The purpose of this study was to improve the quality of Korean traditional sauce products by producing short-term green gochujang with cheongyang pepper powder. To determine its antioxidant compounds and activities, we examined vitamin C, capsaicinoid, total phenolic, and total flavonoid contents, as well as electron-donating, SOD-like, ferrous ion-chelating, and nitrite-scavenging activities. Vitamin C content of short-term green gochujang was higher in CON-M than in the sample, whereas capsaicinoid content increased as the amount of cheongyang pepper powder increased. Total phenolic content, total flavonoid content, electron-donating activity, and nitrite-scavenging activity were higher in the sample than in CON-M, and the contents increased as the amount of meju powder increased. Green gochujang containing the highest amount of cheongyang pepper powder showed the highest SOD-like, and ferrous-ion chelating activities as well as superior nutrient contents, compared to red gochujang.
Bioassay-directed chromatographic fractionation of an ethyl acetate extract from leaves of sweet potato (Ipomoea batatas L.) afforded six quinic acid derivatives: 3,5-epi-dicaffeoylquinic acid (1), 3,5-dicaffeoylquinic acid (2), methyl 3,5-O-dicaffeoylquinate (3), methyl 3,4-dicaffeoylquinate (4), methyl 4,5-dicaffeoylquinic acid (5),4,5-dicaffeoylquinate (6), and two phenolic compounds: caffeic acid (7) and caffeic acid methyl ester (8) together with three flavonoids: quercetin 3-O-${\beta}$-D-glucopyranoside (9), quercetin 3-O-${\beta}$-D-glucopyranoside, isoquercitrin (10) and kaempferol 3-O-${\beta}$-D-glucopyranoside (11). The structures of these compounds were elucidated by the aid of spectroscopic methods. These compounds were assessed for antioxidant activities using three different cell-free bioassay systems. All isolates except 11 showed potent DPPH and superoxide anion radicals scavenging, and lipid peroxidation inhibitory activities. 3,5-epi-DCQA (1) and methyl quinates (3-5) along with flavonoide 9 were isolated for the first time from this plant.
Alnus japonica Steud (Betulaceae) has long been used as a Korean traditional medicine for gastric disorders, hepatitis, and fatty liver. As a part of our study on the identification of secondary metabolites of naturally occurring bioactive compounds, we isolated 1,7-bis(4-hydroxyphenyl)-3,5-heptanediol(1), 5-hydroxy-1,7bis(4-hydroxyphenyl)-3-heptanone(2), 5,3'-dihydroxy-7,4'-dimethoxyflavone(3) and 3,5,7,3',4'-pentahydroxyflavone(4) from the dichloromethane and ethylacetate-soluble fractions of Alnus japonica Steud. These compounds showed significant antioxidant activity in a concentration-dependent manner. The $IC_{50}$ values of compounds 1, 2, 3 and 4 were 30.1, 37.4, 20.2 and 13.7 ${\mu}g/mL$, respectively, through the scavenging capacity of 1,1-diphenyl-2-picrylhydrazyl radical assay.
Arabidopsis mutants deficient in flavonoid and sinapate ester (tt4 and fah1-7, respectively) were evaluated in vivo and in vitro to study the possible role of flavonoid compounds in pollen tube growth. In vivo, we investigated pollen tube growth in the pistils of the mutants and wild type(Ler). The growth of pollen tubes was significantly different among the three genotypes. In the fal1-7 pistils, the tubes grew to a greater length relative to those of the wild type or tt4. To examine in vitro pollen tube growth, a solid medium was devised for pollen germination and subsequent growth. In vitro, the identical result was obtained; fahl-7 pollens developed the longest tubes and elongated most rapidly. Therefore, the growth response of pollen tubes to phenolic compounds was examined by adding quercetin or sinapate ester in various concentrations to the media. Quercetin enhanced both germination rate and tube growth in the pollens of the mutants and the wild type, especially in tt4. In contrast, sinapate ester inhibits pollen germination and pollen tube growth in three genotypes. These results suggest that flavonoids and related phenolic compounds have physiological role in the plant reproductive system.
Antimicrobial activities of plant extractives were investigated to develop a natural fungicide. Two stilbenoids and five flavonoids were isolated from wood extractives of Hovenia dulcis (Rhamnaceae) which had been selected due to its high antifungal activity among the tested species. The chemical structures of isolated compounds were determinded as : 5-hydroxy-7-methoxyflavone, 5,7-dihydroxyflavone (chrysin), 5,7-dihydroxyflavanone (pinocembrin), 3,5,7-trihydroxyflavanone (pinobanksin), 3,4',5,7-tetrahydroxyflavanone (aromadendrin), 3-hydroxy-5-methoxystilbene and 3,5-dihydroxystilbene (pinosylvin) on the basis of Mass and NMR spectroscopic data. According to the results of antifungal test, 3-hydroxy-5-methoxystilbene was evaluated as the strongest antifungal compound among the tested compounds and next were pinocembrin and pinosylvin, but those also had high hyphal growth inhibition activities against C. parasitica, T. versicolor, T. palustris and T. viride. However, pinobanksin, 5-hydroxy-7-methoxyflavone, chrysin and aromadendrin showed very low antifungal activity. In this regard, it could inferred that high antifungal activity of wood extractives of H. dulcis were derived from 3-hydroxy-5-methoxystilbene, pinocembrin and pinosylvin, respectively.
Antioxidative components and activities of the extracts from Pleurotus ostreatus extracted at different ethanol concentration were analyzed and their correlation were investigated. Ergothioneine, total phenolic compounds, and flavonoid contents of the extracts from P. ostreatus extracted with hot water (0% ethanol) were the highest ($2.98{\pm}0.05$, $9.51{\pm}0.45$, and $2.83{\pm}0.03mg/g$, respectively) and the contents were decreased according to increase of ethanol concentration for extraction. DPPH and ABTS radical scavenging activities of the extracts from P. ostreatus extracted with hot water were the highest ($80.41{\pm}0.56$ and $91.47{\pm}0.11%$, respectively). FRAP value also showed the highest reducing power by $8.86{\pm}0.33\;FeSO_4\;eq$. mM in hot water extracts. Positive correlations were found between ergothioneine contents and antioxidative active components and antioxidant activity of the extracts from P. ostreatus. Results indicate that hot water extraction was most efficient for the extracts with high antioxidative activities from P. ostreatus.

Rice (Oryza sativa L.) is one of the most consumed staple food crop which is energy source as carbohydrate and also is considered as the important antioxidant sources including various phenolic compounds. According to the increasing demand of healthy life, the concern to antioxidant also is increasing because of its health-promoting effect. Phenolic compounds are one of the plant secondary metabolites class, which shows various benefits to preventing or treating chronic diseases. In this study, we have measured the total phenol content from total 647 rice samples using the Floin-Ciocalteau method, and then were selected 30 rice genetic resources classified with high, middle, and low group on the basis of total phenol content. The average of the total phenol content of each group was high-group ($6892.9{\pm}488.5{\mu}g\;GAE/g$) > middle-group ($1428.1{\pm}76.0{\mu}g\;GAE/g$) > low-group ($97.6{\pm}11.4{\mu}g\;GAE/g$). The selected rice samples were analyzed with LC-MS/MS to find the composition and concentration of individual phenolic in rice grain. High-group and middle-group contained large amounts of protocatechuic acid and (+)-catechin whereas low-group showed limited amount. Among high-group samples, rice samples with black pericarp color (IT 174089, IT 220079, and IT 259958) had high content of peonidin-3-O-glucoside. Further, these black rice samples were special since polydatin, rarely found stilbenoid in rice grain, was detected. Overall, both the sum of phenolic acid and the sum of flavonoid were high-group > middle-group > low-group. Also, each group exhibited different phenolic compositions; high-group consisted of flavonoid more than phenolic acid, middle-group and low-group was comprised of phenolic acid rather than flavonoid, and non-pigmented rice was composed by fully phenolic acid. The total phenol content had positive relationships with the sum of phenolic compound (r = 0.64), the sum of flavonoid (r = 0.74) at the significance level of p < 0.0001. In addition, protocatechuic acid and quercetin showed positive correlation with above phenolic composition parameters; in order, r = 0.98, 0.65 for protocatechuic acid and r = 0.73, 0.78 for quercetin (p < 0.0001). In conclusion, the total phenol content assay showed the possibility of utilization as a phenolic composition indicator in rice grain. Also, this result was suggested study pigment on other material.

To materialize anti-inflammatory compounds from dandelion leaves, we studied optimal conditions of hot water extraction and concentration. We investigated the properties of extracts on various temperatures between 60 and $90^{\circ}C$. Extraction yields at $60^{\circ}C$ were the highest at $39.2{\pm}1.3%$ and flavonoid contents were increased by heating. However, anti-inflammatory activities were remarkably decreased by increasing temperature. According to the results that the highest anti-inflammatory activities of water extract were demonstrated at $60^{\circ}C$, cell wall degrading enzyme treatment was carried out to increase extraction yield of anti-inflammatory compounds. The enzyme treatment resulted in increased yields and flavonoids from dandelion leaves of 1.5-fold and anti-inflammatory activities were not decreased. Also, column chromatography was used for concentration of anti-inflammatory compounds from extract of dandelion leaves treated with enzyme. Anti-inflammatory activities of adsorbed compounds on Amberlite XAD-2 and Sepabeads SP-850 were significantly increased. However, yields of adsorbed compounds were increased over two-folds on Sepabeads SP-850 compared to Amberlite XAD-2. Thus Sepabeads SP-850 was more effective than Amberlite XAD-2 to concentrate the anti-inflammatory compounds of dandelion leaves.
Various flavonoid derivatives were previously reported to possess the inhibitory activity on cyclooxygenase/lipoxygenase. And these properties of flavonoids might contribute to their anti-inflammatory activity in vivo. In this study, several polyhydroxylated/methoxylated flavonoid derivatives such as oroxylin A. wogonin, skullcapflavone II, tectorigenin and iristectorigenin A were isolated from the medicinal plants. these compounds were evaluated fro their inhibitory effects on cyclooxygenase/lipoxygenase from the homogenate of human platelets in vitro. It was found that isoflavones including daidzein and tectorigenin possessed the inhibitory activity on cycloooxygenase, although the potency of inhibition was far less than that of indomethacin. In addition, oroxylin A, baicalein and wogonin inhibited 12-lipoxygenase activity without affecting cyclooxygenase, which suggested that 5,6,7- or 5,7,8-trisubstitutions of A-ring of flavone gave favorable results. The IC50 values of oroxylin A and NDGA against 12-lipoxygenase were found to be 100 and 1.5 uM, respectively.
Galgeuntang water extract exhibited a strong free radical scavenging activity and reducing power determination. However, a gradual increase in the free radical scavenging activity and reducing power determination was obtained with increasing concentrations. The highest radical scavenging activity was shown by the water extract from Galgeuntang (116.93 ${\mu}g$/mL) and the water extract from Cinnamonum cassia Presl. (95.01 ${\mu}g$/mL). These results of phenolic and flavonoid contents of the extracts indicated that the strong radical scavenging activity of the Cinnamonum cassia Presl. extract might be in part due to the phenolic compounds. The correlation coefficient between TPC and DPPH ($r^2$ = 0.9312), TFC and DPPH ($r^2$ = 0.9677), showed positive correlation among total phenolic/flavonoid contents and antioxidant activity. These results suggest that Galgeuntang has a potential antioxidant activity.
