Measures for a closer-to-real estimate of dietary exposure to total mercury and lead in total diet study for Koreans

  • Previous Korean total diet studies (KTDSs) have estimated dietary exposure to toxic chemicals based on 110-120 representative foods selected from over 500 foods appeared in the Korea National Health & Nutrition Examination Surveys (KNHANES), which would result in a possible underestimation. In order to find measures for a closer-to-real estimate of dietary exposure to heavy metals, this study examined the feasibility of mapping foods to the representative foods in the KTDS by comparing estimates. In mapping, those foods not analyzed in the 2009 KTDS (443 out of 559 foods appeared in the 2007 KNHANES) were mapped to the 114 representative foods used in the 2009 KTDS based on the closeness in regards to biological systematics and morphological similarity. Dietary exposures to total mercury and lead were re-estimated using the content of total mercury and lead in 114 foods analyzed in the 2009 KTDS, food intake, and individual's own body weight for respondents in the 2007 KNHANES instead of mean body weight of Koreans used in the 2009 KTDS. The re-estimates of exposure with mapping were approximately 50% higher than the original estimates reported in the 2009 KTDS. In addition, mapping enabled the comparison of percentile distribution of the exposure among populations of different age groups. In conclusion, estimates via mapping resulted in a more comprehensive estimation of dietary exposure to heavy metals present in foods that Koreans consume.

다년생 근채류 중 중금속 모니터링 및 위해성평가 (Monitoring and Risk Assessment of Heavy Metals in Perennial Root Vegetables)

  • 다년생 근채류인 인삼, 산양삼, 더덕, 도라지의 중금속 함량 실태조사를 통해 우리나라 국민의 식품 섭취로 인한 중금속 노출수준에 따른 위해성을 평가하였으며, 이를 위해 총 214건을 수거하였다. 근채류 중 납, 카드뮴 및 비소 함량분석을 위해 microwave장치를 이용해 전처리한 후 ICP/MS로 측정하는 시험법을 확립하였다. 중금속의 검출한계는 0.010~0.050 μg/kg이었으며 정량한계는 0.035~0.175 μg/kg이었다. 표준용액 첨가법과 CRM을 이용하여 회수율을 실험한 결과, 76~102%의 양호한 결과를 얻었다. 다년생 근채류 중 납 함량은 평균 0.013(인삼)~0.070(산양삼) mg/kg이었고, 카드뮴 함량은 평균 0.009(인삼)~0.034(더덕) mg/kg이었으며 비소 함량은 평균 0.002(인삼)~0.004(도라지) mg/kg이었다. 다년생 근채류 섭취에 따른 중금속 위해성 평가를 수행한 결과, 중금속 노출량은 각각 Pb 0.070 μg/day, Cd 0.041 μg/day, As 0.008 μg/day으로 인체섭취한계량의 0.03%, 0.08%, 0.0003% 수준이었다. 따라서, 국내 유통되는 인삼, 산양삼, 더덕 및 도라지 섭취로 인한 중금속 노출수준은 JECFA 등의 인체노출안전기준보다 매우 낮았으며 우리나라 국민은 다년생 근채류에 존재하는 중금속의 위해성으로부터 안전하였다.

향신료 및 건조과실류 중 총 아플라톡신의 분석 (Analysis of Total Aflatoxins in Spices and Dried Fruits)

  • 총아플라톡신의 분석을 위하여 대상 시료로부터 추출된 용액을 면역친화성칼럼 (immunoaffinity column)을 이용하여 정제하고 트리플루오로초산(TFA, trifluoroacetic acid) 유도체법과 광유도체화 형태인 PHRED (Photochemical reactor enhanced detection) 유도체 법을 비교 검토하여 분석하였다. 결과적으로 재현성 및 편리성은 PHRED유도체법이 우수하였으나 분리도 및 검출한계는 TFA유도체법이 우수하였다. 확립된 시험법은 아플라톡신 B1, B2, G1은 80% 이상, G2는 70% 이상의 회수율을 보였고, 아플라톡신 B1, B2, G1 및 G2 각각 0.05, 0.05, 0.2 및 $0.1\;{\mu}g/kg$의 검출한계를 나타내었다. 확립된 시험법으로 향신료 및 건조과실류 중 총 아플라톡선에 대한 실태 조사를 위하여 향신료 10종 179건 및 건조과실류 9종 137건 총 316건을 분석하였다. 그 결과, 건조과실류 건망고 등 9종 137건 중 27건(19.7%)에서 아플라톡신이 소량 검출되었으며, 건조향신료 육구두 등 10종 179건 중 87건 (48.6%)에서 아플라톡선이 검출되었다.

  • 사람이 섭취하는 식품 내의 항생제, 알레르기 유발 물질, 병원균 및 기타 오염물질의 수준을 모니터링하기 위해서는, 빠르고 정확하며 저렴한 비용으로 테스트 해야 한다. 이러한 문제 중 일부를 해결하기 위해 지난 10-15년 동안 진보된 기술(label-free biosensor assays)이 개발되어 왔다. 이 면역감지키트들은 실시간 측정이 가능하고, 높은 수준의 자동화를 제공하며, 향상된 처리율과 민감도를 가지고 있다. 또한, 기존의 방법과 비교하여 가격이 저렴하고, 덜 복잡하며, 분석 시간을 단축시켜주는 사용자 친화적 키트이다. 이 리뷰에서는 면역감지키트의 장단점, 그리고 미래의 식품안전검사에서의 사용성에 관한 것에 대해 논의해 볼 것이다.

글리코사미노글칸 분해 효소와 핵자기공명을 이용한 오염된 헤파린 시료의 분석 (Analysis of Contaminated Heparins by the Combination of Glycosaminoglycan Degrading Enzymes and Nuclear Magnetic Resonance Spectroscopy)

  • We examined the purity of six heparin samples by using heparinase, chondroitinase, $^{1}H-NMR$, and polyacrylamide gel electrophoresis. To obtain high molecular weight contaminants from heparin samples, heparinase I - digested samples were subjected to the exhaustive microcon filtration. The filtration process removed heparin-derived di- and oligosaccharides effectively. By combining chondroitinase ABC treatment and strong anion exchange - high performance liquid chromatography, the result showed all six samples contained chondroitin sulfate as a contaminant ranging from 1.3 to 14.9%. Among them, sample S3 showed the highest content of 14.9%, which was further analyzed by chondroitinase AC treatment to confirm chondroitin sulfate B (dermatan sulfate). $^{1}H-NMR$ chemical shifts of N-acetyl groups clearly suggested the existence of chondroitin sulfate B (sample S3) and oversulfated chondroitin sulfate (samples S2 and S4) as contaminants. In addition, polyacrylamide gel electrophoresis was useful for qualitative detection on the sample's purity. These results suggest that the tools of heparinase I and chondroitinase ABC in combination with NMR spectroscopy would give very useful information for investigation of heparin contaminants such as oversulfated chondroitin sulfate and dermatan sulfate in heparin samples.

  • Dietary life of Korean population was greatly improved by the increased food production, enlarged food processing and betterment of food distribution system during the last three decades. On the other hand, food pollution issues have arisen from the increased use of chemicals and advancements in analytical techniques and toxicological information. The health risk of hazardous substances in Korean diet was estimated to be in the decreasing order of heavy metals>pesticide residues>food additives>mycotoxins>radionuclides>PCBs whereas consumers do not recognize it in the same order. It is needed to evaluate the food safety more accurately based on scientific evidences and to adopt reasonable regulatory measures in Korea for the benefits of people's health and national interest.

Status and future perspective for soil contamination of arable land in China

  • China is currently facing great challenges in protecting its arable soil from contamination by heavy metals, especially Cd in paddy soil. China enacted the first soil environmental quality standards (SEQS) for ten pollutants in 1995, and the Ministry of Ecology and Environment released the results of the first nationwide soil survey in 2014. The soil survey showed that as much as 16% of China's soil and 19% of the agricultural soils were contaminated mainly with heavy metals and metalloids beyond the environmental quality limits. The exceeded rate of the contaminant limits in food crops was widespread in China, and the most severe regions were East and Southwest China. Heavy metals and metalloids accounted for 82.4% of the contaminants in soils while organic pollutants accounted for 17% of the contaminants in the soil. Among the heavy metals and metalloids exceeding the Ministry of Environmental Protection (MEP) limit, cadmium (Cd) was highest at 7.0%, followed by nickel (4.8%), arsenic (2.7%), cobalt (2.1%), mercury (1.6%) and lead (1.5%). However, all the average concentrations of the pollutants were lower than the recommended values for the contaminants except for Cd for three levels of pH (< 6.5, 6.5 - 7.5, and > 7.5). According to the Action Plan on Prevention and Control of Soil Pollution released by the State Council in 2016, 90% of contaminated farmland will be made safe by 2020 with an increase to 95% by 2030. Therefore, it is necessary to improve the soil quality to meet the environmental quality standard for soils and heavy metal standards for food safety.

  • These days, men live better lives owing to economic growth. They are interested in basic desire such as clothing, food and dwelling. This study investigated food and/or eating. Men like to take better quality food to be healthy. They can hear food problems easily by news to satisfy desire. On October 13, 2014, Dongsuh Food Company was prohibited to distribute a serial product named 'Post Almond Flake' (Statistics Korea). Dongsuh Food was found to produce finished product by mixing contaminants without inspection of colon bacillus, and Crown Confectionery was found to produce 'Organic farming wafer' and 'Organic farming choco wafer' from March 2009 to early August, 2016 cognizing rejection at inspection not to inform Ministry of Health and to sell product amounting to 3.1 billion KRW .

  • The dioxin- or aryl hydrocarbon receptor (AhR) is a member of the Per-Arnt-Sim family of nuclear transcription factors exhibiting a basic helix-loop-helix structure. In its non-ligated state the AhR is associated with hsp 90 and the immunophilin-type XAP2. Upon ligand binding the associated proteins are released, the receptor dimerizes with the AhR nuclear trans locator protein Arnt, and binds to XREs (xenobiotic-responsive elements) in the 5'-flanking region of responsive genes thus modulating their transcription.(omitted)
