Occurrence of Sword bean Scab Caused by Cladosporium cucumerinum in Korea

  • A black scab disease occurred on sword bean (Canavalia gladiata) in plastic film houses around Chinju area during the spring season of 1999. The disease started from flower bud, then moved to flower stalk, pod, petiole, cirrus, stem and leaves. The lesions started with small dark brown spots then were gradually expanded. Severely infected plants reached 37.4% of whole plant covered with scab. Numerous conidia were produced on the diseased flower disk, pod, floral axis, stem and leaves. Most of the conidia were appeared to be readily dispersed in the air, but the mycelia were not suggested causing of sooty mold by ectoparasitism. A fungus was isolated from the diseased stem, and inoculated to healthy plants to satisfy the Koch's postulates and proved the fungus was the causal agent of the disease. The isolated fungus grew on potato dextrose agar, forming greenish black to pale brown colonies. Conidia were ellipsoidal, fusiform or subspherical, mostly one-celled but occasionally septated. The conidia were $3.9{\sim}34.1{\times}2.7{\sim}5.1\;{\mu}m$ in size and formed in long branched chains on the erected conidiophores which were pale olivaceous brown and variable in length between $7.2{\sim}210.7\;{\mu}m$ in size. Ramoconidia were $7.6{\sim}29.2{\times}3.2{\sim}14.4\;{\mu}m$ in size. The fungus was identified as Cladosporium cucumerinum based on the above morphological characteristics. The optimum temperature for mycelial growth and conidial formation was about 15 to $25^{\circ}C$. Cladosporium scab of sword bean caused by the fungi has not been reported in Korea previously.

Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85 (황색종 연초 돌연변이 계통 KF8832-85의 특성)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars. Flowering and Growth of Dendrobium phalaenopsis as influenced by Photoperiod, Temperature, and Plant Growth Regulators (온도, 일장 및 식물생장조절제에 의한 덴파레의 생육 및 개화 반응)

  • This study was conducted to investigate effect of temperature, photoperiod, and plant growth regulators on the growth and flowering of Dendrobium phalaenopsis. Photoperiod treatments of 10 or 16 day length were from Mar. 24 to May 23. After treatments, all plants were exposed under natural photoperiod. Temperature treatments promoted development of new shoots (leads) of Dendrobium phalaenopsis 'Semi Alba' at $30/25^{\circ}C$(day/night) for 12 weeks, and 16 hr-photoperiod treatment promoted development of new shoots and flowering. The 16 hr photoperiod Dendrobium phalaenopsis 'Candistraipe ${\times}$ Tedtakiguz' no difference in the numbers of flower, flower width, and stalk length compared to the 10 hr photoperiod. Days to first flowering from appearance of new shoot were 241 days under natural photoperiod, 243 days under 10 hr, and 216 days under 16 hr, so that 16 hr-photoperiod treatment was faster about 25 days than that of natural or 10 hr treatment. There was not significant difference in growing and flowering of Dendrobium phalaenopsis among plant growth regulators.

Chemical Properties of Cut-flower Rose-growing Soils in Plastic Film Houses (절화장미 시설 재배지 토양의 화학적 특성)

  • Rose is one of the major cut-flower crops in Korea. A survey was conducted to investigate the chemical properties of soils cultivating rose under plastic film house at 72 farms in Korea. Soil texture distribution of the samples were loam(43%), silty clay loam(19%), and clay loam(17%). The average chemical properties of the surface soils were pH 5.85, electrical conductivity $3.61dS\;m^{-1}$, organic matter $37g\;kg^{-1}$, nitrate nitrogen $205mg\;kg^{-1}$, phosphorous as Av. $P_2O_5$ $844mg\;kg^{-1}$, and exchangeable potassium $1.41cmol_c\;kg^{-1}$. Available phosphate and exchangeable potassium were accumulated at the level higher than the optimum content in over 70% of the surveyed farms. The flower stalk length was positively correlated with the organic matter content in soil, but was negatively correlated with the electrical conductivity in Red Sandra cultivar.

  • All the parts of lotus, including the seed, rhizome, leaf, stalk, petal, anther, pericarp, and fruit receptacle, have been used in traditional medicine system as a health beneficial supplement. However the most usually used from lotus plant is only the root. Therefore in this study, it will be discussed more the utilization of other parts of the lotus, namely the flower of lotus. The petals and stamens of lotus actually are also rich in bioactive components such as flavonoids and alkaloids, are used in the treatment of tissue inflammation, cancer, skin disease, and also for us as antidotes. One of the biotechnological process that can be used to improve the nutritional content, sensory, and also antioxidant activities is fermentation process. The final product desired from the fermentation process in this study is vinegar. The microbial strain powder used is Uinkin fermented powder with three variations of fermentation. The variations given in this study were initial sugar 32%, 24%, and 14% with the same fermentation temperature, $35^{\circ}C$ for 3 months. The results obtained showed that the pH value and sugar content of products during the fermentation process were decreasing during the fermentation process, with total polyphenol content of $283.7{\pm}97.6mg/100g\;QAE$, and total flavonoid content of $3.3{\pm}0.0mg/100g\;QAE$. For the DPPH radical scavenging ability of the fermentation product also increased in a concentration dependent manner, with ORAC activity of the product showed a high activity of $20.7{\pm}0.41{\mu}M$ TE. Therefore, fermentation process can be the one of method for improving the product. The efficiency of lotus flower vinegar fermentation can be reached with an initial sugar condition of 25% (sample B).

Cymbidium Hybrid 'Purple Princess' with Dark Purple Flower (진한 자주색계 중형 심비디움 'Purple Princess')

  • $Cymbidium$ 'Purple Princess' ('Show Girl' x 'Eiko') is a new cultivar having dark purple sepal and petal (RHS, RP59A) with purple lip (RHS, RP59A). A cross was made between pink colored $C.$'Show Girl' as maternal line and dark purple colored $C.$ 'Eiko' as paternal line in 1994. It was preliminarily selected as Line No. 94019267 in 1999 considering the characters of flower shape and color, leaf growing shape and growth habit in normal culture condition. Multiplication, cultivation, and characteristic trials were conducted from 2000 to 2003. The line was named as Wongyo F1-7 and phenotype was characterized in 2004 as a new 'Purple Princess'. 'Purple Princess' has about 13.2 flowers per flower stalk and flower size of 5.2 cm. General appearance of petals and sepals is slightly incurved shape. The plant size is intermediate having erect peduncle. Blooming starts from early December under optimal culture condition. Leaf attitude and twisting is erect and very weak respectively. Erect leaf will be suitable for growing and handling in the nursery as pot orchid flowers.

GENETIC ANALYSIS OF QUANTITATIVE CHARACTERS IN $F_2$ POPULATIONS OF KOREA LOCAL AND ORIENTAL TOBACCO VARIETIES (N. TABACUM L.) (재래종과 Orient종 연초 $F_2$세대의 양적 형질에 대한 유전분석)

  • In order to obtain the basic information for the successful breeding of aromatic tobacco (N. tabacum L.), the combining ability and the mode of inheritance in 28 $F_2$ hybrids by diallel crosses between Korea local and oriental varieties were investigated. Estimates of general combining ability were significant for all characters investigated, and the specific combining ability was significant for stalk height, leaves per plant, leaf length, leaf width and days to flower. The partial dominance was exhibited by all characters investigated as well as additive effects were larger than dominance effects for all characters.

Rhizome Growth and Flowering of White Lotus (Nelumbo nucifera) at Muan Hoesan-Lake (무안 회산지(回山池) 백련꽃의 근경생장과 개화특성)

  • This study was conducted to examine characteristics of rhizome growth and flowering of white lotus (Nelumbo nucifera) plant in Muan Hoesan-lake for collecting the basic data on the cultivation and the flowering which is growing in Muan Hoesan-lake of Jeonnam province in Korea. We have planted from the seeds and the enlarged rhizomes of the white lotus which were planted in the plastic containers on April 20 and May 4, 2008, and cultivated under rain shelter condition. Rhizome growth pattern and the flowering characteristics of the white lotus were examined on September 4, 2008. Main rhizomes of the white lotus propagated by the seeds and the enlarged rhizomes were branched by 10% and 15%, respectively. Numbers of nodes in the bloomed and non-bloomed rhizomes of the white lotus were over 14 and below 11, respectively. Flower was not observed in the white lotus when propagated with seeds, however, flowers came out up to 80% by the enlarged rhizomes. First flowers were bloomed from the late July and to the early August, and the number of flowers were 4.0~4.4. The first flower stalks without branching of the main rhizome were emerged in 8.8th node, however, that with branching were 8.0th node. Time to flowering from the emergence of flower stalk on the above-ground parts were required 16 days. White flowers from the lotus were 15.3 cm in height and 28.2 cm in width, their longevity was 3.3 days and the averaged flower fresh weight was 29.2 g. The results conclude that optimum number of nodes on the main rhizomes for blooming the white lotus flowers should be over 8 nodes before the rhizome enlargement.

Morphological characteristics and genetic diversity of Calanthe species native to Korea (한국 자생 새우난초의 형태적 특성 및 유전적 다양성)

  • This study was conducted to research the morphological characteristics and analyze the genetic diversity by using RAPD in Calanthe species native to Korea. Nine samples were selected by flower color and 19 morphological characteristics. In the length and width of leaf, dorsal sepal, the lateral sepal, the petal, the central lip, and the lateral lip, C. discolor was the shortest and narrowest, respectively, but C. sieboldii was the longest and the widest, respectively. The flower stalk length was the shortest in C. discolor, and the longest in C. sieboldii. Three variants were the intermediate between C. discolor and C. sieboldii in the above morphological characteristics, but spur length was the longest in C. discolor, the shortest in C. sieboldii, and intermediate in the variants. The ovary length in C. discolor was shortest and C. sieboldii and variants were similar with each other. The flower color of C. discolor were brownish red, the value of CIE Lab was between 40 and 50. The flower color of C. sieboldii was yellowish, the value of CIE Lab was between 110 and 130. And variants had various colors between 50 to 70 in the value of CIE Lab. By analyzing multiple band patterns of PCR products, 154 bands were selected as polymorphic RAPD markers. The analysis of genetic similarity of Calanther species using RAPD showed that C. discolor and C. sieboldii are more distant from each other than variants, and these results demonstrated that genetic position of variants located between C. discolor and C. sieboldii.

  • This experiment was conducted to examine characteristics of agronomic characters and estimate of gene effect for several mutant characters. The genetic populations were derived from cross between 83H-5 and Hicks. There were significant difference for plant height, stlk height, leaf shape and bacterial wilt disease index except leaf number, leaf length, and what is more, F3 variance is more than Bl and B2 generation from cross 83H-5 X Hicks. Gene actions for stalk height and bacterial wilt disease were estimated by 3-parameter, and by 6- parameter model for all characters except above two characters but stalk height and bacterial wilt disease index are not significant in the additive and dominance effects. Dominant$\times$dominant epitasis for plant height, dominant and dominant$\times$dominant epistasis for leaf length, additive and additive$\times$additive and dominant$\times$dominant epistasis for leaf width, and additive and additive$\times$dominant epistasis for days to flower were appeared significant in gene action.
