마우스에서 Interleukin-2가 RD-995 종양세포에 미치는 항암효과 (Effect of interleukin-2 on antitumor response against intraperitoneal RD-995 tumor in mice)

  • Recombinant interleukin-2(IL-2) has demonstrated as an antineoplastic agent in mice and human, but the relatively low response rates observed in clinical trials. Therefore, the present study was undertaken in order to evaluate therapeutic activities of IL-2 for the establishment of therapeutic applications. At the onset of the experiment, normal C3H/HeN mice were injected with 5$\times$10$\^$6/ RD-995 tumor cells, murine ultraviolet radiation-induced fibrosarcoma, intraperitoneally. Beginning on day 6, experimental groups were treated with a 5-day course of IL-2(subcutaneous injection of 30,000 IU every 12 hours for 5 days). The result of this experiment revealed that body weight gradually decreased from 20th day in control mice. Subcutaneous IL-2 therapy prevented partially decrease body weight, and prolonged survival of mice compared with control group.

Inhibitory effect of DA-125 on cancer metastasis by downregulating MMPs and CAMs

  • Matrix metalloproteinases (MMPs) play an important role in tumor invasion and metastasis by extracellular matrix degradation. To analyze the effect of DA-125, a anthracyclin derivative, on the invasion or metastasis of cancer cells the expression of matrix metalloproteases (MMPs) was investigated in human fibrosarcoma HTl080 cells by RT-PCR or gelatin zymographic methods. As result, DA-125 suppressed the expression of MMP-2 and 9 as well as tissue inhibitor of metalloproteinase-1 (TIMP-1) TIMP-2 and MT1-MMP with a time- and dose-dependent manner. Inaddition, DA-125 inhibited cancer cell migration and colony formation, and also exhibited the inhibitory activities of invasion and motility with a matrigel and type I collagen assay. (omitted)

  • 점액섬유육종은 고령자에서 가장 흔한 연조직 육종 중 하나이며 사지, 몸통 및 후복막에서 흔히 발생한다. 그러나 종격동 점액섬유육종은 매우 드물며 문헌에서도 몇 가지 사례만이 보고되었다. 이에 저자들은 66세 남성에서 수술로 확진된 전종격동 점액섬유육종의 영상 소견을 MRI 소견 중심으로 보고하고자 한다.

  • 개에 자연발생(自然發生)한 다음과 같은 종양(腫瘍)을 수집(蒐集)하여, 그 육안적(肉眼的) 및 조직학적(組織學的) 소견(所見)을 검토(檢討)하고, 이에 관(關)한 간단한 고찰(考察)을 가(加)했다. 피부(皮膚) 및 피하직(皮下織)의 종양(腫瘍) : 섬유종(纖維腫), 지방종(脂肪腫), 표피성(表皮性) 낭종(囊腫), 흑색육종(黑色肉腫), 한선종(汗腺腫), 비만세포종(肥滿細胞腫)(2예(例)), 비만세포육종(肥滿細胞肉腫), 피지선암종(皮脂腺癌腫). 비장(脾臟) 및 임파절(淋巴節)의 종양(腫瘍) : 비피막(脾被膜)의 섬유육종(纖維肉腫), 비(脾)의 평활근육종(平滑筋肉腫), 임파절(淋巴節)의 임파육종(淋巴肉腫)(2예(例)). 폐장(肺臟)의 종양(腫瘍) : 기관지성암종(氣管枝性癌腫)(3예(例)), 각각(各各) 선암종형(腺癌腫型), 편평세포암종형(扁平細胞癌腫型) 및 미분화세포(未分化細胞)(원형(圓形))암종형(癌腫型). 소화관(消化管) 및 간장(肝臟)의 종양(腫瘍) : 위(胃)의 섬유종(纖維腫), 간(肝)의 혈관종(血管腫), 담관암종(膽管癌腫), 간세포암종(肝細胞癌腫), 간(肝)에 출현(出現)한 골수성백혈세포세포(骨髓性白血細胞細胞). 복막(腹膜)의 종양(腫瘍) : 섬유육종(纖維肉腫). 비뇨생식계(泌尿生殖系)의 종양(腫瘍) : 자궁(子宮)의 섬유종(纖維腫), 난소(卵巢)의 난포낭종(卵胞囊腫), 질(膣)의 전염성(傳染性) 성기종(性器腫)(6예(例)), 신암종(腎癌腫), 섭호선종(攝護腺腫)(2예(例)), 포피(包皮)의 섬유종(纖維腫), 고환(睾丸)의 정충종(精蟲腫). 유선(乳腺)의 종양(腫瘍) : 혼합종(混合腫)(2예(例)), 근상피종(筋上皮腫). 신경계(神經系)의 종양(腫瘍) : 대퇴부(大腿部)의 신경섬유육종(神經纖維肉腫).

유건종(類腱腫)의 임상적 고찰 (Clinical Investigation of Desmoid Tumors)

  • A desmoid tumor is a locally aggressive growth of connective tissue origin which infiltrates the surrounding tissue and has a marked tendency for recurrence. And so it was also called as an aggressive fibromatosis, musculofascial fibromatosis or fibrosarcoma etc. Thirteen cases of desmoid tumor was treated since 1980, and their retrospective study was done with 79 months of follow-up after initial surgical excision. The female was involved in 12 cases(one male) with the age ranged from 7-50 years, average 28 years, and seven patients in third decade. A slowly growing mass was excised on average 4 months after first notice of the mass, but their margins are not demarcated clearly in most cases. Wide excision in 12 cases was done, but wide excision and saphenous vein graft was performed in one case because of invasion of posterior tibial artery by tumor mass. The tumor was found on extraabdominal region in 8 cases(61.5%) but 5 cases in abdominal wall(38.5%). The recurrence rate was high(6/13, 46.2%), and 11 times in 6 patients were recurred(average 1.8 times), within 27 months of initial excision. Six cases of recurrence were treated with wide excision again in 3 cases, wide excision combined with radiotherapy(4,000-6,000cGy) in 4 cases and wide excision with chemotherapy in one case. During the follow-up for average 21 months after treatment, no recurrences are found. Tumor remission periods without recurrence are average 67 months in all, and 11 years in longest case. Histologically it was very mimic with fibrosarcoma but could be differentiated with Trichrome stain, and their findings are not changed after recurrence.

MethA Fibrosarcoma Cells Expressing Membrane-Bound Forms of IL-2 Enhance Antitumor Immunity

  • Tumor cells genetically engineered to secrete cytokines are effective in tumor therapy, but various unexpected side effects are observed, which may result from the bulk activation of various bystander cells. In this study, we tested tumor vaccines expressing various membrane-bound forms of IL-2 (mbIL-2) on MethA fibrosarcoma cells to focus antitumor immune responses to CTL. Chimeric forms of IL-2 with whole CD4, deletion forms of CD4, and TNF were expressed on the tumor cell surface, respectively. Tumor clones expressing mbIL-2 or secretory form of IL-2 were able to support the cell growth of CTLL-2, an IL-2-dependent T cell line, and the proliferation of spleen cells from 2C TCR transgenic mice that are responsive to the $p2Ca/L^d$ MHC class I complex. Expression of mbIL-2 on tumor cells reduced the tumorigenicity of tumor cells, and the mice that once rejected the live IL-2/TNF tumor clone acquired systemic immunity against wild-type MethA cells. The IL-2/TNF clone was inferior to other clones in tumor formation, and superior in the stimulation of the CD8+ T cell population in vitro. These results suggest that the IL-2/TNF clone is the best tumor vaccine, and may stimulate CD8+ T cells by direct priming. Expression of IL-2/TNF on tumor cells may serve as an effective gene therapy method to ameliorate the side effects encountered in the recombinant cytokine therapy and the conventional cytokine gene therapy using the secretory form of IL-2.

이하부에 발생한 침습성 섬유종증이 하악체에 침범한 증례 보고 (A CASE OF AGGRESSIVE FIBROMATOSIS INVADED MANDIBULAR BODY ON THE PAROTID REGION)

  • Fibromatosis is benign fibroblastic proliferative lesion with abundant collagenous neo-formation located principally in the abdominal wall and in the upper and lower extremities (Masson & Soule, 1966). Wilkins and Waldron, in 1975, suggested that the title aggressive fibromatosis was a more appropriate term, reflecting the invasive characteristics of the disease. Synonyms listed were extra-abdominal desmoid, juvenile fibromatosis, aggressive infantile fibromatosis and congenital fibrosarcoma. A total of 12% of all fibromatosis arise in head and neck. Fibromatosis of the oral cavity is uncommon and is even more rare when in involve the mandibule. It is a locally aggressive fibrous tissue tumor, generally does not metastasize, but may cause considerable morbility and even death due to local infiltration. The degree of microscopic cellularity is variable, not only from tumor to tumor but also from area to area in the same tumor. Some tumors present with proliferation of mature fibroblasts and a dominating collagenous component : others may show a lack of the tumor in both types. The common histologic denominator appears to be cellular interlacing bundles of elongated fibroblasts, showing little or no mitotic activity and no pleomorphism. Mitosis are not a consistent index of malignancy when found in younger age groups. Fibromatosis still posses difficult problems of diagnosis and treatment. It is frequently recurrent and infliltrates neighbouring tissues. These lesion infliltrate widely and replace muscle, fat, and even bone with fibrous tissue of varying cellularity. Lesion representing fibromatosis in the oral cavity must be carefully evaulated by both surgeon and pathologists to ensure proper diagnosis and treatment planning. When these lesions involve bone, surgeon must be aware of the lesion's potential to perforate the cortex and expand while remaining hidden from the surgeon's view. Careful and precise clinical correlation with histologic appearance is essential to preclude misdiagnosis of fibrosarcoma yet provide surgical treatment plan that provides adequate local excision and long-term follow up. As regards cause, little is known. It is attributed to trauma or alteration in the sex hormone(Carlos, et al, 1986). Clinially, the lesion is reported to be not painful in most cases, but capable of rapid growth. The treatment is essentially surgical excision with wide margin of adjacent uninvolved tissue. Radiotherapy, hormone treatment or chemotherapy are of no use (WIkins et al, 1975 ; Majumudar and Winiarkl, 1978). We report a case of aggressive fibromatosis of 15-year-old with a lesion in the soft tissue of the parotid area that invaded the underlying bone of the mandibular body.

갯메꽃 추출물과 유기용매 분획물의 암전이 억제 효과 (Anti-invasion Effects of Calystegia soldanella Solvent Extracts and Partitioned Fractions on PMA-stimulated Fibrosarcoma Cells)

  • 갯메꽃은 해안 사구에 분포하며 식물로 높은 환경 적응력을 가지고 있으며 갯메꽃은 항산화, 해열, 살균, 이뇨작용 등에 효과가 있는 것으로 알려져 있다. 본 연구에서는 암전이 과정에서 중요한 역할을 한다고 알려진 matrix metalloproteinases (MMPs)의 일종인 MMP-2와 MMP-9의 활성에 대한 갯메꽃의 억제효과를 methylene chloride (MC) 및 methanol (MeOH) 추출물과 조추출물, 유기용매 분획물을 시료로 하여 인체 섬유육종 HT-1080 세포를 이용하여 ELISA (Enzyme-linked immunosorbent), gelatin zymography, Wound healing assay, RT-PCR, western blot 실험을 통해 조사하였다. 갯메꽃의 MC 및 MeOH 추출물, 조추출물과 4가지 유기용매 분획물이 HT-1080 세포에서 MMP-2와 MMP-9 생성 억제 활성을 나타냄을 확인하였다. 그 중에서도 n-hexane, 85% aq.MeOH 분획물이 전반적으로 MMP-2와 MMP-9에 대한 높은 억제활성을 보였다. 이들 결과로부터 85% aq.MeOH과 n-hexane 분획물에 MMP 억제 활성이 높은 물질이 존재할 것으로 추측되어지며, 갯메꽃 추출물 및 분획물이 암침윤 및 전이를 억제하는 효과적인 항암소재로서의 가능성이 있음을 제시한다. 이에 본 연구팀은 85% aq.MeOH과 n-hexane 분획물에 있는 활성물질을 분리하기 위한 연구를 추가적으로 수행할 계획이다.

인간 섬유육종세포에서 비쑥 추출물과 유기용매 분획물의 암전이 억제 효과 (Anti-invasive Effect of Artemisia scoparia Halophyte Extract and its Solvent-partitioned Fractions in Human Fibrosarcoma Cells)

  • 염생식물인 비쑥은 식용이 가능한 약용식물로서 살충, 항염, 항콜레스테롤, 해열, 항균 활성 등이 알려져 있다. 본 연구에서는 phorbol-12-myristate-13-acetate (PMA)로 자극된 인간 섬유육종 HT-1080 세포에서 5가지 활성검색방법 즉 : gelatin zymography, MMPs ELISA, wound healing assay, reverse transcription-polymerase chain reaction (RT-PCR), Western blot을 이용하여 비쑥의 조추출물과 그 용매 분획물의 MMP-2와 MMP-9 효소 활성에 대한 저해 효과를 평가하였다. 비쑥 시료들을 메틸렌 클로라이드(MC)와 메탄올(MeOH)로 각각 2번 추출하여 합한 것을 조추출물로 사용하였으며 이 조추출물은 MMP-2와 MMP-9 효소활성에 대해 유의한 억제 효과를 나타내었다. 이 조추출물은 용매극성에 따라 다시 n-hexane, 85% aq.MeOH, n-butanol 및 물 분획층으로 분획되었으며 이렇게 얻어진 4개의 용매 분획물들중에 n-hexane과 85% aq.MeOH 분획이 gelatin zymography와 MMP ELISA assay에서 MMP-2와 MMP-9의 활성을 효과적으로 억제하였다. 또한 wound healing assay, RT-PCR 및 Western blot assay에서 H2O 분획을 제외한 모든 용매 분획물들이 세포 이동, 그리고 MMP-2 및 MMP-9의 mRNA와 단백질 발현을 유의하게 억제하였다.
