Long-term drought modifies carbon allocation and abscisic acid levels in five forest tree species

  • Background: This study analyzed the drought responses of five forest tree species grown in Korean peninsula, Korean fir Abies koreana (Ak), eastern white pine Pinus strobus (Ps), keyaki Zelkova serrata (Zs), tulip tree Liriodendron tulipifera (Lt), and Japanese elm Ulmus japonica (Uj). Physiological (chlorophyll, root collar diameter [RCD]) and biochemical responses (non-structural carbohydrates, proline, lipid peroxidase and abscisic acid [ABA]) of the plants grown under mild (MD) and severe drought (SD) were compared. Results: In this study, three soil moisture regimes: control (100% precipitation), MD (60% reduction in precipitation) and SD (20% reduction in precipitation) were applied. Soil moisture content showed high water content in control site compared to MD and SD. A decline in RCD was found for Korean fir, keyaki, and tulip plants, with eastern white pine and Japanese elm showing no significant decline to the prolonged drought exposure (both MD and SD). Total chlorophyll showed a significant decline in Korean fir and tulip, with the sugar levels indicating a significant increase in Korean fir and keyaki species under SD compared to control plants. Non-significant decline in sugar level was noted for eastern white pine and Japanese elm. High accumulation of ABA, malondealdehyde and proline was noted in Korean fir, tulip, and keyaki under SD compared to control. Signs of tree mortality was only observed in Korean fir under MD (38%) and SD (43%). Conclusions: The observed findings indicate the drought responses of five tree species. The majority of the morpho-physiological (especially mortality) and biochemical variables assessed in our study indicate superior long-term drought resistance of Ps and Uj compared to the highly sensitive Ak, and moderately sensitive Lt and Zs. The results provided will help species selection for afforestation programs and establishment of sustainable forests, especially of drought-tolerant species, under increased frequency and intensity of spring and summer droughts.

Effects of dietary supplementation with Pediococcus acidilactici ZPA017 on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation

  • Objective: This study was conducted to determine the effects of dietary supplementation with Pediococcus acidilactici (P. acidilactici) ZPA017 as a probiotic on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation. Methods: A total of 94 sows (Large White×Yorkshire, average 4.50 parities) were randomly allotted to two dietary treatments: control diet and the diet supplemented with P. acidilactici ZPA017 (2.40×109 colony-forming unit/kg of diets). The study started at d 90 of gestation and conducted until d 28 of lactation. Results: Compared to sows fed the control diet, supplementation of P. acidilactici ZPA017 increased the number of weaning piglets, weaning weight of litter and piglets, survival rate of piglets at weaning (p<0.05), and decreased diarrhea rate of piglets in lactation (p<0.05). Dietary P. acidilactici ZPA017 increased fecal Lactobacillus populations (p = 0.030) and reduced fecal Escherichia coli and Staphylococcus aureus populations (p<0.05) of sows at weaning. Moreover, the supplementation of P. acidilactici ZPA017 increased serum concentrations of immunoglobulin G, immunoglobulin A and total protein (p<0.05), while decreased serum haptoglobin concentration and alanine aminotransferase activity (p<0.05) of sows at weaning. Conclusion: Administration of P. acidilactici ZPA017 in diets during late gestation and lactation had positive effects on the reproductive performance, intestinal microflora balance and immunity of sows.

Effects of Lactobacillus curvatus and Leuconostoc mesenteroides on Suan Cai Fermentation in Northeast China

  • To investigate the effects of Lactobacillus curvatus and Leuconostoc mesenteroides on suan cai (pickled Chinese cabbage) fermentation, L. curvatus and/or Ln. mesenteroides were inoculated into suan cai. Physicochemical indexes were measured, and the microbial dynamics during the fermentation were analyzed by Illumina MiSeq sequencing and quantitative polymerase chain reaction (qPCR). The results showed that inoculation with lactic acid bacteria (LAB) lowered the pH of the fermentation system more rapidly. The decrease in water-soluble carbohydrates in the inoculated treatments occurred more rapidly than in the control. The LAB counts in the control were lower than in other inoculated treatments during the first 12 days of fermentation. According to the Illumina MiSeq sequencing analyses, Firmicutes, Proteobacteria, Bacteroidetes, Actinobacteria, Cyanobacteria, Fusobacteria, and Verrucomicrobia were present in the fermentations, along with other unclassified bacteria. Generally, Firmicutes was predominant during the fermentation in all treatments. At the genus level, 16 genera were detected. The relative abundance of Lactobacillus in all inoculated treatments was higher than in the control. The relative abundance of Lactobacillus in the treatments containing L. curvatus was higher than in the Ln. mesenteroides-only treatment. The relative abundance of Leuconostoc in the Ln. mesenteroides-containing treatments increased continuously throughout the fermentation. Leuconostoc was highest in the Ln. mesenteroides-only treatment. According to the qPCR results, L. curvatus and/or Ln. mesenteroides inoculations could effectively inhabit the fermentation system. L. curvatus dominated the fermentation in the inoculated treatments.

Influence of a Novel Mold Inhibitor on Mechanical Properties and Water Repellency of Bamboo Fiber-based Composites

  • Effects of a novel mold inhibitor specifically for bamboo, on the properties of composite products have been confirmed in this study. The mechanical and dimensional stability properties of bamboo fiber-based composites (BFBCs) from different bamboo species were also investigated. The results showed that Burmanica Gamble possessed the highest values of modulus of elasticity (MOE) of 33.2 GPa, modulus of rupture (MOR) of 286.9 MPa, compressive strength of 182.6 MPa and shear strength of 24.0 MPa. By contrast, Phyllostochys heterocycla among all of species showed the lowest MOE of 16.3 GPa, MOR of 170.3 MPa and compressive strength of 128.9 MPa were the lowest among all of species. Moreover, there is a remarkable variation in the swelling and water absorption between the samples with 4 h and 28 h water immersion treatment, especially Phyllostachys iridenscens. Overall, the results suggested that TCIT (Tebuconazole and 3(2H)-isothiazolone) had no significant effect on the mechanical properties compared with the control condition, and it would be utilized as an antimould of BFBCs manufacturing.

The Endophytic Bacteria Bacillus velezensis Lle-9, Isolated from Lilium leucanthum, Harbors Antifungal Activity and Plant Growth-Promoting Effects

  • Bacillus velezensis is an important plant growth-promoting rhizobacterium with immense potential in agriculture development. In the present study, Bacillus velezensis Lle-9 was isolated from the bulbs of Lilium leucanthum. The isolated strain showed antifungal activities against plant pathogens like Botryosphaeria dothidea, Fusarium oxysporum, Botrytis cinerea and Fusarium fujikuroi. The highest percentage of growth inhibition i.e., 68.56±2.35% was observed against Fusarium oxysporum followed by 63.12 ± 2.83%, 61.67 ± 3.39% and 55.82 ± 2.76% against Botrytis cinerea, Botryosphaeria dothidea, and Fusarium fujikuroi, respectively. The ethyl acetate fraction revealed a number of bioactive compounds and several were identified as antimicrobial agents such as diketopiperazines, cyclo-peptides, linear peptides, latrunculin A, 5α-hydroxy-6-ketocholesterol, (R)-S-lactoylglutathione, triamterene, rubiadin, moxifloxacin, 9-hydroxy-5Z,7E,11Z,14Z-eicosatetraenoic acid, D-erythro-C18-Sphingosine, citrinin, and 2-arachidonoyllysophosphatidylcholine. The presence of these antimicrobial compounds in the bacterial culture might have contributed to the antifungal activities of the isolated B. velezensis Lle-9. The strain showed plant growth-promoting traits such as production of organic acids, ACC deaminase, indole-3-acetic acid (IAA), siderophores, and nitrogen fixation and phosphate solubilization. IAA production was accelerated with application of exogenous tryptophan concentrations in the medium. Further, the lily plants upon inoculation with Lle-9 exhibited improved vegetative growth, more flowering shoots and longer roots than control plants under greenhouse condition. The isolated B. velezensis strain Lle-9 possessed broad-spectrum antifungal activities and multiple plant growth-promoting traits and thus may play an important role in promoting sustainable agriculture. This strain could be developed and applied in field experiments in order to promote plant growth and control disease pathogens.

Influence of Ectomycorrhizal Colonization on Cesium Uptake by Pinus densiflora Seedlings

  • Radionuclides were deposited at forest areas in eastern parts of Japan following the Fukushima Daiichi Nuclear Power Plant incident in March 2011. Ectomycorrhizal (EM) fungi have important effects on radiocaesium dynamics in forest ecosystems. We examined the effect of colonization by the EM fungus Astraeus hygrometricus on the uptake of cesium (Cs) and potassium (K) by Pinus densiflora seedlings. Pine seedlings exhibited enhanced growth after the EM formation due to the colonization by A. hygrometricus. Additionally, the shoot Cs concentration increased after the EM formation when Cs was not added to the medium. This suggests that A. hygrometricus might be able to solubilize Cs fixed to soil particles. Moreover, the shoot K concentration increased significantly after the EM formation when Cs was added. However, there were no significant differences in the root K concentration between EM and non-EM seedlings. These results suggest that different mechanisms control the transfer of Cs and K from the root to the shoot of pine seedlings.

Effect of Pre-Sowing Treatments on Germination and Initial Seedling Growth of Castanopsis Indica- An Endangered Tree Species in Bangladesh

  • Castanopsis indica (Sil Batna) is an ecologically valuable multipurpose indigenous tree species of Bangladesh. Considering its high value but poor natural regeneration due to seed dormancy, the authors conducted an experiment at the Institute of Forestry and Environmental Sciences Chittagong University (IFESCU) during 2012-2013 to find out effective pre-sowing treatments. Ten pre-sowing seed treatments were applied on mature, even-sized seeds namely-control; sandpaper rubbing; nail clipping; immersion in normal water (at room temperature: 24℃) for 24 hrs., 48 hrs. and 72 hrs.; immersion in hot water (80℃) for 1 minute; soaking in 10% dilute H2SO4; soaking in 10% HCl for 5 minutes; and sowing in propagator house. Seeds sown after sandpaper rubbing at the distal end revealed best performances by providing highest germination percent (66.7%), germination energy (30%), germination index (0.17), germination rate (0.0145), germination value (30%) and plant percent (66.7%) within shortest period (38 days). The treatment also produced most vigor seedlings with 20.9 cm shoot height, 15 node number, and largest leaves (11.1 cm×2.9 cm). Hence, it is recommended to adopt sandpaper rubbing method for maximum germination and quality seedlings.

Development of Nested PCR, Multiplex PCR, and Loop-Mediated Isothermal Amplification Assays for Rapid Detection of Cylindrocladium scoparium on Eucalyptus

  • Eucalyptus dieback disease, caused by Cylindrocladium scoparium, has occurred in last few years in large Eucalyptus planting areas in China and other countries. Rapid, simple, and reliable diagnostic techniques are desired for the early detection of Eucalyptus dieback of C. scoparium prior to formulation of efficient control plan. For this purpose, three PCR-based methods of nested PCR, multiplex PCR, loop-mediated isothermal amplification (LAMP) were developed for detection of C. scoparium based on factor 1-alpha (tef1) and beta-tubulin gene in this study. All of the three methods showed highly specific to C. scoparium. The sensitivities of the nested PCR and LAMP were much higher than the multiplex PCR. The sensitivity of multiplex PCR was also higher than regular PCR. C. scoparium could be detected within 60 min from infected Eucalyptus plants by LAMP, while at least 2 h was needed by the rest two methods. Using different Eucalyptus tissues as samples for C. scoparium detection, all of the three PCR-based methods showed much better detection results than regular PCR. Base on the results from this study, we concluded that any of the three PCR-based methods could be used as diagnostic technology for the development of efficient strategies of Eucalyptus dieback disease control. Particularly, LAMP was the most practical method in field application because of its one-step and rapid reaction, simple operation, single-tube utilization, and simple visualization of amplification products.

느타리버섯 푸른곰팡이병에 관여하는 Trichoderma 속균의 동정 및 검출을 위한 PCR 기반 DNA 마커 개발 (Development of PCR-based DNA markers for identification and detection of Trichoderma species associated with the green mold disease of oyster mushroom)

  • Trichoderma is known as pathogen caused serious green mold disease on commercial production. T. pleuroti and T. pleuroticola were common species in various mushroom media. Many strains of T. pleuroti, known as aggressive species causing major economic losses in Korea, showed benomyl resistance. Accurate identification and detection of Trichoderma species associated with oyster mushrooms is very important for disease control. We developed species-specific primers for T. pleuroticola, T. pleuroti, T. harzianum, and T. atroviride based on species-specific fragments isolated from amplified fragment length polymorphism analysis. PCR products corresponding to the predicted fragment of 500bp, 230bp, 180bp, and 410bp were amplified from T. pleuroticola, T. pleuroti, T. harzianum, and T. atroviride, respectively. Multiplex PCR assay using species-specific primers quickly and accurately identified and detected T. pleuroti from mushroom media in which various species co-exist. Our results can be useful for the effective control of mushroom disease.

Diversity and Antifungal Activity of Endophytic Fungi Associated with Camellia oleifera

  • Endophytic fungi strains (n = 81) were isolated from the leaves, barks, and fruits of Camellia oleifera from Hunan province (China) to delineate their species composition and potential as biological control agents of C. oleifera anthracnose. The fungi were identified by morphological and phylogenetic analyses. Fungal colonization rates of the leaves, barks, and fruits were 58.02, 27.16, and 14.81%, respectively. The isolates were identified as 14 genera, belonging to two subdivisions, Deuteromycotina and Ascomycotina; 87.65% of all isolates belonged to Deuteromycotina. The dominant species, occurring with a high relative frequency, were Pestalotiopsis sp. (14.81%), Penicillium sp. (14.81%), and Fusarium sp. (12.35%). The Simpson's and Shannon's diversity indices revealed the highest species diversity in the leaves, followed by the barks and fruits. The similarity index for the leaves versus barks comparison was the highest, indicating that the number of endophytic fungal species shared by the leaves and barks was higher than barks and fruits or leaves and fruits. Based on the results of dual culture experiments, only five strains exhibited antifungal activity against C. oleifera anthracnose pathogen, with isolate ty-64 (Oidium sp.) generating the broadest inhibition zones. Our results indicate that the endophytes associated with C. oleifera could be employed as natural agents controlling C. oleifera anthracnose.
