Ultrastructures of Germ Cells and the Accessory Cells During Spermatogenesis in Male Gomphina veneriformis (Bivalvia: Veneridae) on the East Sea of Korea

  • The ultrastructures of germ cells and the accessory cells during spermatogenesis and mature sperm ultrastructure in male Gomphina veneriformis, which was collected on the coastal waters of Yangyang, East Sea of Korea, were investigated by transmission electron microscope observations. The morphology of the spermatozoon has a primitive type and is similar to those of other bivalves in that it contains a short midpiece with four mitochondria surrounding the centrioles. Accessory cells are observed to be connected to adjacent germ cells, they contain a large quantity of glycogen particles and lipid droplets in the cytoplasm. Therefore, it is assumed that they are involved in the supplying of the nutrients for germ cell development, while any phenomena associated with phagocytosis of undischarged, residual sperms by lysosomes in the cytoplasm of the accessory cells after spawning was not observed in this study. The morphologies of the sperm nucleus type and the acrosome shape of this species have a cylindrical and modified long cone shape, respectively. In particular, the axial filaments in the lumen of the acrosome, and subacrosomal granular materials are observed in the subacrosomal space between the anterior nuclear fossa and the beginning part of axial filaments in the acrosome. The spermatozoon is approximately $50-55{\mu}m$ in length including a long sperm nucleus (about $7.80{\mu}m$ in length), an acrosome (about $1.13{\mu}m$ in length) and tail flagellum ($40-45{\mu}m$). The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9+2 structure. Some charateristics of sperm morphology of this species in the family Veneridae are (1) acrosomal morphology, (2) the number of mitochondria in the midpiece of the sperm,. The axial filament appears in the acrosome as one of characteristics seen in several species of the family Veneridae in the subclass heterodonta, unlikely the subclass pteriomorphia containing axial rod instead of the axial filament. As some characteristics of the acrosome structures, the peripheral parts of two basal rings show electron opaque part (region), while the apex part of the acrosome shows electron lucent part (region). These charateristics belong to the family Veneridae in the subclass heterodonta, unlikely a characteristic of the subclass pteriomorphia showing all part of the acrosome being composed of electron opaque part (region). Therefore, it is easy to distinguish the families or the subclasses by the acrosome structures. The number of mitochondria in the midpiece of the sperm of this species are four, as one of common characteristics appeared in most species in the family Veneridae.

홍바리, Epinephelus fasciatus 정자의 미세구조 (Ultrastructure of Blacktip Grouper, Epinephelus fasciatus Spermatozoa)

  • 홍바리, Epinephelus fasciatus 정자의 미세구조적 특징을 주사형 및 투과형 전자현미경으로 관찰하였다. 홍바리 정자는 구형의 두부와 cytoplasmic sleeve와 고리형 cytoplasmic canal 입구 등으로 구성된 중편부 그리고 lateral fin을 갖는 1개의 편모로 구성된 미부로 구별된다. 정자의 핵은 구형이며, 첨체 구조물은 존재하지 않으나 이질 염색질이 과립상 덩어리 형태로 조밀하게 분포하였다. Proximal centriole과 distal centriole은 각각 수레바퀴형의 "9+0"의 축사구조를 보이며 서로 수직으로 osmophilic filaments 연결되어 있고, 핵막과도 연결되어 있었다. 중편부는 구형의 8~9개의 미토콘드리아를 포함하며, 빈공간인 cytoplasmic canal과 편모 원형질막에는 7~8개의 구형 입자를 갖는 necklace가 존재하였다. 미부는 1개의 편모를 갖으며 전형적인 "9+2"의 축사구조를 보였고, 편모에는 다양한 형태의 lateral fins을 관찰할 수 있었다. 홍바리 정자의 미세구조적 특징은 바리과 어류의 정자와 상이하여 계통분류학적 동정의 형질이 될 수 있을 뿐만아니라 정자 세포소기관의 기능적 및 생리학적 연구에 이용될 수 있다고 판단된다.

솔수염하늘소(Monochamus alternatus) 성충의 형태 측정과 암수 구분 (Adult Morphological Measurements: An Indicator to Identify Sexes of Japanese Pine)

  • 2001년부터 2002년까지 소나무재선충의 매개체인 솔수염하늘소 성충의 생체중과 체장, 체폭, 두폭, 더듬이의 길이 및 색깔 등을 조사하였다. 소나무재선충에 감염되어 고사된 소나무를 일정한 길이로 잘라 우화망실 안에 두고 우화하는 솔수염하늘소 성충을 채집하여 조사하였다. 분산분석결과 생체중, 체장, 체폭의 측정치는 암컷이 수컷보다 높았고, 두폭은 암수간에 차이가 없었으며, 촉각의 길이는 수컷이 더 길었다. 암컷(n=563)과 수컷(n=601)의 평균 생체중은 각각 0.305g과 0.277g, 체장은 20.97mm와 19.93mm 체폭은 6.52mm 와 6.18mm, 두폭은 3.78 mm와 3.70mm, 촉각의 길이는 31.19mm와 45.49mm이었다. 촉각의 길이나 체장과 촉각 길이의 비율은 암수간에 중복되는 범위가 많아서 이것을 기준으로 암수를 구분할 수는 없었다. 그러나 성충(n=4,033) 촉각의 털 색깔을 조사한 결과, 예외 없이 암컷 촉각의 모든 편절 마디의 기부쪽 절반 정도가 회백색의 미모로 덮여 있으나, 수컷 촉각의 편절 마디는 전체적으로 흑갈색의 미모로 덮여 있었기 때문에, 이 특징으로 암수를 쉽게 구분할 수 있었다.

  • Ultrastructural characteristics of the testis and spermatogenesis of Crassostrea gigas were investigated by Transmission and Scanning Electron microscope observations. The testis is a diffuse organ consisting of branching acini containing differentiating germ cells in a variety of stages. The acinus is surrounded by an intermitent layer of myoepithelial cells andis divided into subcompartments that are partially separated by pleomorphic accessory cells which remain in close contact with germ cells until late stages of development. these accessory cells contain a large quantity of glycogen particles and lipid droplets in the cytoplasm. Therefore, it is assumed that they are involved in the supplying of the nutrients for germ cell development, while any phenomena associated with phagocytosis of undischarged, residual sperms by lysosomes could be find in the cytoplasm of the accessory cells. The morphology of the spermatozoon has a primitive type and is similar to those of other bivalves. Mature spermatozoa consist of broad, cap-shaped acrosomal vesicle, subacrosomal material (containing axial rod embedded in a granular matrix), a oval nucleus showing deeply invaginated anteriorly, two triplet substructure centrioles surrounded by four spherical mitochondria, and satelite fibres appear to the distal centriole and plasma membrane. Spermatozoa of C. gigas resemble to those of other investigated ostreids. In particular, the anterior region of the acrosomal vesicle is transversely banded. It is assumed that differences in this acrosomal substructure are associated with the inability of fertilization between the genus Crassostrea and other genus species in Ostreidae. Therefore, we can use sperm morphology in the resolution of taxonomic relationships within the Ostreidea. The spermatozoon is approximately $42-47{\mu}m$ in length including an oval sperm nucleus (about $0.91{\mu}m$ in length), an acrosome (about $0.42{\mu}m$ in length) and tail flagellum ($40-45{\mu}m$). The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9 + 2 structure. These morphological charateristics of acrosomal vesicle belong to the family Ostreidae in the subclass Pteriomorphia.

Milt Properties of Four Flatfish Species and Fine Structure of Their Cryopreserved Spermatozoa

  • The physico-chemical properties of fresh milt of marbled sole, Limanda yokohamae, brown sole, Limanda herzensteini, starry flounder, Platichthys stellatus and olive flounder, Paralichthys olivaceus among flatfishes, and the fine structure of their cryopresserved spermatozoa were investigated. The highest concentration of sperm among these four species was $3.60\pm1.35\times10^{10}/mL$ in marbled sole. Osmolality and pH of seminal plasma in four flatfish species were approximately 330 mOsm/kg and 7.6-8.1, respectively. Seminal plasma compositions showed interspecific differences. The sperm heads of marbled sole, brown sole and starry flounder were ellipsoidal and that of olive flounder was round. The numbers of mitochondria of these four species were eight in marbled sole, seven in brown sole and starry flounder, and six in olive flounder. Cross-sectional view of flagellum showed typical 9+ 2 structure in all species. Most of sperms cryopreserved with a proper method had no visible ultrastructural changes after freeze-thawing, compared with the fresh sperm, but in a few cases, swelling of their heads and midpiece regions were observed.

Finding of a new freshwater gammarid (Gammarus gageoensis) from South Korea

  • Gammarus gageoensis n. sp., a new species of freshwater gammarid, was collected from Gageodo Island, South Korea. This new species is characterized in terms of its morphology by the flagellum of antenna 2 with calceoli, the pereopods 3 and 4 with numerous long curled setae on the posterior margins, the terminal article of the outer ramus in uropod 3, which is longer than the adjacent spines, and the presence of both rami with long simple setae. The morphological characters of the new species that are different to related species are presented in the text. This paper also compares the new species to related species by the partial sequence of the mitochondrial cytochrome c oxidase subunit 1 (CO1) gene. The sequence difference, consisting of a 39-44% difference relative to closely related species, is also large enough to designate this gammarid as a different species from other freshwater gammarid species. This is the first finding of a new species of Korean freshwater gammarid since 1992. Including this new species, a total of 10 species are known to Korea.

한국산 잉어과어류 칼납자루(Acheilognathus koreensis) 정자의 미세구조 (Ultrastructure of Spermatozoa of a Korean Bitterling, Acheilognathus koreensis (Pisces, Cyprinidae))

  • 칼납자루 Acheilognathus koreensis 정자의 미세구조를 전자현미경으로 관찰하였다. 칼납자루 정자의 미세구조는 잉어과 어류정자의 일반적인 구조와 같이 둥근 두부, 얕은 핵와, 세포의 측면에 위치한 편모구조 그리고 비 대칭적인 편모구조의 특징을 취하고 있었다. 그러나 중편에 위치한 미토콘드리아는 대부분의 잉어류에서와는 달리 융합되어 하나를 형성하고 있으며 그 주위를 막성 구조물들이 둘러싸고 있었다. 하나로 융합된 미토콘드리아를 가지는 정자의 구조는 여러개로 나타나는 것에 비하여 경골어류에서는 파생형질로 알려져 있다. 잉어류 정자에서 미토콘드리아가 융합되어 하나로 나타나는 구조는 Rodeus와 Puntungia에서 보고되어 있으며 그 주위를 막성구조물이 둘러싸고 있는 것은 납자루류에서만 보고되고 있다.

Developmental Phases of the Seminal Vesicle related to the Spermatogenic Stages in the Testicular Lobules of Neptunea (Barbitonia) cumingii (Gastropoda: Buccinidae)

  • Cytological changes of the epithelial cells according to the developmenatal phases of the seminal vesicle related to the spermatogenic stages in the testicular lobules during spermagenesis in male Neptunea (Barbitonia) cumingii (Gastropoda: Buccinidae) were investigated monthly by electron microscopical and histological observations. N. (B) cumingii is dioecious, and an internal fertilization species. The male genital organ is located near the tentacles. The spermatozoon is approximatley $50{\mu}m$ in length. The axoneme of the tail flagellum consists of nine pairs of microtubles at the periphery and one pair at the center. The process of germ cell development during spermatogenesis can be divided into five succesive stages: (1) spermatogonia, (2) primary spermatocytes, (3) secondary spermatocytes, (4) spermatids, and (5) spermatozoa. A considerable amount of spermatozoa make their appearance in the testicular lobules (or acini) and some of them are tranported from the testis towards the seminal vesicles until late July. In this study, the developmental phases of the epithelial cells of the seminal vesicles of N. (B.) cumingii could be classified into four phases: (1) S-I phase (resting), (2) S-IIphase (early accumulating), (3) S-III phase (accumulating), and (4) S-IV phase (spent). However, in case of N. (B.) arthritica cumingii, the developmental phases of the seminal vesicle were devided into three phases: (1) resting, (2) accumulating and (3) spent. Granular bodies in the inner layer of the seminal vesicles are involved in resorption of digestion of residual spermatozoa.

Ultrastructural Studies of Germ Cell Development and the Functions of Leydig Cells and Sertoli Cells associated with Spermatogenesis in Kareius bicoloratus (Teleostei, Pleuronectiformes, Pleuronectidae)

  • The ultrastructures of germ cells and the functions of Leydig cells and Sertoli cells during spermatogenesis in male Kareius bicoloratus (Pleuronectidae) were investigated by electron microscope observation. Each of the well-developed Leydig cells during active maturation division and before spermiation contained an ovoid vesicular nucleus, a number of smooth endoplasmic reticula, well-developed tubular or vesicular mitochondrial cristae, and several lipid droplets in the cytoplasm. It is assumed that Leydig cells are typical steroidogenic cells showing cytological characteristics associated with male steroidogenesis. No cyclic structural changes in the Leydig cells were observed through the year. However, although no clear evidence of steroidogenesis or of any transfer of nutrients from the Sertoli cells to spermatogenic cells was observed, cyclic structural changes in the Sertoli cells were observed over the year. During the period of undischarged germ cell degeneration after spermiation, the Sertoli cells evidenced a lysosomal system associated with phagocytic function in the seminiferous lobules. In this study, the Sertoli cells function in phagocytosis and the resorption of products originating from degenerating spermatids and spermatozoa after spermiation. The spermatozoon lacks an acrosome, as have been shown in all teleost fish spermatozoa. The flagellum or sperm tail of this species evidences the typical 9+2 array of microtubules.

Ultrastructural Characteristics of the Testis, Spermatogenesis and Taxonomic Values of Sperm Morphology in Male Ruditapes philippinarum in Western Korea

The morphology of the spermatozoon is of the primitive type and is somewhat different to those of other bivalves. The morphologies of the sperm nucleus type and the acrosome shape of this species have a cylinderical type and a modified cone shape, respectively. As some ultrastructural characteristics of the acrosomal vesicle, the peripheral parts of two basal rings show electron opaque part, while the apex part of the acrosome shows electron lucent part. These characteristics of sperm belong to the family Veneridae in the subclass Heterodonta, unlike a characteristic of the subclass Pteriomorphia showing all part of the acrosome being composed of electron opaque part. In particular, a cylinder-like nucleus of the sperm is curved. The spermatozoon is approximately $48-51{\mu}m$ in length, including a long acrosome (about $2.4{\mu}m$ in length), a curved sperm nucleus (about $3.40{\mu}m$ in length), and a tail flagellum. The axoneme of the sperm tail shows a 9+2 structure.
