Skin lightening effect of fermented Panax ginseng extract (자연삼 발효 추출물의 미백 활성에 대한 연구)

  • Panax ginseng is known for various pharmacological activities mainly due to saponins. Since minor saponins, generated by the decomposition of major saponins, generally exert higher activities than major saponins, the fermentation may increase the minor saponin contents in ginseng products. In this study, we tested fermented ginseng extract whether or not provide a safe cosmetic ingredient for whitening purpose. In this regard, fermented Ginseng extract was prepared and evaluated the inhibitory activity toward tyrosinase and the melanin synthesis suppression. The safety was tested via cell viability and toxicity test. The skin lightening effect was also evaluated by clinical study. The fermented Ginseng extract exerted higher activities in tyrosine inhibition and in suppressing melanin synthesis compared to Kojic acid and arbutin. In the clinical test, skin lightening effecte of the sample was clearly higher than vehicle or Vitamin C. We thus concluded that the fermented Ginseng extract may provide a safe cosmetic ingredient for skin lightening purpose.

Physicochemical Properties of Carrot Juice Containing Phellinus linteus Extract and Beet Extract Fermented by Leuconostoc mesenteroides SM (상황버섯 추출물과 비트 즙을 첨가한 당근주스 젖산발효 음료의 물리화학적 성질)

  • The physicochemical properties of carrot beverage fermented by Leuconstoc mesenteroides SM isolated from carrot juice were dependent upon the sucrose concentration and beet extract content. The addition of beet extract increased the consistency index and bioconversion of sucrose in the carrot juice fortified with Phellinus linteus extract, showing the highest consistency value at 2% beet extract. The polyphenol content and antioxidant effect of fermented carrot beverage were increased as the content of beet extract increased. Particularly, the consistency of fermented carrot beverage was greatly increased by the cold storage for 120 hr. After the cold storage for 2 weeks, fermented carrot beverage fortified beet extract showed the red-purple color with higher color value (a) and the viable cell counts of $10^9$ CFU/mL.

Changes Observed in Doenjang (Soybean Paste) Containing Fermented-Rhus verniciflua Extract During Aging (발효옻 추출물 첨가 된장의 숙성에 따른 변화)

  • Fermented-Rhus verniciflua (FRV) extract is increasingly used in fermented soy products, fermented vinegars, and certain alcoholic beverages. In this study, we investigated the effects of FRV extract on the physicochemical properties of doenjang (soybean paste). Addition of FRV extract to doenjang resulted in a 28.2-45.4% increase in the amino acid content and a 1.3- to 1.5-fold increase in the concentrations of glutamic acid, which imparts a savory flavor to doenjang. The concentration of biogenic amine (BA) of the sample containing the extract was 5.3-52.6% lower than that of the control. The major components of BA included tyramine (55.1-74.6%), followed by putrescine, spermidine, tryptamine, and cadaverine, in decreasing concentrations. The organic acid concentration of the sample containing the extract was 1.2-1.3-fold higher than that of the control. The total free sugar concentration was 163.4 mg/100 g in the control and 206.6-276.8 mg/100 g in the supplemented sample, showing a 1.3- to 1.9-fold increase as the addition of the extract.

Antimutagenic and Cytotoxicity Effects of Fermented Soybean Extract (발효콩 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was carried out to determine the antimutagenic and anticancer effects of fermented soybean using Ames test and cytotoxicity, respectively. The ethyl acetate fraction (200 g/plate) of fermented soybean in the Salmonella typhimurium TA100 strain showed 86.6% of inhibition rate against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG). In addition, the suppression of ethyl acetate fraction with same concentration of fermented soybean in the Salmonella typhimurium TA98 and TAI00 strains showed 82.4% and 90.8% inhibition against 3-amino-l,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-l), respectively. The cytotoxicity effects of fermented soybean against the cell lines with human lung carcinoma (A549), human gastric carcinoma (AGS) and human breast adenocarcinoma (MCF-7) were inhibited with the increase of the extract concentration. The treatment of 1.0 mg/mL ethyl acetate fraction of fermented soybean showed strong cytotoxicities of 71.6%, 91.5% and 80.7% against A549, AGS and MCF-7, respectively.

Effects of Hibiscus syriacus Extracts on Antioxidant Activities and Blood Circulation Improvement (무궁화 추출물의 항산화 활성 및 혈액 순환 개선에 미치는 효과)

  • This study was worked to investigate the antioxidant activity and the blood circulation improvement effect of two Hibiscus syriacus extracts divided into fermented H. syriacus extract and non-fermented H. syriacus extract. As the results of in vitro experiment, both of fermented H. syriacus extract and non-fermented H. syriacus extract had the DPPH free radical scavenging effect and the high contents of polyphenol and flavonoid known as antioxidant components in dose-dependent manner. Six weeks female C57BL/6 mice were divide into four groups and fed high diets for 28 days. Also fermented H. syriacus extract and non-fermented H. syriacus extract was daily intraperitoneal administration for 28 days at 2 mg/kg of body weight of mice. In vivo C57BL/6 mice experiment of normal diet group (NOR), high-fat diet group (CON), high-fat diet plus fermented H. syriacus extract group (FHS) and high-fat diet plus non-fermented H. syriacus extract group (NFHS), each of total cholesterol, triglyceride and low density lipoprotein (LDL) was decreased in both of FHS and NFHS groups, meanwhile high density lipoprotein (HDL) was increased (p<0.01). Conclusion, the results confirmed that H. syriacus containing the high contents of the antioxidant activity components had the significant effects of blood circulation improvement.

Antioxidant Activity of Solvent Extracts from Korean Fermented Tea (용매별 후발효차 추출물의 항산화 효과)

  • Major components and antioxidant activity of fermented tea were investigated. Contents of total flavonoid and total phenol showed the highest score in ethanol extract ($481.4\;{\mu}g/g$ and 33.5 g/100g), respectively. Their contents in hot water extract ($405.7\;{\mu}g/g$ and 23.6 g/100g) were lower than those of the others extracts. However, their contents of ethyl acetate extract($495.2\;{\mu}g/g$, 39.5 g/100g) from fermented tea stored for 1 year had the highest amount among all tested samples. Catechins of fermented tea were found to be EGC, GC, catechin, catechol and EGCG. However, they were not detected in hot water extract. Scavenging activities of DPPH free radical of all extracts from fermented tea were increased in proportion to their concentration and were $72.4\%$ for fermented tea and $80.9\%$ for fermented tea stored for 1 year. Reducing power of ethanol extract at $500\;{\mu}g/mL$ were 0.78 as OD value of 700 nm for fermented tea and 0.88 for fermented tea stored for 1 year, respectively. Antioxidant activities of ethyl acetate extracts at $3000\;{\mu}g/mL$ were $86.6\%$ for fermented tea and $73.7\%$ for fermented tea stored for 1 year, respectively.

Anti-inflammatory and Anti-oxidative Activities for Extract of Fermented Ligustrum japonicum Fruits (광나무 열매 발효 추출물의 항염 및 항산화 활성)

  • In this study, the anti-inflammatory and anti-oxidative activities were compared for the extracts of non-fermented Ligustrum japonicum fruits (LJF) and fermented counterparts. U se of Latilactobacillus curvatus (L. curvatus) and Weissella minor (W. minor), isolated from the Jeju Chromis notata, as fermented strains led to the extracts of LJF-LC and LJF-WM in this experiment. The yield of each fermented extract (LJF-LC and LJF-WM) was 40.5 ~ 46.0%, higher than 29.5% of non-fermented extract (LJF). As a result of an activity experiment using RAW 264.7 macrophages stimulated by lipopolysaccaride (LPS), it was confirmed that LJF-WM, a fermented extract, has an excellent effect of inhibiting NO production in a concentration-dependent manner without cytotoxicity. Upon the screening of DPPH and ABTS+ radical scavenging activities, the fermented LJF-LC and LJF-WM showed comparable to the non-fermented LJF. In the study of cell protection effect using HaCaT keratinocytes damaged by hydrogen peroxide (H2O2), the fermented LJF-WM indicated protective effect against oxidative stress. In addition, quantitative analysis of a major constituent salidroside by HPLC indicated about 15.6 mg/g for the LJF-LC and 13.9 mg/g for the LJF-WM, which were higher than that of non-fermented LJF (12.0 mg/g). Based on these results, it was suggested that the fermented extract from L. japonicum fruits could be used as a natural cosmetics material with anti-inflammatory and anti-oxidative effects.

  • Objectives : We investigated the anti-aging effects on skin with Saccharomyces fermented modified Kyungohkgo extract (SFKE). Methods : Fermented modified Kyungohkgo extracts by 14 different kinds of microorganism were prepared and most effective Saccharomyces fermented modified Kyungohkgo extract among these was selected for anti-aging agent. We measured various effects related to skin such as scavenging activity against free radical, cell proliferation, collagen synthesis and toxicity of SFKE were evaluated and compared with modified Kyungohkgo extract (KE). Results : Free-radical scavenging activity and cell proliferating ratio of SFKE were 2 and 8 times higher than those of KE. The SFKE could significantly increase the collagen synthetic ratio compared with KE treated group. SFKE showed no toxicity at all tested concentrations. Conclusions : The results of our study propose that SFKE have good anti-aging effects on skin.

  • The purpose of this study was to investigate the antimicrobial activity, antimutagenic and anticancer effects and dyeing properties of the fermented indigo extract. The methanol extract of fermented indigo showed a strong inhibition effect on Trich. mentagrophytes and antimutagenic activities against aflatoxin B$_1$(AFB$_1$) in the Ames test using Salmonella typhimurium TA 100. We also found in vitro anticancer effects of the methanol extract of fermented indigo and it was evaluated by using Clone M-3 mouse melanoma cells and A431 human epidermoid carcinoma cells and exerted little cytotoxity against 3T3-L1 embryo fibroblast cell. In the relationship between the K/S values of cotton and silk fabrics dyed with fermented indigo and dyeing repeating times, the K/S values became higher as the repeating times were increased. The K/S values were high when the fabrics were dyed at low temperture. The K/S values of cotton fabrics were higher than those of silk fabrics. Changes of surface color of silk fabrics were higher than those of cotton fabrics after water fastness test, laundering, irradiation and treatment of acidic and alkaline perspiration.

  • In this study, we evaluated antioxidative effects and skin whitening effects of extract of Caragana sinica fermented by S. cerevisiae KCTC 7913. At first, Caragana sinica was fermented via inoculation of Saccharomyces cerevisiae KCTC 7913 and then extracted for fermented C. sinica. It has shown that more increased of melanogenesis inhibition activity in a dose-dependent manner on B16F10 melanoma cells and elevated the amount of resveratrol contents by HPLC analysis than non-fermented. Furthermore, the extract of fermented C. sinica was inhibitory effects against tyrosinase, a key enzyme of melanogenesis pathway, more than non-fermented C. sinica extract. And it did not show the skin irritation. Therefore, fermented C. sinica extracts might be used as safe cosmetic ingredients.
