Effect of Growth Temperature and Nutritional Components on the Synthesis of Poly-3-Hydroxybutyric Acid by Filamentation-Suppressed Recombinant Escherichaia coli (Filamentation이 억제된 재조합 대장균에 의한 Poly-3-Hydroxybutyric Acid 합성시 배양온도와 영양분의 영향)

  • The effects of growth temperature and nutritional components on the synthesis of poly-3-hydroxybutyric acid, PHB, by filamentation-suppressed recombinant Escherichia coli XL1-Blue (pSYL107) were studied. After culturing XL1-Blue(pSYL107) for 48 hours in complex medium at 30$\circ$C, 7Al g/l of PHB could be obtained with the PHB content and PHB yield of 82% and 0.371 g PHB/g glucose, respectively. Lower concentration of PHB(3.2 g/l) was obtained when cultu- red at 37$\circ$C, which seemed to be due to the instability of this strain having amplified FtsZ activity. The PHB concentration of 3.75 g/l was obtained after culturing 60 hours in R medium supplemen- ted with 20 g/l glucose at 30$\circ$C, which was more than twice higher than that obtained with XL1-Blue(pSYL105). This suggested that the enhancement of PHB synthesis by suppressing filamenta- tion was more significant in a defined medium than complex medium. PHB synthesis could be further enhanced by supplementing a small amount of various complex nitrogen sources. When 5 g/l of beef extract was added to a defined medium, PHB concentration, PHB content, and PHB yield obtained after 60 hours of cultivation at 30$\circ$C were 7.46 g/l, 86%, and 0.375 g PHB/g glucose,respectively.

Roles of Zinc-responsive Transcription Factor Csr1 in Filamentous Growth of the Pathogenic Yeast Candida albicans

  • In the fungal pathogen Candida albicans, the yeast-to-hyphal transition occurs in response to a broad range of environmental stimuli and is considered to be a major virulence factor. To address whether the zinc homeostasis affects the growth or pathogenicity of C. albicans, we functionally characterized the zinc-finger protein Csr1 during filamentation. The deduced amino acid sequence of Csr1 showed a 49% similarity to the zinc-specific transcription factor, Zap1 of Saccharomyces cerevisiae. Sequential disruptions of CSR1 were carried out in diploid C. albicans. The csr1/csr1 mutant strain showed severe growth defects under zinc-limited growth conditions and the filamentation defect under hypha-inducing media. The colony morphology and the germ-tube formation were significantly affected by the csr1 mutation. The expression of the hyphae-specific gene HWP1 was also impaired in csr1/csr1 cells. The C. albicans homologs of ZRTl and ZRT2, which are zinc-transporter genes in S. cerevisiae, were isolated. High-copy number plasmids of these genes suppressed the filamentation defect of the csr1/csr1 mutant strain. We propose that the filamentation phenotype of C. albicans is closely associated with the zinc homeostasis in the cells and that Csr1 plays a critical role in this regulation.

Filamentous growth of Escherichia coli by dephosphorylated NPr (탈인산화된 NPr에 의한 대장균의 섬유상 생장)

  • The nitrogen phosphotransferase (PTS) system is a regulatory cascade present in most Proteobacteria, where it controls different functions. The nitrogen PTS is usually composed of $EI^{Ntr}$ (encoded by the ptsP gene), NPr (encoded by the ptsO gene), and $EIIA^{Ntr}$ (encoded by the ptsN gene). While $EIIA^{Ntr}$ plays a role in a variety of cellular processes, such as potassium homeostasis, regulation of ppGpp accumulation, nitrogen and carbon metabolisms, and regulation of ABC transporters, little information is available for a physiological role of NPr. A recent study showed that dephosphorylated NPr affects adaptation to envelope stresses in Escherichia coli. In this study, we provide another phenotype related to NPr. The ptsP mutant showed a filamentation phenotype. The filamentation phenotype of the ptsP mutant was recovered by additional deletion of the ptsO gene, but not by additional deletion of the ptsN gene, suggesting that an increased level of dephosphorylated NPr in the ptsP mutant renders cells the filamentous growth. This idea was confirmed by the fact that cells with increased levels of dephosphorylated NPr shows the filamentation phenotype. Additionally, we showed that cell size of E. coli increases with incremental dephosphorylated NPr concentrations. These results suggested that dephosphorylated NPr induces morphological change of E. coli.

  • 자체집광(Self-focusing)현상은 매우 높은 집적도를 가지는 광기록매체의 제작에 응용 가능하기 때문에 많은 연구자들에 의해 광범위하게 연구되어왔다. 그동안의 연구에서는 자체집광에 대한 분석방법으로서 빛살의 벡터특성을 무시한 비선형 슈뢰딩거 방정식(nonlinear Schr dinger equation)을 이용하였는데, 이 경우 축상으로 대칭적인 빛살이 입사될 때 발생되는 다중 필라멘테이션(Multiple Filamentation)현상은 입사빛살에 첨가되는 random noise에 기인한다는 것이 유일한 해석방법이었다.   • 1.55 ${\mu}m$multi-quantum well (MQW) broad area laser diodes with different linewidth enhancement factors ($\alpha{-factor}$) of 2 and 4 were fabricated. The far-fields of the laser diodes were measured. It was observed that the full width at half maximum (FWHM) of the far-fields and the filamentations were reduced in the laser diodes for which the value of the $\alpha{-factor}$ was small. As injection current increased, the FWHM of the far-field also increased regardless of the a-factor. This phenomenon was explained by reduction of filament spacing as injection current increased.

  • We have successfully formed filament inside of a transparent soda-lime glass using a Ti:sapphire based femtosecond laser. To make filament form, keeping the laser intensity higher than critical intensity is essential. Also each of the machining parameters plays an important role for the formation of filament. In this paper, we study what parameter can possibly influence for formation of filament, and we introduce an application using filamentation by femtosecond laser for transparent material.

Production of Poly(3-hydroxybutyrate) [P(3HB)] with High P(3HB) Content by Recombinant Escherichia coli Harboring the Alcaligenes latus P(3HB) Biosynthesis Genes and the E. coli ftsZ Gene

  • Filamentation-suppressed recombinant Escherichia coli strain harboring the Alcaligenes latus polyhydroxyalkanoate (PHA) biosynthesis genes and the E. coli ftsZ gene was constructed and cultivated for the production of poly(3-hydroxybutyrate) [P(3HB)] with high concentration and high content. By the pH-stat fed-batch culture of this recombinant E. coli strain XL1-Blue(pJC5), the final cell concentration and P(3HB) concentration obtained in 44.25h were 172.2g cell dry weight/l and 141.9g P(3HB)/l, respectively, resulting in productivity of 3.21g P(3HB)/l-h. More importantly, the P(3HB) content obtained was 82.4 wt %, which was significantly higher than that obtained with the recombinant E. coli harboring only the PHA biosynthesis genes.
