Screening of Seed Treatment Fungicide for Control of Damping-off caused by Rhizoctonia solani on Panax ginseng (Rhizoctonia solani에 의한 인삼 모잘록병 방제용 종자처리 약제 선발)

  • Damping-off is a critical disease on ginseng seedling, which caused by the fungal pathogen Rhizoctonia solani. The disease has been prevented by tolclofos-methyl for the last 20 years. However, the tolclofos-methyl usually detected on the harvested roots of 6-year-old ginseng. Herein, we tried to select an alternative pesticide which not only must be safe but also efficiently inhibits the fungal pathogen. Four fungicides (fludioxonil, flutolanil, pencycuron, and thifluzamide) were applied to their inhibition efficacy against the pathogen. In in vitro test, fludioxonil treatment showed 80% inhibition activity for 25 days. Thifluzamide and flutolanil showed the activity for 10 days. Pencycuron showed for 1 days. In addition, the fludioxonil was more effective to control the pathogen comparing to other three fungicides in field. The incidence of damping-off was reduced to 71% by fludioxonil treatment. The level of the fungicide residue in seedling was 0.44 mg/kg, which value will be a negligible level in final products after 5 years. Consequently, the fludioxonil is a conceivable alternative for tolclofos-methyl to cope with R. solani.

Development and Evaluation of a Patient-Reported Outcome (PRO) Scale for Breast Cancer

  • Background: This study was guided by principles of the theoretical system of evidence-based medicine. In particular, when searching for evidence of breast cancer, a measuring scale is an instrument for evaluating curative effects in accordance with the laws and characteristics of medicine and exploring the establishment of a system for medically assessing curative effects. At present, there exist few tools for evaluating curative effects. Patient-reported outcomes (PROs) refer to outcomes directly reported by patients (without input or explanations from doctors or other intermediaries) with respect to all aspects of their health. Data obtained from PROs provide evidence of treatment effects. Materials and Methods: In accordance with the tenets of theoretical medicine and ancient medical theory regarding breast cancer, principles for developing a PRO scale were established, and a theoretical model was developed and a literature review was performed, items from this pool were combined and split, and an initial scale was constructed. After a pilot survey and additional modifications, a pre-questionnaire scale was formed and used in a field investigation. After the application of statistical methods, the item pool was used to create a formal scale. The reliability, validity and feasibility of this formal scale were then assessed. Results: In a clinical investigation, 479 responses were recovered, with an acceptance rate of 95%. a combination of various methods was employed, and the items that were selected by all methods or more than half of the methods were employed in the questionnaire. In these cases, the screening methods were combined with certain features of the item, A total of four domains and 38 items were reserved. The reliability analysis indicated that the PRO scale was relatively reliable. Conclusions: Scientific assessment proved that the proposed scale exhibited good reliability and validity. This scale was readily accepted and could be used to assess the curative effects of medical therapy. However, given the limited scope of this investigation, the capacity for adapting this scale to incorporate other theories could not be determined.

Screening of Fungicide Resistance of Cucumber Powdery Mildew Pathogen, Sphaerotheca fusca in Gyeonggi Province (경기 지역 오이 흰가루병균(Sphaerotheca fusca)의 살균제 저항성 검정)

  • Fungicide resistance of cucumber powdery mildew was screened among the pathogens isolated from diseased plants in main cucumber productuion areas in Gyeonggi Province. Each fungicide from different activity group for the control of powdery mildew were sprayed on cucumber leaves according to application concentration. Each conidia mixed with sterilized water isolated of pathogens were transferred on the cucumber leaf disks treated with each fungicide. At 7 to 9 days after inoculation of pathogen, disease severity was recorded under the microscope. Most of pathogen isolates showed moderate resistance to difenoconazole belonged to DMI group fungicide while some isolates from Osan were resistant even $300{\mu}g/ml$. Isolates from Pyeongtaek, Osan and Yongin area also showed moderate resistance to fenarimol while one isolate showed resistant to fenarimol even $300{\mu}g/ml$. Most of isolates from Pyeongtaek, Osan and Yongin showed highly resistant to azoxystrobin belonged to strobilurin group fungicide. Standard sensitive isolates the minimum inhibition concentration(MIC) value for azoxystrobin showed $200{\mu}g/ml$ while resistance isolates showed above $2000{\mu}g/ml$. Resistant isloates also showed cross resistance among strobilurin group fungicides and low control efficacy in the field test. These results suggest that treatment of strobilurin fungicides should be reduced for the control of powdery mildew.

Analysis on the Risk-Based Screening Levels Determined by Various Risk Assessment Tools (II): Derivation of Particulate Emission Factor at Former Janghang Smelter Site (다양한 위해성평가 방법에 따라 도출한 토양오염 판정기준의 차이에 관한 연구 (II): (구)장항제련소부지의 기상 및 부지 특성을 반영한 비산계수 결정)

  • This paper presents the short-term and long-term measures to determine the fugitive dust concentration in a contaminated site, which is a crucial step for the determination of particulate emission factor (PEF) for risk assessment. As a long-term measure, USEPA method employing Q/C value (inverse of the ratio of the geometric mean air concentration to the emission flux at center of a 0.5-acre square source) seems to be suitable as it reflects regional-specific meteorological conditions. However, it requires nation-wide database collection and interpretation. Use of ASTM method is an alternative as a short-term measure. The method is readily field-applicable as PEF calculation equation is simple and input parameters can be easily derived at the site of interest as well without the nation-wide efforts. Using ASTM method, PEF at the Former Janghang Smelter Site was determined. According to various mode of aggregate size distribution and fractions of vegetative cover, which are the most important factors in PEF calculation, PEF values at the Former Janghang Smelter Site varied greatly. When the mode of aggregate size distribution was set at 0.25 mm, PEF values at the Former Janghang Smelter Site was 5~20 times higher than the default PEF value (i.e., 35 ${\mu}g/m^3$) shown in the current Korean Soil Contamination Risk Assessment Guidance. On contrast, when the mode was set at 2 mm, PEF values at the Former Janghang Smelter Site was 160~640 times lower than the default PEF value in the Korean Guidance.

Antifungal Activity of Bacillus vallismortis 1A against Phytopathogen (식물병원균에 대한 Bacillus vallismortis 1A 균주의 항진균 활성)

  • In order to isolate novel oligotrophic bacteria exhibiting antifungal activities, soils were collected from pepper-cultivated fields of Yeongyang, Jecheon, Nonsan, Eumsong and Goesan area in Korea. From soils in pepper cultivated area, a total of 9,354 strains were isolated as oligotrophic bacteria by the R2A dilution method. Among 9,354 oligotrohic bacteria candidates, 1A strain was selected by screening against Phytophthora capsici causing phytophthora blight of hot pepper in the greenhouse and field. The strain was identified as Bacillus vallismortis based on its 16S rDNA sequence and key characteristics as compared with those of authentic cultures of B. vallismortis(KACC 12149) and B. mojavensis(KACC 12096). The strain showed broad spectrum of antibiotic activity in vitro test, as revealed in its strong inhibitory activity to the genera Phytophthora, Collectotrichum, Botrytis and Fusarium, but not to Rhizoctonia and Magnaporthe. In pot experiments, infection rate of hot pepper in the non-treated pots was about 89%, while it was only 29% in the pots treated with 1A strain. The result indicated B. vallismortis 1A is a potential biocontrol agent for phytophthora blight of hot pepper

Collection of Soil Actinomycetes from Cheju Island and Screening for their Antibacterial Activities (제주도 토양방선균의 수집과 항균물질 생산균의 선별)

  • Soil actinomycetes of 703 strains were isolated from 25 sampling points in Cheju Island using 4 different media. Arginine glycerol salts agar containing soil extract was found to be the best medium for the isolation of soil actinomycetes. Soil samples from pasture land showed higher population and diversity of the actinomycetes than those from citrus field, forest, island, hill or valley. When the antibacterial activity of the 526 isolates was tested against three bacterial strains, Escherichia coli, Staphylococcus aureus and Pseudomonas solanacearum the frequency of the isolates with antibacterial activity varied much depending upon the media used for isolation and cultivation. BL106Ba, one of the 10 isolates that showed antibacterial activity against all the above 3 test strains, was chosen based upon the pH and heat stability of its antibacterial metabolites, and was identified as Streptomyces sp. based upon its cultural, morphological and physiological characteristics. The partially purified white crystalline substance obtained from the culture supematant of BL1063a through cation exchange chromatography(AG MP-50) and three times consecutive gel filtration(Sephadex G-10) showed high antimicrobial activity against gram positive and negative bacteria, but low activity against yeasts. The partially purified substance was found to contain at least four different compounds with antibacterial activity by both thin layer chromatography and high performance liquid chromatography.

Assessing Frogeye Leaf Spot Resistance on Recommended Soybean Cultivars (콩 주요 품종에 대한 점무늬병 저항성 평가)

  • Soybean frogeye leaf spot caused by the fungus Cercospora sojina Hara, has known to lead a severe reduction of crop yield. Since frogeye leaf spot on soybean has recently become a serious problem in Korea, the susceptibility of recent recommended cultivars against C. sojina had been tested. To standardize the disease severity of soybean, the optimum sporulation condition of C. sojina and the disease index were established in this study. Sporulation was maximized on the 10% V8 juice agar with 12 h light and 12 h dark at $25^{\circ}C$. Spore suspension ($10^5spores/ml$) was sprayed on the leaves of soybean (V6 stage), and the disease responses to each isolate were evaluated on 28 days after inoculation. As a result, Daepung, Shinpaldal2ho, Yeonpung and Cheonga showed the resistance reaction to 8, 7, 6, 6 isolates of C. sojina, respectively, whereas Cheongja, Hwangkeum, Taekwang, Daewon, Cheonsang and Sinhwa showed the susceptible reaction to 8 isolates of C. sojina. Breeding the resistant soybean cultivars against C. sojina requires a uniform resistance for screening technique. The disease index of frogeye leaf spot on soybean developed in this study can be effectively used for the accurate field assay to select the frogeye leaf spot resistant soybean.

  • Korean ginseng(Panax gmseng C.A. Meyer) is very difficult to obtain stable production of qualified ginseng roots because of variable stresses in soil environments. In environment stresses, soil condition is the most important factor, among which nutrients, especially inorganic materials such as N, P, K, Ca, Mg, Fe, etc., influence greatly on the ginseng growth. However, present ginseng field soils in Korea contain so much amount of such inorganic materials that a variety of remarkable disorders were noted in many ginseng plantations, resulting in decrease of qualitative ginseng production. Therefore, it is required to search for genetic resources and genes tolerant to salt stress for the development of ginseng cultivars. Selection of stress-tolerant ginseng lines in fields is very difficult because it is almost impossible to control properly the environmental conditions of soil. On the contrary, it can be studied with ease to search for stress-tolerant ginseng lines through in vitro culture because of easy manipulation of stress conditions. Murashige & Skoog(MS) media with 2.5 folds of $KNO_3,\;NH_4NO_3,\;MgSO_4\;7H_2O,\;KH_2PO_4,\;and\;CaCl_2\;2H_2O$ was established for the selection of ginseng lines tolerant to salt stress under the embryo culture.

Analysis of the Structural Safety of a Wind-Protecting Wall Using ANSYS/CFX (ANSYS와 CFX를 이용한 방풍벽의 구조 안전성 분석)

  • This study was carried out to evaluate the structural safety fur both the attached wind-protecting wall in greenhouse and the detached one installed outside. Regarding the attached wind-protecting wall in greenhouse, the analysis was conducted by doing a fluid-structure coupled field analysis using both CFX-5.7 and ANSYS 8.1 and also under the design condition of an instantaneous maximum wind velocity of $30.9m{\cdot}s^{-1}$. Three kinds of the width ranged from 30 to 90cm were considered in this study. With regard to the detached wind-protecting wall, the structural saffty was analyzed under the pressure difference of 1,117 Pa which corresponded to a wind velocity of $50m{\cdot}s^{-1}$ and the analytical results were also compared with theoretical ones. The result showed that there was little difference in the distribution of velocity overall and total pressure on the lateral side according to the width of the attached wind-protecting wall, but greenhouse with wind-protecting widths of 30 to 60cm has been reinforced to the extent of about 11% when compared with the case of being without the wall. The result also showed that the detached wind-protecting wall with a main-column interval of 3m was not stable so that it was necessary for the detached wind-protecting wall to be adequately reinforced to secure structural stability. Finally, there was great difference between analytical results and theoretical studies. The difference meant that there was some possibility of including errors when a theoretical study was done in three dimensional structure.

Development of Effective Screening Method for Resistance of oyster mushroom to Trichoderma disease in vitro (느타리버섯 푸른곰팡이병 저항성 실내검정 방법의 개발)

  • Trichoderma disease of oyster mushroom has not been effectively detected in the field for testing its resistance against the disease with its varieties. In this study, we investigated the methods to detect its resistance in the laboratory by using media, which enables us to understand the relevant characteristics (e.g., lysis, toxin enzyme, mycelial growth rate). In coculturing with strains of Trichoderma and oyster mushroom, it is possible to observe the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We also observed the inhibition of mycelial growth of oyster mushroom using the dilution method with 48-well plate, but could not observed the inhibition of mycelial growth using the filter paper method of cultural supernatant. In simultaneously culturing both Trichoderma and oyster mushroom, it was possible to detect the inhibition of the mycelial growth of oyster mushroom, but Trichoderma mycelium did not overgrow against oyster mushroom. We found that the pathogenicity was efficient in using solid medium with the phenomena of overgrowth and lysis by inoculating Trichoderma on top of mycelia of oyster mushroom. In conclusion, the methods (e.g., coculture method, dilution method with 48-well plate, post-inoculation method) are recommended to detect the resistance of oyster mushroom against Trichoderma disease.
