Constituents of the Fruits of Rumex japonicus with Inhibitory Activity on Aldose Reductase

  • Five anthraquinones, emodin (1), ${\omega}$-hydroxyemodin (2), chrysophanol-8-O-${\beta}$-D-glucoside (3), emodin-8-O-${\beta}$-D-glucoside (4), and physcion-8-O-${\beta}$-D-glucoside (5), and five flavonoids, kaempferol-3-O-${\beta}$-D-glucoside (6), quercetin (7), quercitrin (8), isoquercitrin (9), and (+)-catechin (10), were isolated from the EtOAc-soluble extract of the fruits of Rumex japonicus. The structures of 1-10 were identified by spectroscopic methods including NMR studies. This is the first report on the isolation of compounds 3-5 from this plant. The isolates were subjected to in vitro bioassays to evaluate their inhibitory activities on the rat lens aldose reductase (RLAR), among which two anthraquinones (1 and 4), and five flavonols (5-9) showed significant activities on RLAR.

  • In order to select superior tree from Korea, five major phenolic compounds including (-)-epicatechin(EC), chlorogenic acid (ChA), hyperoside (HP), isoquercitrin (IQ), and procyanidin B2 (PC-$B_2$) in hawthorn fruit were evaluated. We also compared these hawthorn fruits of five clones with Chinese hawthorn cultivars. HPLC with a diode-array detector was used to determine the contents of the individual compounds. Hawthorn fruits of five clones (selected from different area of Korea), and four Chinese hawthorn cultivars grown in the Korea Forest Research Institute (Suwon) were utilized. With their high functional components, Jungsun is the clone including the highest contents of EC (11.26 mg/g) and PC-$B_2$ (24.46 mg/g). The clone of Chuncheon 15 had highest HP (0.53 mg/g) and IQ (0.41 mg/g). From the results, the clone of Jungsun and Chuncheon 15 can be evaluated to be selected breeding material for cultivar development.

  • Ethylene was released from leaf and fruit but root of Panax ginseng. Root callus showed higher ethylene release (ER) than fruit ER increased with leaf senesence. Fruit during ripening showed decreasing ER in the order of green stage, early stage of reddening and fully ripened stage. between leaves from the plant with fruits in different stages of ripening showed similar trend of fruit in ER but it was about 10 times higher in leaves than in fruits. Leaves of P. quinquefolius showed about 200 times higher ER than that of P ginseng on 22 July Fruits from the plant treated with ethephon showed higher ER after 109 days. Forty-five day-old seedlings grown with various growth regulators showed a significant decrease of stem length and significant increase of ER only in Uniconazole (0.1 ppm) and H-9 (0.0, 5 ppm) solution.

  • We bred a mulberry cultivar named 'Saealchan' through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at four local places (Jeonju, Gongju, Jangseong, and Sangju) for five years from 2013. This variety 'Saealchan' belongs to Morus alba L. selected from seedlings collected from mother mulberry 'Shimheung'. Saealchan was resistant to popcorn disease and high yielding variety in fruit productivity by 194% compared to control cultivar 'Shimheung (Morus alba L.)' for three years. Although fruits of 'Saealchan' was smaller in size and lower in sugar content of mulberry fruits compared to control variety 'Shimheung', it showed strong resistance to sclerotial disease compared to 'Shimheung'.

  • A survey was conducted to examine the food intake of 285 Korean female students attending a secondary school in Seoul. Food intake was assessed using a food frequency based on the Korean Food Tower which consists of five food groups, i.e., grain products, vegetables and fruits, meat, milk, and fats and sweets. To describe food intake descriptive statistics were obtained using SPSS. The food frequency analysis indicated that the participants ate rice, cabbage kimchi, radish, and soybean/red pepper paste most frequently along with milk, ice cream, tea, and seasonal fruits such as citrus fruits, apples, watermelon, and grapes for snacks. Distributions of the total standardized serving frequency for each food group indicated that approximately 84, 83, 45, 60, and 34%, respectively, of the respondents met the recommended serving frequency for grain products, vegetables and fruits, meat, milk, and fats and sweets groups(mean servings per day 5.5, 10.6, 5.5, 2.1, 3.5). Income was the only demographic factor that affected food intake, particularly with respect to meat, milk, and fats and sweets(p < .01).   • Front the production on the farm to the consumer, agricultural products are subject to various physical treatments involving mechanical techniques and devices. It is essential to understand the physical laws governing the response of these biological materials so that the machines, processes, and handling operations can be designed fur maximum efficiency and the highest quality of the end products. A compression test system was developed to test the physical properties of fruits including apple, pear, and peach which may lead to a better understanding of the physical laws. The test system consisted of a digital storage oscilloscope and simple mechanism which can apply quasi-static compression to fresh fruits. Rupture force, energy, and deformation were measured at the five levels of compression speed from 1.25 to 62.5 mm/min for each internal and external tissues. Rupture forces for apple and pear were in the range of 42.2 to 46.2 N and 38.8 to 41.2 N for external and infernal tissues, respectively. Rupture forces fir peach external tissues were in the range of 48.2 to 54.0 N.

  • In order to determine the residual characteristics of pesticides in fruits and their effects on human health, monitoring of pesticide residues were conducted in apples, peaches, pears, grapes and citrus from the major cultivation areas in 2010. Safeties of the pesticides detected from the fruits were evaluated using the amounts of pesticide residue detected and dietary risk index (DRI). Samples were taken from 4-5 major fruit cultivation areas and then pesticide residues in the test fruits were analyzed with a liquid chromatographymass spectrometry (LC-MS/MS). Mean residue levels of the pesticides in fruits were 0.001-0.144 mg/kg. The DRIs were 0.55 for apple, 0.066 for peach, 0.008 for pear, 0.025 for grape, and 0.37 for citrus, respectively. The results indicated that the amounts of pesticide residues in domestic fruits might be safe, considering the regulatory concerning level 1.0.

  • Impact is one of the major cause of damage to fruits druing varios processes from the production on the farm to the consumer. The tissue of fruits are ruptured in a very short period time less than 10ms by impact loading. Mechanical behavior of fruits under impact loading can be analyzed better with high speed sampling data acquisition system and one of them is a digital storage oscilloscope. A impact test system was developed to test the physical properties of fruits including apple, pear, and peach which may lead to a better understanding of the physical laws. The test system consisted of a digital storage oscilloscope and simple mechanism which can apply impact force to fresh produce. Rupture force, energy, and deffrmation were measured at the five levels of drop heights from 4 to 24cm fur each internal and external tissues. Rupture forces for apple and pear were in the range of 72.9 to 87.7 N and 70.8 to 84.1 N for external and internal tissues, respectively. Rupture forces far peach external tissues were in the range of 43.4 to 65.0 N.

Rapid discrimination of commercial strawberry cultivars using Fourier transform infrared spectroscopy data combined by multivariate analysis

  • To determine whether pattern recognition based on metabolite fingerprinting for whole cell extracts can be used to discriminate cultivars metabolically, leaves and fruits of five commercial strawberry cultivars were subjected to Fourier transform infrared (FT-IR) spectroscopy. FT-IR spectral data from leaves were analyzed by principal component analysis (PCA) and Fisher's linear discriminant function analysis. The dendrogram based on hierarchical clustering analysis of these spectral data separated the five commercial cultivars into two major groups with originality. The first group consisted of Korean cultivars including 'Maehyang', 'Seolhyang', and 'Gumhyang', whereas in the second group, 'Ryukbo' clustered with 'Janghee', both Japanese cultivars. The results from analysis of fruits were the same as of leaves. We therefore conclude that the hierarchical dendrogram based on PCA of FT-IR data from leaves represents the most probable chemotaxonomical relationship between cultivars, enabling discrimination of cultivars in a rapid and simple manner.
