  • Quantum yield of photoisomerization of degassed n-hexane solution of all-trans-retinal (1) and all-trans-1, 4-diphenylbutadiene (4) at 25$\circ$C under direct irradiation conditions increases as the concentrations of 1 and 4 increase. Further, fluorescence lifetime of 4 at ambient temperature is also found to increase as the concentrations of 4 are increased. The results are discussed in terms quantum chain process which occurs due to exchange of energy between various conformers of the compounds concerned.

  • This study explores recent research trends and potential applications of ultrasound optical imaging-based multimodal technology. Ultrasound imaging has been widely utilized in medical diagnostics due to its real-time capability and relative safety. However, the drawback of low resolution in ultrasound imaging has prompted active research on multimodal imaging techniques that combine ultrasound with other imaging modalities to enhance diagnostic accuracy. In particular, ultrasound optical imaging-based multimodal technology enables the utilization of each modality's advantages while compensating for their limitations, offering a means to improve the accuracy of the diagnosis. Various forms of multimodal imaging techniques have been proposed, including the fusion of optical coherence tomography, photoacoustic, fluorescence, fluorescence lifetime, and spectral technology with ultrasound. This study investigates recent research trends in ultrasound optical imaging-based multimodal technology, and its potential applications are demonstrated in the biomedical field. The ultrasound optical imaging-based multimodal technology provides insights into the progress of integrating ultrasound and optical technologies, laying the foundation for novel approaches to enhance diagnostic accuracy in the biomedical domain.

Photocycloaddition Reaction of 4',5'-Dihydropsoralen to Tetramethylethylene (4',5'-디히드로소랄렌과 테트라메틸에틸렌의 광고리화 첨가반응에 관한 연구)

  • A psoralen-DNA monoadduct analogue, 4',5'-dihydropsoralen (DHP) is synthesized and its photophysical properties such as fluorescence quantum yield (0.08), phosphorescence quantum yield (0.013), fluorescence lifetime (0.95ns), and phosphorescence lifetime(0.039s) were measured in ether solution and in frozen matrix state at 77K. The photocycloaddition reaction of DHP with tetramethylethylene(TME) was carried out in solution and in the frozen state. The major photoadduct was isolated and characterized by elemental analysis and physical methods such as UV, IR, NMR, and mass spectrometry. The major DHP-TME photoadduct was proved to be an 1 : 1 $C_4$-cycloadduct formed through 2 + 2 cycloaddition of the pyrone double bond of DHP to TME.

Core region and optical properties of Er3+ doped Y3Al5O12 single crystals (Er3+ doped Y3Al5O12 단결정의 core 영역 및 광학적 특성)

  • $Er^{3+}$ doped $Y_3Al_5O_{12}$ (Er:YAG) single crystals, in which the concentrations of $Er^{3+}$ ion were 5, 7.3, 8, and 10 at.%, were grown by the Czochralski method under nitrogen atmosphere. The <111> oriented Er:YAG single crystals with diameters of up to 50 mm were grown at a pulling rate of 1.0 mm/h and rotation rate of 10 rpm. The thick part of the core region was generated mainly when there was a diameter change during the crystal growth. The concentrations of $Er^{3+}$ ion in the crystals were the same as it was in the melt. $Er^{3+}$ concentration of core region was slightly higher than the other regions in the compositional analysis. The fluorescence lifetime was saturated according to the increase of $Er^{3+}$ doping concentrations.

  • The picosecond time resolved fluorescence spectra of Hematoporphyrin Derivative (HPD) in both solutions and cancer cell are measured by a time correlated single photon counting system with a synchronously mode locked dye laser. Two exoponential decay components in the fluorescence spectra were observed. The slow decay(6.3 ㎱)and the fast one(350 ㎰)are attributed to be originated from monomers and dimers, respectively. The absorption and fluorescence measurements in steady state also showed the presence of a monomeric and dimeric forms of HPD molecules. The monomer lifetime in the cancer cell was measured to be longer than that in solution, which was expected from the blue shift and narrowing of the absorption spectra for HPD-treated in vitro. The relative amplitude of the fast component was found to be enhanced in cancer cell, strongly indicating the higher affinity of the dimer for the cancer cell.

  • Fluorscence in situ hybridization with chromosome-specific probe has been shown to be a valid and rapid method for detection of chromosome rearrangements induced by chemical and physical agents. This method is useful for quantifying structural aberrations, expecially for stable ones, such as translocation and insertion, which are difficult to detect with conventional method in human lymphocyte. In order to use the FISH method as a biodosimeter for monitoring human population exposed to various chemical and physical agent, baseline level of chromosome rearragement was established. Blood from forty four healthy adults were collected and analysed with whole chromosome-specific probes by human chromosome 1,2 and 4. The frequencies of stable translocation were 2.45 per 100 cell equivalent and those of insertion, color juction, acentric and dicentric were 0.32, 3.28, 0.23 and 0.27 per 100 cell equivalent respectively. The frequencies of chromosome rearragements increased with age in both sexes except for dicenrics. From above result, stable aberrations accumulate with age and it may reflect integrated lifetime exposure of adverse environment.

Spectroscopic Study on Three States of Water in the Reverse Micelle Using Methylene Blue as a Probe (Methylene Blue를 이용한 역미셀에서 물의 세 가지 상태에 대한 분광학적 연구)

  • In order to find out the microscopic environmental information on the nonionic reverse micelle of Triton X-100/n-hexanol/water in cyclohexane, an absorption and fluorescence spectroscopic study has been conducted using a methylene blue(MB). The information on the microscopic states of water in the polar core of the reverse micelle has been found by investigating complex formation and solvatochromic behavior between MB and Triton X-100. As a result, it was found that there exist three states in the polar core of the reverse micelle. The measured values of $W(=[H_2O]/[Surf])$ for the three states of water are 0.71, 4.98, and 7.26, and the corresponding lifetimes of MB are $15.45 ns{\pm}0.56$, $12.27 ns{\pm}0.79$, and $8.28 ns{\pm}0.82$, respectively.

Relaxation Process of the Photoexcited State and Singlet Oxygen Generating Activity of Water-soluble meso-Phenanthrylporphyrin in a DNA Microenvironment

  • To examine the microenvironmental effect of DNA on the photosensitized reaction, the electron-donor-connecting porphyrin, meso-(9-phenanthryl)-tris(N-methyl-p-pyridinio) porphyrin (Phen-TMPyP), was synthesized. Phen-TMPyP can bind to oligonucleotides with two binding modes, depending on the DNA concentration. The fluorescence lifetime measurement of Phen-TMPyP shows a shorter component than that of the reference porphyrin without the phenanthryl moiety. However, the observed value is much longer than those of previously reported similar types of electron-donor-connecting porphyrins, suggesting that electron-transfer quenching by the phenanthryl moiety is not sufficient. The fluorescence quantum yield of Phen-TMPyP ($5{\mu}M$) decreased with an increase in DNA concentration of up to $5{\mu}M$ base pair (bp), possibly due to self-quenching through an aggregation along the DNA strand, increased with an increase in DNA concentration of more than $5{\mu}M$ bp and reached a plateau. The fluorescence quantum yield of Phen-TMPyP with a sufficient concentration of DNA was larger than that of the reference porphyrin. The singlet oxygen ($^1O_2$) generating activity of Phen-TMPyP was confirmed by the near-infrared emission spectrum measurement. The quantum yield of $^1O_2$ generation was decreased by a relatively small concentration of DNA, possibly due to the aggregation of Phen-TMPyP, and recovered with a sufficient concentration of DNA. The recovered quantum yield was rather smaller than that without DNA, indicating the quenching of $^1O_2$ by DNA. These results show that a DNA strand can stabilize the photoexcited state of a photosensitizer and, in a certain case, suppresses the $^1O_2$ generation.

$Cr^{3+}$ Luminescent centers in $BeAl_6O_{10}$ crystal

  • The fluorescence emission spectrum of Cr$^{3+}$ doped BeAl$_{6}$ O$_{10}$ crystals at 300 K contains a broad band, three R-like lines and another emission lines. It has been identified by a lifetime resolution spectroscopic technique that there are three kinds of single-Cr$^{3+}$ centers, Cr(I), Cr(II), Cr(III), in this crystal. Cr(I) and Cr(II) are high-crystal field centers responsible for the three main "R-lines", and Cr(III) is a low-crystal field center responsible for the main broad band emission. Photosensitized oxidative damage of human serum albumin by water-soluble dichlorophosphorus(V) tetraphenylporphyrin

  • Biomolecular photo-damaging activity of a water-soluble cationic porphyrin was examined using human serum albumin (HSA), a water-soluble protein as a target biomolecule model by a fluorometry. Dichlorophosphorus(V) tetraphenylporphyrin ($Cl_2P(V)TPP$), was synthesized and used as a photosensitizer. This porphyrin could bind to HSA and cause the photosensitized oxidation of HSA through the singlet oxygen generation and the oxidative photo-induced electron transfer (ET). Near infrared emission spectroscopy demonstrated the photosensitized singlet oxygen generation by this porphyrin. Decrement of the fluorescence lifetime of $Cl_2P(V)TPP$ by HSA supported the ET mechanism. Furthermore, the estimated Gibb's energy indicated that the ET mechanism is possible in the terms of energy. Because oxygen concentration in cancer cell is relatively low, ET mechanism is considered to be advantageous for photosensitizer of photodynamic therapy.
