Linarin enhances melanogenesis in B16F10 cells via MAPK and PI3K/AKT signaling pathways

  • In this study, we discovered for the first time that linarin, a flavonoid compound, enhances melanin biosynthesis in B16F10 cells, and subsequently elucidated the underlying mechanism of linarin-induced melanogenesis. Linarin showed no cytotoxicity at a concentration of 42 μM and significantly increased intracellular tyrosinase activity and melanin content in B16F10 cells. Mechanistic analysis showed that linarin increased the expression of tyrosinase, tyrosinase-related protein 1 (TRP-1), and microphthalmia-associated transcription factor (MITF) that are related to melanogenesis. Moreover, linarin decreased the phosphorylation of extracellular signal-regulated kinase (ERK) and protein kinase B (AKT). Finally, we evaluated the effect of the structure-activity relationship of linarin and its aglycone on melanogenesis. The results indicated that linarin enhances the expression of melanogenic proteins by activating MITF expression via the modulation of mitogen-activated protein kinase (MAPK), phosphatidylinositol 3-kinase (PI3K), and protein kinase B signaling pathways in B16F10 cells, thereby enhancing melanogenesis.

Bioactive Effects of Domestic Cherry Tomatoes (국내산 방울토마토의 생리활성 효과)

  • This study was carried out to elucidate bioacive effects of three domestic cherry tomato cultivars. Total polyphenol and flavonoid of Summerking, Qutiquti, and Minichal cultivar were 12.56±1.88, 12.50±1.92, 11.65±1.85 mg/g and 4.58±1.03, 4.19±0.40, 4.30±0.49 mg/g(dry weight) respectively. Domestic cherry tomatoes showed antioxidative activity(DPPH and ABTS radical scavenging activities). All of the cherry tomatoes had no cytotoxicity for normal liver cell, but showed strong inhibitory effect against cervical cancer cell(HeLa) growth. These results revealed that domestic cherry tomatoes can be used as a bioactive food material.

Quality Characteristics, Bioactive Compound Content, and Antioxidant Activity of Butter Cookies Supplemented with Burdock Powder (우엉 분말 첨가 버터 쿠키의 품질특성, 생리활성 물질 함량 및 항산화 활성)

  • We determined physicochemical characteristics and antioxidant activities of cookies prepared by replacing wheat flour with 0, 3, 6, and 9% freeze-dried burdock powder instead of wheat flour. The leavening and loss rates of the cookies decreased in proportion to the amount of burdock powder added to the dough. The moisture content of the cookies increased in proportion to the amount of burdock powder added and the crude protein and hardness were higher in the burdock-added group compared to the control. The lightness and yellowness of the cookies decreased, and the redness increased in proportion to the amount of burdock powder added. The total flavonoid and polyphenol compounds in the cookies increased in proportion to the amounts of burdock powder added. The antioxidant activities also tended to increase in proportion to the amount of burdock powder added. Based on the above results, adding burdock powder to cookies can increase antioxidant activity by increasing the content of antioxidant components, such as polyphenols and flavonoids. In addition, it is believed that adding burdock powder at levels of 3 to 9% of the weight of flour would result in cookies with spreadability, swelling power, and hardness similar to those of regular cookies.

Luteolin sensitizes human liver cancer cells to TRAIL-induced apoptosis via autophagy and JNK-mediated death receptor 5 upregulation

  • The tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a dynamic cytokine that initiates the apoptosis of cancer cells, but exhibits little or no toxicity in normal cells. Luteolin is a flavonoid compound frequently used in the treatment of cancer. In the current study, we demonstrate that treatment with luteolin and TRAIL exerts a synergistic effect and the mechanisms on TRAIL-resistant Huh7 cells. The results demonstrated that luteolin induced an autophagic flux in human liver cancer cells. The attenuation of the autophagic flux by applying the specific inhibitor of autophagy, chloroquine, significantly suppressed DR5 expression. Treatment with genetically modified autophagy-related 5 siRNA abrogated the luteolin-mediated sensitizing effect of TRAIL. Furthermore, pre-treatment with the c-Jun N-terminal kinase (JNK) inhibitor, SP600125, significantly attenuated the luteolin-induced upregulation of DR5 expression, thereby suggesting that JNK activation promotes DR5 expression. Our findings also revealed that Akt phosphorylation was required for TRAIL sensitization. On the whole, the findings of this study indicated that luteolin effectively enhanced TRAIL-initiated apoptosis, and that these effects were likely to be mediated by autophagy and JNK-mediated DR5 expression.

A Study on Antioxidant Activity and Antioxidant Compound Content by the Types of Tea (다류의 유형별 항산화 성분함량 및 항산화 활성 연구)

  • The purpose of this study was to investigate the contents of antioxidant compounds and antioxidant activities in teas. A total of 99 teas were tested for their antioxidant activities based on their ability to scavenge DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical. Antioxidant activity was expressed as mg of ascorbic acid equivalents per 100 g tea sample (L-ascorbic acid equivalent antioxidant capacity, AEAC) and was expressed as mg per 1 serving size (free radical scavenging activity, $FSC_{50}$). The total polyphenol contents of the extracts was quantified using the Folin-Ciocalteu method and the total flavonoid contents of the extracts was determined using a modied method of Davis. Vitamin C was analyzed by HPLC method. According to the AEAC value, mate tea, green tea, black tea, oriental raisin tea, chamomile tea and burdock tea showed relatively high antioxidant activities. Polyphenolic compounds were the major naturally occurring antioxidant compounds found in teas and the high concentrations of total polyphenol compounds were observed in black tea, green tea and mate tea. The high concentrations of total flavonoid compounds were observed in mate tea, black tea and yam tea and the amount of vitamin C contents were found to be high in citron tea and green tea. As a result, steeping tea was much higher in antioxidant activity than either powdered tea or liquefied tea. According to the study, a high correlation was demonstrated between the total polyphenol contents and antioxidant activities in teas (r = 0.846) and correlations between the total flavonoid contents and antioxidant activities was statistically significant in teas (r = 0.625). It was found that also the proportional relationship established among the total polyphenol content and antioxidant activities. That is, antioxidant activity of teas has been confirmed to have been caused by the total polyphenol.

The antioxidative and cytoprotective effect of Lonicerae japonicae Flos water extracts on the ultraviolet(UV)B-induced human HaCaT keratinocytes (금은화 물추출물의 항산화 효과와 Ultraviolet(UV)B로 유도된 사람 각질형성세포 손상에 대한 보호효과)

  • Objectives : Lonicerae japonicae Flos(LJF) has been reported to exhibit anti-oxidant, anti-inflammatory, anti-viral, anti-rheumatoid properties. However, it is still largely unknown whether LJF inhibits the ultraviolet(UV)B-induced oxidative damage in human HaCaT keratinocytes. Therefore in this paper, we investigated the anti-oxidative capacity and protective effect of LJF against UVB-induced oxidative demage in human HaCaT keratinocytes. Methods : To evaluate the anti-oxidative activity of LJF extracts, we measured total phenolic contents, total flavonoid contents, antioxidant capacity, and superoxide scavenging activity. To give an oxidative stress to HaCaT cells, UVB was irradiated with $200mJ/cm^2$ to HaCaT cells. To detect the protective effect of LJF against UVB, we measured cell viability, DNA fragmentation and reactive oxygen species (ROS) production. In addition, we performed high-performance liquid chromatography (HPLC) analysis to find a major component of LJF. Results : LJF contained phenolic and flavonoid contents, and showed the anti-oxidant and superoxide scavenging activity. The UVB-induced oxidative conditions led to the cell death, DNA fragmentation and reactive oxygen species (ROS) production. However, pretreatment with LJF reduced oxidative conditions, including inhibition of cell death, DNA fragmentation and ROS production. In addition, we found out chlorogenic acid as major component of LJF. Conclusions : These results could suggest that LJF contained anti-oxidative contents and exhibited protective effects against UVB on human HaCaT keratinocytes. And the effective compound of LJF which could show protective activities against UVB is chlorogenic acid. Thus, LJF and chlorogenic acid would be useful for the development of drug or cosmetics treating skin troubles.

Comparison of physiological activities and of useful compounds between new and waste bulbs of different lily (Lilium davidii) varieties

  • Lily (Lilium davidii) is a high-yielding flowering plant. Besides roses and chrysanthemums, lily bulbs have long been used as food and in oriental medicine. However, the usage and value of cut lily bulbs has not been recognized. A bulb whose yield has been decreased is called a waste bulb, and a large amount of such bulbs is discarded every year. In this study, the functionality of waste bulbs from cut lilies was investigated to explore their potential use as a value-added product. We divided lily bulbs into two groups, one group with six varieties of new bulbs (Medusa, Siberia, Woori Tower, Yelloween, Le Reve, and Morning Star) used for cultivation and the other group with six varieties of waste bulbs (Medusa, Siberia, Woori Tower, Yelloween, Sorbonne, and Sheila). Physiological activities (${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl: DPPH) and 3-ethlbenzthiazolne-6-sulfonic acid (ABTS) radical scavenging capability and tyrosinase inhibiting activity), the amount of total as well as eight individual phenolic compounds (chlorogenic acid, epicatechin, rutin hydrate, p-coumaric acid, kaempferol 3-O-${\beta}$-rutinoside, phloridzin dihydrate, myricetin, and quercetin), and total flavonoid content were measured in the bulbs by high performance liquid chromatography. We detected high amounts of total phenol and total flavonoid as well as high DPPH and ABTS radical scavenging ability. More tyrosinase inhibiting activity was detected in the new bulbs than in the waste bulbs. However, both the new and waste bulbs showed a higher inhibitory activity than the standard (100 ppm ascorbic acid). Although the content of phenolic compounds differed among varieties, under the conditions of the experiment, the most abundant phenolics were epicatechins, followed by chlorogenic acid, and rutins. Overall, the waste bulbs had a higher content of these compounds than the new bulbs. Based on these results, we concluded that bulbs from cut lilies could be used as functional foods in the future and farmers could expect economic gain from the hitherto neglected waste bulbs.

Evaluation of the Anti-obesity Activity of Platycodon grandiflorum Root and Curcuma longa Root Fermented with Aspergillus oryzae (도라지, 울금의 Aspergillus oryzae 발효에 의한 항비만효과 연구)

  • In the present study, the phenolic compound level, antioxidant activity, and inhibition of lipid accumulation in Aspergillus oryzae-fermented water extracts of the Platycodon grandiflorum (PG) root and the Curcuma longa (CL) root were determined. Total polyphenol and flavonoid contents were decreased after fermentation. However, the flavonoid content of the fermented PG (FPG) was increased by 2.9-fold that of the PG before fermentation. In addition, the antioxidant activities were significantly decreased following fermentation. The potential anti-obesity activity was assessed by determining lipid accumulation and mRNA expression of sterol regulatory element-binding protein 1c (SREBP-1c) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) in 3T3-L1 cells. Aspergillus-fermented extracts of PG and CL roots decreased lipid accumulation, and mRNA expression of SREBP-1c and $PPAR{\gamma}$ in 3T3-L1 cells. These results indicate that Aspergillus fermentation augments the anti-obesity activity of PG and CL by regulating the expression of the genes involved in lipid accumulation and cell differentiation of 3T3-L1 cells.

Inhibitory Effect of Methanol Extract and Its Fractions from Grape Seeds on Mushroom Tyrosinase (포도씨 추출물과 분획물의 Tyrosinase 저해활성)

  • The objectives of this study were to determine inhibitory effect on tyrosinase activity and antioxidant activity of the methanolic extract from grape seeds and to investigate relationships between tyrosinase inhibitory activity and antioxidant activity of the extract. The 80% methanol extracts of grape seeds were fractionated subsequently with hexane, chloroform, ethyl acetate and water. The methanolic extract and fractions from grape seeds inhibited tyrosinase activity in a concentration dependent manner. The methanolic extracts showed the highest inhibitory effects on tyrosinase activity. The inhibitory effects of the ethyl acetate fraction from the methanolic extract on tyrosinase activity was higher than other fractions. The ethyl acetate fraction from methanolic extracts showed higher antioxidant activity and contained higher polyphenolic and flavonoid contents compared to other fractions. The correlation coefficients among the polyphenoilc content of methanolic extracts, ABTS radical cation scavenging activity and inhibitory effect of tyrosinase were relatively high. These results suggest that grape seeds may have potential as a depigmentation agent for cosmetics and functional food products.

Bioactive Materials and Antioxidant Properties of Fermented Rice-bran Extract (쌀겨발효추출물의 이화학적 특성 및 항산화 작용)

  • This study suggests that fermented rice bran extract contains natural antioxidants. The contents of bioactive materials (e.g., polyphenolic compounds and flavonoids), antioxidative properties (DPPH (α,α'- diphenyl-β-picrylhydrazyl) free radical scavenging activity, Fe reducing, Cu reducing power, peroxidation of linoleic acid and rat hepatocyte microsome) were tested by in vitro experimental models using fermented rice bran (FRB) extract. The concentrations of phenolic compound and flavonoid were 19.92 mg/g and 11.56 mg/g, respectively. In oxidation in vitro models using DPPH free radical scavenging activity, (free radical scavenging activity 69.8%) Fe reducing power and Cu reducing power (effect of dose-dependent manner), Fe2+/ascorbate induced linolenic acid peroxidation by ferric thiocyanate and thiobarbituric acid (TBA) methods (inhibition activity 81%), and autooxidation of rat hepatic microsomes membrane (lipid peroxidation inhibition activity 38%), antioxidative activities were stronger in FRB extract than FRS (Fermented Rice and Soybean, positive control) extract and, these effects were dose-dependent manner. From these results, FRB extract was shown to have the most potent antioxidative properties and contain the highest amounts of antioxidative compounds such as phenolic compounds and flavonoids. Overall, these results may provide the basic data to understand the antioxidative properties of fermented rice bran for development of functional foods.
