발효천마의 총 폴리페놀 함량 변화 및 라디칼 소거능 (Change of Total Polyphenol Content of Fermented Gastrodia elata Blume and Radical Scavenging)

  • 효모를 이용한 발효천마와 비발효천마의 폴리페놀 함량과과 지표성분인 p-hydroxybenzyl alcohol(HBA) 함량 변화 및 항산화활성을 비교 분석하고자 본 연구를 수행하였다. 발효 전과 발효 후의 총 폴리페놀 함량은 각각 108.65, 389.99 mg/mL으로 나타내었으며, 지표성분인 HBA 함량은 발효 전 2.46 mg/g에서 발효 후 7.94 mg/g로 증가되었다. 비발효천마추출물과 발효천마추출물의 DPPH, ABTS, superoxide 라디칼 소거능을 비교한 결과 추출물의 농도가 증가할수록 비발효천마추출물보다 발효천마추출물의 활성능이 높음을 확인할 수 있었다. 특히, superoxide 라디칼 소거능을 비교한 결과 발효천마추출물에서 20배 이상의 활성능이 증가되었다. 이상의 결과로부터 건천마를 발효할 경우 전자공여능 및 라디칼 소거능이 증가됨을 확인하였으며 향후 항산화소재로서의 이용가능성을 확인하였다.

  • Citrus sunki Hort. ex Tanaka is one of several Jeju-native citrus fruits. A number of biological properties for citrus fruits have been suggested, however little is known about those of C. sunki. The present study tested whether fermented product of C. sunki peel (FSP) might affect the activities of enzymes regulating ethanol metabolism. Effects on body weight gain as well as on fatty liver formation were also investigated. The activities of alcohol dehydrogenase and aldehyde dehydrogenase were stimulated remarkably by FSP. Excessive ethanol-induced cytotoxicity was also prevented by FSP in HepG2 cells. FSP decreased the weight gain and fatty liver formation induced by a high-fat diet in mice. From these results, FSP might be a potent source of neutraceuticals useful for preventing ethanol-induced health problems.

솔잎 발효액(醱酵液)의 항산화효과(抗酸化效果) (Antioxidative effects of the fermented extract of Pinus densiflora)

  • This study was performed to investigate the effects of fermented extract of Pinus densiflora (FPD) on oxygen radicals and $H_2O_2$-induced damage. The results are as follows: 1. The 1.1-diphenyl-2-picrylhydrazyl (DPPH) free radicals were considerably reduced by FPD and $IC_{50}$ value was showed the concentration of 20 ㎍/㎖. 2. The cytotoxicity did not observe by FPD treatment in A548 cells. 3. The $H_2O_2$-induced cell damage was recovered by FPD pretreatment in A549 cells. These results suggest that FPD, as a natural antioxidant, has scavenging effect of free radicals and protection effect from $H_2O_2$-induced cytotoxicity.

GBCK25, fermented ginseng, attenuates cardiac dysfunction in high fat diet-induced obese mice

  • The fermentation of medicinal herbs facilitated by microbes is assumed to exert promising therapeutic efficacy on the absorption, bioavailability, and pharmacological effects by speeding up the making or conversion of active constituents into their metabolites. We examined the cardioprotective potential of fermented ginseng, GBCK25, against high-fat diet (HFD)-induced metabolic and functional illnesses as following the essential analysis such as electrocardiographic parameters, alterations of body and organ weights, and echocardiographic studies. The results exhibited that body weights were significantly reduced and the gain of different organ weights were partly eased by GBCK25 treatment. Echocardiography results proposed the amelioration of heart function through normalized levels of left ventricle systolic pressure, ejection fraction, and fractional shortening. These outcomes deliver straight confirmation that GBCK25 could be a potential nutraceutical source for the relief of HFD-induced obesity mediated cardiac dysfunctions.

황칠 발효액을 첨가한 된장의 품질 특성 (Quality Characteristics of Doenjang Prepared with Fermented Hwangchil (Dendropanax morbifera) Extract)

  • 황칠 추출물에 미생물 균주를 달리 접종하여 제조한 황칠 발효액을 스타터로 첨가하여 제조한 된장의 발효 및 품질 특성을 조사하였다. 황칠 발효액 접종 시 총균수가 log 8 CFU/mL 이상으로 스타터로서 적용 가능성을 확인하였고, 발효 후에는 염도 8%, 10%의 경우 황칠 발효액을 첨가하여 제조한 된장의 총균수가 황칠 발효액을 첨가하지 않은 된장보다 낮았다. pH의 경우 염도 8%, 10% 된장이 12% 된장에 비하여 낮은 pH 값을 보였으며, 이와 반대로 산도는 12% 된장이 낮았다. 총유리아미노산 함량은 발효 후 2,162.5~2,872.2 mg/L로 발효 전보다 함량이 증가하였고 염도가 낮아질수록 총 유리아미노산 함량은 높았으며, Bacillus methylotrophicus S8을 이용하여 제조한 저염 된장(염도 8%, 10%)의 경우 같은 염도에서 다른 균주를 이용한 된장보다 glutamic acid 함량이 유의적으로 높았다. 황칠 발효액을 첨가하여 제조한 20일 숙성한 된장의 아미노태 질소는 413.4~700.7 mg%로 염도가 낮을수록 높았으며, 특히 B. methylotrophicus S8을 이용하여 제조한 된장에서 높은 함량을 보였다. 관능평가 결과 B. methylotrophicus S8과 Saccharomyces bayanus를 접종하여 제조한 된장에서 좋은 평가를 보였으며 8% 염도의 된장에서 B. methylotrophicus S8의 전체적 기호도가 가장 높게 평가되었다.

유산발효에 의한 발효한약의 기능분석 (Functionality Analysis of Korean Medicine Fermented by Lactobacillus Strains)

  • 본 연구에서는 중풍, 고혈압, 동맥경화, 염증성 피부질환에 사용되고 있으며, 다양한 질환들에서 연구되어 있는 방풍통성산을 선정하여 4가지 약재를 가미한 후 기존의 기능성 물질과 Lactobacillus 균주에 의해 생성되거나 증가된 기능성 물질을 확인하고자 하였다. 한약을 이용한 복합배양은 공시균주의 수 및 접종시기를 달리하여 여러 조합으로 수행하였다. 전자공여능 및 superoxide dismutase 유사 활성 측정을 통해 한약의 항산화 활성은 각각 31.7%와 36.3%로 나타났으며, 복합배양을 통해 발효된 한약의 항산화 활성은 각각 77% 이상, 42% 이상으로 나타나 한약보다 항산화 활성이 더 높게 나타났다. 항균 활성은 한약의 경우 모든 시험균주에 항균 활성이 나타나지 않았으나, 복합배양 된 한약의 경우에는 B. subtilis PCI 219, P. aeruginosa KCTC 2004, S. aureus subsp. aureus KCTC 1916에 항균 활성을 보였으며, 특히 여드름을 유발시키는 P. acnes KCTC 3314에 항균 활성을 보였다. RBL-2H3 세포주에서 알레르기 억제 효과를 확인한 결과 한약은 60% 알레르기 억제 효과를 보였으며, 발효한약은 57% 알레르기 억제 효과를 보였다. 발효한약은 알레르기 억제 효과와 함께 항균 활성과 항산화 활성의 증가로 인하여 알레르기 개선, 피부노화 및 병원성 미생물에 의한 피부질환의 개선에 효과가 있을 것으로 사료된다.

A comparative study on chemical composition of total saponins extracted from fermented and white ginseng under the effect of macrophage phagocytotic function

  • In this study, white ginseng was used as the raw material, which was fermented with Paecilomyces hepiali through solid culture medium, to produce ginsenosides with modified chemical composition. The characteristic chemical markers of the products thus produced were investigated using rapid resolution liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (RRLC-QTOF-MS). Chemical profiling data were obtained, which were then subjected to multivariate statistical analysis for the systematic comparison of active ingredients in white ginseng and fermented ginseng to understand the beneficial properties of ginsenoside metabolites. In addition, the effects of these components on biological activity were investigated to understand the improvements in the phagocytic function of macrophages in zebrafish. According to the established RRLC-QTOF-MS chemical profiling, the contents in ginsenosides of high molecular weight, especially malonylated protopanaxadiol ginsenosides, were slightly reduced due to the fermentation, which were hydrolyzed into rare and minor ginsenosides. Moreover, the facilitation of macrophage phagocytic function in zebrafish following treatment with different ginseng extracts confirmed that the fermented ginseng is superior to white ginseng. Our results prove that there is a profound change in chemical constituents of ginsenosides during the fermentation process, which has a significant effect on the biological activity of these compounds.

고체발효 숙지황의 지표성분 함량분석 (Quantitative Analysis of Marker Substance in Fermented Rehmanniae Radix Preparata)

  • Medicinal plant Rehmanniae Radix has long been used as traditional herbal medicine in Korea. In this study, we investigated quantitative changes of marker substance 5-hydroxymethyl-2-furaldehyde (5-HMF) in Rehmanniae Radix Preparata fermented with Ganoderma lucidum, Paecilomyces japonica, honey, and Nuruk, using HPLC according to the Korean Pharmacopoeia. As a result, 5-HMF was decreased in SDT and SYT, and increased in SST and SNT, comparing with control group. Quantitative increasing of 5-HMF is not exactly in direct proportion with fermentation, and it is need further studies to elucidate mechanism of quantitative changes by converted and newly produced substances with fermentation.

꽃송이버섯 기부 발효물 첨가 사료가 장어의 면역반응에 미치는 영향 (Effects of Fermented Sparassis crispa Stipe Extract Supplemented Diet on the Immune Responses of Philippines Eel, Anguilla bicolor)

  • 본 연구에서는 폐기되고 있는 꽃송이버섯 기부 발효에 적합한 균주 선발을 위해 Lactobacillus plantarum, Saccharomyces cerevisiae, Bacillus subtilis를 활용하여 발효를 진행하였으며, 생균수 실험 결과 L. plantarum이 발효 48시간에 1.486의 흡광도를 보여 빠른 발효속도와 생육능을 보였다. 총 페놀 함량은 L. plantarum과 S. cerevisiae를 활용하여 발효한 실험구에서 301.68 ppm, 299.35 ppm으로 발효 전보다 높은 함량을 보였으며, B. subtilis 발효 실험구는 큰 차이를 보이지 않았다. DPPH 라디칼 소거 활성은 L. plantarum이 63.9%로 가장 우수한 항산화 활성을 보여 L. plantarum을 이용하여 꽃송이버섯 기부를 발효한 후 장어에 사료 첨가제로 급이하였다. 6주간 장어의 폐사는 대조구에서 60마리로 가장 많은 폐사를 보였으며, 꽃송이버섯 기부 추출물이 24마리로 가장 적은 폐사를 보였다. 장어 혈청의 lysozyme 용균 활성 및 E. coli 살균능은 꽃송이버섯 기부 추출물과 발효물 모두 대조구에 비해 높은 활성을 보였다.

Molecular identification of Bacillus licheniformis isolates from Korean traditional fermented soybean by the multilocus phylogenetic analysis

  • In this study, Bacillus licheniformis which has been used as probiotics was isolated from Korean traditional fermented soybean. A total of 69 strains were presumptively identified as B. licheniformis by phenotypic methods. Based on PCR amplification and 16S rRNA gene sequencing, the multilocus sequence typing of gyrA and rpoB, followed by phylogenetic analysis was performed. The isolates were distinctly differentiated and found to be closely related to B. amyloliquefaciens, B. subtilis, and B. aerius. The partial 16S rRNA gene sequences of those strains matched those of B. sonorensis (97%) and B. aerius (98%) in the phylogenetic tree. In contrast, multilocus phylogenetic analysis (MLPA) showed that only 61 (86.9%) out of 69 strains were B. licheniformis. The rest of those strains were found to be B. subtilis (5.8%), B. amyloliquefaciens (2.9%), and B. sonorensis (2.9%), respectively. Therefore, our results suggested that since the 16S rRNA gene sequencing alone was not sufficient to compare and discriminate closely related lineages of Bacillus spp., it was required to analyze the MLPA simultaneously to avoid any misleading phenotype-based grouping of these closely related species.
