식이섬유가 고콜레스테롤혈증 흰쥐의 혈청 지질대사에 미치는 영향 (Effect of Fiber on Serum Lipid Metabolism in Rats with Diet-Induced Cholesterolemia)

  • The study was conducted to investigate the effect of cholesterol and fiber on serum lipid metabolism in rats. Male Sprague Dawley rats were administrated 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia and were fed on diet containing three concentrations(0%, 5%, 10%) of cellulose and pectin, respectively. The rats were sacrified after 5 weeks of feeding periods. Serum triglyceride concentration was significantly decreased in proportion to level of fiber. Phospholipid concentration was more decreased in 10% fiber groups than 5% fiber groups but did not show any difference by fiber types. Free cholesterol and cholesteryl ester concentrations in serum were significantly decreased by fiber supplement and pectin was more effective than cellulose in cholesterol lowering effect. Whereas HDL cholesterol concentration and HDL C/TC were increased in proportion to level of fiber and the effect of pectin was not more significant than that of cellulose. Serum chylomicron cholesterol concentration also showed significant decrease by pectin supplement and LDL , VLDL cholesterol concentrations were more significantly decreased in 10% fiber groups than those of 5% fiber groups. The results indicate that 10% pectin may have beneficial roles in hypercholesterolemia.

식이섬유 첨가식이 Streptozotocin-유도 당뇨쥐의 장기능과 지질 및 당질대사에 미치는 영향 (The Effects of Dietary Fiber Feeding on Gastrointestinal Functions and Lipid and Glucose Metabolism in Streptozotocin-induced Diabetic Rats)

  • This study was conducted to compare the effects of four types of dietary fiber supplementations (cellulose, pectin, guar gum, and polydextrose) on gastrointestinal function, diabetic symptom amelioration and lipid & glucose metabolism in streptozotocin-induced diabetic rats. Six groups of male rats were fed ad libitum dietary fiber-free control diet or one of experimental diets containing 5% dietary fiber for four weeks. All types of dietary fiber supplementation seemed to protect the diabetic animals from the loss of body weight. The primary diabetic symptoms such as polydipsia, polyphasia, polyuria and urinary glucose excretion were ameliorated by cellulose, pectin, and guar gum, but not by polydextrose. Gastrointestinal transit time was significantly shortened and fecal dry weight was significantly increased in all the dietary fiber-supplemented groups except the polydextrose group. Large intestine was significantly lengthened by dietary fiber feeding. The serum triglyceride and total cholesterol levels were effectively lowered by pectin, guar gum and polydextrose. Regardless of their types, the fiber supplementation had no effect on serum HDL-cholesterol. Whereas fasting blood glucose level was significantly lowered by all types of fiber supplementations, glucose tolerance was more effectively improved by pectin and guar gum.

Effects of ruminally degradable starch levels on performance, nitrogen balance, and nutrient digestibility in dairy cows fed low corn-based starch diets

  • Objective: This trial was performed to examine the effects of ruminally degradable starch (RDS) levels in total mixed ration (TMR) with low corn-based starch on the milk production, whole-tract nutrient digestibility and nitrogen balance in dairy cows. Methods: Eight multiparous Holstein cows (body weight [BW]: $717{\pm}63kg$; days in milk [DIM]: $169{\pm}29$) were assigned to a crossover design with two dietary treatments: a diet containing 62.3% ruminally degradable starch (% of total starch, low RDS) or 72.1% ruminally degradable starch (% of total starch, high RDS). Changes to the ruminally degradable levels were conducted by using either finely ground corn or steam-flaked corn as the starch component. Results: The results showed that dry matter intake, milk yield and composition in dairy cows were not affected by dietary treatments. The concentration of milk urea nitrogen was lower for cows fed high RDS TMR than low RDS TMR. The whole-tract apparent digestibility of neutral detergent fiber, acid detergent fiber and crude protein decreased, and that of starch increased for cows fed high RDS TMR over those fed low RDS TMR, with no dietary effect on the whole-tract apparent digestibility of dry matter and organic matter. The proportion of urinary N excretion in N intake was lower and that of fecal N excretion in N intake was higher for cows fed high RDS TMR than those fed low RDS TMR. The N secretion in milk and the retention of N were not influenced by the dietary treatments. Total purine derivative was similar in cows fed high RDS TMR and low RDS TMR. Consequently, estimated microbial N flow to the duodenum was similar in cows fed high RDS TMR and low RDS TMR. Conclusion: Results of this study show that ruminally degradable starch levels can influence whole-tract nutrient digestibility and nitrogen balance in dairy cows fed low corn-based starch diets, with no influence on performance.

  • This study was performed to investigate the influence of various dietary fiber sources(rice bran, Chinese cabbage, radish, apple, laver, sea mustard) on the intestinal microflora in rats. Eight groups of rats were fed each experimental diets containing 5% of total dietary fiber for 4 weeks. Total viable counts and the numbers of Bifidobacterium, Lactobacillus, Clostridium, E. coli and Staphylococcus were determined by nonselective medium and various selective media. The number of Bifidobacterium(p<0.01) was higher in the apple and sea mustard groups than those in the other groups and the number of Lactobacillus(p<0.01) was lower in the radish group. A decrease in the intestinal population of clostridium was shown in the sea mustard group. The E. coli(p<0.01) and Staphylococcus(p<0.01) populations decreased in the apple group compared with other groups. These findings suggest that the apple fiber and sea mustard fiber are effective in improving intestinal flora of rats.

EFFECTS OF CALCIUM SALTS OF LONG-CHAIN FATTY ACIDS ON RUMINAL DIGESTIBILITY, MICROBIAL PROTEIN YIELD AND LACTATION PERFORMANCE

  • Four sheep per treatment were fed either control or 3% calcium salts of long-chain fatty acids (Ca-LCFA) in a total mixed ration (TMR). Feed and free water intakes were not different, but digestibilities of crude protein and crude fiber were lower (p<0.05) and that of crude fat was higher (p<0.05) for sheep fed Ca-LCFA than for control sheep. Dry matter digestibility, ruminal pH and microbial protein yield were not different between treatments and ammonia-N concentration in the rumen was higher for sheep fed Ca-LCFA than for control sheep. A 60-day milk production trial was conducted with thirty lactation Holstein cows. Fifteen cows per treatment were fed TMR containing either control or 3% Ca-LCFA ad libitum. Feed intake was not different between treatments, but milk yield was significantly higher (p<0.05) for cows fed Ca-LCFA than for control cows. Milk fat percentage was slightly higher and milk protein was lower for cows fed Ca-LCFA than for control cows. Lactose and total solid contents in milk were not different between treatments.

EFFECTS OF CIMATEROL ON CARCASS AND SKELETAL MUSCLE CHARACTERISTICS UNDER AD LIBITUM AND RESTRICTED FEEDING CONDITIONS IN LAMBS

  • Fifty-two wether lambs weighing 30 kg were randomly assigned to 5 treatment groups; 1) initial slaughter. 2) control-maintenance (CON-MT), 3) control-ad libitum (CON-AL), 4) cimaterol-maintenance (CIM-MT) and 5) cimaterol-ad libitum (CIM-AL). Ad libitum-fed animals had free access of a high-concentrate diet, whereas maintenance animals were restricted in feed intake to maintain the initial weight of 30 kg for 90 days. Cimaterol was administered in the feed at 10 mg/kg. Regardless of feeding level, the administration of CIM improved carcass weight (p < .05), dressing % (p < .01), longissimus muscle area (p < .01), leg conformation and muscling (p < .01), USDA yield and quality grades (p < .01) and protein concentration (p < .01) in carcass as well as in muscle. Cimaterol feeding decreased organ wt (p < .01), baekfat depth (p < .01), intramuscular fat and overall fatness. Cimaterol was effective for muscle accretion even under restricted feeding condition. The greater accretion of muscle was the result of the hypertrophy of both type I and type II muscle fibers but the hypertrophy of type II fiber (110%) was much greater than that of type I fiber (37%). Cimaterol feeding decreased muscle DNA concentrations but the number of nuclei per muscle fiber was not changed, indicating that the lower DNA concentration was due to the dilution effect caused by the hypertrophy of muscle fiber. As evidenced by lower flank streaking, lower marbling and darker muscle, CIM feeding adversely affected meat quality. Meat tenderness was also adversely affected, resulting in significantly (p H .01) tougher meat in CIM-fed animals.

  • 우렁쉥이 추출 불용성 식이섬유를 5%, 10%, 20% 첨가한 식이 후 실험쥐 집단의 영양생리학적 특성변화를 조사하였다. Cellulose 급여에 따른 총 식이량에는 차이가 없었으나 식이 중 cellulose의 함량이 높아짐에 따라 체중은 감소하였으며 식이효율의 조절, 배변량 증가, 변의 수분 증가, 식이의 장 통과시간 단축, 장 길이의 증가 등의 유의적인 효과를 나타내었다. 우렁쉥이 식이섬유를 급여한 실험쥐의 혈청분석 결과 total cholesterol, LDL-cholesterol, 중성 지질 및 인지질, 혈당은 식이섬유의 함량이 높아짐에 따라 감소하였으며 HDL-cholesterol은 식이섬유의 함량이 높아짐에 따라 증가하였다. 따라서 우렁쉥이 식이섬유를 첨가한 가공식품을 섭취할 경우 배변량의 조절, 변비 및 비만조절, 혈청 지질의 감소와 같은 유용한 생리적 효과가 있을 것으로 기대된다.

Effects of Rice Bran and Wheat Bran on Intestinal Physiology and Small-bowel Morphology in Rats

  • The present study was aimed at investigating the nutritional and physiological significance of rice bran as a source of dietary fiber as compared to pectin and wheat bran. The parameters for comparison included hypertrophy and morphology of intestines, stool weights and villus marker enzyme activity. For 6 weeks, 10 Sprague Dawley male rats were given one of six experimental diets: 1% cellulose control (CC), 5% pectin (P5), 5% rice bran(RB5), 10% rice bran(RB10), 5% wheat bran (WB5) or 10% wheat bran (WB10) based on the level of dietary fiber. Among experimental groups, food efficiency ratio and body weight gain was comparable. RB10 increased cecal and colonic tissue weights and content weights of cecum and colon as much as P5 did. Stool weight was positiviely correlated with colonic tissue weight (r=0.727, P<0.001), with colonic content weight(r=0.647, P<0.001). Small intestine length increased most in the P5 group, followed by the RB10 group. The scanning electron micrograph of jejunal villi from rice bran groups showed a leaf-shaped, smooth and regular pattern, whereas that of CC group produced a rather long shape. The wheat bran groups showed an irregular leafshaped pattern, and the pectin group typically produced leaf-shaped villi with surface damage. The activities of villus marker enzymes (maltase and sucrase) were higher in the bran-fed rats than in the control or pectin-fed rats. The results indicate than not only dietary fiber amounts but also fiber sources are closely related to the physiology and morphology of the large and small intestines in rats. Rice bran exerted effects on fecal output and trophic effects on the intestines similar to those of pectin.

Effect of Protein and Fiber Levels on Ethanol-Induced Brain Damage in Rats

  • The purpose of this study was to investigate the effect of protein and dietary fiber levels on the activities of ehanol metabilizing enzymes of the brain in acute and chronic ethanol-treated rats. Male Sprague-Dwley rats were fed on diets containing two levels of protein(7%, 20%)) with two levels of fiber(5%, 105) for 5 weeks. Rats were orally administered 40% (v/v) ethanol(5g/body weight) 90 min before decapitation in the acute ethanol-treated groups and 25% (v/v) ethanol (5g/kg body weight) once a day for 5 weeks in the chronic ethnol-treated groups. Cytosilic alcohol dehydrogenase (ADH) activities were higher than those of mitochondrial ADH. The ADH activities were increased by 20% protein and %% fiber levels in the diet in two fractions , but were decreased by chronic ethanol treatment. Mitochondrial aldehyde dehydrogenase (ALDH) activities did not change by ethanol treatment but were increased by the 20% protein level. However, cytosilic ALDH activities were decreased by chronic ethanol treatment at the 5% fiber level and did not change with protein levels. Both ALDH activities were higher in the 10% fiber groups than the 5% fiber groups. Cytochrome P-450 contents were significantly increased in the chronic ethanol-treated groups but xanthine oxidase (XO) activities did not change. P-450 contents and XO activities were significantly decreased in both the low protein and fiber groups.
