Changes in Some Physico-Chemical Properties and γ-Aminobutyric Acid Content of Kimchi during Fermentation and Storage

  • In this study, we investigated changes in some physico-chemical and biochemical properties of Kimchi during fermentation and storage. After fermenting Kimchi at $15^{\circ}C$ for 23, 36, 40, 44, and 60 hrs during the first week, we stored it at $-1^{\circ}C$ in a Kimchi refrigerator until the end of 8th week. The pH of samples fermented for 36 hr, 40 hr, 44 hr and 60 hr sharply decreased during the first seven days and then slowly decreased. Acidities of samples fermented for 36 hr, 40 hr and 44 hr sharply increased for the first seven days. According to measured changes of lactic acid bacteria number, samples fermented for 60 hr revealed the largest augmentation in the number of lactobacilli for the first seven days. The $\gamma$-aminobutyric acid (GABA) content of the sample fermented for 40 hr was the most superior, with an early increase and maintenance of GABA content, which maintained a maximum 20 mg per 100 g of Kimchi sample on the seventh, fourteenth, and twenty eighth days. These results suggest that relatively enhanced levels of GABA in Kimchi samples can be produced and maintained by controlling the fermentation and storage processes, as with the 40 hr fermented sample conditions.

Antimicrobial activity of fermented Maillard reaction products, novel milk-derived material, made by whey protein and Lactobacillus rhamnosus and Lactobacillus gasseri on Clostridium perfringens

  • Objective: The objective of this study was to evaluate the antimicrobial effects of fermented Maillard reaction products made by milk proteins (FMRPs) on Clostridium perfringens (C. perfringens), and to elucidate antimicrobial modes of FMRPs on the bacteria, using physiological and morphological analyses. Methods: Antimicrobial effects of FMRPs (whey protein plus galactose fermented by Lactobacillus rhamnosus [L. rhamnosus] 4B15 [Gal-4B15] or Lactobacillus gasseri 4M13 [Gal-4M13], and whey protein plus glucose fermented by L. rhamnosus 4B15 [Glc-4B15] or L. gasseri 4M13 [Glc-4M13]) on C. perfringens were tested by examining growth responses of the pathogen. Iron chelation activity analysis, propidium iodide uptake assay, and morphological analysis with field emission scanning electron microscope (FE-SEM) were conducted to elucidate the modes of antimicrobial activities of FMRPs. Results: When C. perfringens were exposed to the FMRPs, C. perfringens cell counts were decreased (p<0.05) by the all tested FMRPs; iron chelation activities by FMRPs, except for Glc-4M13. Propidium iodide uptake assay indicate that bacterial cellular damage increased in all FMRPs-treated C. perfringens, and it was observed by FE-SEM. Conclusion: These results indicate that the FMRPs can destroy C. perfringens by iron chelation and cell membrane damage. Thus, it could be used in dairy products, and controlling intestinal C. perfringens.

Biogenic Amine Degradation by Bacillus Species Isolated from Traditional Fermented Soybean Food and Detection of Decarboxylase-Related Genes

  • Biogenic amines in some food products present considerable toxicological risks as potential human carcinogens when consumed in excess concentrations. In this study, we investigated the degradation of the biogenic amines histamine and tyramine and the presence of genes encoding histidine and tyrosine decarboxylases and amine oxidase in Bacillus species isolated from fermented soybean food. No expression of histidine and tyrosine decarboxylase genes (hdc and tydc) were detected in the Bacillus species isolated (B. subtilis HJ0-6, B. subtilis D'J53-4, and B. idriensis RD13-10), although substantial levels of amine oxidase gene (yobN) expression were observed. We also found that the three selected strains, as non-biogenic amineproducing bacteria, were significantly able to degrade the biogenic amines histamine and tyramine. These results indicated that the selected Bacillus species could be used as a starter culture for the control of biogenic amine accumulation and degradation in food. Our study findings also provided the basis for the development of potential biological control agents against these biogenic amines for use in the food preservation and food safety sectors.

자리젓 중 N-Nitrosamine 생성에 관한 연구 (Studies on the Formatiion of N-Nitrosamine in the Salt-Fermented Damsel fish Chromis notatus)

  • N-Nitrosamines have been known to be strong carcinogens and are formed by the reaction of nitrous acid with amines. In this experiment the changes in the contents of nitrate nitrite trimethylaminoxide(TMAO) trimethylamine(TMA) and dimethylamine(DMA) during femen-tation of damsel fish were analyzed periodically and N-nitrosamines in a commercial products. N-Nitrosamines were determined by mineral oil distillation methods using gas chromatography-thermal energy anlyzer(GC-TEA) in a commerical product. Nitrate nitrite and amines were quantitate by colorimetric methods. Level of nitrate-N were gradually decreased but nitrite-n was not detected or trace. Contents of dimethlamine(DMA) and trimethlamine were mar-kedly increased while trimethylaminoxide nitrogen was decreased during the fermentation of damsel fish. The change of pH was in the ranges of 5,5-7.0 during fermentation of salted damsel fish. It was out of the optimum pH(3.0-3.4) for the formation of nitrosamine. N-Nitrosamines were not detected in salt-fermented damsel fist but much N-nitrosodimethyla-mine(NDMA) could be detected in salt-fermented damsel fish after adding 0.05M NaNo2 in the acidic condition. The identifaction of NDMA in it was confirmed by mass spectrophotometry. Nitrate decrea-sed during the fermentation of damsel fish. however nitrite was trace level and nitrosamines were not formed in its. This could be supposed that it was due to the rapid consumption of nitrite by amino acid and bacteria.

Optimization of Enzymatic Pretreatment for the Production of Fermented Ginseng using Leaves, Stems and Roots of Ginseng

  • This study sought to optimize the extraction and enzymatic treatment conditions of Panax ginseng leaves, stems, and roots for the production of fermented ginseng. The optimization enhanced the extraction of total saccharide, a nutrient and growth-activating factor for Lactobacillus bacteria. The hydrolysis of ginseng leaves, stems, and roots was tested with eight enzymes (Pentopan, Promozyme, Celluclast, Ultraflo, Pectinex, Ceremix, Viscozyme, and Tunicase). The enzymatic hydrolysis conditions were statistically optimized by the experimental design. Optimal particle size of ginseng raw material was <0.15 mm, and optimal hydrolysis occurred at a pH of 5.0-5.5, a reaction temperature of 55-$60^{\circ}C$, a Ceremix concentration of 1%, and a reaction time of 2 hr. Ceremix produced the highest dry matter yield and total saccharide extraction. Ginseng leaves were found to be the most suitable raw material for the production of fermented ginseng because they have higher carbohydrate and crude saponin contents than ginseng roots.

Physiological Characteristics and Immunomodulating Activity by Lactobacillus paracasei subsp. paracasei BFI46 Isolated from New-Born Infant Feces

  • In order to develop a new starter for fermented milk, Lactobacillus paracasei subsp. paracasei BFI46 (BFI46) obtained from new-born infant feces was investigated for physiological characteristics. Good immunomodulating activity was evident compared with commercial lactic acid bacteria starter cultures. The optimum growth temperature of BFI46 was $40^{\circ}C$ with 12 h required to reach pH 4.3. Testing with 13 different antibiotics revealed greatest sensitivity of BFI46 to penicillin- G and chloramphenicol, and heightened resistance to neomycin, kanamycin and polymyxin. BFI46 displayed higher esterase activities compared to 18 other enzymes, was comparatively tolerant to bile juice and able to survive at pH 2 for 3 h, and displayed high resistance against Escherichia coli and Salmonella Typhimurium with a survival rate of 57.14% and 96.36%, respectively. The results indicate that BFI46 could be an excellent starter culture for fermented milk with high level of immunomodulating activity.

Lactobacillus plantarum으로 발효한 천년초 선인장 열매의 발효특성 및 in vitro 항비만 효과 (Fermentation Characteristics and Anti-Obesity Effects of Cheonnyuncho (Oputia Humifusa) Fruit Fermented with Lactobacillus plantarum in 3T3-L1 Cells)

  • This study was conducted to investigate the fermentation characteristics and anti-obesity effects of Cheonnyuncho (Oputia Humifusa) fruit fermented with Lactobacillus plantarum SRCM 100320 (FC). The pH gradually decreased from 4.77 to 3.63 at 72 hours during fermentation. Counts of lactic acid bacteria, total polyphenol and flavonoid contents and DPPH scavenging activity were the highest at 48 hours during fermentation. Evaluation of the composition of polyphenols and flavonoids of FC fermented at 48 hours by HPLC revealed hyperoside (quercetin 3-galactoside), luteolin and kaempferol were the major components. The hyperoside content of FC was decreased, while the luteolin and kaempferol contents of FC were increased compared to unfermented Cheonnyuncho (NFC). Evaluation of the anti-obesity effects of FC in 3T3L-1 cells revealed that the accumulation of triglyceride was inhibited by about 27.3% in cells treated with FC at $150{\mu}g/mL$ compared to NFC. These findings indicate FC has the potential for use as an anti-obesity material.

Effects of Autochthonous Yeast Cultures on Some Quality Characteristics of Traditional Turkish Fermented Sausage "Sucuk"

  • The objective of this study was to determine the effects of yeast cultures (Candida zeylanoides and Debaryomyces hansenii) isolated from traditionally dry fermented Turkish sucuks, on some physicochemical and microbiological properties of the product. Eight different batches of the sucuks were produced by the inoculation of yeast and lactic acid bacteria (LAB) cultures (Lactobacillus curvatus, Lactobacillus plantarum and Lactobacillus sakei) in different combinations. The sucuks were ripened for 12 days and analyzed at 1st, 6th, and 12th days of ripening. Percent moisture content, pH, water activity (aw) and residual nitrite values of the sucuk inoculated with the yeast cultures were higher at the end of the ripening. The use of yeast cultures decreased hardness, gumminess, and chewiness values of the sucuk while increased adhesiveness values. Major volatile groups were aldehydes, terpenes, and sulphur compounds in the sucuk samples. The most noticeable results were for sensory properties of the sucuk that were positively improved by the yeast cultures.

국내산 시판 어리굴(Crassostrea gigas)젓의 미생물학적 평가 (Microbiological Evaluation of Commercial Eorigul-jeot, Salt-fermented Oyster Crassostrea gigas with Seasoning, Produced in Korea)

  • We investigated the quality of 10 commercial eorigul-jeot, salt-fermented oysters with seasoning, by measuring their chemical composition and bacterial concentrations. The Eorigul-jeot had 5.07-6.06 pH (mean, 5.63), of 1.92-4.74% salinity (mean, 3.36%), 7.01-14.70 mg/100 g volatile basic nitrogen (mean, 11.91 mg/100 g), 139.22-267.11 mg/100 g amino acid nitrogen (mean, 212.69 mg/100 g), and 1.02-1.65 g/100 g total acidity (mean, 1.24 g/100 g). The total viable and lactic acid bacterial counts were 5.7×104-8.7×105 and 2.7×103-2.0×105 CFU/g, respectively, and fecal coliform was detected in only one Eorigul-jeot sample. Bacillus cereus and Clostridium perfringens were detected in two samples, and all Eorigul-jeot samples were negative for Staphylococcus aureus and Vibrio parahaemolyticus. These results strongly suggest the need to monitor food-poisoning bacteria in commercial Eorigul-jeot to ensure consumer health.

젖산균과 청국균으로 발효시킨 용안육의 항산화, 항암효과 및 일산화질소생성 (Antioxidant, Anticancer Activities and Nitric Oxide Production of Euphoria longana Fermented with Lactic Acid Bacteria and Bacillus subtilis)

  • 태국산과 베트남산의 용안육 원료 및 그 발효물의 전자공여능, 일산화질소(NO) 생성효과, 인체암 세포주인 자궁경부암세포(HeLa) 및 간암세포(HepG2)의 항암활성 효과를 조사하였다. 원료 용안육의 총 유기산은 태국산 용안육(473.49 mg/g)이 베트남산(148.48 mg/g) 용안육에 비하여 3.2배 높은 함량을 나타내었고, 전체 유기산의 각각 89.2%와 86.8%를 나타내었으며, 주요 유기산은 formic acid와 malic acid였다. 원료 용안육의 총 유리당은 베트남산(434.63 mg/g)이 태국산(378.77 mg/g)에 비하여 1.2배 높은 함량을 나타내었으며, 주요한 유리당은 sucrose, glucose와 fructose였다. 단핵세포 세포주인 RAW264.7 세포에서 발효용안육의 NO 생성은 베트남산이 태국산에 비하여 약간 낮은 농도를 나타내었고, 원료 용안육과 비교하면 고농도 보다 저농도 추출물 첨가에서 그 함량의 차이가 크게 나타났다. 원료 용안육의 추출물 $600\;{\mu}g$에서 전자공여능은 베트남산이 $41.72{\pm}3.59%$로 태국산 $30.20{\pm}4.8%$에 비하여 1.4배 높았으며, 발효 용안육의 전자공여능은 B. subtilis로 발효시킨 베트남산 용안육의 추출물 $600\;{\mu}g$에서 $43.57{\pm}2.07%$로 가장 높게 나타났다. 원료 용안육의 자궁경부암세포에 대한 항암활성은 베트남산이 태국산 용안육보다 대체로 비슷하였지만, 간암세포의 경우는 태국산 용안육 추출물 $200\;{\mu}g$ 농도에서 저해율이 $46.13{\pm}4.80%$로 베트남산 $33.07{\pm}0.92%$보다 높게 나타났다. 그리고 발효 용안육의 자궁경부암세포 및 간암세포에 대한 항암활성은 각각 B. subtilis로 발효시킨 베트남산 및 태국산 용안육 추출물 $200\;{\mu}g$에서 저해율 $39.21{\pm}1.46%$$48.07{\pm}1.63%$로 가장 높게 나타났다.
