  • In this study, a 12-week feeding experiment was conducted to characterize the effects of exogenous α-amylase on the growth, feed utilization, digestibility, plasma α-amylase activity, feed degradation rate, and fecal particle size of olive flounder (Paralichthys olivaceus). Diet was supplemented with 0 (AA0; control), 100 (AA100), 200 (AA200), or 400 (AA400) mg/kg of α-amylase, respectively. Fish (273.1 ± 2.3 g) were stocked into 12 tanks (25 fish/1,000-L tank) and 3 tanks were randomly selected for each diet group. As a result, α-amylase was found to have no significant effects (p ≥ 0.05) on the growth, feed utilization parameters, and whole-body proximate compositions. α-Amylase-treated fish exhibited only a significant increase in the apparent digestibility coefficient of carbohydrates compared to the controls. In addition, in vitro analyses revealed that α-amylase dose-dependently increased (p < 0.05) the feed degradation rate, while photographs of the intestinal content after 2, 4, and 8 h of feeding demonstrated an improved degradation rate in the α-amylase-treated groups. Plasma α-amylase content was higher in the AA200 and AA400 groups, whereas the control group produced significantly larger-sized fecal particles (90% size class) than these two groups. In the intestine, no changes were observed in the expression levels of the immune-related TNF-α, IL-1β, IL-2, immunoglobulin-M, HSP-70, lysozyme, and amylase alpha-2A. However, growth-related genes IGF-1, IGF-2, TGF-β3, and growth hormone genes were upregulated in muscle tissues. Collectively, exogenous α-amylase has positive roles in the modulation of the digestibility coefficient, blood α-amylase concentration, growth-related gene expression, and diet degradation for improved digestion in olive flounder.

  • Two experiments were conducted to investigate the effects of dietary moisture content on the growth and gastrointestinal evacuation of sub-adult olive flounder (Paralichthys olivaceus) in the summer and winter seasons. In the first experiment, three experimental diets containing different moisture levels (7%, 15%) and additives were prepared by adding water and commercial additives (1% nutrients, 1% digestives) to commercial extruded pellets. Fish (initial weight: $332{\pm}9.2$ g) were distributed randomly into six 3000 L tanks (25 fish/tank) in a flow-through tank system. Two replicate groups of fish were fed the experimental diets to satiation for 14 weeks during the summer season ($21.4{\pm}2.5^{\circ}C$). Survival, weight gain, feed efficiency, and daily feed intake did not differ significantly among groups. In the second experiment, two experimental diets containing different moisture levels (8%, 23%) were prepared by adding water to commercial extruded pellets. Fish (initial weight: $646{\pm}6.7$ g) were randomly distributed into six 3000 L tanks (20 fish/tank) in a flow-through tank system. Three replicate groups of fish were fed the experimental diets to satiation for 19 weeks during the winter season ($12.2{\pm}1.2^{\circ}C$). Weight gain, feed efficiency, and daily feed intake did not differ significantly among groups. Gastrointestinal evacuation rates were determined after the end of the winter feeding trials. The stomach contents of fish fed diets containing different moisture levels peaked within 3 h after feeding and then decreased gradually over 40 h to approach pre-feeding levels. Intestinal contents began to accumulate 3 h after feeding and reached a maximum at 32 h, then declined until nearly complete evacuation around 56 h. The stomach contents reached moisture levels of approximately 70% within 3 h after feeding and then increased gradually. No considerable difference in moisture levels was observed in the stomach contents of fish fed diets with different moisture contents. The results of this study suggest that the gastric evacuation of sub-adult olive flounder was not affected by dietary moisture level and the addition of water to the diet had no beneficial effect on the growth of sub-adult olive flounder in the summer and winter seasons.

  • This study was designed to evaluate the economic analysis of channel catfish production in 1998 based on fish value and total feed cost. Catfish received higher protein feeds with lesser amount based on the dietary protein levels, but received the constant total protein input for all treatments. Weight gain per pond for treatment 1 $(28\%\;protein,\;100\%\;of\;satiation)$ was higher (P<0.10) than for treatment 3 $(36\%\;protein,\;77.8\%\;of\;satiation)$, but not significantly higher than for treatment 2 $(32\%\;protein,\;87.5\%\;of\;satiation)$ at constant DE. At constant DE/P (treatments 4, 2 and 5), weight gain per pond for treatment 5 $(36\%\;protein,\;77.8\%\;of\;satiation)$ was lower (P<0.10) than for treatment 2, but not significantly lower than for treatment 4 $(28\%\;protein,\;100\%\;of\;satiation)$. At constant DE, feed conversion slightly improved as dietary protein level increased from $28\%\;to\;32\%$ and feed allowance decreased by $12.5\%$, but did not improve further as dietary protein level increased from $28\%\;to\;36\%$ and feed allowance decreased by $22.2\%$. At constant DE/P, feed conversion improved as dietary protein level increased from $28\%\;to\;32\%$ increased and feed allowance decreased by $12.5\%$, but did not improve as dietary protein level increased from $28\%\;to\;36\%$ and feed allowance decreased by $22.2\%$ Total feed cost for treatment 1 was slightly, but not significantly higher than for treatments 2 and 3 at constant DE. At constant DE/P, total feed cost for treatment 5 was higher (P<0.05) than for treatment 2, but not significantly higher than for treatment 4. Total value of fish ($ /ha) produced for treatment 1 was highest and lowest was for treatment 5. Return above feed cost was highest for treatment 1 and nearly the same as treatment 2. Return over feed cost for treatments 3 and 4 were slightly lower than for treatments 1 and 2. Economic analysis showed that feeding fish the diet containing $28\%$ protein and 3.08 kcal/g DE to satiation and the diet containing $32\%$ protein and 3.08 kcal/g DE to $87.5\%$ of satiation produced the highest profit to farmer.

  • This study was conducted to estimate the optimum dietary DHA oil level and replacement level of enzyme treated fish meal (EFM) with sardine fish meal for juvenile Atlantic bluefin tuna Thunnus thynnus. Four diets were used: 1) EFM75 in which 75% EFM and 4% DHA oil were applied, 2) EFM60, with 60% EFM and 15% sardine fish meal, 3) DHA2 with 2% of DHA oil, and 4) SL as a raw fish feed. In a feeding trial, juvenile bluefin tuna (body weight 30.1 g) were randomly stocked into four experimental tanks (69 tones) and fed the experimental diets for 13 days. Fish weight gain was higher in the EFM75 and SL groups than in the DHA2 and EFM60 groups. The feed conversion ratio was lower in the EFM75 and DHA2 groups than in the EFM60 and SL groups. Survival was higher in fish fed the formulated diet groups (EFM75, EFM60 and DHA2) than in fish fed SL. This study clearly indicates that up to 10% dietary sardine fish meal can be used in juvenile T. thynnus diets, with an optimum dietary DHA oil level of approximately 3%.

  • This study was conducted to estimate the optimum dietary protein to energy (P/E) ratio in juvenile river puffer. Nine experimental diets were formulated with three energy levels and three protein levels at each energy levels. Three energy levels of 3000, 3750 and 4500 kcal diets were included at 45, 50 and 55% crude protein (CP) levels, respectively (45P3000, 45P3750, 45P4500, 50P3000, 50P3750, 50P4500, 55P3000, 55P3750 and 55P4500). Fish averaging $3.43{\pm}0.02$ g randomly were fed the experimental diets in triplicate groups for 8 weeks. Weight gain of fish fed the 50P4500 diet were significantly higher than that of fish fed the 45P3000, 45P3750, 45P4500, 50P3000 and 50P3750 diets (P<0.05), but there was not significantly different from that of fish fed the 50P4500, 55P3750 and 55P4500 diets. Feed efficiency of fish fed the 50P4500 diet were significantly higher than that of fish fed the 45P3000, 45P3750, 45P4500, 50P3000 and 50P3750 diets (P<0.05), but there was not significantly different from that of fish fed the 50P4500, 55P3000, 55P3750 and 55P4500 diets. Protein efficiency ratio of fish fed the 45P3000 and 45P3750 diets was higher than that of fish fed 50P4500 and 55P4500, but there was not significantly different from that of fish fed the 45P3000, 45P3750, 45P4500, 50P3000, 50P3750, 55P3000 and 55P3750 diets. Based on weight gain, feed efficiency and specific growth rate, diets containing energy levels 4500 kcal/kg diet had an optimum P/E ratio of approximately 111 mg protein/kcal (50% crude protein) in juvenile river puffer.

  • The chemical composition of fish feeds(mackerel, gizzard shad, shrimp and formula feed) were analyzed and compared with those of untreated and irradiated (5 kGy) feeds, and the effects of gamma irradiation (5 kGy) and butylated hydroxyanisole (BHA, 200ppm) on feeds spoilage were investigated by determination of thiobarbituric acid (TBA) value and peroxide value for 14 days at $20^{\circ}C$, $4^{\circ}C$ or $-20^{\circ}C$. Crude protein, crude lipid, crude fiber, crude ash and nitrogen free extract were not significantly changed by gamma irradiation with the dose of 5 kGy. Immediately gamma irradiation increased the initiative oxidation of feeds as expected. But BHA was found to be greatly effective in minimizing the radiation-induced peroxidation (p<0.05). The TBA value and peroxide value increased with the elapse of the storage period. The level of lipid oxidation was increased depending on the rise in storage temperature. But the additions of BHA inhibited the lipid oxidation (p<0.05).

  • The effect of feeding frequency on growth performance, body nutrient composition, and hematological parameters of juvenile chum salmon Oncorhynchus keta was investigated. The juveniles (10.9±0.3 g) were fed commercial feed for 30 days with feeding frequencies of 1, 2, 3, 4, or 5 meal (s)/day (n=2 tanks per treatment) in a recirculating aquaculture system (RAS). Fish were fed to satiation at each of the feeding frequencies. At the end of the feeding trial, final body weight, weight gain, and specific growth rate of fish fed 2 meals/day were significantly higher than those of fish fed 1 meal/day(P<0.05); however, no difference was detected among the other feeding frequencies. Daily feed intake significantly increased with increasing feeding frequency, whereas feed and protein utilization efficiencies continuously decreased with increase in the feeding frequency. Among the treatments, fish fed 1 meal/day showed the lowest value in whole-body lipid content and total protein concentration in plasma. Taken together, the optimal feeding frequency for growth performance and feed utilization efficiency in chum salmon juveniles reared in the RAS was determined to be 2 meals/day.

  • The aim of this study was to evaluate a new Bacillus amyloliquefaciens B-1895 probiotic as a feed additive for farmed trout. Final weight, absolute and average daily gain of fish, and average daily growth rate were higher in the group that received the probiotic than in the control group (p<0.05). Moreover, the probiotic-fed trout had more intense growth rates than the control group (higher by 15.7%; p<0.05). A decrease in feed ratio was also observed in the group that received probiotic (25% decrease; p<0.05), indicating more efficient digestion and assimilation of feed. In general, the introduction of probiotic in the feed did not adversely affect the functional status of the fish. In young trout of the control group, when assessing the general chemical composition of the organism in the muscle tissue revealed significantly (p≤0.001) higher level of moisture content by 5.1% and lower by 11.0% dry matter content. In muscle, the protein content was higher by 1.33% (p≤0.001) and fat content by 2.1% (p≤0.001) in experimental fish. Generally, Lactobacilli, Enterococcus, Vibrio, Bacillus, and coliform bacteria were found in the intestinal samples of rainbow trout. Significant reliable difference (p≤0.05) between the samples of experimental and control groups was noted in the content of Bacillus bacteria. In the control group, 5.0±0.4×103 CFU/g was detected, while in the experimental group 8.4±0.8×104 CFU/g. Overall, the data indicate that probiotic bacteria B. amyloliquefaciens B-1895 has no adverse effect on selected microorganisms in the study fish.

  • A four-week feeding trial was conducted to investigate the effects of dietary soybean meal (SBM) and powdered Cheongkukjang (CKJ) on non-specific immune responses of parrotfish Oplegnathus fasciatus. Three isonitrogenous (42% crude protein) and isocaloric (17.1 MJ/kg) diets were formulated to replace fish meal by 0, 25% SBM or 25% CKJ (designated as FM, 25SBM and 25CKJ, respectively). Ninety fish (initial body weight 122 g) were randomly allotted into nine 150 L tanks. One of the three experimental diets was fed to triplicate groups of fish for 4 weeks. After the feeding trial, no differences were observed in growth performances and feed utilization among fish groups. Liver superoxide dismutase activity of the fish fed CKJ containing diet was significantly higher than that of the control groups. DPPH radical scavenging and $Fe^{2+}-chelating$ activities of the experimental diets containing SBM or powdered CKJ were significantly higher than that of the control diet. The results of the present study suggest that dietary inclusion of powdered 25CKJ significantly increased liver superoxide dismutase activity and did not affect the growth performances, feed utilization, morphological parameters, as well as hematological values of parrotfish.

  • The lysine requirements of juvenile yellowtail flounder (Pleuronectes ferrugineus) having 19.5 g initial body weight were estimated by feeding six practical-type diets containing graded levels of lysine (1.21 to 2.69% of dry diet). Dietary amino acid profile simulated that of whole body of yellowtail flounder. Most of amino acids in the diets were provided by corn gluten meal, herring meal and gelatin. Protein efficiency ratio (PER) improved significantly until lysine level increased up to 2.1% (4.3% of protein). Same trend was observed in feed:gain ratio (FGR) which maintained constant in fish groups fed diets containing lysine above 2.1%. The highest nitrogen gain (0.34 g/fish) in whole body was found in fish fed 2.1% lysine, though the value was not different from those of fish fed above the level of lysine. Fish fed 2.1% lysine also showed the best nitrogen retention efficiency of 24.6%. The broken-line analysis of protein efficiency ratio and body nitrogen gain against dietary lysine level yielded an estimated lysine requirement of 2.2% (4.5% of protein) and 2.3% (4.7% of protein), respectively.
