A review on advanced flow visualization techniques is presented particularly for applications to micro scale heat and mass transport measurements. Challenges, development and applications of micro scale visualization techniques are discussed for the study of heating/evaporating thin films, a heated micro channel, and a thermopneumatic micro pump. The developed methods are (1) Molecular Tagging Fluorescence Velocimetry (MTFV) using 10-nm caged seeding molecules (2) Micro Particle Velocimetry (MPIV) and (3) Ratiometric Laser Induced Fluorescence (LIF) for micro-resolution thermometry. These three methods are totally non-intrusive techniques and would be useful to investigate the temperature and flow characteristics in MEMS. Each of these techniques is discussed in three-fold: (1) its operating principle and operation, (2) its application and measurement results, and (3) its future challenges.
The recently developed correlative super-resolution fluorescence microscopy (SRM) and electron microscopy (EM) is a hybrid technique that simultaneously obtains the spatial locations of specific molecules with SRM and the context of the cellular ultrastructure by EM. Although the combination of SRM and EM remains challenging owing to the incompatibility of samples prepared for these techniques, the increasing research attention on these methods has led to drastic improvements in their performances and resulted in wide applications. Here, we review the development of correlative SRM and EM (sCLEM) with a focus on the correlation of EM with different SRM techniques. We discuss the limitations of the integration of these two microscopy techniques and how these challenges can be addressed to improve the quality of correlative images. Finally, we address possible future improvements and advances in the continued development and wide application of sCLEM approaches.
We have investigated quantitative measurement techniques of the degree of dispersion of single-walled carbon nanotubes (SWNTs). SWNTs were suspended in aqueous media using a sodium dodecyl sulfate (SDS) surfactant. SWNTs with different dispersion states were prepared by controlling the intensity and time of sonication and centrifugation. The laser spectroscopic techniques were employed to characterize the dispersion state; i.e., raman fluorescence and absorption spectroscopic techniques. Raman spectroscopy has been used to probe the dispersion and aggregation state of SWNTs in solution. Individually suspended SWNTs show increased fluorescence peaks and decreased roping peaks at a raman shift 267 $cm^{-1}$ compared with the samples containing bundles of SWNTs. The ultraviolet-visible-near infrared (UV-vis-NIR) absorption spectrum of decanted supernatant samples show sharp van Hove singularity peaks
An intercomparison experiment was implemented to assess the uncertainties and precisions om atmospheric SO$_{2}$ measurement techiques including a pulsed fluorescence (P-F), a diffusion-scrubber /ion chromatography (D-I), and a mist-chamber/ion chromatography (M-I). Each of those three techniques was investigated by researchers at Seoul National University, Yonsei University, and Hankuk University of Foreign Studies, respectively. The concentrations of atmospheric SO$_{2}$ were determined concurrently using three independent measurement techniques at the Seoul National University campus, Korea during Nov.22 to Dec.2, 1995. While the results from the P-F and D-I techinques showed the very close agreements (within +-5%) throughout the experiment period, M-I technique showed systematically smaller values (up to 30%) than the other two techniques. Although sources of larger discrepancy between different techniques were mot identified, the lower SO$_{2}$ values of the M-I method may be related to the errors associated with sample collection effciency, mass flow rate measurements, and standardization of ion chromatography.
Carbon monoxide (CO) is not only an important intermediate species in chemical reaction mechanisms of hydrocarbon fuel combustion, but also a crucial pollutant species emitted from automotive engines. To better understand the physical processes impacting CO emissions, the development of laser-based measurement techniques that can visualize in-cylinder CO distributions is desirable. Among these techniques, Laser-Induced Fluorescence (LIF) is a sensitive and species-selective detection technique capable of good spatial resolution. However, some technical matters such as deep UV excitation, severe pressure dependency of the LIF signal, and potential interference from other species have been major challenges for CO LIF application. This study is focused on investigating the feasibility of CO two-photon LIF in a direct-injection diesel engine operating at typical pressure and temperature conditions with commercial grade diesel fuel. Spectroscopic analysis shows that the CO fluorescence signal can be separated from $C_2$ Swan band or broadband fluorescence from PAHs when the signal is collected near 483 nm. The signal-to-noise ratio of CO LIF deteriorate rapidly as pressure is increased, following $P^{-1.49}$ which matches the theoretical signal pressure dependency.
Laser induced incandescence and laser induced fluorescence techniques have been investigated to measure the concentrations of soot and PAH, respectively. The Nd:YAG and dye lasers were used to form a sheet beam, and its wavelength were modulated to obtain a optimized signals of soot and PAH. Results showed that the relative size groups of soot and PAH can be measured by using our laser techniques.
The constituents of a watershed control a wide range of ecosystem processes, such as, carbon sequestration, nutrient retention, and biodiversity preservation. Maintenance of a healthy watershed is advantageous to humans in many direct and indirect ways. Dissolved organic matter fluorescence analysis is one of the most commonly utilized parameters for water quality measurement, pollution source tracking, and determination of the ecological state of a watershed. Throughout the recent decades, the advancement in data processing, instrumentation, and methods has resulted in many improvements in the area of watershed study with fluorescence analysis. The current trend of coupling advanced instrumentations and new comparative parameters, such as, microplastics of different types, antibiotics, and specific bacterial contaminants have been reported in watershed studies. However, conventional methodologies for obtaining fluorescence excitation emission matrices and for calculating the fluorescence and spectral indices are preferred to advanced methods, due to their easiness and simple data collection. This review aims to gain a general understanding of the use of dissolved organic matter fluorescence analysis for diagnosis and source identification of watershed pollutions, by focusing on how the studies have utilized fluorescence analysis to improve existing knowledge and techniques in recent years.

Recently the water management department advised the water treatment industry to focus on deploy the chemical free and the environmentally responsible process to adopt on water treatment plants in every country. In this objective, water treatment process started using ultrasonic based phycocyanin extraction with fluorescence measurement techniques to detect the change in the yield of phycocyanin. This paper propose the design of optical filter model for fluorescence technique based immersive optical phycocyanin measurement sensor design. The proposed design uses the multi-wavelength sensor module for irradiating part, and this plays a role of removing a wavelength band other than 590 ~ 620 nm. The preliminary study on immersed phycocyanin sensor, the fluorescence value of picocyanin according to the ultrasonic intensity, treatment time and number of cells was measured using JM phycocyanin module to emulate the proposed design, and were compared performance of the proposed sensor emulation. In this design, the phycocyanin fluorescence value increased about 2.1 ~ 4.7 times as the ultrasonic treatment time increased as compared with JM phycocyanin module, and the phycocyanin fluorescence value within the analysis range was obtained by ultrasonic treatment within one minute.
2D fluorescence sensors produce a great deal of spectral data during fermentation processes, which can be analyzed using a variety of statistical techniques. Principal component analysis (PCA) and a self-organizing map (SOM) were used to analyze these 2D fluorescence spectra and to extract useful information from them. PCA resulted in scores and loadings that were visualized in the score-loading plots and used to monitor various fermentation processes with recombinant Escherichia coli and Saccharomyces cerevisiae. The SOM was found to be a useful and interpretative method of classifying the entire gamut of 2D fluorescence spectra and of selecting some significant combinations of excitation and emission wavelengths. The results, including the normalized weights and variances, indicated that the SOM network is capable of being used to interpret the fermentation processes monitored by a 2D fluorescence sensor.
Fluorescence imaging is considered as one of the most powerful techniques for monitoring biomolecule activities in living systems. Near-infrared (NIR) light is advantageous for minimum photodamage, deep tissue penetration, and minimum background autofluorescence interference. Herein, we have developed a new NIR fluorescent dye, namely, RB-1, based on the Rhodamine B scaffold. RB-1 exhibits excellent photophysical properties including large absorption extinction coefficients, high fluorescence quantum yields, and high photostability. In particular, RB-1 displays both absorption and emission in the NIR region of the "biological window" (650-900 nm) for imaging in biological samples. RB-1 shows absorption maximum at 614 nm (500-725 nm) and emission maximum at 712 nm (650-825 nm) in ethanol, which is superior to those of traditional rhodamine B in the selected spectral region. Furthermore, applications of RB-1 for fluorescence imaging in living cells and small animals were investigated using confocal fluorescence microscopy and in vivo imaging system with a high signal-to-noise ratio (SNR = 10.1).
