The Effect of Change in Meat Quality Parameters on Pig Longissimus dorsi Muscle by the Addition of Fermented Persimmon Shell Diet

  • The objective of the present study was to investigate the effects of growth performance, chemical composition and meat quality parameters by the addition of fermented persimmon shell diets (FPSD). The experimental animals were ninety-six Berkshires. The pigs were allotted at 8 per pen in front-open building with three replicate pens per treatment. Until $61{\pm}1kg$ live weight at 140 days, the animals were fed growing diet, after which, experimental samples were fixed at 0, 3, 5 and 7% FPSD as C, T1, T2 and T3 in the finishing diets. Pigs of $103{\pm}1kg$ live weight were slaughtered by electrical stunning. In growth performance, ADG increased more (p<0.05) in T2 than C and T1. ADFI (kg/day) was higher (p<0.05) in T2 than in other groups. Feed conversion ratio was lower (p<0.05) in T2 than in other groups. On longissimus dorsi muscle (LM), the content of moisture was lower (p<0.05) in T3 than in other treatments. Crude fat increased (p<0.05) by addition of FPSD. pH at 24 h ($pH_{24}$) decreased more (p<0.05) in T2 and T3 than C and T1 by addition of FPSD. WHC decreased (p<0.05) by addition of FPSD. In meat, Hunter $L^*$ and $a^*$ were lower (p<0.05) in C than in other treatments. Hunter $b^*$ was higher (p<0.05) in T3 than in other treatments. In back-fat, Hunter $L^*$ was higher (p<0.05) in C than in other treatments. Hunter $a^*$ was not different in C and T1 but increased (p<0.05) in treatments by addition of FPSD. Hunter $b^*$ increased (p<0.05) by addition of FPSD. In sensory evaluation scores, for fresh meat, the value of meat color was higher (p<0.05) in T2 and T3 than in C and T1. The value of marbling was lower (p<0.05) in C than in other treatments. The value of drip loss was higher (p<0.05) in C than in other treatments and was lower (p<0.05) in T2 and T3 than in other treatments. The results of cooked meat, the value of tenderness was not different in C and T1 but increased (p<0.05) in other treatments. The value of flavor was significantly higher (p<0.05) in T2 and T3 than in C and T1. The value of overall acceptability was increased (p<0.05) by addition of FPSD. Hence we can conclude that addition of FPSD affected growth performance and meat quality parameters, in particular, it improved crude fat, hunter $L^*$ and $b^*$, and sensory evaluation on pork and was also seen to affect $pH_{24}$ and WHC. FPSD can be used for improvement of meat quality parameters.

고추장 추출물을 급여한 Sprague-Dawley-Rat에서의 혈중지질 농도의 변화 양상 (Variation Patterns of the Blood Lipid Levels on the Sprague-Dawley-Rats Fed with Kochujang Extracts)

  • 오늘날 현대인들의 식생활방식 중 과다한 열량의 영양소 섭취로 인하여 초래되는 비만의 유발 원인 중의 하나로 알려진 고지혈증(hyperlipidemia)에 대한 예방효과가 있는 것으로 알려진 전통방식으로 제조된 고추장의 생리활성 기능성 효과를 분석하고자 하였다. 이 연구에서는 실험동물로서 Sprague-Dawley(SD)-Rat에 대해서 고추장 추출물의 건강증진 효과에 대해서 실험동물을 사용하여 직접 확인하고 시도하였다. 실험군은 실험동물용 일반사료만을 먹인 음성대조군 그룹(N-group)과 일반사료와 고추장 추출물을 동시에 먹인 시험그룹(K-group) 그리고 양성대조군으로 고콜레스테롤 사료만을 먹인 그룹(H-group)으로 구분하여 5주간 사육하면서 건강증진효과의 발현에 대해서 분석하였다. 3그룹의 실험동물을 5주간의 사육 후 해부했을 때 H-group의 모든 동물(100%)에서 공통적으로 심한 지방간(fatty liver) 소견이 관찰되었으나, N-group이나 K-group의 어떠한 동물에서도 지방간 소견은 관찰되지 않았다. 한편, 실험동물의 혈장성분에 대한 혈액생화학적 성적의 비교에서는 고지혈증의 예방에 유익한 성분으로 알려진 HDL의 경우, 고추장 추출물 투여 그룹($25{\pm}8.43mg/d{\ell}$)은 H-group($22.6{\pm}4.16$) 보다 다소간 상승된 수치를 보였으나, 특히 고지혈증을 악화시키는 원인으로 알려진 LDL 경우에는 H-group ($55.8{\pm}44.8mg/d{\ell}$)이 고추장 추출물 투여 그룹($8.6{\pm}1.52mg/d{\ell}$)과 음성대조군인 N-group($9.8{\pm}2.39$)의 성적보다도 유의적으로 높은 수치를 나타냈다. 결과적으로 이 연구에서 SD-Rat에 투여했던 고추장 추출물은 실험동물에서 건강기능 개선효과의 하나인 저지방혈증 효과를 발현한 것으로 사료되었다.

잣나무 生枝葉사일리지 급여가 한우의 영양소 소화율, 사료요구율 및 도체성적에 미치는 영향 (Effect of Pine Silage Feeding on Nutrient Digestibility, Feed Conversion and Carcass Traits of Korean Native Cattle)

  • 본 시험은 산림 간벌 부산물인 잣나무 생지엽을 반추가축 사료로 이용하기 위하여 제조된 발효 생지엽(pine silage)의 한우 반추위내 in situ 분해율 및 in vivo 소화율, 사양성적 및 도체분석에 있어서 볏짚 대체효과를 규명하기 위하여 수행되었다. In situ 시험은 nylon bag 시료를 처리 당 4반복으로 하여, 반추위내에서 48시간 동안 발효시켰으며, in vivo 소화시험은 볏짚 급여구와 생지엽 silage구로 나누어 incomplete double turn over design에 의해 처리 당 5반복으로 수행되었다. 사양시험에서는 생지엽 silage에 대한 볏짚 대체비율을 4수준(0%, 25%, 50%, 75%)으로 나누어 한우수소 48두를 처리당 12두씩 완전임의로 배치하였으며, 시험종료 후, 도체분석을 하였다. 반추위내 in situ 조단백질 분해율은 생지엽 silage가 볏짚보다 높았으나(P<0.01), NDF 분해율은 오히려 낮았다(P<0.01). In vivo 건물, 조단백질 및 NDF 소화율은 처리간 차이가 없었다. 일당 증체량은 볏짚 급여구와 비해 생지엽 silage 25% 대체구를 제외한 모든 생지엽 silage 대체구에서 유의적(P<0.01)으로 높았다. 생지엽 silage 급여 및 급여수준은 사료 효율에 영향을 주지 않았다.도체성적에 있어서 통계적 유의차는 없었으나 근내지방도는 50% 생지엽 silage 대체구가 타 처리구보다 수치적으로 높고, 등지방 두께는 생지엽 silage 대체구가 볏짚 급여구보다 다소 두꺼운 경향을 보였다. 이상의 결과로부터 생지엽 silage를 한우의 조사료로서 사용할 경우, 볏짚과 50%씩 혼합해서 급여하는 것이 건물섭취량과 일당증체량에서 좋은 효과를 기대할 수 있는 것으로 조사되었다.

양파껍질 추출물 급여가 비만 유도쥐의 소장내 변화에 미치는 영향 (Supplementation Effect of Onion Peel Extracts on Small Intestine of Obese Mice)

  • 본 연구는 양파껍질을 발효주정 에탄올 70%와 95% 농도로 추출하여 항균활성, 항산화활성 및 비만쥐의 소화기관에 미치는 영향에 대하여 알아보고자 실시하였다. 각 주정 추출물 5%는 E. coli와 Listeria monocytogens에서 유의적인 감소를 나타내었다. 각 주정 추출물 내 총 페놀함량은 각각 $166.89{\pm}0.03$ mg/g과 $160.89{\pm}0.13$ mg/g을 나타내었다. 항산화효과를 살펴본 결과 DPPH 라디칼 소거능과 SOD 유사 활성은 100 ug/ml의 농도에서 높은 수준을 나타내었다. 고지방식이를 급여하여 유도한 비만을 유도한 마우스에 70%와 95% 주정으로 추출한 양파껍질 추출물을 1, 3, 5%의 농도로 4주간 급여한 후 체중, 사료섭취량, 사료효율, 소장무게, 길이 장내 융모의 길이, 소장 내 미생물수를 측정하였다. 각 주정으로 추출한 양파 추출물 5%의 농도로 급여하였을 때 대조군에 비해 체중이 유의적으로 낮은 수준을 나타내었으나(p<0.05) 사료섭취량은 증가함을 나타내었다. 실험동물 마우스 소장의 무게와 길이는 양파 껍질추출물 급여에 따른 유의적인 변화는 보이지 않았으나 소장 융모의 길이는 대조군보다 유의적으로 증가함을 나타내었다. 총균수, Cl.perfringenes와 E.coli는 5%의 농도로 양파껍질 주정 추출물을 처리했을 때 유의적으로 감소함을 나타내었으나 젖산균은 증가함을 나타내었다. 이러한 결과는 추후 부가적인 추가 연구가 필요하겠지만 70%와 95%의 주정으로 추출한 양파껍질 추출물을 5% 수준으로 이용하면 체중조절 첨가제로서 그리고 장의 건강을 증진시키기 위한 첨가제로 사용할 수 있는 잠재성이 있는 것으로 기대된다.

일제하(日帝下)(1920년대) 조선인수형인(朝鮮人受刑人)의 식생활상황(食生活狀況)과 그 배경(背景) (Dietary Life Status of Korean Prisoners' and the Background during the Period of Japanese Ruling)

  • The objects of this study are to find out (1) real situation of the food supply in prison under Imperial Government of Japan more cleary (historical meaning) and (2) which might help in understanding wrong present food consumption patterns in Korea which causes environmental as well as health problems. It is generally known that the length of the Japanese occupation for Korea is 36 years. However, it is concluded in this study that it was longer (70 years ; from 1875 to 1945 from when Japanese Army attacked and occupied Yungjongdo and Kanghwado island to e time when they were defeated on World War II.) Korea was annexed by Japan in 1910 then the Imperial Government of Japan dismissed the Korea Army, controlled the Office of Justice and the management of prison by force. Since then about 50% of all land was fell into Japanese Government ownership and 80% of Korean farmers became as tenant. After this change, Korea farmers were forced to pay extremely high rent (up to 80% of its harvest). Forced immigration, low price procurement of grain by Japanese government up to more than 30%of their production, was practiced. Accordingly, the food situation of Korean farmers became miserable, which may caused more violations of Imperial Japanese Law. Malnutrition, epidemic diseases, mortality rate of infants soared and average life expectancy shortened to 20-30 years old. This was the period of World Economic Crises and Food Crises in Japan. It was said then that if one Japanese comes to Korea then 200 Koreans will starved to death. Meanwhile, Proconsul Bureau of Chosun requested to the Department of Medicine, the Imperial University of Kyungsung to survey food supply situation of Koreans in prison throughout Korea. Objectives of the survey then was not only to find out scientifically whether it is agreeable in maintaining prisoner's health and also find out the possibility to save food during food crisis. Survey was started from 1923 and ended in 1945, and it focussed on prisoners in the Seodaemoon Prison. This report is the outcome of the first survey. They concluded that the food supplied was nutritionally (had) no problem, in compare with those of workers in the factory, students in the dormitory in Japan and with those of prisoners in Taiwan, France and Germany. Amount of grain supplied were different according to their work lord and was divided into 9 different levels. Total grain was consisted of 50% millet, 30% soybean and 20% indica rice(variety). However, there were no difference in the amount of supply of side dishes between work groups. For the highest working group, 3280g of boiled grain per day was supplied to make stomach full but as a side dishes, salty fermented bean paste, fermented fish and salty soups, etc. was supplied. Deficiency of animal protein were observed, however, high intake of soybean may possibly caused animal protein deficiency problem. On the contrary, the intake of water soluble vitamins were insufficient but the level of calcium and iron intake seems to be sufficient, however, imbalance of intake of nutrition may caused low absorbtion rate which might caused malnutrition. High intake of dietary fiber and low intake of cholesterol may possibly prohibited them from so called modem disease but may caused the defect in disease resistancy againist epidemics and other traditional disease. Over intake of salt(20-30g per day) was observed. Surveyors who attended in this survey, mentioned that the amount of food intake may nutritionally be sufficient enough but the quality of food(and possibly, the taste of food) were like that of animal feed. For the officials who received this report might consider that considering the war situation and food crisis, the supply situation of food in the prison may considered to be good enough(because they are not starving). But as a Korean who studied this report, one feel extremely pity about those situation because (situation of) those period were very harsh under the Imperial Law and keeping the Law by Koreans were almost impossible, therefore, about one third adult violated the Law and were put into jail. And they were treated like animals.

Bacillus coagulans NRR1207이 생산하는 α-galactosidase에 의한 대두박 비소화성분의 가수분해 (Hydrolysis of Non-digestible Components of Soybean Meal by α-Galactosidase from Bacillus coagulans NRR1207)

  • 본 연구는 한국전통약용식물(구기자, 오미자 잎)의 발효물로부터 분리한 Bacillus coagulanse NRR1207의 발효 특성을 파악하고 Bacillus coagulans NRR1207의 ${\alpha}$-galactosidase의 활성과 이를 통한 대두박의 비소화성분의 분해를 확인하였다. Bacillus coagulans NRR1207이 생산하는 효소 중 ${\alpha}$-galactosidase, ${\beta}$-galactosidase, ${\alpha}$-glucosidase가 가장 높은 40 nmol 이상의 활성을 나타내었다. Bacillus coagulans NRR1207의 발효 특성은 10% skim milk에서 배양했을 때, pH는 급속히 감소했고 적정 산도는 1.9%까지 증가했고 생균수도 발효 24시간에 8.8 log CFU/ml로 증가했다. 유당은 배양 72시간째 완전히 고갈되었고 유산 생산 능력도 탁월했다. Bacillus coagulans NRR1207을 대두박에 접종 후 배양시간에 따른 생균수의 변화는 배양 시작 시 7.6 log CFU/ml, 배양 16시간에 최고에 도달하여 9.0 log CFU/ml이었고 배양 72시간에 8.3 log CFU/ml로 Bacillus coagulans NRR1207이 왕성하게 잘 성장하였다. 대두박의 비소화성분 분해는 Bacillus coagulans NRR1207 접종 후 발효 24, 48, 72시간이 경과하면서 이 균이 생산한 ${\alpha}$-galactosidase에 의해 비소화성분인 stachyose와 raffinose가 대부분 분해되고 galactose가 생성되었다. 따라서 Bacillus coagulans NRR1207은 대두박의 비소화성분을 분해하는 생균제(Probiotics)로써 이용하여 식품 및 가축 사료 이용성 증대에 활용이 가능할 것으로 사료된다.

Bacillus licheniformis 분리균 2종의 α-Galactosidase 생산성과 효소특성 (Production and Characterization of α-Galactosidases from Two Bacillus licheniformis Isolates)

  • 전통 발효된장으로부터 α-galactosidase를 분비 생산하는 두 종류 Bacillus licheniformis 균주로 YB-1413과 YB-1414를 분리하여 당 이용능을 비롯한 생화학적 특성 및 16S rRNA 염기서열과 중합효소 연쇄반응에 의한 임의증폭 DNA 다형성의 유전학적 특성을 비교한 결과 두 균주는 매우 유사하였지만 동일 균주는 아닌 것으로 확인되었다. 탄소원으로 wheat bran을 사용하였을 때는 두 균주의 α-galactosidase 생산성이 증가하였으나, melibiose, raffinose 또는 sucrose는 효소 생산성이 급격하게 감소하였다. 질소원으로 yeast extract가 첨가된 배지에서 효소 생산성이 높았으며, YB-1413은 1.87 U/ml, YB-1414는 1.69 U/ml의 효소 생산성을 보였다. 이들 균주가 생산하는 α-galactosidases는 pH 6.0과 45℃에서 모두 최대활성을 보였으며, 낮은 농도의 ribose와 galalctose에 의해서도 효소활성이 급격하게 저해되었다. 한편 이들 효소는 대두와 콩과식품에 존재하는 항영양인자인 raffinose와 starchyose를 완전히 가수분해 하였는데, 이로 보아 분리균 YB-1413과 YB-1414는 콩 발효식품과 대두 사료의 영양가를 개선하는데 활용할 가치가 있다고 여겨진다.

영년초지에서 돈분뇨의 시용형태가 목초생산, 토양성분 및 용탈수의 성분에 미치는 영향 (Effects of Swine Manure on The Production of Forage, Soil Properties, and the Chemical Characteristics of Leaching Water in Mixed Grassland)

  • 본 연구에서는 돈분뇨의 처리형태가 초지의 생산성, 토양의 지력증진 효과 및 양분의 용탈량을 조사하여 친환경적 조사료를 생산하기 위하여 전라북도 축산 진흥연구소내의 초지시험포의 영년초지에서 2002년 10월부터 2005년 10월까지 3개년에 걸쳐 수행하였다. 시험구는 돈분뇨의 처리형태별로 화학비료 표준구, 돈분액비 100%+N 50%구 및 돈분퇴비 100%+N 50%구로 배치하여 수행하였다. 돈분액비 및 돈분퇴비의 시용에 따른 건물수량은 화학비료 시용구와 차이가 거의 없었다. 조단백질, NDF, ADF 및 TDN 함량은 돈분액비 및 돈분퇴비, 화학비료구간에 거의 차이를 나타내지 않았다. 처리별 토양성분 변화는 돈분퇴비 및 돈분액비구에서 화학비료구보다 유기물, 유효인산 함량, Ca 함량 등은 증가하는 경향을 보였다(p<0.05). 영년초지에서 화학비료, 돈분액비 및 돈분퇴비의 시용에 의한 용탈수중 $NO_3$-N 및 $NH_4$-N의 농도는 돈분뇨와 화학비료 시용에 의한 영향은 거의 없었으며 $NO_3$-N 및 $NH_4$-N 용탈량은 5 ppm 미만으로 변이가 크지 않았다.

천연물질을 이용한 반추동물 항생제 대체용 친환경 펠렛코팅오일의 개발 (Development of an Environmental Friend Pellet Coating Oil for Replacement of Antibiotics for Ruminant Aninmal)

  • This study investigated the effect of coated feed using mixed oil on rumen fermentation characteristics. Two experiments were conducted based on materials that were mixed. First, cashew nut and soybean oils were mixed with white mineral oil. And second, different plant extracts were mixed with white mineral oil. At first experiment, inclusion levels of mixed oil on diet (0.03%, 0.1%, 0.5%) were applied as variables. A coated diet was fermented with rumen inoculum according to in vitro rumen fermentation and its parameters were investigated. In the result of first experiment, no negative effects on rumen pH were found. Significantly decreased dry matter digestibility was detected at 0.5% treatment (P<0.05). Total gas productions in control and 0.03% were significantly greater than those of others (P<0.05). Significantly reduced methane productions were found in all treatments compared to the control (P<0.05). Inclusion of mixed oil did not affect on ammonia production. Total volatile fatty acid production was also not influenced by coating with mixed oils. Rumen fermentation parameters were greatly changed according to introduced plant extracts at second experiment. The significantly lowest and greatest ammonia productions were found at treatments with Ixeris dentata and Plantago asiatica, respectively (P<0.05). The significantly greatest acetate and propionate productions were detected at treatments with Crucuma longa and Zizyphus vulgaris 1, respectively (P<0.05). All treatments, except Chrysanthemum idicum, Euyale ferox seed, Moringa leaf and fruit and Zizyphus vulgaris 1, showed significantly increased total volatile fatty acid production compared to the control (P<0.05). Only Paeonia lactiflora showed significantly lesser gas production than the control (P<0.05). In methane production, Ceramium, Zizyphus, Paeonia, Agrimonia, Torilis, Mugwort, Foeniculum, Euphorbia, Taraxacum, Artemisia, Momordica, Curcuma and Moringa reduced methane significantly compared to the control (P<0.05).

Effect of Applying Molasses and Propionic Acid on Fermentation Quality and Aerobic Stability of Total Mixed Ration Silage Prepared with Whole-plant Corn in Tibet

  • The objective of this study was to evaluate the effects of molasses and propionic acid on the fermentation quality and aerobic stability of total mixed ration (TMR) silages prepared with whole-plant corn in Tibet. TMR (354 g/kg DM) was ensiled with four different treatments: no additive (control), molasses (M), propionic acid (P), and molasses+propionic acid (PM), in laboratory silos (250 mL) and fermented for 45 d. Silos were opened and silages were subjected to an aerobic stability test for 12 days, in which chemical and microbiological parameters of TMR silages were measured to determined the aerobic deterioration. After 45 d of ensiling, the four TMR silages were of good quality with low pH value and ammonia/total N (AN), and high lactic acid (LA) content and V-scores. M silage showed the highest (p<0.05) LA content and higher dry matter (DM) recovery than the control and P silages. P silage had lower (p<0.05) LA content than the control silage. During aerobic exposure, lactic acid contents decreased gradually in the control and M silages, while that of P and PM silages increased, and the peak values were observed after 9 d. M silage had similar yeast counts with the control silage (> $10^5$ cfu/g FM), however, it appeared to be more stable as indicated by a delayed pH value increase. P and PM silages showed fewer yeasts (< $10^5$ cfu/g FM) (p<0.05) and were more stable than the control and M silages during aerobic exposure. It was concluded that M application increased LA content and improved aerobic stability of TMR silage prepared with whole-plant corn in Tibet. P application inhibited lactic acid production during ensiling, and apparently preserved available sugars which stimulated large increases in lactic acid during aerobic exposure stage, which resulted in greater aerobic stability of TMR silage.
