Ascophyllum nodosum and its symbionts: XI. The epiphyte Vertebrata lanosa performs better photosynthetically when attached to Ascophyllum than when alone

  • Vertebrata lanosa is an abundant and obligate red algal epiphyte of Ascophyllum nodosum that forms part of a complex and highly integrated symbiotic system that includes the ascomycete, Mycophycias ascophylli. As part of ongoing studies to resolve interactions among species in the symbiosis, we used pulse amplitude modulation fluorimetry of chlorophyll a fluorescence, from photosystem II (PSII), to measure the maximum quantum yield ($F_v/F_m$) of PSII [$QY(II)_{max}$] and relative photosynthetic electron transport rates (rETR), as a function of light intensity, in order to evaluate the photosynthetic capacity of the two algal symbionts in the field and in the laboratory under different treatments. Our primary question was 'Is the ecological integration of these species reflected in a corresponding physiological integration involving photosynthetic process?' In the laboratory we measured changes in $QY(II)_{max}$ in thalli of V. lanosa and A. nodosum over one week periods when maintained together in either attached or detached treatments or when maintained separated from each other. While the $QY(II)_{max}$ of PSII of A. nodosum remained high and showed no significant variation among treatments, V. lanosa showed decreasing performance in the following conditions: V. lanosa attached to A. nodosum, V. lanosa in the same culture, but not attached to A. nodosum, and V. lanosa alone. These results are consistent with observations in which rETR was reduced in V. lanosa maintained alone versus attached to A. nodosum. Values for $QY(II)_{max}$ in V. lanosa measured in the field in fully submerged thalli were similar to those measured in the laboratory when V. lanosa was attached to it obligate host A. nodosum. Our results provide evidence of a physiological association of the epiphyte and its host that reflects the known ecology.

Possibility of Drought stress Indexing by Chlorophyll Fluorescence Imaging Technique in Red Pepper (Capsicum annuum L.) (고추의 엽록소 형광 이미지 분석법에 의한 한발스트레스 지표화 가능성)

  • The objectives of this study focused on measuring chlorophyll fluorescence related to drought stress comparing some parameters. Almost parameters were declined although they were not significant on the basis of mean values of fluorescence of total leaf area. While the ratio of fluorescence intensity variable chlorophyll ($F_V$) to fluorescence intensity maximal chlorophyll ($F_M$) was not changed, the effective quantum yield of photochemical energy conversion in photosystemII (${\Phi}PSII$) and chlorophyll fluorescence decrease ratio ($R_{fd}$) were slightly reduced, indicating inhibition of the electron transport from quinone bind protein A ($Q_A$) to quinone bind protein B ($Q_B$). Some parameters such as non-photochemical quenching rate ($NPQ_{_-LSS}$) and coefficients of non-photochemical quenching of variable fluorescence (qN) in mid-zone of leaf and near petiole zone leaf were significantly enhanced within 4 days after drought stress, which can be used as physiological stress parameters. Decrease in ${\Phi}PSII$ could was significantly measured in all leaf zones. In conclusion, three parametric evidences for chlorophyll fluorescence responses such as ${\Phi}PSII$, NPQ, and qN insinuated the possibility of photophysiological indices under drought stress.

  • Photoinhibition of photosystem (PS) n was induced in primary leaves of 25 day-old peppers grown $100\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1},\;at\;25^{\circ}C$. The modulation of PSII functionality in vivo was induced by varying both irradiance ($0-3000\;{\mu}molm^{-2}{\cdot}s^{-1}$) and duration (0-70 min) of light treatment. The functionality of PSII was investigated in terms of photochemical efficiency of PSII (Fv/Fm) and quantum yield of $O_2$ evolution, and expressed as a function of photon exposure [$mol\;photons{\cdot}m^{-2}$, the product of irradiance and duration of light treatment (Bell and Rose, 1981)]. Contrary to the linear decline of Fv/Fm ratio showing 50% decreases by absorption of $10\;mol\;photons{\cdot}m^{-2$, quantum yield of $O_2$ evolution decreased biphasically with increasing photon exposure, showing 50% decreases by absorption of $5.5\;mol\;photons{\cdot}m^{-2}$. Treatment of low temperature at $15^{\circ}C$ for 30 min alone did not affect the functionality of PSII, but high temperature ($45^{\circ}C$) significantly inactivated PSII activity. However, when Jeaves of pepper were subjected to low or high temperature in the presence of light, PSII was substantially photoinactivated. These results suggest the presence of different photoinhibitory mechanisms at low and high temperature.rature.

Quantitative Estimation of Binding Site Polarities of Water-Soluble Polyparacyclophanes in Aqueous Solution by Spectrofluorometry (형광광도법에 의한 수용액중 수용성 폴리파라시클로판류의 결합부위의 극성 측정)

  • The fluorescence characteristics of l-anilinonaphthalene-8-sulfonate (ANS) and 2-p-toluidinylnaphthalene-6-sulfonate (TNS) made the dyes useful probes for the determination of the polarity at the binding sites of several water-soluble polyparacyclophanes. Polyparacyclophanes used were 1,6,20,25-tetraaza[ 6.1.6.1]paracyclophane (CPM 44), 1,7,21,27 -tetraaza[7.1.7.1]paracyclophane (CPM 55). 1,7,21,27 -tetraaza-14,34-dioxa[7.1.7.1]paracyclophane (CPE 55) and 1,8,22,29-tetraaza-15,36-dioxa[8.1.8.1] paracyclophane (CPE 66). The fluorescence quantum yield, emission maximum, and half bandwidth of ANS or TNS obtained in a variety of solvent systems were plotted as a function of four kinds of empirical solvent polarity scales such as dielectric constant (D), (D-l)/(2D+1). Y and Z values. It was found that the Z-value-emission maximum $(\overline}V_F,\;cm^{-1})$ profile showed the most reliable linearity. ANS and TNS interacted with CPM 44, CPM 55, CPE 55. CPE 66. ${\alpha}-cyclodextrin$ (CyD) and ${\beta}-CyD$ in the aqueous solution, and from the emission maxima the polarities (Z-value) of their binding sites were calculated to be 92.65, 87.50, 93.35, 84.52, 94.36, and 90.48 for ANS, respectively. and 91.07, 89.68, 85.44, 86.74 and 87.6 for TNS except for ${\alpha}-CyD$, respectively.

Photosynthetic Response and Protective Regulation To Ultraviolet-B Radiation In Green Pepper (Capsicum annuum L.)Leaves

  • The deteriorative effect of ultraviolet-B(UV-B) radiation on photosynthesis was assessed by the simultaneous measurement of O$_2$ evolution and chlorophyll(Chl) fluorescence in green pepper. UV-B was given at the intensity of 1 W$.$m$\^$-2/, a dosage often encountered in urban area of Seoul in Korea, to detached leaves. Both Pmax and quantum yield of O$_2$ evolution was rapidly decreased, in a parallel phase, with increasing time of UV-B treatment. Chl fluorescence parameters were also significantly affected. Fo was increased while both Fm and Fv were decreased. Photochemical efficiency of PSII(Fv/Fm) was also declined, although to a lesser extent than Pmax. Both qP and NPQ were decreased similarly with increasing time of UV-B treatment. However, PS I remained stable. The addition of lincomycin prior to UV-B treatment accelerated the decline in Fv/Fm to some extent, suggesting that D1 protein turnover may play a role in overcoming the harmful effect of UV-B. The amount of photosynthetic pigments was less affected than photosynthetic response in showing decline in Chl a and carotenoids after 24 h-treatment. Presumptive flavonoid contents, measured by changes in absorbance at 270 nm , 300 nm and 330nm, were all increased by roughly 50% after 8 h-treatment. Among antioxidant enzymes, activities of catalase and peroxidase were steadily increased until 12h of UV-B treatment whereas ascorbate perxidase, dehydroascorvate reductase and glutathione reductase did not show any significant change. The results indicate that deteriorative effect of UV-B on photosynthesis precedes the protection exerted by pigment synthesis and antioxidant enzymes.

Application of Chlorophyll Fluorescence Imaging Technique to Estimate Fresh Weight in Kiwifruit (엽록소 형광이미징 기술을 이용한 키위과일의 생체중 예측)

  • BACKGROUND: Fresh weight is one of the major quality measurement factors in determining the quality of fresh fruits. A practical method has been developed for rapid and non-destructive measurement using the Chlorophyll Fluorescence Image (CFI) technique to estimate changes in fresh weight of post-harvest products. METHODS AND RESULTS: Kiwifruit (Actinidia deliciosa) was used and measured for the fresh weight and CFI under different temperature conditions at 0, 10, and 20℃, from 0 to 21 days after storage (DAS). We observed the fresh weight of kiwifruit and measured the surface image for determining Fv/Fm value in terms of maximum quantum yield on each day. To estimate freshness of kiwifruit we applied linear regression between the measured fruit weights and Fv/Fm values. Results showed that fruit weights were reduced by 4% at 0℃, 6% at 10℃, and 14% at 20℃ for 21 days, respectively. And also, the value of Fv/Fm was shown as decreasing trend at all temperature conditions, especially at 20 ℃. Fv/Fm values showed highly significant correlation (R2>0.9) with fresh weight of kiwifruit at all different storage temperatures. CONCLUSION: Thus, CFI technique can be useful to estimate the fresh weight of kiwifruit.

Effect of Calcium Chloride($CaCl_2$) on Chlorophyll Fluorescence Image and Photosynthetic Apparatus in the Leaves of Prunus sargentii (염화칼슘 처리가 산벚나무 엽의 엽록소형광반응과 광합성기구에 미치는 영향)

  • There is a little information on the effect of calcium cloride ($CaCl_2$) which is used as deicing salt in Korea on the physiological responses of the street trees. Prunus sargentii is one of the most widespread tree species of street vegetation in Korea. In this study, the effect of $CaCl_2$ on photosynthetic apparatus such as chlorophyll fluorescence image and light response curve of P. sargentii in relation to their leaf and root collar growth responses were investigated. To study the effect of $CaCl_2$ treatment in the early spring, we irrigated twice in rhizosphere of P. sargentii (3-year-old) planted plastic pots with solution of 0.5%, 1.0%, 3.0% $CaCl_2$ concentration before leaf expansion. Results after treatments, total chlorophyll contents and the chlorophyll a/b, photosynthetic rate, quantum yield, dark respiration decreased with increasing $CaCl_2$ concentration. On the contrary, light compensation point increased with increasing $CaCl_2$ concentration. Through the linear regressions of correlation of photosynthetic rate with photosynthetic parameters (quantum yield, dark respiration and light compensation point), we found a significant relationship (p<0.05) between photosynthetic rate and quantum yield and light compensation point except dark respiration. Calcium cloride ($CaCl_2$) induced inhibition of photochemical efficiency ($F_v/F_M$) and non-photochemical quenching (NPQ) were found in treatments of $CaCl_2$, and these reduction rates between control and CaCl2 treatments were drastically showed at 80 days. We suggest that physiological activities are limited from treatment of $CaCl_2$. These reductions of photosynthetic apparatus ability caused eventually the reduction of leaf and diameter at root collar growth.

Effects of NaCl on the Growth and Physiological Characteristics of Crepidiastrum sonchifolium (Maxim.) Pak & Kawano (NaCl 처리가 고들빼기의 생장과 생리적 특성에 미치는 영향)

  • Background: This study was conducted to investigate the effects of NaCl concentration on the photosynthetic parameters, chlorophyll fluorescence and growth characteristics of Crepidiastrum sonchifolium. Methods and Results: As treatments, we subjected C. sonchifolium plants to four different concentrations of NaCl (0, 50, 100 and 200 mM). We found that the photosynthetic parameters maximum photosynthesis rate (PN max), net apparent quantum yield (Φ), maximum carboxylation rate (Vcmax), and maximum electron transport rate (Jmax) were significantly reduced at an NaCl concentration greater than 100 mM. In contrast, there was an increase in water-use efficiency with increasing NaCl concentration, although in terms of growth performances, leaf dry weight, root dry weight, stem length, and total dry weight all decreased with increasing NaCl concentration. Furthermore, leakage of electrolytes, as a consequence of cell membrane damage, clearly increased in response to an increase in NaCl concentration. Analysis of the polyphasic elevation of chlorophyll a fluorescence transients (OKJIP) revealed marked decrease in flux ratios (ΦPO, ΨO and ΦEO) and the PIabs, performance index in response to treatment with 200 mM NaCl, thereby reflectings the relatively reduced state of photosystem II. This increase in fluorescence could be due to a reduction in electron transport beyond Q-A. We thus found that the photosynthetic parameters, chlorophyll fluorescence and growth characteristics of C. sonchifolium significantly increased in response to treatment with 200 mM NaCl. Conclusions: Collectively, the findings of this study indicate that C. sonchifolium shows relatively low sensitivity to NaCl stress, although photosynthetic activity was markedly reduced in plants exposed to 200 mM NaCl.

Different modes of antibiotic action of homodimeric and monomeric bactenecin, a cathelicidin-derived antibacterial peptide

  • The bactenecin is an antibacterial peptide with an intramolecular disulfide bond. We recently found that homodimeric bactenecin exhibits more potent antibacterial activity than the monomeric form and retains its activity at physiological conditions. Here we assess the difference in the modes of antibiotic action of homodimeric and monomeric bactenecins. Both monomeric and dimeric bactenecins almost completely killed both Staphylococcus aureus and E. coli within 10-30 min at concentrations of $8-16\;{\mu}M$. However, exposure to liposomes elicited an increase in the fluorescence quantum yield from a tryptophan-containing monomeric analog, while the homodimeric analog showed a significant reduction in fluorescence intensity. Moreover, unlike the monomer, the homodimer displayed apparent membrane-lytic activity enabling release of various sized dyes from liposomes, and rapidly and fully depolarized the S. aureus membrane. Together, our results suggest that homodimeric bactenecin forms pores in the bacterial membrane, while monomeric one penetrates through the membrane to target intracellular molecules/organelles.

Synthesis and Spectral Properties of 1,2-Bispyrazyl Ethylene (1,2-비스피라질 에틸렌의 합성과 분광학적 성질에 대한 연구)

  • A new compound, 1,2-bispyrazyl ethylene,is synthesized starting from pyrazine carboxylic acid and methyl pyrazine. The compound is characterized utilizing UV-VIS, IR, NMR and mass spectra along with elemental analysis. Spectroscopic properties are studied from UV-VIS and fluorescence spectra. From unusual salt effects on fluorescence spectra, it is believed that $(n,\;{\pi}^*)$ state has about the same energy as $({\pi},\;{\pi}^*)$ state. The compound fluoresces from $({\pi},\;{\pi}^*)$ state with the quantum yield of 0.025 at $77^{\circ}K$ compared to near unity for stilbene at the same temperature indicating the efficient intersystem crossing to triplet state, because of strong $(n,\;{\pi}^*)$ and $({\pi},\;{\pi}^*)$ mixing in the lowest excited state.
