Effects of water extract of Paeoniae Radix Alba on a thioacetamide induced acute liver injury rat model (Thioacetamide 유발 급성 간손상 동물모델에 백작약 열수 추출물이 미치는 효능)

  • Purpose: Paeonia Radix Alba is a traditional herbal medicine used to treat the liver and the spleen. Many studies have reported that Paeonia Radix Alba extract (PR) affects liver injury, but there has been no study on liver injuries induced by thioacetamide (TAA). Therefore, we aimed at evaluating the effect of PR on a TAA-induced acute liver injury (ALI) model. Methods: The antioxidant activity of PR was assayed by the content of total polyphenol, total flavonoid, 1,1-diphenyl-2'-picrylhydrazyl (DPPH), and 2,2'-azino-bis (3-ethylbenzo-thiazoline-6-sulfonicacid) (ABTS) radical scavenging activities in vitro test. ALI was induced via-intraperitoneal injection of TAA (200 mg/kg body weight) for three consecutive days. Also, silymarin (100 mg/kg body weight) and PR (100 or 200 mg/kg body weight) were administered at 1 hours 30 minutes prior to TAA treatment. The levels of ammonia, glutamic oxaloacetic transaminase (GOT), and glutamic pyruvic transaminase (GPT) were analyzed using an assay kit. The expressions of antioxidant proteins including Nrf2, Keap1, HO-1, SOD, catalase, and GPx-1/2 and oxidative stress-related proteins including NOX2, p47phox, and p22phox were evaluated by the western blot analysis. Results: PR showed excellent antioxidant activity in vitro. TAA administration increased the levels of ammonia, GOT, and GPT in the ALI control group compared to the normal group, whereas it was significantly reduced by PR pretreatment. Moreover, NADPH oxidase protein expressions were upregulated after TAA treatment, while the elevated expressions were inhibited by PR pretreatment. The expressions of antioxidant protein were downregulated in the ALI control group, whereas Nrf2 activation in the PR group was accompanied by increased levels of antioxidant enzymes. Conclusion: PR administration increased the antioxidant enzymes via activation of the Keap1/Nrf2 pathway and inhibited the protein levels of NADPH oxidase factors. Taken together, these results showed that PR treatment may be considered to ameliorate acute liver injury induced by TAA.

Antioxidant Activity of Korean Gomchwi (Ligularia fischen) Extracts (국내산 곰취(Ligularia fischen) 추출물의 항산화 활성)

  • In this study, Gomchwi (Ligularia fischen) derived from Taebaek-si, Gangwon-do was extracted with 70% ethanol (LFE) and antioxidant activity was measured. The following experimental techniques were used to evaluate the antioxidant efficacy of LFE. Total phenolic contents, ABTS/DPPH radical scavenging analysis, cell viability assay, NO assay, and quantitative real-time PCR technique. The content of polyphenol and flavonoid was each 113.97±0.37 mg GAE/g or 29.22±2.06 mg QE/g in LFE. DPPH radical scavenging activity was measured to be 25 ㎍/㎖ 11.26±0.95%, 50 ㎍/㎖ 17.12±0.63%, 100 ㎍/㎖ 29.54±0.36%, 250 ㎍/㎖ 68.31±0.28%, 500 ㎍/㎖ 75.12±0.05%, and 1000 ㎍/㎖ 75.75±1.57%. In addition, ABTS radical scavenging activity was identified as LFE 25 ㎍/㎖ 13.75±0.21%, 50 ㎍/㎖ 26.71±0.20%, 100 ㎍/㎖ 56.92±0.22%, 250 ㎍/㎖ 91.30±0.12%, 500 ㎍/㎖ 93.40±0.02, and 1000 ㎍/㎖ 93.19±0.04%. There was no significant cytotoxicity of LFE. NO production was significantly decreased to LFE 50 ㎍/㎖ 79.40±2.64%, 100 ㎍/㎖ 55.01±5.36%, and 200 ㎍/㎖ 30.93±3.11%. Also, the NOS2 gene expression was significantly reduced to LFE 50 ㎍/㎖ 0.94±0.11, 100 ㎍/㎖ 0.59±0.05, and 200 ㎍/㎖ 0.32±0.04. This result objectively confirmed the antioxidant effect of Gomchwi. We will continue to conduct in-depth research. Therefore, it is believed that the possibility of using Gomchwi as a cosmetic and functional food material can be established.

Physicochemical Properties and Antioxidant Activities of gochujang with lotus leaf powder

  • This study examined that have excellent antioxidant effect and expand the base of consumption of lotus leaves. We added 1%, 3%, and 5% of lotus leaf powder to traditional gochujang. pH was the highest in the gochujang with lotus leaf powder 1%(p<0.001). The moisture was significantly lower as the addition of lotus leaf powder increased(p<0.01). The viscosity was the lowest with lotus leaf powder 5% was found to be 15.61 dPa·s(p<0.001). In chromaticity, the L value was the highest in the control, the viscosity of gochujang became darker and lessened(p<0.001). The a value and b value showed the highest in the control(p<0.001). The salinity was the lowest in the gochujang with lotus leaf powder 3%(p<0.001). The sugar content decreased as the more amount of lotus leaf. Total phenol and total flavonoid contents was the highest in the gochujang with lotus leaf powder 5%. The DPPH radical scavenging ability was higher as the more amount of lotus leaf. Reducing power and α-glucosidase inhibitory activity was the highest in the gochujang with lotus leaf powder 3%. The gochujang with lotus leaf powder can be expected to have higher antioxidant activities and to be a health functional food.

Antioxidant Activities of Protaetia brevitarsis Larvae Fermented by Lactobacillus acidophilus (유산균 발효가 흰점박이꽃무지(Protaetia brevitarsis) 유충의 항산화활성에 미치는 영향)

  • This study was carried out to evaluate the effect of fermentation by Lactobacillus acidophilus on the antioxidant activity of Protaetia brevitarsis larvae fed with mushroom substrates (king oyster mushroom). The total polyphenol content of the P. brevitarsis larvae extracts (PLEs) (93.33±0.98 mg GAEs/extract g) was higher than that of the fermented P. brevitarsis larvae extracts (FPLEs) (65.02±1.32 mg GAEs/extract g). The flavonoid contents of the PLEs and FPLEs were 18.3±1.57 QEs mg/extract g and 17.69±0.95 QEs mg/extract g, respectively. The DPPH radical scavenging activity showed no significant difference between the PLEs and FPLEs at a concentration of 2-4 mg/ml. However, at a concentration of 8 mg/ml or more, the DPPH radical scavenging activity of the FPLEs was higher than that of the PLEs. The reducing power of the FPLEs was also higher than that of the PLEs, and more than twice as high at a concentration of 1.6 mg/ml or more. The ORAC value of the FPLEs (79.22±0.72 μM TEs/extract g) was higher than that of the PLEs (74.34±0.37 μMTEs/extract g). A WST-1 assay of the RAW 264.7 cells indicated that the PLEs and FPLEs showed no cytotoxicity.

Protective Effect of Tongyuhwalhyeol-tang on Liver Injury in Thioacetamide-induced Rat (Thioacetamide 유발 간손상모델에서 통규활혈탕의 간보호효과)

  • Objectives : Liver disease is an inflammatory reaction caused by oxidative stress, viral, alcohol, and drug properties. Inflammatory reaction causes hepatitis and chronic hepatitis is persistent, it progresses to liver fibrogenesis and liver cancer. The aim of this study was to confirm the hepatoprotective effect of Tongyuhwalhyeol-tang(Tongqiaohuoxue Decoction) (TH) and Gamtongyuhawlhyeol-tang(GTH) in TAA-induced liver injury animal model. Methods : The antioxidant activities were evaluated through in vitro experiments, such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and 2, 2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) radical scavenging assays, total polyphenol and total flavonoid content measurement. To confirm the liver protective effect, induced by Thioacetamide (TAA) for 3 days injection at 200 mg/kg rats. TH and GTH were treated 3 days at 200 mg/kg/day. The changes of reactive oxygen species (ROS), peroxynitrite ($ONOO^-$), alanine aminotransferanse (ALT) and aspartate aminotransferase (AST) in serum were analyzed after experiment. Also, expression of anti-inflammation, anti-oxidant related proteins were investigated by western blot analysis. Results : TH was inhibited the antioxidant activities. In the TAA-induced rat, TH decreased ROS, $ONOO^-$, ALT, AST level in serum. Inflammation related protein expressions increased in TAA-induced rat compared to normal rat. However, TH group inhibited the down expression of these proteins. Also, anti-oxidant related protein expressions increased in TH group compared TAA-induced rat. Conclusion : Therefor, these results suggested that TH provided hepatoprotective effects on the hepatic injury leading to the reduction of inflammatory response. In addition, the effect of TH was superior to that of GTH.

Anti-oxidative, Acetylcholinesterase Inhibitory Activities and Acute Toxicity Study of Nepeta sibirica L.

  • Nepeta sibirica L. or Siberian catmint is a medicinal plant species used in Mongolian traditional medicine for curing human different disorders and veterinary practices. The previous study of the whole plant concentrated on the determination of its essential oil composition and reported that the major ones are sesquiterpenes, including nepetalactone. The aim of this study was to reveal a new biological activity of the above-ground parts of N. sibirica L. and compare the activity of different extracts correlating with the content of biologically active compounds and evaluate their toxicity. For this purpose, anti-oxidative and acetylcholinesterase inhibitory activities of the above-ground parts of N. sibirica L. aqueous and ethanol (EtOH) (40%, 70%) extracts were assayed spectrophotometrically. The aqueous extract showed positive anti-oxidative activity by both tested DPPH and FRAP assays with IC50 134.24 ± 1.42 mg/mL and FRAP value 1385.15 ± 8.12 µmol/L at 200 ㎍/mL, in contrast to 40% and 70% EtOH extracts. The 70% EtOH extract presented the highest acetylcholinesterase inhibitory activity (IC50 77.29 ± 0.38 mg/mL) followed by 40% EtOH extract (176.72 ± 0.35 mg/mL) and aqueous extract (275.41 ± 0.23 mg/mL). Total phenolics were found to be gallic acid equivalent, % 3.74 ± 0.05 (70% EtOH), 3.94 ± 0.04 (40% EtOH), and 3.79 ± 0.16 (aqueous), whereas the total flavonoids as a rutin equivalent, % as 2.01 ± 0.12, 1.44 ± 0.17 and 1.99 ± 0.02, each. The aqueous extract showed the best anti-oxidative and lowest activity against the acetylcholinesterase; however, the 70% EtOH extract showed the opposite effects than that of the aqueous. No mortality incidence was visible at various doses, indicating that the oral median lethal dose of aqueous and 70% EtOH extracts were considered greater than 5000 mg/kg. N. sibirica L. belongs to the non-toxic category of the OECD 423 classification.

A Study on the Prediction Model for Bioactive Components of Cnidium officinale Makino according to Climate Change using Machine Learning (머신러닝을 이용한 기후변화에 따른 천궁 생리 활성 성분 예측 모델 연구)

  • Climate change has emerged as a global problem, with frequent temperature increases, droughts, and floods, and it is predicted that it will have a great impact on the characteristics and productivity of crops. Cnidium officinale is used not only as traditionally used herbal medicines, but also as various industrial raw materials such as health functional foods, natural medicines, and living materials, but productivity is decreasing due to threats such as continuous crop damage and climate change. Therefore, this paper proposes a model that can predict the physiologically active ingredient index according to the climate change scenario of Cnidium officinale, a representative medicinal crop vulnerable to climate change. In this paper, data was first augmented using the CTGAN algorithm to solve the problem of data imbalance in the collection of environment information, physiological reactions, and physiological active ingredient information. Column Shape and Column Pair Trends were used to measure augmented data quality, and overall quality of 88% was achieved on average. In addition, five models RF, SVR, XGBoost, AdaBoost, and LightBGM were used to predict phenol and flavonoid content by dividing them into ground and underground using augmented data. As a result of model evaluation, the XGBoost model showed the best performance in predicting the physiological active ingredients of the sacrum, and it was confirmed to be about twice as accurate as the SVR model.

Anti-inflammatory activities of carrot(Daucus carota) leaf Fermented by Weizmannia coagulans KK7 (김치에서 분리한 Weizmannia coagulans KK7으로 발효한 당근 잎 추출물의 함염증 활성 연구)

  • In this study, the extracts of carrot (Daucus carota var. sativa) leaf fermented with Weizmannia coagulans KK7 strain were investigated for the anti-inflammatory activities and component changes. The KK7 strain was isolated from kimchi, a Korean fermented vegetable. The high-performance liquid chromatography was performed to analyze the changes in the components of the carrot leaf extracts before and after fermentation. It was confirmed that the content of luteolin, a kind of flavonoid, was significantly increased after fermentation. The anti-inflammatory activities of the carrot leaf extracts and the fermented carrot leaf extracts were evaluated by the inhibition of NO (nitric oxide) production in LPS-induced RAW 264.7 cells. The NO scavenging ability of the fermented carrot leaf extracts was higher than the other extracts. The protein expression of iNOS, an enzyme responsible for the NO production was significantly reduced in a concentration-dependent manner by treatment with the fermented carrot leaf extracts. In conclusion, we found that the anti-inflammatory effect of carrot leaf was increased by microbial fermentation, suggesting that carrot leaf generally discarded could be used as new food and cosmetic materials through fermentation.

Performance, blood and antioxidant status in dual-purpose laying hens supplemented with aqueous extract of Christ's thorn jujube (Ziziphus spina-christi L.) leaves as phytogenic agent in subtropical conditions

  • Objective: The potential of aqueous extract of Christ's thorn jujube (Ziziphus spina-christi) leaves (SLAE) to reduce the negative impacts of heat stress on production performance and physiological traits was investigated in dual-purpose layers under subtropical farming. Methods: A total of 200, 25-week-old laying hens (Inshas strain) were randomly assigned to four dietary treatments including SLAE at 0, 2.5, 5.0, and 7.5 mL/kg, respectively. The average temperature-humidity index value was 26.69 during the experimental period. The SLAE contained saponin (0.045%), total flavonoid content of 17.9 mg of quercetin equivalent/100 g and overall antioxidant capacity concentration of 17.9 mg of ascorbic acid equivalent/100 g. Results: The maximum final body weight (BW), BW gain, egg weight, number, and mass occurred at the level of SLAE7.5 inclusion. The egg quality was significantly higher in SLAE groups than in control, and overall, SLAE7.5 had the most favorable influence at 28 and 32 weeks. Liver and kidney function, as well as lipids profile, improved significantly by SLAE inclusion; the lowest concentrations of these parameters were in SLAE7.5 hens. Treatment with SLAE7.5 increased total antioxidant capacity and endogenous antioxidant enzymes compared to control, whereas no effect on superoxide dismutase was noticed. Conclusion: The addition of SLAE at 7.5 mL/kg diet improved egg laying performance and quality, metabolic profiles, and antioxidant status during hyperthermia conditions.

Study on the Anti-Aging Activity of Chrysanthemum lucidum Exosomes (울릉국화 엑소좀의 항노화 활성 효과 연구)

  • Chrysanthemum lucidum (C. lucidum), a perennial herb in the Asteraceae family, is an endemic species found only on Ulleung island in Gyeongsangbuk-do, South Korea. Previous studies have reported that the extract of C. lucidum exhibits excellent antioxidant activity due to its high polyphenol and flavonoid content. However, the anti-aging effects of C. lucidum extract, such as wrinkle improvement and cell regeneration, are not well known, and there has been no research on the activity of C. lucidum-derived extracellular vesicles (ClDEVs). Therefore, this study aimed to verify the anti-aging effects of ClDEVs through in vitro and clinical analyses. In cell experiments, ClDEVs promoted cell regeneration, increased the expression of COL1A1, a gene involved in collagen synthesis, and enhanced the expression of FLG and LOR, a biomarker related to skin barrier improvement. Additionally, ClDEVs suppressed the expression of aging-related biomarkers, such as the CDKN2A (encodes p16) and TP53 (encodes p53) genes, in cells induced to age. In a human clinical trial, after using a cosmetic product containing ClDEVs for 4 weeks, significant improvement in wrinkles around the eyes and nasolabial folds was observed. In conclusion, ClDEVs have demonstrated high potential as a bio-cosmetic ingredient for wrinkle improvement and anti-aging.
