해조류 중의 anti-tumor initiator 및 promoter의 해석-3 : 곰피 추출물중의 발암 promotion억제 인자 (Elucidation of Anti-tumor Initiator and Promoter Derived from Seaweed-3 : Anti-tumor Promoters of Ecklonia stolonifera Extracts)

  • 갈조류중의 발암 promotion 억제인자를 밝히기 위하여 곰피, 미역 및 다시마의 용매추출물을 이용하여 발암 promotion 억제효과를 조사한 결과, 곰피. 미역 및 다시마의 수용성 추출물을 제외한 유기용매 분획물에서 우수한 억제효과를 나타내었다. 이 중에서 억제효과가 강한 곰피를 이용하여 발암 promotion 억제인자를 살펴본 결과, 곰피중의 bromophenol, phloroglucinol 등의 phenol 을 비롯하여 chlorophyll 및 carotenoid 화합물에서 억제 효과가 높게 나타났다. 곰피의 chlorophyll a 및 pheophorbide a에서 5 $\mu$M 첨가시 $77.4\%$$66.6\%$의 억제효과가 나타났으며, 곰피의 carotenoid 화물 중에서는 lutein, $\alpha$-cryptoxanthin 및 $\beta$-carotene이 주요 억제인자로 밝혀졌다. Lutein 및 $\alpha$-cryptoxanthin 의 발암 promotion 억제효과는 20$\mu$g 첨가에 의해 각각 $76.9\%$$84.4\%$로 우수하였고, 세포 생잔율도 높게 나타났다.

Isolation and genetic characterization of canine adenovirus type 2 variant from raccoon dog (Nyctereutes procynoide koresis) in Republic of Korea

  • Canine adenovirus type 2 (CAV-2) is a common causative agent of respiratory disease in canines. There have been no reports of CAV-2 variants isolated from raccoon dogs. This study aims to investigate the biological and genetic characteristics of a novel Korean CAV-2 variant. Madin-Darby canine kidney cells were used to isolate the CAV-2 variant from 45 fecal swab samples. Diagnostic tools such as the cytopathic effect (CPE) assay, electron microscopy, polymerase chain reaction, and immunofluorescence and hemagglutination assays were used to confirm the presence of the CAV-2 isolate. A cross-virus neutralization assay was performed to verify the novelty of this CAV variant. Genetic analysis was performed using nucleotide sequences obtained through next-generation sequencing. The isolate was confirmed to be a CAV-2 variant based on the aforementioned methods and designated CAV2232. The number of bases in the fiber and E3 genes of CAV2232 were 1,626 and 414, respectively. Phylogenetic analysis of the fiber and E3 genes confirmed that CAV2232 was classified into a different clade from the known CAV-1 and CAV-2 strains. Mice inoculated with the CAV2232 vaccine developed high virus neutralization antibody titers of 1,024 (210) against CAV2232, while mice inoculated with CAV-1 and CAV-2 vaccines had low virus neutralization antibody titers of 12.9 (23.7) and 6.5 (22.7), respectively, against CAV2232. CAV2232 isolated from wild raccoon dog feces was classified as a novel CAV-2 variant. CAV2232 may therefore be used as an antigen for new vaccine development and serological investigations.

메주 유래 Enterococcus faecium 균주의 기능적 특성 및 안전성 (Technological Characteristics and Safety of Enterococcus faecium Isolates from Meju, a Traditional Korean Fermented Soybean Food)

  • γ-Aminobutyric acid 생성에 관여하는 glutamate decarboxylase 유전자 gadA/B를 보유한 메주 유래 Enterococcus faecium 88균주를 대상으로 기능적 특성 및 안전성 평가를 진행하여 메주 유래 E. faecium의 발효식품용 종균으로써의 활용 가능성을 검토하였다. 6% NaCl 농도(w/v)에서 모든 균주의 생장 및 산 생성이 확인되었다. 7% NaCl 농도에서는 21균주(24%)가 낮은 생장을 나타냈으며, 72균주(82%)가 약한 산 생성 활성을 나타냈고, 16균주(18%)는 산생성을 나타내지 않았다. 4% NaCl 농도에서는 모든 균주가 단백질 분해 활성을 나타냈지만, 5% NaCl 농도에서 86균주(98%)가 약한 활성을 나타냈고, 1균주(1%)는 활성을 나타내지 않았다. 지방 분해 활성은 모든 균주에서 나타나지 않았다. 모든 균주가 7종의 항생제(ampicillin, chloramphenicol, ciprofloxacin, gentamicin, penicillin G, tetracycline, vancomycin)에 대해 획득형 항생제 내성을 나타내지 않았다. Enterococcus 심내막염 항원 유전자 efaA 및 tyramine 생성에 관여하는 tyrosine decarboxylase 유전자 tdc가 메주 유래 88균주에서 발견되었지만, 사람 기원 E. faecium 균주가 특이적으로 보유하고 있는 Enterococcus surface protein 유전자 esp는 발견되지 않았다.

Sprague Dawley 흰쥐에서 테스토스테론에 의하여 유발된 전립선 비대증에 미치는 산수유 추출물의 영향 (Effects of Corni Fructus on Testosterone-induced Benign Prostatic Hyperplasia in Sprague Dawley Rats)

  • 노년기 비뇨기 계통에 가장 흔한 증상의 하나인 전립선 비대증은 요도를 둘러싼 전립선의 주위의 평활근과 상피세포의 과다 증식에 의한 것이다. 산수유는 강력한 항산화 효과로 인하여 다양한 질병의 예방 및 치료에 효과적이라고 보고되었지만 전립선 비대증에 대한 효능은 아직 알려지지 않았다. 본 연구에서는 산수유 열수 추출물이 testosterone에 의하여 유도되는 전립선 비대증에 미치는 영향을 조사하였다. 실험동물 내재성 testosterone의 영향을 배제하기 위해 거세를 하였으며, 전립선 비대증을 유도하기 위해, testosterone propionate (TP)를 피하 주사하였다. 산수유 추출물은 TP 주입과 함께 매일 경구 투여하였고, $5{\alpha}$-reductase type 2의 선택적 억제제인 finasteride를 양성 대조군으로 사용하였다. 본 연구의 결과에 의하면, 산수유 추출물 투여군에서는 finasteride 처리군에서와 마찬가지로 혈청 내 dihydrotestosterone 농도가 억제되었으며 전립선 무게 증가와 조직병리학적 변화가 유의하게 감소되었다. 산수유 추출물은 또한 전립선 조직 및 혈청에서 각각 TP에 의해 증가된 $5{\alpha}$-reductase type 2의 발현 및 농도를 유의적으로 억제하였다. 아울러 산수유 추출물은 TP에 의하여 유도된 AR, AR의 co-activator 및 세포증식 마커 단백질들의 발현 증가뿐 만 아니라 prostate-specific antigen의 수치와 발현도 감소시켰다. 결론적으로 산수유 추출물은 전립선 비대억제를 위한 식의약 소재로서의 개발 가능성이 매우 높음을 의미한다.

Coxiella burnetii 보체결합항체 검사에 의한 경기지역 젖소에 있어서의 Q열에 대한 혈청학적 조사연구 (Serological Study on Q Fever by Detection of Complement Fixation Antibodies to Coxiella burnetii in Dairy Cattle in Kyuuggi Province, Korea)

  • Regarding to Q fever which is one of the most important zoonoses in Food (Milk) Hygiene and in Environmental Public Health, a sero-epidemiological study was carried out to detect the complement fixation antibodies to Coxiella burnetii Nine Mile strain phase II antigen among the milking cows in Kyunggi Province. The results obtained were summarized as follows: 1. The overall prevalence of the CF antibodies to Q fever among 989 milking cows of 75 dairy farms in Kyunggi Province was revealed as high as 58.7% by the farms and 27.8% by the individual cows with higher prevalence in Kyunggi Central and Kyunggi East regions than any other regions in the Province. 2. Anticomplementary reactions were appeared as 7.5% (74/989) and it ranged from 1.0% to 16.0% according to the regions investigated. 3. In the titration of the positively reacted sera, the figures of 16.7%, 37.5%, 29.8%, 9.5%, 2.9% and 3.6% at the serum dilutions of 1: 10, 1: 20, 1: 40, 1: 80, 1: 160 and higher than 1: 160, respectively. 4. It was recognized that the relatively higher cumulated frequency distribution of the CF antibodies was shown in the sera collected from the regions with higher prevalence of Q fever. 5. There was a high correlation between the prevalence of Q fever CF antibodies and the age which is usually equivalent to one year older than the calving history of the milking cows.

  • Leptin is a circulating non-glycosylated protein that is mainly produced in adipocytes. Leptin acts in the brain to regulate food intake and energy expenditure. Previously we reported our success in the isolation of a partial cDNA of the long form of the leptin receptor, OB-Rb, from rat spleen, and showed that leptin might also play a role in peripheral immune organs. In the present study, for the first time, the complete coding region of OB-Rb cDNA was cloned from rat splenocytes, and its nucleotide sequence was determined. The cDNA was then further expressed in E. coli and mammalian cells, thereby confirming the functional integrity of this receptor. Prokaryotically overexpressed OB-R protein was then used as an immunizing antigen in BALE/c mice to produce leptin receptor-specific antibodies. By using them, we confirmed the cell surface expression of OB-Rb in transfected CHO cells. It is our belief that the reagents, as produced in this study, will be of great use in further studies of the biological role of rat leptin.

경구백신의 효율적인 적용을 위한 면역 보조제 개발 (Development of adjuvant for effective oral vaccine application)

  • Vaccine is one of the best known and most successful applications of immunological theory to human health and it protects human life through inducing the immune response in systemic compartment. However, when we consider the fact that mucosal epithelium is exposed to diverse foreign materials including viruses, bacteria, and food antigens and protects body from entry of unwanted materials using layer of tightly joined epithelial cells, establishing the immunological barrier on the lining of mucosal surfaces is believed to be an effective strategy to protect body from unwanted antigens. Unfortunately, however, oral mucosal site, which is considered as the best target to induce mucosal immune response due to application convenience, is prone to induce immune tolerance rather than immune stimulation. Since intestinal epithelium is tightly organized, a prerequisite for successful mucosal vaccination is delivery of antigen to mucosal immune induction site including a complex system of highly specialized cells such as M cells. Consequently, development of efficient mucosal adjuvant capable of introducing antigens to mucosal immune induction site and overcome oral tolerance is an important subject in oral vaccine development. In this review, various approaches on the development of oral mucosal adjuvants being suggested for effective oral mucosal immune induction.

한우의 주요 조직 적합성 항원 규명 (Characterization of major histocompatibility complex antigen on Korean native cattles)

  • The characterization of the MHC of domestic animals may constitute a first step towards increasing the efficiency of food production through improved disease resistance. In order to study the role of the MHC in regulating immune response it is first necessary to identify the different MHC alleles. In this research we try to investigate the possible associations between BoLA of Korean native cattles and infectious cattle disease. For this purpose we used one approach, serology. The results were summarized as follows : 1. Korean native cattle's lymphocyte reacted with alloantisera which recognized seven official BoLA allele. Korean native cattle's lymphocytes were reacted same as European breeds(especially with 673/3(W20)). 2. Korean native cattle's lymphocytes reacted with alloantisera 773/2, 673/3, 638/3, 773/3, 602/2, 639/2 and 639/3 at high reaction frequency. But alloantisera 642/1 was not expressed on Korean native cattle. If this allele, recognized by alloantisera(642/1), officially certificate In BoLA workshop it will be characterization factor of Korean native cattle. 3. According to cellular similarity index, we can presume on genetic relativity which has no family relationship.

Possible Protective Effects of Quercetin and Sodium Gluconate Against Colon Cancer Induction by Dimethylhydrazine in Mice

  • Micronutrients in food have been found to have chemopreventive effects, supporting the conclusions from epidemiologie studies that consumption of fresh fruits and vegetables reduces cancer risk. The present study was carried out to evaluate the role of querctin (Q) and sodium gluconate (GNA) supplementation separately or in combination in ameliorating promotion of colon tumor development by dimethyl-hydrazine (DMH) in mice. Histopathological observation of colons in mice treated with DMH showed goblet cell dysplasia with inflammatory cell infiltration. This pathological finding was associated with significant alteration in oxidative stress markers in colon tissues and carcinoembryonic antigen (CEA) levels in plasma. Mice co-treated with GNA and Q showed mild changes of absorptive and goblet cells and inflammatory cell infiltration in lamina properia, with improvement in oxidative stress markers. In conclusion, findings of the present study indicate significant roles for reactive oxygen species (ROS) in pathogenesis of DMH-induced colon toxicity and initiation of colon cancer. Also, they suggest that Q, GNA or the combination of both have a positive beneficial effect against DMH induced colonic cancer induction in mice.

Toxicity Study of Streptococcus pneumoniae Vaccine Administrated Subcutaneously in Rats

  • This study was performed to evaluate the toxicity of polysaccharide-based Streptococcus pneumoniae vaccine in Specific Pathogen Free (SPF), Sprague-Dawley (SD) rats. S. pneumoniae vaccine was administrated subcutaneously each dose level of high (560 ${\mu}g$/rat), medium (280 ${\mu}g$/rat) and low (140 ${\mu}g$/rat) on days 0, 14, 28. The rats were observed for 2 weeks or 4 weeks after the final injection. During this test, there were no significant dose-dependent changes in body weight, water and food consumption. In urinalysis and serum chemistry, dose-related changes were not detected. In hematology, the percent of neutrophils and lymphocytes in white blood cells were changed significantly. According to the measurement of organ weight, only spleen weight was significantly increased in all groups of administration compared to the control group. In the histopathological examination, an antigen-deposit, vacuolated macrophages, infiltrated inflammatory cells and a formation of granulation tissue were observed at the site of an administration. These results are considered as an outcome by immune responses through a vaccination. Consequently, the results of this study demonstrated that S. pneumoniae vaccine has no toxicity when it was administrated subcutaneously three times in 2-week interval at a high dose of 560 ${\mu}g$/rat.
