Suppressive Effect of Administrated Glutathione-Enriched Saccharomyces cerevisiae FF-8 on the Oxidative Stress in Alcoholic Fatty Liver (알코올 투여 흰쥐의 간 조직 산화스트레스에 미치는 글루타티온 고함유 효모 Saccharomyces cerevisiae FF-8 균체의 영향)

  • Glutathione is a well known chemotherapeutic agent for liver disease and is a popular nutritional supplement in the United States. Previous our studies reported the suppressive effects of glutathione-enriched Saccharomyces cerevisiae FF-8 strain (FF-8GY) on carbon tetrachloride- and alcohol-induced hepatotoxicity. The primary objective of this study was to investigate the comparative effects of FF-8GY and commercially available glutathione-enriched yeast extract (GYE) against the oxidative stress in alcohol-induced fatty liver of rats. The lipid peroxidative index (thiobarbituric acid-reactive substances, TBARS) and antioxidant status (reduced glutathione level) were used to monitor those protective roles of FF-8GY or GYE treatment. When the rat was treated alcohol, the TBARS levels in the whole liver and the subfractions of microsomal and mitochondria were significantly increased but these were significantly decreased by FF-8GY treatment and tended to be lowered by GYE treatment. The concentration of hepatic glutathione is known to be closely associated with antioxidant system and this was slightly deplete in the alcohol-induced rats, but this was recovered by treating with FF-8GY. However, the glutathione concentration was more significantly decreased in the GYE supplementation in alcohol feeding rats. Alcohol treatment also negatively affected the serum total protein and albumin, but these were significantly increased near normal levels in FF-8GY coadministered rats. These results suggest that glutathione-enriched Saccharomyces cerevisiae FF-8 strain may have positively mediate the alcohol-induced oxidative stress, and this effect was more pronounced in FF-8GY compared to GYE.

Effects of Development and Viability of Pig Oocytes Matured in Defined Medium Containing PVA, PVP and pFF (PVA, PVP 및 pFF를 첨가한 체외성숙 한정배지가 미성숙 돼지 난포란의 성숙과 배발달에 미치는 영향)

  • This study was conducted to develop a serum-free, defined medium of IVM of pig oocytes. The TCM-199 with supplemented with polyvinylalcohol(PVA), polyvinylpyrrollidone(PVP) and porcine follicular fluid(pFF) were used as basal medium. The effects of the these additives on the rates of maturity and development under in-vitro fertilization and in vitro culture were examined and subsequently considered on the possibilities be sustituted for the bovine serum albumin(BSA). Maturation rate of pig oocytes in IVM media containing PVA(82.4%), pFF(89.4%) and BSA(90.0%) were significantly higher(P<0.05) than that of PVP(78.6%). Cleavage rate after IVF of PVP(64%) was significantly lower(P<0.05) than these of PVA(73%), pFF(77%) and BSA(73%) supplements. in vitro development rates to morulae and blastocyst on PVP(54%) were also significantly lower(P<0.05) than these of the supplements of PVA(63%), pFF(69%) and BSA(65%). In comparison of maturation and fertilization rates of pig oocytes in each supplements, the maturity rates of PVA(82.4%), pFF(89.4%) and BSA(90.0%) were significantly lower(P<0.05) than that of PVP(72.4%) and while, the fertilization rates of pFF(87.1%) and BSA(89.1%) were significantly higher(P<0.05) than these of PVA(78.0%) and PVP(70.6%). It may be concluded that PVA and pFF can be substituted far BSA in medium for culturing pig oocytes; however, it may be considered that PVP were limited to for BSA in the in vitro culture of the embryos.

Effect of Treatment and Samples of Human Follicular Fluid on Development In Vitro of Mouse Embryos (사람 난포액의 처리 방법과 Sample이 생쥐 수정란의 체외 발달율에 미치는 영향)

  • The present studt was performed to investigate the effect of treatment and samples of human follicular fluid (hFF) on the development in vitro of mouse embryos. The two cell stage embryos collected at 40 h post-hCG injection were cultured in the modified human tubal fluid (m-RTF) containing 15% synthetic serum substitute (SSS) or human tubal fluid (hFF) for up to 3 days at $37^{\circ}C$ in 5% $CO_2$ incubator. Also the composition of hormone, total protein and protein pattern of hFF samples were analyzed. The developmental rate of mouse embryos developed to blastocyst were not significant difference in the m-RTF containing 15% hFF filtered with 0.22 or 0.8 ${\mu}m$ syringe filter, however, the embryos cultured in the m-RTF containing inactivated hFF were significantly (p<0.05) developed at the high rate to blastocyst than those containing fresh hFF and SSS. The in vitro developmental rate to blastocyst and hatched blastocyst in the m-RTF containing 15% hFF sample A (90.5 and 85.4%, respectively) and SSS (79.4 and 75.3, respectively) were significantly (p<0.05) increased, compared with hFF sample B (64.2 and 54.1 %, respectively). The hFF sample A tended to be higher concentration of LH, FSR, total protein and the ratio of progesterone/$E_2$ and lower concentration of $E_2$ and progesterone than the hFF sample B, but there were no differences in the protein pattern between the two hFF samples. The results of these study suggest that the addition of hFF to the culture medium enhances the development in vitro to blastocyst and hatched blastocyst, but the in vitro developmental rate of mouse embryos is different between hFF samples.

Fenofibrate decreases radiation sensitivity via peroxisome proliferator-activated receptor ${\alpha}$-mediated superoxide dismutase induction in HeLa cells

  • Purpose: The fibrates are ligands for peroxisome proliferator-activated receptor (PPAR) ${\alpha}$ and used clinically as hypolipidemic drugs. The fibrates are known to cause peroxisome proliferation, enhance superoxide dismutase (SOD) expression and catalase activity. The antioxidant actions of the fibrates may modify radiation sensitivity. Here, we investigated the change of the radiation sensitivity in two cervix cancer cell lines in combination with fenofibrate (FF). Materials and Methods: Activity and protein expression of SOD were measured according to the concentration of FF. The mRNA expressions were measured by using real time reverse-transcription polymerase chain reaction. Combined cytotoxic effect of FF and radiation was measured by using clonogenic assay. Results: In HeLa cells total SOD activity was increased with increasing FF doses up to 30 ${\mu}M$. In the other hand, the catalase activity was increased a little. As with activity the protein expression of SOD1 and SOD2 was increased with increasing doses of FF. The mRNAs of SOD1, SOD2, $PPAR{\alpha}$ and $PPAR{\gamma}$ were increased with increasing doses of FF. The reactive oxygen species (ROS) produced by radiation was decreased by preincubation with FF. The surviving fractions (SF) by combining FF and radiation was higher than those of radiation alone. In Me180 cells SOD and catalase activity were not increased with FF. Also, the mRNAs of SOD1, SOD2, and $PPAR{\alpha}$ were not increased with FF. However, the mRNA of $PPAR{\gamma}$ was increased with FF. Conclusion: FF can reduce radiation sensitivity by ROS scavenging via SOD induction in HeLa. SOD induction by FF is related with $PPAR{\alpha}$.

Composition and Changes of Gastrointestinal Microflora in Breast-fed and Formula-fed Infant before and after Weaning (모유영양아의 인공영양아의 장내균총 조성과 이유보충식 도입에 따른 변화)

  • Microflora in 14 breast-fed(BF) and 15 formula-fed(FF) infants were investigated before (2-3 months after birth) and after weaning (5-7 months birth) to find the floral differences between the two lactation groups and the floral changes after weaning. Bifidolbacteria showed the highest count among the species of microflora in the BF group before and after weaning, but in the FF group, streptococci showed the highest count before weaning and bifidobacteria after weaning. Before weaning , the count of bifidobacteria, lactobacilli and total acaerobes were significantly higher in the BF group than in the FF. However, the frequencies of clostridia and klesidella , along with the counts of streptococci and eubacteria, were higher in the FF group. After weaning , the only difference was that the total count of aerobic bacteria was higher in the BF group. The counts of streptococci in the BF group and lactobacilli and total anaerobes in the FF group after weaning were significantly higher than before weaning. However, the count of total aerobes in the FF group after weaning was significantly lower than the count before weaning.

  • In this talk, I will introduce a reactive force field (ReaxFF) molecular dynamics (MD) simulation. In contrast to common MD simulations with empirical FFs, we can predict chemical reactions (bond breaking and formation) in large scale systems with the ReaxFF simulation where all of the ReaxFF parameters are from quantum mechanical calculations such as density functional theory to provide high accuracy. Accordingly, the ReaxFF simulation provides both accuracy of quantum mechanical calculations and description of large scale systems of atomistic simulations at the same time. Here, I will first discuss a theory in the ReaxFF including the differences from other empirical FFs, and then show several applications for studying chemical reactions of SiHx radicals on Si surfaces, which is an important issue in Si process.

  • In this paper, based on the relative logical storage cost of each movie by the difference of service request probability, we propose and implement the SDU(Separate Data Use) method and the EDU(Existing Data Use) method to provide FF(Fast Forward) and FR(Fast Reverse) function in efficiently through analysing MPEG bit stream and frame characteristics. By selecting and applying the FF/FR function providing method based on the service request probability of movie, the proposed method can reduce the cost of FF/FR functions for total serviced movies.

  • Objectives: The purpose of this study is to observe the suppressive effect of Asthma and Immune response improvement of FF(Farfarae Flos) Herbal-acupuncture into Joksamni(ST36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks. Experimental group was treated with concentration (1%) of FF-HA at Joksamni(ST36) for the later 8 weeks(3times/week). Results: 1. The weight and total cells in the mice lung treated with FF-HA decreased significantly compared with that of control group. 2. Total leukocytes and eosinophils in BALF of the mice group treated with FF-HA decreased remarkably compared with those of control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with FF-HA decreased significantly compared with those of Control group. 4. The concentrations of IgE, IL-4, IL-5 in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with FF-HA decreased significantly compared with those of control group 5. The number of Gr-l+/CDllb+, CDll+ cells, CCR3+ cells, CD4+ cells, and CD3e+/CD69+ in the lungs of the mice group treated with FF-HA decreased significantly compared with those of control group. 6. The mRNA expression of TNF-a, IL-4, IL-5, IL-13 in the mice group treated with FF-HA with RT-PCR decreased remarkably compared with those of control group. Conclusion : These result suggests that Farfarae Flos Herbal-acupuncture at Joksamni(ST36) in C57BL/6mice may be an effective part to OVA-induced asthma in C57BL/6 mice.

Antimicrobial Activity of Florfenicol Against Fish Pathogenic Bacteria and Pharmacokinetics in Blood of Cultured Olive Flounder by Oral Administration (Florfenicol의 어류 병원성 세균에 대한 항균 효과와 경구 투여후 넙치 혈액에서의 약물동태학적 특성)

  • The pharmacokinetics of florfenicol (FF) after oral administration was studied in the cultured olive flounder, Paralichthys olivaceus, using high performance liquid chromatography (HPLC). After single administration of FF (20 mg/kg body weight) by oral route in olive flounder ($700{\pm}50$ g, $23{\pm}1.5^{\circ}C$), the concentration in the serum was determined at 1, 5, 10, 15, 24, 30, 50 and 168 h post-dose. The kinetic profile of absorption, distribution and elimination of FF in serum were analyzed fitting to a two-compartment model by WinNonlin program. The area under the concentration-time curve (AUC), maximum concentration ($C_{max}$), time for maximum concentration ($T_{max}$) and elimination time were 22.51 ${\mu}g{\cdot}h/mL$, 0.84 ${\mu}g/mL$, 8.62 h and 447 h, respectively. The results of this study related to dosage and withdrawal times could be used for prescription of FF in field for the treatment of bacterial diseases in olive flounder.

Effect of Follicular Fluid on Attraction and Motility of Human Spermatozoa (사람 정자에 대한 유인능과 운동성에 미치는 난포액의 영향)

  • Follicular fluid has pivotal effects on attraction and motility of spermatozoa for successful fertilization. The effect of samples of human follicu1ar fluid(hFF) on attraction and motility of spermatozoa was investigated. Capillary tubes loaded with one of these samples, hFF sample A collected from patients with tubal factor, hFF sample B collected from patients with male factor, m-HTF and heated hFF sample were used for assessment of attraction and motility of spermatozoa following culture of 1, 2, and 4 hrs. Number and motile rate of spermatozoa in the tubes loaded with hFF sample A were significantly(P<0.05) higher than those of m-HTF, hFF sample B and heated hFF. Although the fresh hFF tended to increase the attraction of spermatozoa as compared to inactivated hFF, there was no significant difference between treatments.
