Effect of Seed Leachates of 'Vernal' Alfalfa on Inhibition of Alfalfa Germination and Root Growth

  • Most parts of alfalfa plant have been reported to contain autotoxic substances that inhibit seed germination and early seedling growth, however, the chemical(s) is not still studied much. Effect of seed leachates of 'Vernal' alfalfa (Medicago sativa L.) was evaluated for inhibition of alfalfa germination and root growth through bioassay. Alfalfa seeds were extracted in 1 L deionized water for 1 h after soaking and the leachates caused to reduce root length of alfalfa significantly as the soaking time increased. Crude seeds at 4 g L$L^{-1}$ exudated autotoxins that reduce significantly root length by 34 % compared to the control, when the seeds soaked in deionized water for 24 h. However, the extracts did not affect final germination as well as speed of germination. Extracts from ground seeds significantly reduced speed of germination (GT 50) and root length. The results indicate that release of autotoxic substances from seeds during seed imbibition was increased with increase of soaking time and seed amount, and that autotoxicity was more occurred in ground seeds than in crude seeds.

ISTA 변온조건에서 퍼레니얼 라이그래스 신품종 8종류의 발아특성 및 일일 발아패턴 (Germination Characteristics and Daily Seed Germinating Pattern of 8 New Varieties of Perennial Ryegrass under Alternative Conditions Required by ISTA)

  • Research was initiated to investigate early establislment characteristics and germination pattern of perennial ryegrass (PR, Lolium perenne L.). Eight varieties of PR were evaluated in the study. An alternative environmental condition for a PR germination test required by International Seed Testing Association (ISTA) was applied in the experiment, consisting of 8-hr light at $25^{\circ}C$ and 16-hr dark at $15^{\circ}C$ (ISTA conditions). Daily and cumulative germination pattern were measured and analyzed daily for 30 days. Significant differences were observed in germination pattern, days to the first germination, days to 50% germination, days to 75% germination, and final germination rate. Germination rates were variable among the varieties, being 66.00 to 93.75% at the end of study. There were considerable variations in early germination characteristics and germination pattern among PR varieties. The first germination in all entries tested was initiated from 3 to 5 DAS (days after seeding) under ISTA conditions, being 2 days in differences among the varieties. It was 4 to 7 DAS in days to the 50% germination, being 1 to 3 days later from the first germination and three days were variable among them. Days to the 75% germination were 5.93 to 10.50 DAS under ISTA conditions, being 4.57 days in variety differences. Pennant II and Brightstar II were the fastest varieties. The slowest ones were Revenge GLX and Brightstar SLT. Considering days to the first germination, days to 75% germination, and germination pattern etc, Pennant II and Brightstar II were regarded as excellent varieties under ISTA conditions. From tills study, information on differences in germination characteristics and pattern would be usefully applied for golf course design and construction, when established with PR.

종자 발아율 지표 및 초기 묘조 성장에 의한 다환방향족탄화수소의 식물 독성 측정 (Phyto-toxicological Effects of PAHs on the Germination and Growth of Alfalfa, Barley, and Tall Fescue)

  • The test of germination has been used as a good indicator to assess the toxicity of chemicals to plant. This study was carried out to investigate the effect of PAHs, anthracene and phenanthrene, on germination ratio, germination index and germination time of barley, tall fescue and alfafa. Anthracene and phenathrene induced to decrease the seedling length and increase the germination time of the experimental plants. But they did not affect the maximum germination ratio. While it was not correlated between seedling length and final germination ratio, it was shown a close relationship between seedling length and mean germination time of the plants. These results suggested that the mean of germination time and seedling growth could be useful to exmine the phyto -toxicological effect of PAHs. The mixture of anthracene and phenanthrene was examined to investigate the combined effect on seedling length; the mixture induced the reduction of seedling. This was meant that the mixture toxicity of PAHs might be not shown in the sum of each chemical toxicity.

  • The effects of some chemicals for the germinative stimulation of Clostridium chauvoei spore studied. The results obtained were as follows: 1. Cooked meat medium (CMM) was superior to Liver-liver bouillon (LLB) for cultivation of the organisms. 2. Heating the organisms at $70^{\circ}C$ for 30 minutes prior to cultivation in CMM stimulated the germination rate. 3. The addition of 0.5mM L-alanine to CMM was found to be most effective for the rapid germination of the spores. 4. The addition of 0.2 mM D-alanine to CMM inhibited the germination of the spores even if the spores were heat treated. 5. The addition of 0.1mM manganese retarded the germination time, but the final germination rate was greater with 0.1mM manganese than with 0.5mM L-alanine. 6. The addition of both 0.5mM L-alanine and 0.1mM manganese stimulated the germination rate to 100 per cent. 7. The germination rate was greater with the addition of germination stimulants than without any stimulants. The germination rate was approximately 6 per cent greater with prior heat treatment than without heat treatment. 8. The optimum time for the harvest of vegetative forms of the organisms was 15 hours after cultivation in the media which contain suitable germination stimulants.

Enhancement of Seed Germination and Uniformity in Triploid Watermelon (Citrullus lanatus (Thunb.) Matsum. and Nakai)

  • One of the main factors restricting production of triploid seedless watermelon is poor germination due to weak embryos, thick seed coats, and larger air spaces. This study was carried out to investigate the priming effects of different concentrations of chemicals, including hydrogen peroxide ($H_2O_2$), fusicoccin, and gibberellic acid (GA) on germination and seedling uniformity of triploid watermelon (Citrullus lanatus). Three commercial triploid cultivars, Seedless Plus, Sinus, and Sizero, were pretreated with water and different levels of $H_2O_2$ (2 and 4%), fusicoccin (FC: 1, 5, and $10{\mu}M$), and GA (1, 5, and $10{\mu}M$). The present findings helped to find optimal priming conditions for improving germination of triploid watermelon. Treatment with $5{\mu}M$ GA and hydropriming helped to break seed dormancy, enhancing the final germination percentages in all triploid cultivars and increasing the germination index in Sizero. These seed-priming treatments could be used on large scale for industrial applications. Moreover, hydropriming provides a simple, effective, and costless method to improve seed germination and seedling vigor of Sinus and Sizero varieties.

봉래꼬리풀의 종자 발아에 미치는 온도 조건과 화학적 처리의 영향 (Effect of Temperature Conditions and Chemical Treatments on Seed Germination of Pseudolysimachion kiusianum var. diamantiacum (Nakai) T.Yamaz.)

  • 산림청 지정 희귀·특산식물 봉래꼬리풀(Pseudolysimachion kiusianum var. diamantiacum (Nakai) T.Yamaz.)을 대상으로 광조건, 온도와 전처리에 따른 발아 특성을 조사하였다. 광조건에 따른 발아실험결과, 봉래꼬리풀은 암조건에서 전혀 발아하지 않는 광발아성 종자로 판단되었다. 봉래꼬리풀의 최적 발아온도는 최종 발아율과 발아세를 고려하여 20-25℃임을 확인하였다. 봉래꼬리풀 종자의 발아율 향상을 위해 생장조절제 3종(IAA, GA3, kinetin)과 무기염류 2종(KNO3, KCl)을 전처리하였다. 생장조절제 IAA와 kinetin은 봉래꼬리풀 종자의 발아율 향상에 유의미한 영향을 미치지 못했다. 반면 GA3는 농도와 관계없이 최종 발아율과 발아세를 유의미한 수준으로 증가시켰고, 특히 20℃에서 500 mg·L-1 이상 처리한 것은 무처리구에 비해 4배 이상 효과적이었다. 무기염류 KNO3과 KCl는 저농도에서 봉래꼬리풀 종자에 유의미한 영향을 미치지 못했지만 고농도(각40 mM, 300 mM) 이상에서는 발아율과 발아세를 무처리구에 비해 1.5배 이상 향상시켰다. 본 연구의 결과는 복원용 자생식물로 활용 가치가 있는 봉래꼬리풀의 대량증식에 유용한 자료가 될 것이다.

탄닌 함유 사초식물의 발아 및 생육특성 (Germination and Growth Characteristics of Tannin Containing Forage Crops)

  • In an effort to investigate the germination and growth characteristics of tannin containing forage crops (such as chicory, birdsfoot trefoil, Lotus pedunculatus, sulla, lespedeza, crownvetch, sainfoin), germinativity, seedling vigor, and productivity were studied. Although the cumulative germination percentage (CGP) of chicory and birdsfoot trefoil was low, the germination rate(GR) and days to reach 50% of final germination(Gt50) were excellent. Crownvetch, lespedeza and sainfoin showed poor germination. Chicory seeds were light but the rate of emergence and establishment were high. Seeds of Lotus pedunculatus and birdsfoot trefoil were also small, but their rates of emergence and establishment were quite good. On the other hand, seeds of sainfoin and crownvetch were heavy but their emergence and establishment rate were very low. Lespedeza was the best in getting over the hot summer, and crownvetch also survived well during summer. Chicory was good in productivity and early growing but its persistence was not good. In case of birdsfoot trefoil and Lotus pedunculatus their early growing were slow but their productivity and persistence were fairly well. Lespedeza, crownvetch, and sainfoin showed poor growth in Korea. Based on the results of this study it can be concluded that chicory would be better to use as short term high productive forage crop, and Lotus pedunculatus and birdsfoot trefoil could be used as continuous functional forage crops in Korea. 分布地域에 따른 지침개 ( Hemistepta lyrata Bunge ) 個體群의 發芽 習性 및 幼植物 生長의 地理的 變異 (Geographical variations in the seed germination response and the seedling growth of hemistepta lyrata bunge by distrbution areas)

  • The geographical variation of the germination response and the seedling growth of the seed populations of hemistepta lyrata bunge distributed in the southern area of korean peninsula (seoul, yongwol, andong, $Ch^{\prime}angwon, \;Sunch^{\prime}on$)was investigated. Five populations were divided into two principal groups according to the phases of their seed germination. The first group consisting of the seoul and yongwol population was 62% and 64%, respectively, in the final germination percentage of 32%, 24% and 28%, respectively, were Andong, $Ch^{\prime}angwon\;and\;Sunch^{\prime}on$ populations. The seed populations of hemistepta lyrata bunge which hardly germination. the germination of seoul and yongwol populations located at the higher latitudes took plase synchronously in early autumn. On the other hand, andong, $Ch^{\prime}angwon\;and\;Sunch^{\prime}on$ populations located at the lower latitudes showed a tendency to germinata asynchronously in late autumn. the speed of the seedling growth of the populations located at the higher latitudes was greater than that of the populations at the lower latitudes. Therefore the geographical variation in the germination response and the seedling growth of the seed populations of hemistepta lyrate bunge appeared to be an important ecological strategy to maintain their existence in the extreme environmental variations.

Physicochemical and textural properties of germinated brown rice according to rice varieties

  • Germination is one of the techniques used to enhance the texture properties and nutritional value of the brown rice (BR). Therefore, germinated BR (GBR) has received significant attention during the last decade. Physicochemical and cooking properties of brown rice were examined before and after germination. Germination raised the cooking properties, such as water absorption, expanded volume and soluble solid of cooked BR (brown rice). The texture, measured using tensipresser, was significantly improved by germination. The hardness of cooked BR was decreased by germination, but the GBR was sticker. In RVA, all viscosity value (peak viscosity, break down, set back, and final viscosity) of germinated rice flour was also reduced while gelatinization temperature did not change. Amylose content and amylopectin chain length distribution of BR starch were slightly changed by germination. Overall results revealed that germination was an effective tool to improve texture and cooking properties of BR.

Seed Germination Response to Temperature, Cold Stratification Period, and Gibberellin Treatment in Spiraea fritschiana

  • To improve the germination of Spiraea fritschiana seeds for mass propagation, we evaluated the effect of a range of temperatures, cold stratification periods, and gibberellic acid ($GA_3$) treatments on three germination characteristics. Final germination percentage (FGP) increased as the temperature for seed germination increased, up to $30^{\circ}C$, while the mean germination time (MGT) and the mean number of days to 30% germination ($T_{30}$) decreased when seeds were germinated at $25-30^{\circ}C$. The optimum germination temperature of S. fritschiana seeds is approximately $30^{\circ}C$ considering FGP, MGT, and $T_{30}$ together. FGP increased with the duration of cold stratification up to a period of 6 weeks, but declined after 8 weeks of cold stratification, as prolonged cold stratification can induce dormancy with a resultant decline in germination. Pretreatment with 6-8 weeks of cold stratification or soaking seeds in distilled water or $500mg{\cdot}L^{-1}$ $GA_3$ for 24 h accelerated and increased the germination of S. fritschiana seeds, regardless of temperature. However, further study might be required to evaluate the effect of $GA_3$ concentrations lower than $500mg{\cdot}L^{-1}$ on the promotion of germination in S. fritschiana seeds.
