Background: CD11c, also known as integrin alpha x, is one of the optimum markers of dendritic cells. However, the regulation of the CD11c expression in mouse has not been identified yet. In this study, in order to analyze the regulation of CD11c expression, the promoter of CD11c was cloned and characterized. Methods: To identify the promoter portion, various sizes of what are considered to be CD11c promoter fragments was amplified by polymerase chain reaction (PCR), using mouse genomic DNA as a template. After sequence was obtained, these fragments were transfected into various cell lines including mouse dendritic cell lines such as JAWSII and DC2.4 and L929 as control cell line.. The promoter activity of three promoter fragments was measured and compared by luciferase activity in the transfected cells. Results: Three clones with size of 1kb, 3kb and 6kb were obtained from mouse genomic DNA. Flow cytometry analysis of JAWSII cells revealed that 52% of the cells expressed CD11c, which was confirmed by RT-PCR analysis. On the contrary, L929 and DC 2.4 cells did not express CD11c. The CD11c+ JAWSII cells were enriched from 52% to 90% with cell sorter. The comparative luciferase activity analyisis demonstrated that the region responsible for tissue specific expression was contained within -3 kb and the clone with size of 3 kb particularly showed higher luciferase activity than 6 kb and 1 kb clones. Conclusion: The CD11c promoter region containing the region responsible for tissue specificity was successfully cloned and -3 kb region showed the highest activity.
This study was carried out to observe the direct effect of hydrocortisone on renal function by infusing it into a renal artery. Hydrocortisone (5mg/kg) or saline (0.5 ml/kg) was infused directly into the left renal artery of the rabbit, the right kidney was left intact to serve as a control for general action of acetazolamide (10 mg/kg) or aminophylline (10 mg/kg), which was administered intravenously 30 minutes after the direct infusion of pretreated drugs (hydrocortisone or saline). The changes of urine volume, pH, urinary excretion rates of $Na^+,\;K^+\;and\;Cl^-$, and the clearances of inulin and PAH were measured at an interval of 10 minutes for half an hour after the direct infusion of hydrocortisone or saline, and for one hour after intravenous administration of acetazolamide or aminophylline. The results of the experiment were as follows: 1. Significant changes in urine volume and urinary electrolytes (excreted rates of $Na^+,\;K^+\;and\;Cl^-$) were observed in the hydrocortisone-infused group 10 minutes after the administration of acetazolamide, compared with the saline-infused group. Especially, the effect was more potent on the infused (left) side than on the contralateral (right) side. 2. Significant changes in urine volume and urinary electrolytes were also observed in all the aminophylline-treated groups, but no remarkable difference was noticed between the hydrocortisone-infused group and the saline-infused group, nor between the left and right sides. 3. No signicant changes in the clearances of inulin and PAH were in the infused (left) side of all the experimental groups, as compared with the contralateral (right) side. From the above results, it is obvious that hydrocortisone infused into a renal artery exerts diuretic action when administered in combination with acetazolamide, and the mechanism of action rests not on its hemodynamic change for renal blood flow, but on the potentiation of carbonic anhydrase inhibiting action. However, the exact mode of action remains yet to be clarified.

The effects of recirculated exhaust gas on the characteristics of $NO_x$ and soot emissions under a wide range of engine load have been experimentally investigated by a water-cooled, four-cylinder, indirect injection, four cycle and marine diesel engine operating at two kinds of engine speeds. The simultaneous control of $NO_x$ and soot emissions in diesel engines is targeted in this study. The EGR system is used to reduce $NO_x$ emissions, and a novel diesel soot removal device with a cylinder-type scrubber for the experiment system which has 6 water injectors(A water injector has 144 nozzles in 1.0 mm diameter) is specially designed and manufactured to reduce the soot contents in the recirculated exhaust gas to intake system of the engines. The intake oxygen concentration and the mean equivalence ratio calculated by the intake air flow and fuel consumption rate, and the exhaust oxygen concentration measured are used to analyse and discuss the influences of EGR rate on $NO_x$ and soot emissions. The experiments are performed at the fixed fuel injection timing of $15.3^{\circ}$ BTDC regardless of experimental conditions. It is found that $NO_x$ emissions are decreased and soot emissions are increased owing to the drop of intake oxygen concentration and exhaust oxygen concentration, and the rise of equivalence ratio as the EGR rate rises.
Sedimentation basin plays an important role in urban water treatment, and there are many complicated phenomena which need to be understood for efficient design and control of it. Especially, the study on the improvement of settling efficiency is required. In this study, commercial CFD (Computational Fluid Dynamics) program, FLUENT, and particle tracking method were used to simulate the flow in sedimentation basin, and to predict the settling efficiency. Computational domain of real scale was made, and detail factors such as porous wall, and outlet trough were considered instead of being simplified. The simulation results were compared with the experimental data to calibrate the parameters of particle tracking method. Sensitivity analysis showed that the particle diameter had more significant effects on settling efficiency than the particle density. The computation results gave the best agreements with the experimental data, when the value of particle diameter was 26.5 ${\mu}{\textrm}{m}$.
Prediction of exact soil loss yield has as important engineering meaning as prediction of exact flow measurement in a stream. The quantity of soil loss in a stream should be considered in planning and management of water resources and water quality such as design and maintenace of hydraulic structures : dams, weirs and seawalls, channel improvement, channel stabilization, flood control, design and operation of reservoirs and design of harbors. In this study, the soil loss of Sap-gyo reservoir watershed is simulated and estimated by RUSLE model which is generally used in the estimation of soil loss. The parameters of RUSLE model are selected and estimated using slope map, landuse map and soil map by GIS. These parameters are applied to RUSLE's estimating program. And soil loss under probability rainfall in different frequencies are estimated by recent 30 years of rainfall data of Sap-gyo reservoir watershed.

Objective: We aimed to perform in-vitro evaluation to compare 1) shear bond strength (SBS), adhesive remnant index (ARI), and color change between self-etched and acid-etched primers; 2) the SBS, ARI and color change between direct and indirect bonding; and 3) the enamel roughness (ER) between 12-blade bur and aluminum oxide polisher debonding methods. Methods: Seventy bovine incisors were distributed in seven groups: control (no bonding), direct (DTBX), and 5 indirect bonding (ITBX, IZ350, ISONDHI, ISEP, and ITBXp). Transbond XT Primer was used in the DTBX, ITBX, and ITBXp groups, flow resin Z350 in the IZ350 group, Sondhi in the ISONDHI group, and SEP primer in the ISEP group. SBS, ARI, and ER were evaluated. The adhesive remnant was removed using a low-speed tungsten bur in all groups except the ITBXp, in which an aluminum oxide polisher was used. After coffee staining, color evaluations were performed using a spectrophotometer immediately after staining and prior to bonding. Results: ISONDHI and ISEP showed significantly lower SBS (p < 0.01). DTBX had a greater number of teeth with all the adhesive on the enamel (70%), compared with the indirect bonding groups (0-30%). The ER in the ITBX and ITBXp groups was found to be greater because of both clean-up techniques used. Conclusions: Direct and indirect bonding have similar results and all the primers used show satisfactory adhesion strength. Use of burs and polishers increases the ER, but polishers ensure greater integrity of the initial roughness. Resin tags do not change the color of the teeth.
Cell behavior of the transformed cells is known to affect by interaction with extracellular matrix (ECM) proteins and integrin. To investigate the alterations of both integrin expression and cell-matrix interaction during neoplastic conversion of human oral kerationcytes, we studied expression levels of integrin subunits by flow cytometry and cellular responses to the ECM proteins in normal human oral keratinocytes (NHOKs), HPV-immortalized HOK-16B line, and three oral cancer cell lines established from HOK-16B line, CTHOK-16B-BaP, CTHOK-16B-DMBA, and CTHOK-16B-Dexa lines. The expression levels of ${\alpha}\;and\;{\beta}$ integrin subunits were shown decreased tendency in human oral keratinocytes undergoing immortalization and tumorigenic transformation except CTHOK-16B-DMBA line tested. Although ${\alpha}v{\beta}6$ integrin is known to be highly expressed in squamous cell carcinomas, and the altered integrin expression is suspected to be associated with cellular carcinogenesis, ${\alpha}v$ integrin subunit and ${\alpha}v{\beta}6$ integrin did not express in oral cancer cell lines tested. Cell behavior to the ECM proteins in HOK-16B line was generally similar to that of exponentially proliferating NHOKs. The adhesion activity profiles of type I collagen were very similar to that of its laminin counterparts, but fibronectin showed minimal adhesion activity under our conditions compared to the BSA control. The ability of the CTHOK-16B-BaP line to spread upon type I collagen and laminin markedly decreased, but migration was notably increased on type I collagen. In contrast, CTHOK-16B-DMBA and CTHOK-16B-Dexa lines spread less but migrated more upon type I collagen than immortalized HOK-16B line. These data indicate that downregulation of integrin subunits causes the changes of cellular responses to the ECM proteins during neoplastic conversion of human oral keratinocytes, and that cellular responses to the ECM proteins in oral cancer cell lines established by exposing different carcinogens are variable according to chemical carcinogens treatment.

In this talk, a reduced-order modeling methodology based on centroidal Voronoi tessellations (CVT's)is introduced. CVT's are special Voronoi tessellations for which the generators of the Voronoi diagram are also the centers of mass (means) of the corresponding Voronoi cells. The discrete data sets, CVT's are closely related to the h-means clustering techniques. Even with the use of good mesh generators, discretization schemes, and solution algorithms, the computational simulation of complex, turbulent, or chaotic systems still remains a formidable endeavor. For example, typical finite element codes may require many thousands of degrees of freedom for the accurate simulation of fluid flows. The situation is even worse for optimization problems for which multiple solutions of the complex state system are usually required or in feedback control problems for which real-time solutions of the complex state system are needed. There hava been many studies devoted to the development, testing, and use of reduced-order models for complex systems such as unsteady fluid flows. The types of reduced-ordered models that we study are those attempt to determine accurate approximate solutions of a complex system using very few degrees of freedom. To do so, such models have to use basis functions that are in some way intimately connected to the problem being approximated. Once a very low-dimensional reduced basis has been determined, one can employ it to solve the complex system by applying, e.g., a Galerkin method. In general, reduced bases are globally supported so that the discrete systems are dense; however, if the reduced basis is of very low dimension, one does not care about the lack of sparsity in the discrete system. A discussion of reduced-ordering modeling for complex systems such as fluid flows is given to provide a context for the application of reduced-order bases. Then, detailed descriptions of CVT-based reduced-order bases and how they can be constructed of complex systems are given. Subsequently, some concrete incompressible flow examples are used to illustrate the construction and use of CVT-based reduced-order bases. The CVT-based reduced-order modeling methodology is shown to be effective for these examples and is also shown to be inexpensive to apply compared to other reduced-order methods.
Dignosis of migraine is only based on the medical history, and objective methods to aid the clinical diagnosisare absent. Although transcranial Doppler ultrasonography (TCD) abnormalities in headache-free migraineurs have been reported previously, diagnostic criteria for migraine is still lacking and this may limit the practical application of TCD for migraine. We prospectively studied several abnormal TCD indices in interictal migraineurs and their sensitivity and specificity to define the optimal diagnostic criteria. Young (20 yrs$age=29.0{\pm}6.1yrs$) were compared to 69 controls (M:F=25:44, Mean $age=31.2{\pm}5.5yrs$). Elevated MFV (> 2SD)was observed in 63% of migraineurs while n 12% of control (p<0.01). High AI (>25%) or high HI (>3.0) was present in 17% of migraineurs, while 3% and none in controls (p<0.01). Sensitivity of elevated MFV, high AI, and high HI was 63%, 17%, 17% and specificity was 88%, 97%, 100%, respectively. If all these indices were combined, sensitivity and specificity reached 69% and 86%. These preliminary results suggest pathophysiological implication of vasospasm in interictal migraineurs, and TCD may be practically applicable for migraine. Optimal diagnostic criteria and therapeutic options for patients with abnormal TCD findings remain to bo determined.

This study is intended to clarify the effects of heated effluents on intertidal benthic marine algal community in Korea. The species composition and biomass of marine algae at the discharge canal of Gori nuclear power plant on the southeastern coast of Korea were investigated seasonally from February 2001 to October 2006. As a result, 54 species (7 blue-green, 12 green, 9 brown and 26 red algae) of marine algae were found at the discharge canal during the past six years. In general, the number of species observed was abundant during winter to spring and less in autumn. Enteromorpha compressa, E. intestinalis, E. prolifera and Caulacanthus ustulatus were common species found more than 80% frequency during the study period. Seasonal fluctuations of mean biomass were 1-440 g dry wt m–2 and dominant species in biomass were Enteromorpha spp. (contribution to a total biomass proportion 28%), Sargassum horneri (14%) and Amphiroa beauvoisii (14%). It is evident from the floristic composition and biomass data that unique micro-environment of the discharge canal support different communities from those on the intake or control area. Results from the large numbers of surveys before and during plant operation showed that, in the regions influenced by thermal effluents such as the discharge canal of power plants, the process of ecological succession has been proceeded. It is assumed that the uni-directional water flow and the time of overhaul largely affect the development and succession of benthic marine algal communities of the discharge canal.
