A STUDY ABOUT THE CHILDREN'S COMCEPTION OF MOVEMENT AND SPEED (아동의 운동과 속력개념형성에 관한 연구)

  • In order to research into the devel of the concept of movement and speed in our own way and compare with the Piaget's method, and then find out the formative period of such development, we have put the 192 primary school children as the model and obtained the following results. 1. As their grade upyards, all the experimental subjects showed the gradual progress generally regardless of sex or regions, while only the relative speed showed irregular progress. 2. There was no experimental subject which showed any remarkable sexual characteristics. But in the relative movement, the lower grade boys and higher grade girls made progress. In the subjects of circulation movement, intuition of speed, relative speed and speed of simultaneous movement, the boys marked better progress, whilst the girls were advanced a little in the speed of continuous movement with, different distance and time. 3. There was no conspicuous difference between the urban and rural areas, except a slight tendency that the urban children made better progress in the change of progressing direction, native continuous procedure of periodical movement, travelling road, and the speed of continuous movement with different distance and time whereas the rural children were more advanced in the relative movement, relative speed and intuition of speed. But it should be 'taken into consideration that the rural regions in our case were relatively developed in comparison with the traditional rural communities, which may explain about little regional difference. 4. Comparing our research results with the Piaget's theory, we have reached below results. Our research reports that the formative period of the conception of the problems of diverse progressing direction and a native continuous procedure of periodical movement was simultaneous, at the Piagetian level(at the fourth grade) which coincides with Piagetian theory. The travelling road should be made up a little lower at 2A/2B according' to Piagetian Level, whereas it was formed at the 4th grade as shown in our previous research. Intuition of speed should belong to the prior stage to concrete operation, but our research shows it was formed late at the 4th grade. Composition of displacement was made at the 6th grade, and it was almost equal to the first stage of formal operation(3A). But in the subjects of relative movement relative speed, the speed of continuous movement with different distance and time, relation and preservation of invariable speed, and accelerated motion, even 6th grade children marked a poor record. Summed up, the procedure conception as a basic movement conception coincides with the Piagetion level. But as for speed intuition, relative speed and speed fixation, it was Jar behind Piagetian level. Therefore it is required that we have to concentrate on the systematic training in these parts on the spot.

Genetic signature of strong recent positive selection at interleukin-32 gene in goat

  • Objective: Identification of the candidate genes that play key roles in phenotypic variations can provide new information about evolution and positive selection. Interleukin (IL)-32 is involved in many biological processes, however, its role for the immune response against various diseases in mammals is poorly understood. Therefore, the current investigation was performed for the better understanding of the molecular evolution and the positive selection of single nucleotide polymorphisms in IL-32 gene. Methods: By using fixation index ($F_{ST}$) based method, IL-32 (9375) gene was found to be outlier and under significant positive selection with the provisional combined allocation of mean heterozygosity and $F_{ST}$. Using nucleotide sequences of 11 mammalian species from National Center for Biotechnology Information database, the evolutionary selection of IL-32 gene was determined using Maximum likelihood model method, through four models (M1a, M2a, M7, and M8) in Codeml program of phylogenetic analysis by maximum liklihood. Results: IL-32 is detected under positive selection using the $F_{ST}$ simulations method. The phylogenetic tree revealed that goat IL-32 was in close resemblance with sheep IL-32. The coding nucleotide sequences were compared among 11 species and it was found that the goat IL-32 gene shared identity with sheep (96.54%), bison (91.97%), camel (58.39%), cat (56.59%), buffalo (56.50%), human (56.13%), dog (50.97%), horse (54.04%), and rabbit (53.41%) respectively. Conclusion: This study provides evidence for IL-32 gene as under significant positive selection in goat.

Antioxidative Activities of Mixture of Schisandrae Fructus (SF) and Perilae Folium (PF) using Natural Color fixation technic (천연 색소 고정화 기술을 활용한 오미자와 자소엽 추출물 함유 음료의 항산화 활성 연구)

  • Objectives : This study focused on Schisandrae Fructus (SF) and Perilae Folium (PF), traditional medicine herbs and health functional food in Korea, Japan and China. We investigated various pharmacological activities that include a potential source of free radical scavenging, anti-viral, anti-microbial, anti-allergy and anti-inflammatory activities. Methods : We conducted an investigation of total contents of phenolic and flavonoid compounds in these single herbal extraction with/without combined to mixture. We also measured antioxidant activities such as DPPH free radical scavenging, SOD-like scavenging, nitrite scavenging and hydroxyl radical scavenging activities, xanthine oxidase inhibition, linoleic acid peroxidation inhibition, and reducing power. Results : As the results, contents of total phenoilc compounds and flavonoids were higher in those of PF than those of SF. Those of SF+PF mixture showed the synergy effects compared with those of SF and PF single extractions. Activities of DPPH free radical and SOD-like scavenging in 1 mg/mL concentration increased in dose dependent manners. That of SF increased compared with that of PF. That of SF and PF mixture also increased compared with that of BHA as a positive control. The other antioxident activities also showed similar to patten of activity of DPPH free radical scavenging. When combined to SF and PF extractions, there was showed synergic effect compared with those of BHA, excepted activities of xanthine oxidase inhibition and reducing power. Taken together, SF and PF have high phenolic and flavonoid compounds content furthermore, antioxidant activities in SF and PF mixture showed more synergy effect compared with those of BHA. Conclusions : Therefore, these findings suggest that SF and PF mixture may offer functional materials potential for development of functional beverage. But further studies are needed for the identification of the active compounds.

An Electron Microscopy of Oogenesis and Fertilization in Clonorchis sinensis (간(肝)디스토마 자성생식세포(雌性生殖細胞) 분화과정(分化過程)의 전자현미경적(電子顯微鏡的) 연구(硏究))

  • The morphological and fine structural changes during the oogensis of Clonorchis sinensis were studied on the developing ovums in the ovary and ootype with electron microscope. Adult worms were removed from the hepar of the which and previously infected with metacercariae of Clonorchis sinensis. The ovary including the Mehlis' glands and an ootype from adult worm was prefixed for 1-2 hours in 1.25% glutaraldehyde buffered with 0.2M cacodylate at PH 7.2, secondarily fixed for 30 minutes in potassium bicromate and postfixed for an hour in 1% osmic acid buffered with 0.4M cacodylate at PH 7.2. After fixation tissues were dehydrated in an alcohol series, embedded in Epon 812 from propylene oxide and stained with saturated uranyl acetate and $Pn(NO_3)_2$ solution. Material was examined with a Hitachi HS-7S electron microscope. The periphery of the ovary, except for the posterior region, is made up of oogonia. As the oogonia divide they proliferate primary oocytes toward the central part of the ovary. After a period of growth the primary oocyte leaves the ovary and is penetrated by a sperm in the ootype. Sperm penetration immediately activates the primary oocyte to resume its meiotic activity. After the oocytes meiotic activity is completed, the pronuclei fuse to form a single cleavage nucleus which possesses two nucleoli. As the oocytes develop their cytoplamic materials are abundant; small mitochondria are abundant and often their profiles are more unmerous in one part of the cytoplasm than elsewhere; the granular endoplasmic reticulum becomes alveolar-sac form after it leaves the ovary it becomes stratified form. The reticulate Golgi apparatus is apparent in the developed oocyte. A little of cortical granules are distributed inside of the plasma membrane I oogonia and large quantity of cortical granules are arranged just inside of the plasma membrane of the primary oocyte and after fertilization they are disappeared with broken out.

Verification of Target Position in Stereotactic Radiosurgery Based on Photon Knife System (Photon Knife 시스템에 근거한 뇌정위 방사선수술에서 표적위치 확인)

  • This study was performed to prepare the verification film for localizing beam-target position with the Photon Knife radiosurgery system (PKRS) using linear accelerator(Mitsubishi, Model ML-15MDX). We developed a laser calibration system using a reticle of transparent lucite to detect Inlet and outlet beams. We verified fixation of the second collimator with film mounted on a holder in the shape of an octagon block 5cm apart from the isocenter. The film was exposed to photon beams of linear accelerator at an interval of 45 degrees during the gantry movement. There were no shifts in the beam of the second collimator during gantry movement. We used a position marker which is designed a head-shaped small lead block and a 10 mm in diameter of steel bead in the plastic tube. The position marker helped to verify the beam directions with patient position in multi-arc and trans-multi-arc of PKRS The verification of beam alignments showed an average 0.8$\pm$0.26 mm discrepancy in LINAC-gram images of PKRS. In our study, the couch movement was $\pm$5 mm laterally, while it shook $\pm$ 2 mm toward the couch axis. The couch, however, was immediately returned to the initial site after shaking. Thus, we postulate that the beam-target position(s) should be verified with LINAC-gram in a stereotactic radiosurgery system to achieve the accuracy of beam-target alignment.

Genetic diversity and structure of Pulsatilla tongkangensis as inferred from ISSR markers (ISSR 표지자에 의한 동강할미꽃(Pulsatilla tongkangensis)의 유전다양성과 구조)

  • The genetic diversity and structure of P. tongkangensis in 5 populations from 3 regions was investigated using 56 markers derived from 6 ISSR primers. Genetic diversity at the species level (P=94.6, SI=0.377, h=0.240) was substantial considering the limited distribution and small size of populations. Genetic differentiation among regions (12%) and among populations (13%) in the region was not clearly evident, which suggested a moderate level of gene flow among adjacent populations. The Mantel test revealed a significant correlation between genetic differentiation (${\Phi}_{ST}$) and geographic distance among populations. This was supported by cluster analysis and principal coordinate analysis (PCoA). The significant difference in marker band frequency at many loci and their fixation in opposite directions in the smallest and most isolated population SC were considered due to genetic drift. Therefore, the genetic diversity of P. tongkangensis could be compromised if the distribution area or the size of the population was further reduced. In particular, small and isolated populations could be at great risk of extinction. Considering this, the unique habitats of P. tongkangensis should be protected and the reduction of population size should be closely monitored. Conservation efforts including the seeding and planting of seedlings should be done carefully based on their genetic and ecological traits. Our data support the argument that establishing an integrated management system for the efficient conservation of P. tongkangensis is critical.

Reason and Prevention of Color Reversion of Corn Oil in Summer (옥수수기름의 하절기 변색현상 원인규명 및 방지대책)

  • Crude corn oil (CCO) was obtained through the expression-extraction process from corn germ. The CCOs of final process at $90^{\circ}C,\;70^{\circ}C\;and\;50^{\circ}C$ were stored in outdoor storage tanks. From the samples, refined CCO (RCCO) were prepared with $0\%,\;10\%\;and\;20\%$ excessive of phosphoric acid, caustic solution and acidic clay were used in degumming-alkali refining-bleaching process. RCCOs were stored at room temperature in dark places. The color change was not effected by the amount of phosphoric acid, caustic solution and acidic clay, but temperature of process affects the color change. Finally, the prevention for color reversion of RCCO could be obtained by lowering the temperature of final process and optimal temperature of RCCO in summer was found about less than $50^{\circ}C$.

Potential Seizurogenic Effect of Tramadol in a Dog with Necrotizing Meningoencephalitis (괴사성 뇌척수막염을 가진 진돗개에서 Tramadol에 의한 잠재적 경련발생 증례)

  • A Jindo dog (8-month-old, intact male) was referred for hind limb lameness on the right side. The dog was diagnosed with a simple femoral fracture by radiological examination. After surgical fixation of the femoral fracture, tramadol: a narcotic-like synthetic analgesic was intravenously administrated for post-operative analgesia. After injection of the tramadol, generalized tonic clonic seizure was immediately occurred in the dog. Seventeen hours later, the dog died despite intensive care. We suspected that tramadol might induce the seizurogenic effect resulted in death. A necropsy was performed to examine the cause of the death. In consequence, the dog was diagnosed as necrotizing meningoencephalitis (NME) based on histopathological examination. We would be concerned that tramadol may be related to seizure activity in the NME patient. From this case, it is known that although tramadol has been proven to be a safe and effective agent for the control of pain in veterinary medicine, it would be carefully used to patient with history of neurological diseases including meningoencephalitis, hydrocephalus, and encephalopathy.

A Case of Intradiscal Oxygen-ozone Injection Therapy for Cervical Herniated Intervertebral Disc in a Dog (개에서 탈출된 경추디스크에 대한 디스크내 오존가스 주입 치료)

  • A 6-year-old castrated male Shih-tzu was presented because of a 1-month history of progressive tetraplegia. On the basis of the findings of neurological examination, radiographs and magnetic resonance imaging, herniated intervertebral disc of C4-C5, C5-C6 and C6-C7 compressed spinal cord. The oxygen-ozone was intra-operatively injected into the C4-C5 and C6-C7 intervertebral disc and a ventral decompression performed at C5-C6 intervertebral space. Immediately after the surgery, neck pain was disappeared and three weeks after surgery, there was complete resolution of the neurological deficits with normal gaits. On magnetic resonance images at 2-month after surgery, herniated disc materials of C5-C6 and C6-C7 were completely removed and that of C4-C5 remarkably reduced. Consequently decompression which was performed at all of the three lesions resulted in good clinical outcome without additional internal fixation for minimizing postoperative instability.

Factors Regulating the Nitrogen Fixation Activity and Growth of Anabaena variabilis ATCC 29413 (Anabaena variabilis ATCC 29413 의 생장과 질소고정활성의 조절요인)

  • Anabaena variabilis A TCC 29413. a photoautotrophic and nitrogen fixing cyanobacteria. was investigated on the environmental factors regulating the growth and nitrogen lixation activity. A good growth of cyanobacteria] cells was observed due to nitrogen t1xation by the heterocyst differentiation in nitrogen free Allen and Arnon (]/8) medium. The nitrogenase activity was appeared to be in proportion to the cell growth lor 6 days then drastically decreased in the later growth period when the nitraTe was accumulated to high level in the culture to cause the inhibition. The optima] conditions lilr the cell growth and nitrogenase activity of A. varillbili.l were anaerobic. IO.OO0 lux. $30^{\circ}C$ and pH 8 with the nitrogen Cree minimal medium. The activity was significantly inhihited by the low concentrations of ammonium and nitrate. but was stimulated b) the ]ow Ieve] of phosphate and carbonate sources. The treatments of several toxic heavy metals showed strong inhibition of the cell growth and nitrogenase activity by O.3~10 ppm in the order of $Hg^{2+}$ > $Cd^{2+}$ > $Co^{2+}$ > $Zn^{2+}$ > $Ph^{2+}$, and the concentrations for 50% inhibition of the maximum activity were 0.41. 0.47. 0.5 L 0.66 and 8.1 ppm. respectively. The addition of carbohydrates (0.5~ 1.0%) in the dark condition stimulated the growth and activity in the order of sucrose > fructose > glucose.
