Growth Inhibitory Effect of Fermented Kimchi on Food-borne Pathogens

  • The effect of kimchi, traditional Korean fermented vegetables, on inactivating food-borne pathogens and the kimchi factors affecting the antimicrobial activity were investigated. More cells of Listeria monocytogenes, Staphylococcus aureus, Escherichia coli O157:H7, and Salmonella typhimurium were inactivated in the kimchi that had low pH and high titratable acidity. Of the raw ingredients in kimchi, raw garlic showed the strongest antimicrobial activity against the pathogens. When kimchi was fermented at 0, 4, 10, or $20^{\circ}C$ to pH 4.4, higher kimchi fermentation temperature resulted in higher titratable acidity. The greatest inactivation of S. typhimurium occurred in kimchi fermented at $20^{\circ}C$, while L. monocytogenes were inactivated in kimchi fermented at $0^{\circ}C$ in situ. This study showed that appropriately fermented kimchi can inactivate various food-borne pathogens and that the fermentation temperature of the kimchi is an important factor in determining the ability of the kimchi to inactivate specific pathogens. Lactic acid bacteria (LAB) multiplication and organic acids produced according to LAB metabolism play a role in inactivating food-borne pathogens in kimchi.

FUNCTIONAL PROPERTIES OF SAENGSIK (SUBSTITUTE MEAL) FORMULATED WITH UNCOOKED CEREALS FERMENTED BY LACTIC ACID BACTERIA

  • The objective of this study was to improve the qualities of Saengsik (substitute meal formulated with uncooked cereal powder) such as functionality, safety and sensory preference by using Lactic Acid Bacteria (LAB). The optimum formulation of fermented Saengsik (substitute meal) was verified as $40\%$ of fermented brown rice, $13\%$ of fermented job tears, $5\%$ of yellow soybean, $5\%$ of waxy rice, $5\%$ of sorghum, $3\%$ of Seumok bean, $1\%$ of sesame, $3\%$ of barley, $2\%$ of red bean, $5\%$ of prosomillet, $2\%$ of foxtail millet, $9%$ of black rice, $3\%$ of buckwheat, $1\%$ of chestnut, $2\%$ of pumpkin, $3\%$ of peanut. Fermented Saengsik decreased significantly serum glucose in STZ-induced diabetic rats. However, serum lipid levels were not affected by fermented Saengsik. In rats fed with high-fat diet, serum triglyceride level was slightly decreased and the contents of liver triglyceride were significantly decreased by supplementation of fermented Saengsik.

블루베리 유산균 발효물의 이화학적 특성 및 저장안정성 (Physicochemical properties and storage stability of blueberry fermented by lactic acid bacteria)

  • 본 연구에서는 다양한 생리활성을 가진다고 알려져 있는 블루베리의 식품 산업에서의 이용 증대를 목적으로 블루베리 유산균 발효물을 제조하였으며 이화학적 특성 및 저장 안정성을 조사하였다. 생균수는 발효 72시간에서 가장 높은 5.96 log CFU/mL를 나타내었으며, pH 및 산도는 각각 발효 시간 경과에 따라 유의적으로 감소 및 증가하였다. 총 안토시아닌 및 cyanidin-3-glucoside(C3G) 함량은 발효 72시간에서 각각 100 g 당 31.52 mg 및 5.41 mg으로 가장 높은 함량을 나타내어 생균수, 총 안토시아닌 및 C3G 함량을 고려하여 72시간을 최적 발효 구간으로 선정하였다. 블루베리 발효물의 L값 및 a값은 발효 후 감소하고, b값 및 ${\Delta}E$값은 증가하였으며, 총 폴리페놀 및 총 플라보노이드 함량은 100 g당 2.21 g 및 0.91 g으로 발효 전에 비해 높게 나타났다. 항산화활성은 DPPH 및 superoxide radical 소거 활성에서는 블루베리 발효물에서 각각 30.74% 및 52.76%를 나타내어 무발효물 보다 높은 소거활성을 나타내었으며, FRAP에서도 $256.42 {\mu}M$을 나타내어 블루베리 발효물에서 높은 항산화 활성을 나타내었다. 블루베리 발효물의 저장안정성은 총 안토시아닌 및 C3G 함량의 감소율로 확인하였는데, 42일 경과 후 48.03% 및 38.71% 감소하여 무발효물에 비해 우수하였다. 따라서, 블루베리 발효물은 유용물질 증가로 인한 안정성 향상을 통해 식품가공 산업에서의 활용증진이 기대된다.

Quality Characteristics of Low-Salt Myungran Jeotkal Fermented by Vegetable-Origin Lactic Acid Bacteria and Salt from Deep Sea Water

  • In this study, the physicochemical and sensory characteristics of low-salt Myungran jeotkal (Alaskan pollock roe) were evaluated after fermentation at $4^{\circ}C$ and $20^{\circ}C$ with or without the addition of deep sea water, salt from deep sea water, and vegetable-origin lactic acid bacteria (Lactobacillus fermentum JS, LBF). When fermented at $20^{\circ}C$, the addition of LBF to Myungran jeotkal resulted in a slow increase in lactic acid content, followed by an abrupt increase after five days of fermentation. However, when fermented at $4^{\circ}C$, the lactic acid content did not change significantly. Further, when Myungran jeotkal fermented at $4^{\circ}C$, the pH decreased as lactic acid production increased. The salinity of Myungran jeotkal fermented at $4^{\circ}C$ and $20^{\circ}C$ was 7% and was not affected by fermentation period. When fermented at $20^{\circ}C$, volatile basic nitrogen and amino nitrogen contents increased with increasing duration of fermentation. Further, volatile acid content decreased, however, the content of amino nitrogen increased after 11 days of fermentation with LBF and no salt effects were observed. When fermented at $20^{\circ}C$ for 13 days, preference (sensory evaluation) was the highest in all experimental groups after 9 days of fermentation, and then decreased as the fermentation period increased. The free amino acid content was highest (1,648.8 mg/100 g) in Myungran jeotkal when sun-dried salt and LBF were added, 2.3 times higher than in the control.

박테리오신 생성 젖산균을 이용한 항충치 활성을 지닌 발효이온음료 개발 (Development of Fermented Isotonic Beverage with Anticariogenic Activity using Bacteriocin-Producing Lactic Acid Bacteria)

  • 박테리오신 생성 젖산균을 이용하여 과실채소 혼합발효액을 제조하고, 발효액의 이화학적 특성과 항균특성을 조사하여 항균 이온음료로서의 가능성을 고찰하였다. 박테리오신 생성 젖산균을 이용한 발효시 발효종균으로 Leu. mesenteroides와 Lc. lactis ATCC l1454를 동시 접종하였을때는박테리오신이 생성되지 않았으며, Leu mesenteroides에 의한 젖산발효 후에 pH를 조정하고 박테리오신 생성균을 접종하였을 때 적정량의 산과 항균물질이 생성되었다. 발효액의 당함량과 무기염류를 분석한 결과, 발효액에는 시판되고 있는 이온음료에 비해 Na과 P의 함량은 높은 편이나, K,Ca, Mg 등의 농도 차이는 크지 않은 것으로 나타났으며, 당함량도 약간 낮은 것으로 나타났다. 발효액은 충치 원인균인 Streptococcus mutans에 대한 안정된 항균효과를 지니고 있으며, 발효액의 색상은 저장 중에도 안정된 것으로 나타났다. 따라서 과실채소 혼합발효액의 무기질 발란스와 기호도를 고려한 당농도 조절 등의 보완이 이루어진다면 우리고유의 이온음료로 개발 가능성이 있다고 판단된다.

청국장 제조과정에서 Bacterial Phytase에 의한 Phytic Acid의 분해 (Degradation of Phytic acid in Chungkookjang Fermented with Phytase-producing Bacteria)

  • 재래식 발효 청국장으로부터 분리한 8균주 중 phytase 생산성이 높은 3균주는 Bacillus subtilis, Bacillus licheniformis, Staphylococcus epidermidis로 동정되었다. 이들 phytase 생산균주를 단독 또는 혼합균주로 하여 35-$40^{\circ}C$와 pH 7.0의 최적조건에서 5일 동안 청국장을 발효시켰을 때 최대의 phytase 생산성을 보였으며 glutamic acid와 leucine 등의 일부 아미노산과 riboflavin의 함량이 증가되고 phytic acid 의 분해율도 재래식 청국장에서 보다 훨씬 높았다. 특히 B.subtilis와 B.licheniformis의 혼합균주로 발효시켰을 때 phytic acid 함량의 감소가 현저하였다.

RecA 유전자 특이적 PCR을 이용한 전통 침채류 유래 유산균의 검출 (PCR-Based Detection of Lactic Acid Bacteria in Korean Fermented Vegetables with recA Gene Targeted Species-Specific Primers)

  • recA 유전자를 특이적으로 증폭하는 PCR을 이용하여 우리나라 전통 침채류 발효에 관여하는 유산균의 다양성을 검토해 보았다. 김치에서 많이 검출되는 유산균 7종 및 대조군으로 Lactobacillus acidophilus를 검출할 수 있는 특이적 PCR primer pair을 이용하여 전통 침채류 5증(갓김치, 동치미, 배추김치, 오이소박이, 총각김치로부터 추출한 DNA를 template로 PCR을 수행한 결과, 5종의 침채류 모두에서 Lactobacillus plantarum과 Lactobacillus sakei가 검출되었지만, Lactobacillus paraplantarum, Lactobacillus pentosus와 대조군인 Lb. acidophilus는 검출되지 않았다. Lactobacillus brevis와 Leuconostoc citreum은 배추김치에서만 검출되었으며, Leuconostoc mesenteroides의 경우 갓김치, 동치미, 배추김치, 오이소박이에서 검출되었다. 주재료의 종류에 따라서 발효에 관여하는 유산균은 차이가 있는 것으로 추정되며, Lb. plantarum과 Lb. sakei가 우리나라 침채류 발효에 가장 널리 관여하는 것으로 사료된다.

Effects of lactic acid bacteria fermented feed and three types of lactic acid bacteria (L. plantarum, L. acidophilus, B. animalis) on intestinal microbiota and T cell polarization (Th1, Th2, Th17, Treg) in the intestinal lymph nodes and spleens of rats

  • Objective: In this study, we investigated the effects of Rubus coreanus-derived lactic acid bacteria (LAB) fermented feed (RC-LAB fermented feed) and three types of LAB (Lactobacillus plantarum, Lactobacillus acidophilus, Bifidobacterium animalis) on the expression of transcription factors and cytokines in Th1, Th2, Th17, and Treg cells in the intestinal lymph nodes and spleens of rats. In addition, the effect on intestinal microbiota composition and body weight was investigated. Methods: Five-week-old male rats were assigned to five treatments and eight replicates. The expression of transcription factors and cytokines of Th1, Th2, Th17, and Treg cells in the intestinal lymph nodes and spleens was analyzed using real-time reverse transcriptase polymerase chain reaction assays. Intestinal tract microbiota compositions were analyzed by next-generation sequencing and quantitative polymerase chain reaction assays. Results: RC-LAB fermented feed and three types of LAB increased the expression of transcription factors and cytokines in Th1, Treg cells and Galectin-9, but decreased in Th2 and Th17 cells. In addition, the intestinal microbiota composition changed, the body weight and Firmicutes to Bacteroidetes (F/B) ratio decreased, and the relative abundance of LAB increased. Conclusion: LAB fermented feed and three types of LAB showed an immune modulation effect by inducing T cell polarization and increased LAB in the intestinal microbiota.

Productivity increase and odor reduction effect of fermented barley sprout extract in broiler farms

  • The current study examined the impact of fermented barley sprout extract prepared using lactic acid bacteria (Lactobacillus sp.) in decreasing odor and increasing livestock productivity and measured the difference in the polyphenol and flavonoid contents of the extract after fermentation. Furthermore, the effectiveness of fermented barley sprout extract was evaluated through order level and production index of livestock by supplying it to a broiler house. The results showed that with fermented barley sprout extract, the polyphenol and flavonoid contents were increased significantly by 174% and 562%, respectively. When the extract was applied as an additive to drinking water in the test farm, the productivity improved by about 10%, the mortality rate was reduced by about 66%, and there was a significant decrease in odor by about 80%. Compared with the control group, the production index increased by about 21%, the feed requirement decreased by about 8%, the odor showed a decrease in the NH3 level, and no other gas was detected. It was observed that lactic acid bacteria settle in the intestine, suppress the proliferation of bacteria that cause diarrhea and enteritis, and help digestion. The lactic acid bacteria effectively remove bad odor gases such as NH3, Amines, H2S and CH4S. Such odor reduction improves productivity. Our findings provide valuable information for quality water supply, production optimization and livestock management.

Effect of Different Salt Concentrations and Temperatures on the Lactic Acid Fermentation of Radish Juice

  • The characteristics of natural lactic acid fermentation of radish juice were investigated at different salt concentrations (0~2%) and temperatures (10~3$0^{\circ}C$). Major lactic acid bacteria isolated from the radish juice fermented at 2% slat concentration were Leuconostoc mesenteroides, Lactobacillus plantarum, and Lactobacillus brevis. The percentage of lactic acid bacteria against total microbe in the fermented radish juice was over 80% at 0~1% salt concentrations, suggesting the possibility of fermentation even at low salt concentration, but was still active even at 1$0^{\circ}C$. The time to reach pH 4.0 during fermentation of juice of 1% salt concentration was 281~301 hrs at 1$0^{\circ}C$ and 50-73 hrs at 3$0^{\circ}C$. The concentrations of sucrose and glucose in the fermented juice were low at high temperatures and were the lowest at a 1.0% salt concentration. However, the content of mannitol showed the opposite trend. Although sour taste, ripened taste, and acidic odor of the fermented juice showed no significant differences among various temperatures and salt concentrations, sensory values of ripened taste and sour acidic were high at high temperatures. The overall quality was the best at 1.0% salt concentration, irrespective of the temperature.
