Fluoroquinolones are the most common group of antibacterial agents currently used in the Korean aquaculture industry, and use of these agents has been increasing steadily. High performance liquid chromatography (HPLC) with fluorescence detection was used for the simultaneous determination of five fluoroquinolones in fish and shellfish: ofloxacin (OFL), pefloxacin (PEF), norfloxacin (NOR), ciprofloxacin (CIP), and enrofloxacin (ENRO). Fish and shellfish muscle was homogenized, and protein, lipid, and low molecular weight pigments were then excluded from the homogenate. The final eluates were analyzed by HPLC equipped with a Shiseido UG-120 type C18 reverse-phase column ($4.6{\times}250 mm$, $5{\mu}m$) and a fluorescence detector (excitation at 280 nm, emission at 450 nm). The mobile phase was 0.1 M phosphoric acid and acetonitrile solution (91:9, v/v) and tetrahydrofuran (THF) was added to it at a rate of 5 mL per a liter of the mobile phase. Adequate chromatography separation was obtained using the above method. Average recoveries of fortified samples at levels from 0.05 to 0.5 mg/kg were $72.3{\pm}2.5-84.5{\pm}1.2%$ for OFL, $82.7{\pm}3.3- 109.3{\pm}7.5%$ for NOR, $85.3{\pm}6.6-116.0{\pm}7.9%$ for PEF, $76.0{\pm}4.3-109.3{\pm}12.4%$ for CIP, and $78.7{\pm}5.9-100.0{\pm}9.8%$ for ENRO. The limit of detection of OFL was $5{\mu}g/L$, the others were $1{\mu}g/L$. We concluded that the new analytical method was suitable for the determination of fluoroquinolones in fish and shellfish.
The aim of this study was to acquire level on the occurrence of five fluoroquinolones(FQs) residue among cultured eels in Jeonbuk province. Samples were collected from the four local area(Kunsan, Gochang, Wanju and Osu) and surveyed on the distribution of FQs residual level in edible muscle using HPLC method. A total of 90 samples of eel was monitored for detection of the FQs residue. Among them, 30 eels(33.3%) were detected the FQs single or paired, respectively. The detection ratios were showed Osu(40%, 8/20), Gochang(33%, 13/44), Wanju(30%, 6/20) and Kunsan(30%, 3/10) in order, respectively. The ratio was equivalent to over 30% of overall in sampled area. The most frequently detected FQs were perfloxacin(PF) followed by enrofloxacin(EF), ciprofloxacin(CF), ofloxacin(OF) and norfloxacin(NF). Eight different combinations of FQ residual patterns were observed as single(5 type, 21%) or as paired(3type, 12,5%) respectively and frequently encountered patterns were PF(37%) and PF-EF(23%). The residual level of FQs was evaluated in the range of 1-75 ug/kg and the most prevalent levels of FQs was found in 1-25 ug/kg(35 fish, 85%) followed by 26-50 ug/kg(5 fishes, 11%) and 51-75 ug/kg(1 fishes, 3.3%). These results could be possible that the cultured eels were mostly exposed with PF, EF and CF, and might be treated with a variety of fluoroquinolones for prevention of bacterial disease.
Deep-sea fishery in the Antarctic Ocean has been actively progressed by the developed countries including Korea. In order to prevent the environmental destruction of the Antarctic Ocean, related countries have established the Commission for the Conservation of Antarctic Marine Living Resources (CCAMLR) and have monitored any illegal unreported or unregulated fishing. Fishing of tooth fish, an expensive fish, in the Antarctic Ocean has increased recently and high catches per unit effort (CPUE) of fishing boats, which is suspicious for an illegal activity, have been frequently reported. The data of CPUEs in a fishing area of the Antarctic Ocean often show an extreme Distribution or a mixture of two extreme distributions. This paper proposes an algorithm to detect an outlier of CPUEs by using the mixture of two extreme distributions. The parameters of the mixture distribution are estimated by the EM algorithm. Log likelihood value and posterior probabilities are used to detect an outlier. Experiments show that the proposed algorithm to detect outlier of the data can be adopted instead of simple criteria such as a CPUE is greater than 1.
In the face of escalating utilization of the marine spatial domain, conflicts have emerged among stakeholders, necessitating effective management strategies beyond conventional government permits and regulations. Particularly within the domain of aquaculture, operational oversight relies on a localized licensing system, posing challenges in accurately assessing the prevailing circumstances. This research employs synthetic aperture radar (SAR) imagery as a tool to monitor coastal aquaculture fish farms, aimed at enhancing insights into management protocols. Leveraging Sentinel-1A imagery and time series SAR data integration, a superimposition technique is utilized, facilitating noise reduction while retaining crucial information regarding smaller-scale facilities, such as fish farms. Through analysis of VH polarization data, a detection overall accuracy of approximately 88% for coastal fish farms was achieved. The findings of this study offer potential applications in the continuous monitoring of aquaculture farms in correspondence with seasonal variations in aquaculture yields, thereby proposing frameworks for the establishment of effective management cycles for marine space utilization.
Purpose : Fluorescence in situ hybridization (FISH) on uncultured amniotic fluid cells offers the opportunity for rapid screening of aneuploidies and has become an integral part of the current practice in many clinical cytogenetics laboratories. Here, we retrospectively analyzed the results of interphase FISH in 943 amniotic fluid samples and assessed the efficiency of FISH for rapid detection of aneuploidies. Methods : Interphase FISH for chromosome 13, 18, and 21 was performed in 943 consecutive amniotic fluid samples for rapid diagnosis of aneuploidies referred from 2004 to 2006. Karyotypes from standard cytogenetic analysis were compared to the FISH results. Results : A total of 45 chromosomal rearrangements (4.8%) were found after conventional cytogenetic analysis of the 943 amniotic fluid. After exclusion of known familiar chromosomal rearrangements and inversions (2.1%, 20/943), 2.7% (25/943) were found to have chromosomal abnormalities. Of this group, 0.7% (6/943) were chromosomal abnormalities not detectable by FISH and 2.0% (19/943) were numerical abnormalities detectable by FISH. All 14 cases of Down syndrome (Classic type, 13 cases; Robertsonian type, 1 case) and 5 cases of trisomy 18 were diagnosed and detected by FISH and there were no false-positive or -negative results (specificity and sensitivity=100%). Conclusion : The present study demonstrates that FISH can provide a rapid and sensitive clinical method for prenatal identification of chromosome aneuploidies. However, careful genetic counseling is essential to explain the limitations of FISH, including the inability to detect all chromosomal abnormalities and the possibilities of uninformative or false-negative results in some cases.
Ergasilus sieboldi, a parasitic Copepoda, was detected from the gill of Silurus asotus fingerling(7~10 cm long) cultured at a fish farm in Chungnam Province (Fig. 1 and 2). The infected fish did not show any particular external symptoms except a feeble swimming around the boarder of the pond. The isolated parasite was of 1.07 mm in body length with two egg sacks of 1.05 mm and one pair of hook-shaped secondary tentacle. Bychowsky(1963) described that this parasite infected the gill of Salmonidae, Serranidae, Cyprinidae and Siluridae and that the fish in the Baltic Sea, Black Sea. Caspian Sea. Japanese sea zones, Siberian rivers and Lake Bikal were infectd. Bychowsky further pointed out that heavily infected fish with this parasite become emaciated and slow in growth. and often die. As this parasite infected on the peri-branchial area. the infected fish showed a severe disturbance of respiration. Urawa et ai.(1980) studied on the development of Neoergasilus japonicus. an Ergasilidae, which infects Japanese freshwater fish. In Korea, Chun(1985) reported the identification of Ergasilus sieboldi from imported carp. and Neoergasilus and Pseudogasilus japonicus from bluegills. respectively. Chun also identified Pseudoergasilus zacconis in cultured ayu(Plecoglossus altivelis). However. this report deals with the detection of Ergasilus sieboldi from cultured catfish(Silurus asotus) for the first time in Korea.
Minimun-detectable levels to 28 antibacterial agents used for the prevention and the treatment of fish diseases were determined to establish optimal detective method of bioassay in fish by the EEC 4-plate method, the modified method of EEC 4-plate and the standard method of analysis in food safety regulation. The test organisms used in the methods of bioassay were as follows: Bacillus subtilis BGA (B. subtilis) and Micrococcus luteus ATCC 9341 (M. luteus) in the EEC 4-plate method, B. subtilis, M. luteus and Bacillus cereus var. mycoides ATCC 11778 (B. cereus) in the modified of EEC 4-plate, and B. subtilis, M. luteus, B. cereus and Bacillus stearothermophilis var. calidolactis C-953 (B. stearothermophilis) in the standard method. The standard method showed predominant sensitivity in the detection of penicillins (PCs), and was also highly sensitive to aminoglycosides (AGs). The sensitivity of standard method in the detection of tetracyclines (TCs), marrolides (MLs), nitrofuran derivatives(NFs) and quinolones (QNs) was very low, and against sulfonamides (SAs), however, was extremely low. The modified method of EEC 4-plate showed very high sensitivity to TCs. Both the EEC 4-plate and the modified method of EEC 4-plate showed competitively high sensitivity in the detection of PCs, MLs, NFs, QNs and SAs. All the methods studied in the experiment showed very low sensitivity against chloramphenicol (CMs). Consequently, the modified method of EEC 4-Plate was the best bioassay method with a wide range of sensitivity for the optimal detection of the residual antibacterial agents in fish.

In order to establish an effective management strategy for invasive species early detection and regular monitoring are required to assess their introduction or dispersal. Environmental DNA (eDNA) is actively applied to evaluate the fauna including the presence of invasive species as it has high detection sensitivity and can detect multiple species simultaneously. In Korea, the applicability evaluation of metabarcoding is being conducted mainly on fish, and research on other taxa is insufficient. Therefore, this study identified the feasibility of detecting invasive species in Korea using eDNA metabarcoding. In addition, to confirm the possibility of detection by taxa, the detection of target species was evaluated using four universal primers (MiFish, MiMammal, Mibird, Amp16S) designed for fish, mammals, birds, and amphibians. As a result, target species (Trachemys scripta, 3 sites; Cervus nippon, 3 sites; Micropterus salmoides, 7 sites; Rana catesbeiana, 4 sites) were detected in 17 of the total 55 sites. Even in the selection of dense sampling sites within the study area, there was a difference in the detection result by reflecting the ecological characteristics of the target species. A comparison of community structures (species richness, abundance and diversity) based on the presence of invasive species focused on M.salmoides and T.scripta, showed higher diversity at the point where invasive species were detected. Also, 1 to 4 more species were detected and abundance was also up to 1.7 times higher. The results of invasive species detection through metabarcoding and the comparison of community structures indicate that the accumulation of large amounts of monitoring data through eDNA can be efficiently utilized for multidimensional ecosystem evaluation. In addition, it suggested that eDNA can be used as major data for evaluation and prediction, such as tracking biological changes caused by artificial and natural factors and environmental impact assessment.
In the present study, liquid chromatography tandem-mass spectrometry was used to conduct a risk assessment of ethoxyquin and ethoxyquin dimer residues in 288 farmed fish sold in wholesale and traditional markets in Seoul. The detection range (detection rate) of ethoxyquin and ethoxyquin dimer was 0.005~0.309 mg/kg (4.9%) and 0.001~2.828 mg/kg (69.8%), respectively. Ethoxyquin was detected only in freshwater eels, loaches, catfish, and flatfish, whereas ethoxyquin dimer was present in all fish species. To estimate the dietary exposure levels of ethoxyquin and ethoxyquin dimer, the hazard index, calculated using the acceptable daily intake and estimated daily intake, ranged from 0.001 to 0.300%. Our results indicate that there is no significant exposure risk to humans from these farmed fish despite 10 farmed fish samples exceeding the maximum residue level (1.0 mg/kg as the sum of ethoxyquin and ethoxyquin dimer).

Several molecular biological techniques have been used to detect virus rapidly and accurately, but these methods have limitations in the early stage of viral infection with very low concentration of virus. We compared the detection limits of IMS-PCR, Western blot and ELISA with infectious hematopoietic necrosis virus OHNV). Four antibodies, rabbit anti-IHNV polyclonal antibody, anti-IHNV nucleocapsid protein monoclonal antibody, anti-IHNV nucleocapsid protein polyclonal antibody, and anti-IHNV glycoprotein polyclonal antibody, were tested to find out the most effective antibody for each method. The detection limit with IMS- PCR was $2\times10^6$ pfu when the viral RNA was extracted before RT-PCR. In the western blot with rabbit antiIHNV polyclonal antibody one pfu of virus could be detected. In ELISA, 10 pfu of virus particles were detected with the same antibody.
