Changes in Infarct Size after Reperfusion with Time in a Reversible Cerebral Ischemic Model in Rats (백서의 가역성 뇌허혈 모형에서 재관류 시간에 따른 뇌경색 크기의 변화)

  • Objective : The purpose of the present study was to determine the appropriate time of clinical intervention by observing and analyzing the changes in the size of infarct, penumbra and cerebral edema and the extend of neurological deficit due to reperfusion damage according to time in a reversible cerebral ischemic model of reperfusing blood flow after inducing ischemia by maintaining middle cerebral artery occlusion for 2 hours(h) in rats. Methods : The rats were divided according to reperfusion time into control group(0 h reperfusion time) and experimental groups(0.5, 1, 2, 3, 4, 5, 6, 12, and 24 h of reperfusion time). Results : Changes in the size of infarction due to reperfusion damage were 0.93, 1.48 and 1.16% at 0.5, 1 and 2 h after reperfusion, respectively, and although a statistical significance was not present compared to 1.35% of the control group, damages increased drastically up to 6 h(6.64%), and the size increased were 6.65 and 6.78% at 12 and 24 h, respectively. Also there was no significant difference after 6 h up to 24 h in the size of infarction. In the areas where infarction occurred, reperfusion damage increased significantly with time in cortex than in subcortex. Accordingly, the size of penumbra area also showed a statistically significant decrease from 2 h up to 6 h after reperfusion, and 6 h after reperfusion, the area almost disappeared, becoming permanent infarction. Thus, reperfusion damage showed a significant increase from 2 h up to 6 h after reperfusion, and became steady thereafter. As for the mean ratio of the extend of cerebral edema, the control group and reperfusion 0.5 h group were 1.073 and 1.081, respectively ; up to 2 h thereafter, the ratio decreased to 1.01 but increased again with time ; and in reperfusion 12 h and reperfusion 24 h, the ratios were 1.070 and 1.075, respectively, showing similar size with that of control group. As for neurological deficit scores, the score of the control group was 2.67, that of reperfusion 2 h was 2, those of reperfusion 3 h and 6 h groups were 3.2 and 3.8, respectively, and those of reperfusion 12 h and 24 h groups were 4.2 and 4.6, respectively. Thus, as for the test results, the neurological deficit increased with time 2 h after reperfusion, and in reperfusion 12 and 24 h groups, almost all the symptoms appeared. Conclusion : As shown in these results, although the changes in the size of infarction due to reperfusion damage did not increase up to 2 h after reperfusion in the experimental groups compared to the control group, damage increased significantly thereafter up to 6 h, and the size remained about the same from 6 h to 24 h after reperfusion, becoming permanent infarction ; thus, the appropriate time of intervention according to the present study is at least 6 h before after maintaining reperfusion, including the time of cerebral artery occlusion.

The Experimental Study for Myocardial Preservation Effect of Ischemic Preconditioning (허혈성 전조건화 유발이 심근보호에 미치는 영향에 관한 실험적 연구)

  • Decrease in cardiac function after open heart surgery is due to an ischemia induced myocardial damage during surgery, and ischemic preconditioning, a condition in which the myocardial damage does not accumulate after repeated episodes of ischemia but protects itself from damage after prolonged ischemia due to myocytes tolerating the ischemia, is known to diminish myocardial damage, which also helps the recovery of myocardium after reperfusion, and decreases incidences of arrythmia. Our study is performed to display the ischemic preconditioning and show the myocardial protective effect by applying cardioplegic solution to the heart removed from rat. Material and Method: Sprague-Dawley male rats were used, They were fixed on a modified isolated working heart model after cannulation. The reperfusion process was according to non-working and working heart methods and the working method was executed for 20 minutes in which the heart rate, aortic pressure, aortic flow and coronary flow were measured and recorded. The control group is the group which the extracted heart was fixed on the isolated working heart model, recovered by reperfusion 60 minutes after infusion and preserved in the cardioplegic solution 20 minutes after the working heart perfusion and aortic cross clamp, The thesis groups were divided into group I, which ischemic hearts that were hypoxia induced were perfused by cardioplegic solution and preserved for 60 minutes; group II, the cardioplegic solution was infused 45 seconds (II-1), 1 minutes (II-2), 3 minutes (II-3), after the ischemia induction, 20 minutes after working heart perfusion and aortic cross clamp; and group III, hearts were executed on working heart perfusion for 20 minutes and aortic cross clamp was performed for 45 seconds (III-1), 1minute (III-2), 3 minutes (III-3), reperfused for 2 minutes to recover the heart, and then aortic cross clamping was repeated for reperfusion, all the groups were compared based on hemodynamic performance after reperfusion of the heart after preservation for 60 minutes. Result: The recovery time until spontaneous heart beat was longer in groups I, II-3, III-2 and III-3 to control group (p<0.01). Group III-1 (p<0.05) had better results in terms of recovery in number of heart rates compared to control group, and recovered better compared to II-1 (p<0.05). The recovery of aortic blood pressure favored group III-1 (p<0.05) and had better outcomes compared with II-1 (p<0.01). Group III-1 also showed best results in terms of cardiac output (p<0.05) and group III-2 was better compared to II-2 (p<0.05). Group I (p<0.01) and II-3 (p<0.05) showed more cardiac edema than control group. Conclusion: When the effects of other organs are dismissed, protecting the heart by infusion of cardioplegic solution after enforcing ischemia for a short period of time before the onset of abnormal heart beats for preconditioning has a better recovery effect in the cardioplegic group with preconditioning compared to the cardioplegic solution itself. we believe that further study is needed to find a more effective method of preconditioning.

Lymphocyte Subpopulations and Proliferation of T cells, Phagocytic Activity of Leukocytes on Alcoholics (알코올중독자의 백혈구탐식능, 림프구아형 및 증식능)

  • Alcoholics increased susceptibility to microbial infection that is associated with decreased immunity. but there has been little experimental evidence to support alcoholics-induced increase of microbial infection directly in non-specific immunity. Therefore, we were used the method of phagocytic-plaque including all the stimulating factors for the phagocytosis, subtypes of lymphocytes and T-lymphocyte proliferation. The experimental groups were divided into 3 groups: (1) alcoholics who were hospitalized less than 1 week (newly hospitalized alcoholics), (2) alcoholics who were hospitalized more than 2 weeks (old hospitalized alcoholics), (3) healthy blood donors. We have studied 98 alcoholics and 35 healthy blood donors and control groups. A physician has checked the biological markers and diagnosed the body-condition alcoholics. The immunity and non-specific immunity on the alcoholics were analyzed by using the simultest kit and flow cytometry. Proliferation of the lymphocytes was analyzed by the phytohemmagglutinine mitogen. Phagocytosis and migration properties of leukocytes were identified on the layer formed by Staphylococcus aureus Cowan I strain. Biological markers of alcoholics and control groups, by such as blood glucose, ${\gamma}$-glutamyl transpeptidase and mean corpuscular volumes of red blood cells, were determined by biochemical and hematological methods. Compared with control groups, cluster of differentiation (CD)3+, CD8+ and CD19+ in alcoholic were more decreased except CD4+/CD8+ ratio. Proliferation of the T-lymphocytes, phagocytosis and migration properties of the leukocytes in alcoholics were decreased compared with those of control groups. According to the results observed in our experiment, they can be summerized as follows: 1, Cellular, humoral and non-specific immunities, are markedly decreased in alcoholics than those in control groups. 2. It is inferred that Phagocytic plaque formation is a very useful method to evaluate phagocytosis and migration properties of the alcoholic leukocytes 3. It is thought that the subtypes of lymphocytes, especially CD4+/CD8+ ratio, are essential methods to analyzed the alcoholic immunity. 4. Specific and non-specific immunity on the old hospitalized alcoholics was slightly increased, which depends upon the alcoholic medication.

Effect of Low-dose Aspirin on Implantation and Pregnancy Rates in Patients Undergoing Frozen-thawed Embryo Transfer (동결보존 배아 이식에서 저 용량 아스피린 투여가 임신율과 착상율에 미치는 영향에 관한 연구)

  • Objective: Low-dose aspirin have been proposed to improving endometrial receptivity and pregnancy rate in COH-IVF by increasing endometrial perfusion. However, the effect of low-dose aspirin in COH-IVF could be negligible because there have been large quantity of other important factors responsible for changing endometrial perfusion accompanied by COH procedure. In contrast, in frozen-thawed embryo transfer cycles which were not accompanied by COH procedure, the effects of low-dose aspirin in endometrial blood flow seems to be more certain than in COH-IVF cycles. In this study, we analyzed the effect of low-dose aspirin treatment on implantation and pregnancy rates in patients undergoing frozen-thawed embryo transfer Methods: From January 2003 to December 2003, total 264 cycles from 264 patients who attended infertility clinic at Samsung Cheil Hospital were enrolled in this study. All cases included in this study, embryos were frozen and thawed at the pronuclear stage and three days after incubation, at least 2 or more good quality embryos were transferred into uterus. In study group, low dose aspirin (100 mg/day) was administrated from the first or second date of menstrual day to 9 days after embryo transfer. On the other hand, control group did not take any medicine except estradiol valerate for endometrial priming. Several variables including implantation and pregnancy rates were compared in both groups. After then, each groups were stratified by endometrial thickness checked at embryo transfer (ET) day such as (28 mm versus <8 mm) and same variables above described were compared between study and control groups. Results: The mean age, infertility duration, endometrial thickness at embryo transfer day and mean number of transferred embryo were not significantly different in both groups. Also, implantation rates (study group: 15.8%, control group: 20.5%) and pregnancy rate (study group: 45.1%, control group: 43.5%) were not significantly different between two groups. (p>0.05) After we analyzed same variables stratified by endometrial thickness checked at embryo transfer day, we could not found any significant difference between study and control groups. Conclusions: Low-dose aspirin treatment seems to have no advantage of improving implantation and pregnancy rates in patients undergoing frozen-thawed embryo transfer.

Guaranteeing delay bounds based on the Bandwidth Allocation Scheme (패킷 지연 한계 보장을 위한 공평 큐잉 기반 대역할당 알고리즘)

  • We propose a scheduling algorithm, Bandwidth Allocation Scheme (BAS), that guarantees bounded delay in a switching node. It is based on the notion of the GPS (Generalized Processor Sharing) mechanism, which has clarified the concept of fair queueing with a fluid-flow hypothesis of traffic modeling. The main objective of this paper is to determine the session-level weights that define the GPS sewer. The way of introducing and derivation of the so-called system equation' implies the approach we take. With multiple classes of traffic, we define a set of service curves:one for each class. Constrained to the required profiles of individual service curves for delay satisfaction, the sets of weights are determined as a function of both the delay requirements and the traffic parameters. The schedulability test conditions, which are necessary to implement the call admission control, are also derived to ensure the proposed bandwidth allocation scheme' be able to support delay guarantees for all accepted classes of traffic. It is noticeable that the values of weights are tunable rather than fixed in accordance with the varying system status. This feature of adaptability is beneficial towards the enhanced efficiency of bandwidth sharing.

Development of a Rapeseed Reaping Equipment Attachable to a Conventional Combine (I) - Design and Construction of a Prototype - (보통형 콤바인 부착용 유채 예취장치 개발(I) - 시작기 설계 및 제작 -)

  • Bio-diesel applications seem to be extended due to bio-diesel policies and changes of agricultural environment. This study was conducted to develop a rapeseed reaping equipment attachable to the conventional combine. This paper was intended to report concept design, process and manufacturing of the prototype rapeseed reaping equipment. For concept design, physical properties of "SUNMANG", which is a typical rapeseed as bio-diesel materials, were considered. The designed prototype rapeseed reaping equipment consisted of wide-width plates, finger type knifes, side cutter knifes and drive equipments. The wide-width plate is 2.1 m wide, 0.7 m long, and 0.002 m thick. The finger type cutter knifes have 14.5 fingers, 30 knifes, and the specification was 7.6 cm of pitch, 8.3 cm of length and $21^{\circ}$ of cutting angle. The side cutter knifes consisted of a hydraulic pump, a hydraulic motor, a flow control and a relief valve, a hydraulic hose, a driving equipment and a reciprocating cutter knife. The 18 reciprocating cutter knifes were 137 cm long and knife pitch, knife length and cutting angle were 7.7 cm, 10.5 cm, and $18^{\circ}$. Prototype weight of the rapeseed reaping equipment was heavier by 272 kg when compared with the manual reaping equipments. Load distributions of left and right side showed 50% and 49%, and those of front and rear side showed 64% and 36%. Static turn-over angles in left and right of the prototype were $38.1^{\circ}$ and $38.7^{\circ}$, respectively. The designed prototype rapeseed reaping equipment was properly mounted at the front of a conventional combine.

Effects of Phamacopuncture Therapy Using Cervi Pantotrichum Cornu at BL23.BL52 on the Cerebral Hemodynamics in Rats (신유.지실 녹용약침이 뇌혈류역학에 미치는 영향)

  • The present study was designed to investigate the effects of Phamacopuncture therapy using Cervi Pantotrichum Cornu (PC) at BL23 BL52 on the regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, then the related mechanisms were also investigated. In addition, the present author also investigated the effects of phamacopuncture therapy at BL23.BL52 on the rCBF in cerebral ischemic rats. The results in normal rats were as follows; PC (3 mg/kg and 5 mg/kg) at BL23 BL52 significantly increased rCBF but decreased MABP. This result suggests that PC at BL23 BL52 significantly increased rCBF by dilating pial arterial diameter. Increase of PC (5 mg/kg)-induced rCBF was significantly inhibited by pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase and methylene blue ($10{\mu}g/kg$, i.p.), an inhibitor of guanylate cyclase. Decrease of PC (5 mg/kg)-induced MABP was significantly increased by pretreatment with methylene blue but was decreased by pretreatment with indomethacin. These results suggested that the action of PC (5 mg/kg) was mediated by guanylate cyclase. The results in cerebral ischemic rats were as follows ; The rCBF was significantly and stably increased by PC (5 mg/kg) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in Control group. In conclusion, these results suggest that phamacopuncture therapy using Carthami flos at BL23 BL52 can increase rCBF in normal state, and improve stability of rCBF in ischemic state. In addition, the present author also suggest that related mechanisms are involved in guanylate cyclase pathway.

Evaluation of pore water pressure on the lining during tunnel operation (운영 중 터널에 작용하는 간극수압 평가기법)

  • Control of ground water is one of the most important factors for long-term operation of tunnel because most of tunnel is located in the ground. In case of leakage tunnel, there is no pore water pressure on the lining when the drainage system is properly working. After long-term operation, however, the pore water pressure can be developed on the lining due to the deterioration of the drainage system. The increased pore water pressure on the lining is termed here as 'residual pore water pressure'. Residual pore water pressure can be measured by piezometer, but it is generally not allowed because of damages of drainage system. Therefore, an indirect and nondestructive method is required for evaluating the residual pore water pressure. Moreover, understanding of pore water pressure is needed during healthy operation of the lining. In this study, a new method for evaluation of pore water pressure on the lining during operation is proposed using theoretical and numerical analysis. It is shown that the method is particularly useful for stability investigation of pore water pressure on the lining during operation using theoretical analysis with normalized pore water pressure curve.

A STUDY ON THE FLOW CHARACTERISTICS OF AIR-KNIFE USING A CONSTANT EXPANSION RATE NOZZLE (팽창률이 일정한 노즐을 사용한 AIR-KNIFE 유동에 관한 연구)

  • In the process of continuous hot-dip galvanizing, it is well known that the gas wiping through an air knife system is most effective because of its uniformity in coating thickness, possibility of thin coating, workability in high speed, and simplicity of control. However, gas wiping used in the galvanizing process brings about a problem of splashing at the strip edge above a certain high speed of process. It is also known that the problem of edge splashing is more harmful than that at the mid strip surface. For a given liquid(of a certain viscosity and surface tension), the onset of splashing mainly depends upon the strip velocity, the gas-jet pressure, and the nozzle's stand-off distance. In these connections in the present study, we proposed three kinds of air knife system having nozzles of constant expansion rate, and compared the jet structures issuing from newly proposed nozzle systems with the result by a conventional one. In numerical analysis, the governing equations are consisted of two-dimensional time dependent Navier-Stokes equations, and the standard k-${\varepsilon}$ turbulence model is employed to solve turbulence stress and so on. As the result, it is found that we had better use the constant expansion-rate nozzle which can be interpreted from the point view of the energy saving for the same coating thickness. Also, we better reduce the size of separation bubble and enhance the cutting ability at the strip surface, by using an air-knife having constant expansion-rate nozzle.

PERFORMANCE OF TWO-PHASE UASB REACTOR IN ANAEROBIC TREATMENT OF WASTEWATER WITH SULFATE

  • Two phase UASB reactors for treating wastewater with sulfate were operated to assess the performance and competition of organics between sulfate reducing bacteria(SRB) and methane producing bacteria(MPB), and the change of characteristics of microorganisms. The reactors were fed in parallel with a synthetic wastewater of 4,000-5,000 mgCOD/L and sulfate concentration of $800-1,000\;mgSO_4/L$. In the MPR(methane producing reactor) and CR(control reactor), COD removal efficiencies were 90% and 60%, respectively, at the OLR(organic loading rate) of 6 gCOD/L, while the amount of biogas and methane content were 6.5 L/day and 80%, and 3 L/day and 50%, respectively. However, the portion of electron flow used by SRB at the OLR of 6 gCOD/L day in MPR and CR was 3% and 26%, respectively. This indicated that the increase of OLR of wastewater containing high sulfate like CR resulted in activity decrease and cell decay of MPB, while SRB was adapted immediately to new environment. The MPB activities in MPR and CR were 2 and $0.38\;kgCH_4-COD$/gVSS day at the OLR of 6 gCOD/L. This indicated hat SRB dominated gradually over MPB during long-term operation with wastewater containing sulfate as a consequence of outcompeting of SRB over MPB. In addition, the solution within AFR was maintained around pH 5.0, the MPB such as Methanothrix spp. which was very important to formation of granules was detached from the surface of granules due to the decrease of activity by limitation of substrate transportation into MPB. Therefore, a significant amount of sludge was washed out from the reactor.
