근권미생물과 토양병방제 -유용길항균이 인삼근부병원에 미치는 영향- (Establishment of rhizosphere microbes for plant protection on soil-borne diseases -Benificial antagonist and its mode of action toward ginseng root rot pathogen-)

  • 토양으로부터 분리한 380개체의 균주 중 Fusarium solani에 대해 길항능력이 있는 방선균 및 세균 42개 균주로부터 길항작용이 큰 균주로 CHA 1과 5-PFHR 6을 공시하여 동정한 결과 각각 Promicromonospora sp.와 Pseudomonas pseudoalcaligenes로 동정되었다. PDA 평판배지상에서 두 균주는 모두 F. solani의 저해작용으로 균사의 길이 생장억제, 비정상적인 균사의 분지, 세포벽 분해에 의한 돌기형성 등이 관찰되었다. F. solani 포자발아에 미치는 길항균주의 배양여과액의 작용으로 감자즙액 배지나 nutrient broth에 배양한 여과액은 포자발아 억제력이 높아 14.3%의 낮은 발아율을 보였으며, 토양침출액 단독 또는 두 영양배지에 토양침출액을 첨가하여 준비한 배양 여과액에서의 발아율은 85% 이상으로 포자발아가 촉진되었으나 S-PFHR 6의 경우 두 영양배지에 소량의 토양침출액을 첨가한 배앙여과액에서 4%의 발아율을 보였다. 자연토양에서 대형포자는 19.4%, 후막포자는 17.7%가 발아하였으나 증기살균 토양에서의 발아율은 79.7% 이상으로 증가하였다. 자연토양에 공시길항균 CHA 1과 S-PFHR 6의 균체를 처리하면 토양 정균력이 증가되어 F. solani의 대형 포자 발아율은 각 처리구에서 각각 14.7%, 11.7%로 감소하였다. 길항균을 처리한 토양에 glucose와 asparagine을 처리하면 토양의 정균력이 점차 해소되어 대형포자의 발아율은 48.0% 이상으로 증가하였다. 공시균주의 2차 대사산물에 의한 길항작용으로 두 균주의 배양여과액을 토양에 처리한 경우 CHA 1의 배앙여과액을 처리한 토양에서의 F. solani 대형포자의 발아율은 9.3%, 5-PFHR 6 처리구에서의 발아율은 38.0%가 되었다. F. solani의 후막포자는 공시길한균주의 균체나 배양여과액 처리구에서의 발아율이 자연토양에서의 발아율보다 낮았으며 이러한 현상은 영앙원을 첨가해 주어도 변하지 않았다.

  • 수종의 식물병원균(흰비단병균, 균핵병균, 좀검은균 핵병균)이 생산하는 가수분해산소(Cellulase Cx, Invertase, $\beta-amylase$, Xylanase, PMG, PG, Phosphatase, Protease)를 균계체내의 효소 및 배양여액내의 효소와 균핵내의 효소(좀검은균핵병균은 제외)로 나누어 그의 생산량과 pH에 따른 활성의 변화를 검토한 결과를 요약하면 다음과 같다. 1. 배양 10일후의 Cx활성은 균핵병균이 다른 균에 비하여 활성이 높았고 흰비단병균과 균핵병균은 산성측(pH 3.0 부근)에서, 좀검은균핵병균은 중성측(pH6.0 부근)에서 활성이 높았다. 2. 흰비단병균의 Invertase는 다른균에 비하여 약 20배정도 높은 활성을 보였고 3균주 모드 배양여액과 균계체간에 효소활성의 차가 인정되지 않았다. 3. Xylanase의 활성은 3균주 모두 균계체, 균핵 및 배양여액에 따라 또 pH에 따라 아주 다양한 변화를 나타냈고 균핵내에서 활성이 높았다. 4. $\beta-amylase$의 활성은 공시균중 좀검은균핵병균의 균계체, 배양여액이 가장 높았다. (약 12.0ug/min) 흰비단병균과 균핵병균ㅇ서는 균계체나 균핵에서 보다 배양여액에서 높았는데 활성최적pH는 균계체, 균핵 모두 pH 6.2였으나 배양여액에서는 흰비단 병균과 균핵병균이 pH3.0이었는데 반해 좀검은균핵병균은 pH6.2였다. 5. PMG의 활성은 공시균 모두 배양여액에서 높았고 균계체에서는 균핵병균과 좀검은균핵병균이 높았으며 활성최적 pH는 균에 따라 또는 측정부분에 따라 다양하게 나타났다. 6. PG의 활성은 흰비단 병균과 균핵병균의 균계체에서 각각 9,1ug/min. 9.5ug/min으로서 가장 높았고 활성최적 pH는 흰비단병균이 pH4.5부근 균핵병균이 pH.3.0부근이었다. 7. 흰비단병균과 균핵병균의 Phosphatase는 산성측(최적 pH3.5)에서 활성이 높았고 좀검은 균핵 병균은 산성, 중성, 알카리측에서 각각 Peak가 나타났으나 최적 pH는 9.5였다. 8. 공시균주 모두 Protease는 pH 10.0에서 최고활성을 나타냈고 특히 좀검은균핵 병균의 배양여액내 효소활성이 높았다.

Purification During Crossflow Electromicrofiltration of Fermentation Broth

  • The present study was to investigate the purification of a fermentation broth by an electromicrofiltration membrane. Microfiltration runs with a crude and a centrifuged broth, with solution of particles recovered from centrifugation and with permeates from microfiltration experiments were thus compared. Microfiltration performances were governed by colloids and small particles that induced sharp initial flux declines. For these results, the evolution of the overall membrane resistance was increased by $80\%$ in comparison with the electromicrofiltration membrane. The main focus of this study was set on the enhancement of the filtrate flux by an electric field. This pressure electrofiltration leads to a drastic improvement of the filtration by $100\%$ and the filtration time was thereby reduced. Pressure electrofiltration serves as an inter­esting alternative to the cross-flow filtration and it effectively separates advantageous constitu­ents such as amino acids and biopolymers from a fermentation broth. They were equally main­tained during the microelectrofiltration, although they were significantly reduced by $45\%$ by the microfiltration without the application of an electric field. Accordingly, since the electrofiltration membrane was provided more permeability, this study experimentally demonstrates that the permeability inside a membrane can be controlled using an electric field.

방선균이 생산하는 아미노산 대사길항물질, YS-460의 분리 정제 및 특성

  • An amino acid antimetabolite named YS-460 was isolated from the culture filtrate of a newly isolated Actinomycetes identified as Streptomyces sp. Fermentation was carried out in the synthetic medium at 30$\circ$C for 5 days. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 38 mg of pure active substance per liter of the broth. YS-460, an amino acid like substance, has the molecular formula of C$_{7}$H$_{11}$NO$_{3}$- Its structure determined to be furanomycin by spectral analysis. It is active against some bacteria on a chemically defined medium and reversed competitively by L-isoleucine and non-competitively by L-leucine and L-valine.

Micromonospora sp. SA-246 균주가 생산하는 Isochromanequinone계 항생물질

  • In the course of screening for new bioactive compounds from oligotrophs in soil, a microorganism, designated as SA-246 and now identified as Micromonospora sp., has been shown to produce a strong antibacterial compound. The active compound was purified from broth filtrate by ethylacetate extraction, silica gel column chromatography, preparative TLC and HPLC, and was identified as crisamicin A based on mass and NMR spectral data. The compound SA- 246 exhibited not only strong antibacterial activity against Gram-positive bacteria but also cytotoxicity against cancer cell lines such as A549 (lung), SK-OV-3 (ovarian), SK-MEL-2 (melanoma), XF498 (central nervous system) and HCT15 (colon).

Enterobacter cloacae MG82에 의한Triphenylmethane흡수 특성과 탈색효소의 세포내 위치

  • Triphenylmethane was decolorized rapidly by enterbacter cloacae MG 82 at initial reaction time. The spheroplast showed higher activity of triphenylmentane decolorization than that of intact cell suspension. The outer part of the bacterial cell envelope and the peptidoglycan are important for the function of transport barrier of triphenylmethane. In intact cell, decolorization activity was higher at 37$\circ $C than at $\circ $C, indicating that triphenylmethane decolorization is due to the enzyme reaction. Culture filtrate showed no decolorization activity, while cell-free extract appeared high activity of 1.45 units, clearly showing that decolorization activity was due to the cell-free extract. Comparing decolorization activities of cell fractions, it was found that decolorization activity was located at the compartment of cytoplasmic membrane. The enzyme activity was also shown to be Mg$^{++}$-dependent. The optimum pH and temperature of enzyme activity were 7.0 and 50$\circ $C, respectively. The thermostability of this enzyme at 35$\circ $C was kept to 58% for 3 hours.

  • With the introduction of high-tech industries, these days textile products used as advanced materials of semiconductor, transport and medical fields. It was concerned with environmental problem in textile industry lately. So the problem is receiving careful study for high effectiveness and high efficiency of filter media made by nonwoven fabric to get rid of a pollutant.   • 1. 5% 밀기울 기본배지에다 inducer로서 CMC 혹은 Avicel을 첨가하여 Myriococcum albomyces를 접종하여 진탕배양할 때의 cellulase 유도 생성을 검토하였다. 2. DEAE-Sephadex A-25 column chromatography 법으로 배양액에서 cellulase 활성이 다른 3개의 부분, fraction I, fraction II 와 fraction III를 분리하였다. 3. Inducer로서 CMC첨가의 경우는 CMCase가 강해지고 반대로 Avicel을 가한 경우는 Avicelase가 강하였다.

  • Glucoamylase from the culture filtrate of Aspergillus nicer was purified by ammonium sulfate precipitation, aceton precipitation, DEAE-cellulose ion exchange chromatography and Sephadex G-50 gel fillration. Glucoamylase was secreted into the medium upon growth on glucose, sucrose or a variety of other hexose sugars or hexose sugar polymers and little or no glucoamylase activity was found when glycerol or xylose was used as the carbon source. The optimum pH and temperature (or the maximum enzyme activity were found to be 5.0 and $50^{\circ}C$, respectively. The enzyme was considerably thermostable, for no loss of activity was observed when the enzyme was preincubated at $60^{\circ}C$ for 30 min. The enzyme activity was inhibited by 20 mM of $Hg^{2+}$, $Fe^{2+}$. The km value for starch was 0.045%.
