MTS) assay. Intracelluar oxidation was analysed by DCF-DA assay. The nitric oxide(NO) production was measured by Griess reagent system. The levels of iNOS and COX-2 expression were confirmed by western blot. And pro inflammatory cytokines were measured by ELISA kit. Results: Our results indicated that fractionated extracts, especially dichloromethane and ethyl acetate extracts, significantly inhibited free radical generation, the LPS-induced $H_2O_2$, NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of TNF-${\alpha}$, IL-1${\beta}$ and IL-6 formation in macrophages. Conclusions: These results indicate that dichloromethane and ethyl acetate extracts of MT have potential as an agent of chronic inflammatory diseases.

Inhibitory Effects of Herbal Medicines on Hyaluronidase Activity

  • Inhibitory effects of 130 medicinal plants on hyaluronidase activity were analyzed. The medicinal plants are clinically used as herbal medicines for korean traditional prescriptions. Six out of the 130 herbal medicines exhibited more than 50% of inhibition on hyaluronidase activity by their total methanol extracts with 5mg/ml as a final concentration. The active total methanol extracts were prepared from cortex of Acantbopanax gracilistylus, lignum of Caesalpinia sappan, radix of Glycyrrhiza uralensis, radicis cortex of Morus alba, herba of Prunella vulgaris, and radix of Sanguisorba officinalis. These active total methanol extracts were sequentially fractionated with dichloromethane, ethyl acetate, n-butanol, and then water. Among the solvent-fractionated extracts, the butanol fractions of Acantbopanax gracilistylus and Glycyrrhiza uralensis with 1 mg/ml as the final concentration exhibited more than 50% of inhibition on hyaluronidase activity, and the other fractions with the same concentration did less than 20% of inhibition.

  • Yields of various solvent extracts for Zanthoxylum schinifolium and Z. piperitum leaves were higher in ethanol extract layer. Ethanol Extract in sancho and chopi leaves was fractionated with different solvents, such as hexane, chloroform, ethyl acetate, butanol and water, yields of hexane fraction was higher. In the solvent extracts using methanol, ethanol, hexane, chloroform, and water, ethanol extract showed the most effective antimicrobial activities. Ethanol extract in sancho and chopi leaves was fractionated, the most antimicrobial activities of sancho leaf were ethyl acetate fraction, and chopi leaf were chloroform fraction.

  • The methanol, dichloromethane, petroleum ether, 90% methanol and 1-butanol fractions of 40 Malaysian medicinal plants belonging to 25 families were tested for brine shrimp lethality. Various parts and fractions of eight (20%) plants, viz. Annona muricata, Cerbera odollam. Calophyllum inophyllum, Entada phaseoloides, Pithecellobium jiringa, Crotolaria retusa, Morinda elliptica and Sellaginella willdenovii showed very strong toxicity $(LC_{50}:$<$100\;ppm)$. The methanol extract of the seed of Calophyllum inophyllum showed exceptionally toxic activity $(LC_{50}:$<$5\;ppm)$.

  • The present study was conducted to investigate antioxidant, nitrite scavenging and alcohol degradation effects of extracts from leaf and fruit stalk of Hovenia dulcis. Yields of various solvent extracts for Hovenia dulcis leaf and fruit stalk of were higher in water and methanol extract layer, respectively. Ethanol extracts of Hovenia dulcis leaf and fruit stalk of were fractionated with different solvents such as hexane, chloroform, ethyl acetate, butanol and water, yields of water fraction were highest. In the solvent extracts using methanol, ethanol, hexane, chloroform and water, ethanol extracts showed the most effective antioxidant and nitrite-scavenging effects. Ethanol extracts from Hovenia dulcis leaf and fruit stalk of were fractionated, the most natioxidant and nitrite-scavenging effects were ethyl acetate fraction. Alcohol degradation effects from different parts of Hovenia dulcis showed higher leaf and fruit stalk than xylem, branch and seed. Alcohol degradation effects from leaf and fruit stalk increased as time passed.

The Physiological Activity of Crude Polysaccharide Solvent Extracted from Herbal Medicine Mixture (생약복합물 용매추출물과 조다당획분의 생리활성)

  • After ethanol (BM-E and RW-E) and hot-water (BM-HW and RW-HW) extracts were fractionated from two herbal mixtures (BM and RW), their physiological activities were investigated. All extracts consisted of more than 50% of neutral sugar, with their total polyphenol levels higher than flavonoid levels. Radical scavenging activities of EtOH extracts remained significantly higher compared to that of hot-water extracts, and in particular, RW-E showed consistently higher antioxidant activity than BM-E. When anti-inflammatory activities of the extracts were evaluated by LPS-stimulated RAW 264.7 cells at 10~500 μg/mL non-cytotoxicity doses, BM-E showed significantly higher levels of TNF-α, IL-1β, IL-6, and nitric oxide inhibitory activity than those of hot-water extracts and RW-E. Murine peritoneal macrophage cells were shown to be enhanced in crude polysaccharides (BM-CP and RW-CP fractionated from BM-HW and RW-HW) compared to hot-water extracts and polysaccharide K (PSK, positive control). Especially, RW-CP exhibited higher activity than BM-CP, and component sugar analysis showed that BM-CP mainly contained galacturonic acid, glucose, arabinose, galactose, and xylose (34.5%, 33.9%, 16.1%, 7.1%, and 6.3%, respectively), whereas RW-CP showed different measurements (29.5%, 59.2%, 5.0%, 4.5%, and 0.2%). In conclusion, two herbal mixtures could contain varying sets of physiological activities dependent on different extraction and fractionation methods.
