Antimicrobial Effect of Indigofera kirilowii Extracts on Food-borne Pathogens (산두근 추출물의 식중독성 미생물에 대한 항균효과)

  • This study was performed to investigate the antimicrobial effect of the Indigofera kirilowii extracts against food-borne pathogens. The Indigofera kirilowii was extracted with methanol at room temperature, and fractionation of the methanol extract from Indigofera kirilowii was carried out by using petroleum ether, chloroform, ethyl acetate, and methanol, respectively. The antimicrobial activity of the Indigofera kirilowii extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extract of Indigo/era kirilowii showed the highest antimicrobial activity against Staphylococcus aureus and Shigella dysenteriae. The water extract of Indigofera kirilowii showed relatively low antimicrobial activity against microorganisms used in this experiment. The synergistic effect has been found in combined extracts of Indigofera kirilowii and Pulsatilla koreana as compared to each extract alone. The growth inhibition curve was determined using ethyl acetate extracts of Indigofera kirilowii against S. aureus and S. dysenteriae. The ethyl acetate extract of Indigofera kirilowii showed strong antimicrobial activity against S. aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate extract from Indigofera kirilowii retarded the growth of S. aureus more than 24 hours and S. dysenteriae up to 48 hours. This study showed the possibility of using ethyl acetate extract of Indigofera kirilowii as a material of food preservative.

Antimicrobial and Antioxidative Activities of Solvent Fractions of Quercus mongolica Leaf (신갈나무 잎의 용매분획별 항균 및 항산화 효과)

  • The ethanol extract of Quercus mongolica leaf was fractionated by various organic solvents, and their antimicrobial and antioxidative activities were investigated against several microorganisms. The ethanol extract of Quercus mongolica leaf at two thousand $\mu\textrm{g}$ per disc showed 17~21mm inhibition zone against Gram postive and Gram negative bacteria. Among the various solvent fractions from ethanol extract of Quercus mongolica leaf, the hexane fraction showed the strongest antimicrobial activity. Minimal inhibitory concentration (MIC) of hexane fraction was 250$\mu\textrm{g}$/mL against Bacillus cereus, 250~500$\mu\textrm{g}$/mL against Listeria monocytogenes, 500$\mu\textrm{g}$/mL against Staphylococcus aureus, Salmonella typhimurium, Escherichia coli O157, and Pseudomonas aeruginosa. Also, the hexane and chloroform fraction had the similar antioxidative activity compared to that of butylated hydroxy toluene(BHT).

Isolation of Growth Inhibition Substance on Food borne Microorganisms from Hypericum ascyron L. and Application to Food Preservation (물레나물(Hypericum ascyron L.)의 식중독 미생물 증식 억제 물질의 분리 및 식품적용)

  • The ethanol extract and n-hexane fraction from Hypericum ascyron L. showed strong growth inhibition at 25 ppm on 5 strains of Listeria monocytogenes for 72 hr at $32^{\circ}C$. The purified substance, H2-5-2 fraction, was isolated by silica gel column and preparative thin layer chromatography from n-hexane fraction of Hypericum ascyron L. The H2-5-2 fraction showed a strong bacteriostatic activity on 5 strains of L. monocytogenes at 10 ppm in tryptic soy broth, and the viable cell was reduced 1 log cycle compared to initial cell number. The n-hexane fraction of Hypericum ascyron L. showed strong growth inhibition at 25 ppm on Bacillus cereus and Staphylococcus aureus, and at 50 ppm on Vibrio parahaemolyticus for 72 hr. The purified antimicrobial substance, the H2-5-2 fraction, was assumed as high unsaturated sterol by $^1H-NMR$ and $^{13}C-NMR$. On application test using minced Alaska pollack and ground beef, the n-hexane fraction of Hypericum ascyron L. at the level of 250 ppm was applied at $32^{\circ}C$ and $5^{\circ}C$. At $32^{\circ}C$ storage condition, the antimicrobial substances did not reduced L. monocytogenes ATCC 19113, meanwhile at $5^{\circ}C$ storage condition, L. monocytogenes ATCC 19113 was reduced in viable number.

Antibiotic Resistance and Assessment of the Food-borne Pathogenic Microorganisms in Ready to Eat Meals (식사대용 식품(Ready-to-eat meals) 중 병원성 세균의 분포와 항생제 감수성 양상)

  • This study was performed in order to measure the level of food-borne pathogenic bacteria and antibiotic resistance pattern of found ready to eat meals such as Him-bap, Cho-bap, Hamburger, Sandwich and packed lunch boxes. A total of 497 samples were collected from supermarket and department of Seoul, Kyung-ki, Inctleon, Kang-won, Chung-Cheong from November, 2005 to March, 2006. The contaminated microorganisms were in most cases tract relative strain like E. coli and S. aureus. Result have shown E. coli was detected 4 strains and S. aureus was detected 22 strains. 26 strains were also tested the antibiotic resistance pattern. 26 strains were shown to be relatively susceptible to synercid, vancomycin, teicoplanin, ciprofloxacin, gentamycin, lincomycn, cefotaxime, meropenem, cephalosporin, amoxicillin-clavulanic acid by the MIC dilution method, but E. coli 1 strain was resistant to amoxicillin-clavulanic acid.

Prevalence and Microbial Flora of Chicken Slaughtering and Processing Procedure

  • This study has been performed to measure the prevalence and microbial flora on chicken slaughtering as well as the processing process from the months of October to November. Whole-chicken rinsing technique was used in order to analyze the incidence of microorganisms on chicken carcass at the stage before chilling (after evisceration), after chilling and after cutting. The swab technique was used on processing the processed samples, such as working plates and cutting knives. Brine and cooling water from four cooling tubs were taken from each processing processes and were used as samples. Furthermore, the matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) for whole cell fingerprinting in combination with a dedicated bioinformatic software tool was used to identify the isolated microorganisms. Of the tested samples and processes, brine ($4.50{\pm}0.64$ Log CFU/mL) and chicken carcass before chilling ($4.15{\pm}0.46$ Log CFU/mL) showed the highest population of microorganisms; the predominant microbial flora of them were Moellerella wisconsensis (54.84%), a member of the Enterobacteriaceae family, and Escherichia coli (60.36%), respectively. However, the predominant microbial flora of cut carcass was changed to Staphylococcus aureus (27.32%), which is a kind of pathogenic microorganism that can cause a food-borne illness. Therefore, the slaughtering and processing procedure of chicken are needed to be controlled more hygienically.

Isolation and Antimicrobial Action of Growth Inhibitory Substance on Food-borne Microorganisms from Dryopteris crassirhizoma Nakai (관중(Dryopteris crassirhizoma Nakai)의 식중독 미생물 증식 억제 물질의 분리 및 항균작용)

  • The ethanol extract of Dryopteris crassirhizoma Nakai showed strong growth inhibition against 5 strains of Listeria monocytogenes at the concentrations of $100{\sim}500$ ppm and the minimum inhibitory concentration of n-hexane fraction was under 50 ppm. The D8-2-5 fraction isolated from n-hexane fraction of Dryopteris crassirhizoma Nakai showed a strong bactericidal activity on 5 strains of L. monocytogenes at 20 ppm level in tryptic soy broth medium. At the level, the viable count was reduced $4{\sim}6$ log cycle compared to initial cell number. Observation by the measurement of adenosine triphosphate (ATP) contents and transmission electron microscope showed that disruptions of the cell wall and elution of intracellular ATP are assumed to be due to the bactericidal activity. In addition, the n-hexane fraction of Dryopteris crassirhizoma Nakai showed the strong growth inhibitions at 50 ppm on Vibrio parahaemolyticus and Bacillus cereus, and at 25 ppm on Staphylococcus aureus.

Antimicrobial Activity of Lysimachia clethroides Duby Extracts on Food-borne Microorganisms (식중독 미생물에 대한 큰까치수영(Lysimachia clethroides Duby)의 항균활성)

  • The ethanol extract of 77 species of edible and medicinal plants were examined antimicrobial activity against 5 strains of Listeria monocytogenes ATCC 19111, ATCC 19112, ATCC 19113, ATCC 19114 and ATCC 15313 by optical density using Bioscreen C. The ethanol extract of Siegesbeckia glabrescens Makino, Jeffersonia dubia Benth, Aquilaria agallocha Roxburgh, Lysimachia clethroides Duby and Nardostachys chinensis Batal. exhibited comparatively strong growth inhibition effect on 5 strains of L. monocytogenes at 1000 ppm level in broth. The minimum inhibitory concentration (MIC) of ethanol extract of Lysimachia clethroides Duby was $100{\sim}500\;ppm$ on 5 strains of L. monocytogenes. The MIC of the n-hexane and chloroform fraction of the extract were same concentration as $50{\sim}100\;ppm$. The n-hexane fraction of Lysimachia clethroides Duby showed strong growth inhibition at 25 ppm on Vibrio parahaemolyticus for 72 hr at $30^{\circ}C$ and at 50 ppm on Bacillus cereus and at 500 ppm on Staphylococcus aureus and Escherichia coli.

Antimicrobial Effect of Methanol Extracts from Some Medicinal Herbs and the Content of Phenolic Compounds (약용식물 추출물에 대한 항미생물 활성 검색과 폴리페놀 함량)

  • Methanol extracts were prepared from 32 medicinal herbs of the extracts were tested their microbial inhibition activities against food borne pathogens and/or food poisoning microorganisms, food-related bacteria and yeast. Methanol extracts of Cinnamomum cassia, Paeonia suffruticosa, Alnus japonica, Eugenio caryophyllata and Illicium verum exhibited antimicrobial activity for the microorganisms tested, except lactic acid bacteria and yeast. Minimum inhibitory concentrations(MIC) were about 5 mg/mL for Bacillus subtilis, Staphylococcus epidermidis and Pseudomonas aeruginosa. Cell growth of lactic acid bacteria was inhibitied, but greatly on Leuconostoc mesenteroides. The phenolic compound contents were 10.98 mg/g, 10.31 mg/g, 8.55 mg/g and 6.69 mg/g in Thea sinensis, Eugenia caryophyllata, Alnus japonica and Artenisia capillaris, respectively. Antimicrobial activity appeared to be related to phenol compound content in medicinal herbs. The methanol extracts of medicinal herbs could be suitable for the development of a food preservative.

Antimicrobial Activities of White, Red, and Extruded Ginsengs with Different Extraction Conditions

  • White, red, and extruded ginsengs were studied against 8 strains of food-borne pathogens and/or food spoilage microorganisms. The ginseng powders were extracted with different extractants and screened for antimicrobial activity using the disc diffusion and broth dilution techniques. The results showed that the yield of extraction was higher with increase of aqueous solution content and temperature. Preliminary screening revealed that the red ginseng extracts were most active, that has been found to be highly effective against all tested microbe except Listeria monocytogenes. Moreover, Bacillus subtilis has shown highly susceptible, which the diameters of inhibition zone values of 28 extracts were between 7 and 14 mm. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) recorded for the different crude ginseng extracts against microorganism using ranged from 6.25 to 100 mg/mL, indicated that the methanol extract of ginseng were more effective than ethanol and water extracts. The 60% methanol extract of red ginseng had the greatest effects against B. subtilis with MIC and MBC values at 6.25 mg/mL.

Anthocyanins from Clitoria ternatea Attenuate Food-Borne Penicillium expansum and its Potential Application as Food Biopreservative

  • Clitoria ternatea or Commonly known blue pea, is a perennial climber crop native to Asian countries. The current study was aimed to evaluate the antimicrobial activity C. ternatea extract on food borne microorganisms and its antifungal effect on Penicillium expansum. The extract showed significant antimicrobial activity against 3 Gram positive bacteria, 2 Gram negative bacteria and 1 filamentous fungus on disc diffusion assay. The extract also showed good biocidal effect on all Gram positive bacteria tested and P. expansum. However, the kill curve analysis revealed that the fungicidal activity of the extract against P. expansum conidia was depend on the concentration of the extract and the time of exposure of the conidia to the extract. The scanning electron micrograph of the extract treated P. expansum culture showed alterations in the morphology of fungal hyphae. The germination of P. expansum conidia was completely inhibited and conidial development was totally suppressed by the extract, suggesting the possible mode of action of anthocyanin. Besides, the extract also exhibited 5.0-log suppression of microbial growth relative to control in the rice model. The results indicate the potential use of the C. ternatea anthocyanin as food biopreservative.
