The Effect of Dietary Calcium Level on Bone Mineral Density and Bone Mineral Content in Ovariectomized Female Rats (난소절제한 흰쥐에서 식이칼슘량이 골밀도에 미치는 영향)

  • This study was done to evaluate the effect of dietary calcium level (a diet which met 100% or twice the calcium level in AIN-76 diet) on preventing bone loss in ovariectomized rats. Forty Sprauge-Dawley female rats(body weight 200$\pm$5g)were divided into two groups. One group were ovariecotomized (Ovx) while the others received sham operation(Sham). Thereafter, each rat group was further divided into normal calcium diet(0.52%) and high calcium diet(1.04%) subgroups. All rats were fed on experimental diet and deionized water ad libitum for 8 weeks. The total body, spine and femur bone mineral densities and bone mineral contents were measured by Dual Energy X-ray Absorptiometry, Eight weeks following operation, ovariectomized rats fed a high calcium diet had a significantly higher total bone mineral content, total bone calcium content, spine bone mineral density, spine bone mineral content and femur bone mineral content than ovariectomized rats fed control calcium diet. The correlation between dietary calcium intake level and spine bone mineral density were positive, but there was no correlation between dietary calcium intake and femur bone mineral density. The findings from the present study demonstrated that bone loss due to ovarian hormonal deficiency can be partially prevented by a high calcium diet. Futhermore, these findings support the strategy of the use of a high calcium diet in the prevention of estrogen depleted bone loss(postmenopausal osteoporosis)

Effect of Dietary Calcium Levels on Peak Bone Mass Formation in Growing Female Rats (칼슘 섭취 수준이 성장기 암컷 흰쥐의 최대골질량 형성에 미치는 영향)

  • The present study was designed to examine how Ca intake contributes to the increase of peak bone mass with growing female rats. Weaned rats were fed experimental diets consisting in five levels of Ca; very low(0.1%), low(0.2%), moderate(0.5%), high(1.0%) and very high(1.5%) for 4, 8 and 12 weeks. Bone growth, metabolism and Ca metabolism were determined. As for the rats fed for 4 weeks, the bone weight, length and breaking force and bone metabolism were not significantly affected by dietary Ca levels, whereas the current intakes of Ca were observed to have significantly affected the rats fed for 8 or 12 weeks with regard to the bone weight, length and breaking force and bone metabolism. The bone ash and Ca contents of the rats were affected by dietary Ca levels for the total period of feeding. It is suggested that dietary Ca itself affected the mineralization process either during the growth or later, although the resulting bone mass is not a linear function of dietary Ca content.

Four-week Oral Toxicity Study of DA-9601, an Antiulcer Agent of Artemisia spp. Extract, in Rats (애엽추출물 항궤양제 DA-9601의 랫드에 대한 4주 경구 반복투여 독성연구)

  • This study was conducted to investigate the repeated dose toxicity of DA-9601, an antiulcer agent of Artemisia app. extract, in rats. DA-9601 was administered orally once a day for 4 weeks to 10 males and 10 females per group at doses of 0(vehicle control), 125, 500 or 2000 mg/kg/day. Throughout the study, no treatment-related deaths and clinical signs were observed. In female rats receiving 125 mg/kg of DA-9601, water consumption increased slightly on day 4, 11 and 25. Hematological examination showed a decrease of MCV and an increase of PLT in male rats at the doses of 500 and 2000 mg/kg groups. Blood biochemistry revealed slight decreases of cholesterol, BUN and Na in male rats and decreases of total bilirubin and creatinine and slight increases of globulin and Cl in female rats. The organ weights at the end of 4 weeks showed slight changes in some organs of treated groups. But, all these changes were not considered to be of toxicological importance, because they did not show dose-response relationship and relevance to gross and microscopic findings. Histopathologically, abnormal treatment-related changes were not observed in any organ and target organs were not detected. On the basis of these results, the NOAEL(no-observed-adverse-effect level) of DA-9601 was estimated to be more than 2000 mg/kg/day under the conditions tested.

  • Gami-Samhwang-San, a herbal prescription for obesity treatment, is composed of seven crude herbs such as Rehmanniae Radix Preparata, Ephedrae Herba, Scutellariae Radix, Acori Gramineri Rhizoma, Polygalae Radix, Typhae Pollen, Armeniacae Semen, Menthae Herba. In this study, marker substances in n-butanol fraction (SH-21-B) from Gami-Samhwang-San were analyzed by high performance liquid chromatography (HPLC) and acute toxicity of standardized SH-21-B was evaluated by good laboratory practices (GLP) guideline of Korea Food and Drug Administration. Therefore we confirmed that there were baicalin of 15.92%, amygdalin of 6.57% and ephedrine of 2.49% in SH-21-B. SH-21-B was administered in rats at dose of 0 mg/kg, 2,000 mg/kg, and 5,000mg/kg. Clinical signs of both sexes of rats were observed daily for 14 days after single oral administration. Two female rats one administered at 2,000 mg/kg and the other administered at 5,000 mg/kg, died, but no dead animal was observed among male rats. Therefore $LD_{50}$ in the female rat is observed to be 8,710 mg/kg, and MLD (Minimun Lethal Dose) of the male rat is observed to be more than 5,000 mg/kg.

Interaction of Dietary Wheat Bran and Dietary Calcium Levels ell Calcium Utilization and Bone Mass in Post-breeding Female Rats

  • This study was conducted to determine the interaction of dietary wheat bran and dietary calcium levels n)n calcium utilization in post-breeding female rats. It was designed to compare the effects of four different levels (2.5, 5, 10 and 20%) of wheat bran and two different levels (0.5 and 1%) of calcium on bone and calcium balance in post-breeding female rats over a ten-week period. The effects of diet on animal weight gain, serum calcium, femur weight, femur calcium concentration, bone mass and calcium balance were determined and statistically analyzed. The addition of 20% wheat bran significantly (p$\leq$0.05) decreased the weight gain of rats. Serum calcium and bone calcium contents were more affected by dietary calcium level than by dietary wheat bran level. There was no significant difference in fat-free solid, ash, percentage of ash to fat-free solid and percentage of calcium to ash among groups. Groups fed the 1% calcium diet had a higher percentage of calcium to fat-free solid. All rats were in positive calcium balance during the three-4ay experimental period. The average calcium balance of the rats fed 1% calcium diet ranged from 25.34 to 53.90mg and the average calcium balance of the rats fed the 0.5% calcium diet ranged from 26.71 to 32.90mg. In rats receiving 2.5% wheat bran, the difference in calcium balance between the group fed the 1% calcium diet and the group fed the 0.5% calcium diet was only 1.37mg, which was not significantly (p$\leq$0.05) different. In rats receiving 20% wheat bran, the difference in calcium balance between the group fed the 1% calcium diet and the group fed the 0.5% calcium diet was 19.S7mg, which was significantly (p$\leq$0.05) different. The addition of wheat bran caused an increase in the calcium balance of the rats adminslesed the 1% calcium diet. On the other hand, the addition of wheat bran had no effect on the calcium balance of the rats adminislesed the 0.5% calcium diet. In conclusion, calcium utilization of rats wire more positively affected by the interaction of both dietary wheat bran levels and dietary calcium levels than either dietary wheat bran levels or dietary calcium levels alone. Moderate wheat bran consumption did not interfere with the calcium metabolism of rats when calcium intake was high enough.

Effect of Postnatal Angelicae Gigantis Radix Herb-acupuncture on Cell Proliferation in Offspring Rats with Prenatal Noise Stress during Pregnancy (당귀약침(當歸藥鍼)이 소음 Stress를 받은 새끼 쥐의 신경세포 생성에 미치는 영향)

  • Methods : 40 of Female rats were allowed to mate with 40 of male rats. Then, female rats were delivered of offspring rats, After birth 28 days, offspring rats were divided 8 groups, The normal group(Group A), the 10 mg/kg Angelicae gig antis radix~treated group(Group B), the 50 mg/kg Angelicae gigantis radix-treated group(Group C), the 100 mg/kg Angelicae gig antis radix-treated group(Group D), The control (noise-treated) group(Group E). the noise -10 mg/kg Angelicae gigantis radix-treated group(Group F), the noise-50 mg/kg Angelicae gigantis radix~treated group(Group G), and the noise-100 mg/kg Angelicae gigantis radix-treated group(Group H)(n = 5 in each group), From the 15th day of pregnancy, all rats were subcutaneously injected with 50 mg/kg BrdU once a day 30 min before the starting of experimental treatment. Rats of the prenatal noise-treated group were applied with 95 decibel supersonic machine sound for 1h once a day until delivery, After birth 28 days, offspring rats intraperitoneally injected with 50 mg/kg of BrdU and offspring rats were treated Angelicae gigantis radix Herb-acupunture on chungwan(CV12) for 7 consecutive days. For the detection of BrdU-positive cells and Ki-67 positive cells in hippocampus, immunohistochemistry was performed. Results : 1. The number of BrdU-positive cells in the dentate gyrus of noise-treated group was significantly decreased to normal group, and the Group F, G, H were significantly increased to control group. 2. The number of Ki-67 positive cells in the dentate gyrus of noise-treated group was significantly decreased to control group, and the Group G, H were significantly increased to control group. Conclusion : We concluded that postnatal Angelicae gigantis radix administration has effect on cell proliferation in offspring rats with prenatal noise stress during pregnancy.

The Effect of Hypophysectomy and Subsequent Administration of Sex Hormone on Several Endocrine Glands and Plasma Components in Rats (흰쥐의 내분비선 및 혈장성분에 미치는 뇌하수체척출의 영향과 이에 대한 성 Hormone의 효과)

  • The present experiments were carried out to elucidate the effects of hypophysectomy and subsequent administration of sex hormone on thyroid, adrenal gland, gonads and blood plasma components in the rat. The jresults obtained were summarized as follows: The weight of the thyroid gland of both male and female hypophysectiomzed rats decreased markedly from 7 days up to 56 days after the hypophysectomy as compared to the control group. The administration of sex hormone (6 mg of testosterone propionate to male and 6 mg of hexestrol to female) to the hypophysectomized rat gave on effect on the change in the weight of the thyroid gland. The hopophysectomy decreased the uptake of radioactive iodine in the thyroid gland in both male and female rats with time. Subsequent administration of the sex hormone caused no effect. With regard to the histological changes of the thyroid gland, the hypophysectomy caused significant changes in the gland showing a remarkable degeneration. The function of the gland seemed to disa, pp.ar almost completely on 56th day after the hypophysectomy. Upon the administration of sex hormone after the hypophysectomy, however. the epithelia of the follicle which has changed to flat from has partly returned to its functional cubicfrom and nuclei recovered as nearly as normal. These recovery were more remarkable in the female than in the male. The hypophysectomy kept causing a significant decrease in the weight of the adrenal gland in male and female rats during the period of observation (up to 56 days) as in the case of thyroid gland. The administration of sex hormone has on effect in this respect either. The hypophysectomy also caused a marked morphological change in the gland: zona fasciculata and zona reticularis were dicreased in size quichly after the hypophysectomy. The administraton of the sex hormone to the hypophysectomized rat resulted in clear distinction among the three layers of the adrenal cortex which otherwise very diffused. In the male, this phenomenum was more remarkable than in the female and the pattern of the cell arrangements and the thickening of each layer became similar to those of normal rats. The gonads of both sexes have also kept decreasing in the weight and degenerated in morpohology after the hypophysectomy. However, the degenerate follicle became enlarged after the administration of hexestrol in the female. Furthermore, the vacuoles found in interstitial cells of hypophysectomized rat disa, pp.ared after the administration of testosterone in the male and the formation of spermatocytes seemed to be recovered. Hypophysectomy also caused a gradual increase in the contents of total protein, non-protein nitrogen, total lipid, cholesterol and calcium in the blood plasma with time. The concentrations of sodium, potassium and chloride in the blood did not change after the hypophysectomy. Sex hormone caused practically no change in above tendency.

Toxicological effects of perfluorooctanoic acid in rats (랫드에서 Perfluorooctanoic Acid의 독성에 관한 연구)

  • Perfluorooctanoic acid (PFOA), a member of the perfluoroalkyl acids that have wide commercial applications, is persistent organic pollutants widely spread throughout the environment and human population. But little is known about the adverse biological effects of the PFOA. In the present study, the toxicological effects of PFOA were investigated in rats. Sprague-Dawley rats (N = 10 in each group) were orally administered with PFOA in drinking water for 4 weeks (0, 100, 200, or 400 ppm in male, and 0, 200, 400, or 800 ppm in female). Three female rats given 800 ppm died during the study. PFOA treatment decreased the body weight gain and increased the liver weights in both genders. Serum biochemical investigations revealed significant increases in the aspartate aminotransferase, alanine transaminase, alkaline phosphatase, blood urea nitrogen, total cholesterol, and total bilirubin in male but in female. Serum estradiol (E2) levels were increased in all treated rats. Histopathologically, hepatocellular hypertrophy around central vein was noted in the liver of treated rats. No significant histopathological changes were noted in other organs. In conclusion, PFOA induced toxicological changes in the liver and increased serum E2 level which was not related to histopathological changes of endocrine and reproductive system.

Single and Five-Week Oral Dose Toxicity Studies of Calcitriol and Alendronate Mixtures in Rats

  • The purpose of this study was to assess the single and 5 week oral dose toxicity of calcitriol and alendronate combination (1 : 10,000) treatment for osteoporosis or Paget's disease in male and female rats. In single dose oral toxicity study, the values of $LD_{50}$ of calcitriol and alendronate mixture were 750.075 mg/kg in male rats and 775.0775 mg/kg in female rats, respectively. Body weight and food consumption were continuously increased after adminstration of calcitriol and alendronate mixtures, and there was no significant changes in body weight and food consumption in all groups. In five-week oral toxicity study of calcitriol and alendronate mixture at a dose of 0.2 $\mu\textrm{g}$ + 2 mg, 1 $\mu\textrm{g}$ + 10 mg, 5 $\mu\textrm{g}$ + 50 mg and 25 $\mu\textrm{g}$ + 250 mg, respectively, there was no mortality, abnormal behavior and appearance in all groups throughout the administration period (5 weeks) and recovery period (2 weeks). Dose-dependent changes in parameters of urinalysis and hematological analysis were not observed in male and female rats treated with calcitriol and alendronate mixtures. All the values of the parameters appeared to be in the normal range. These data indicate that both calcitriol and alendronate are drugs having low toxicity in rats. NOAEL of calcitriol and alendronate mixtures were 50.005 mg/kg in 5-week oral toxicity.

Effects of Ovariectomy and Administration of Estradiol Benzoate on Body Growth and Development of Endocrine Glands in Female Rats (암 흰쥐에 있어서 난소척출 및 Estradiol Benzoate 투여가 체성장 및 내분비선 발육에 미치는 영향)

  • This experiment was conducted to investigate the effects of ovariectomy and estradiol benzoate administration on body growth and development of endocrine glands in female rats. One hundred and twenty female rats, 30 days old weighing 72.0${\pm}$3.0g, were divided into control and ovariectomized groups and divided again into two groups, treated and untreated with estradiol benzoate, respectively. Each group was composed of 30 rats. Hormone was injected every two week interval. The body weight was measured weekly from the beginning of treatment to 63 days after treatment and 6 rats per group were sacrificed at 7, 21, 35, 49 and 63 days after treatment for weight change of endocrine gland. The results obtained were as follows: 1. the changes in the body weight during the experimental period were not signifilcantly different between two groups, although the body weight of ovariectomized groups were heavier than that of control groups and the body weight of treated group with estradiol benzoate was heavier than that of untreated group. 2. The body lengths in all groups were increased proportionally to body weight until 63 days after treatment and there were not significantly different among the experimental groups. 3. The weight of hypophysis treated with estradial benzoate in ovariectomized groups was continuously increased in the whole period and was highly significant at 49 and 63 days after treatment as compared with those of untreated in ovariectomized group. 4. Adrenal gland weights of the control and ovariectomized group treated with estradiol benzoate were distinctly light at 21 days after treatment, but there were not significantly different among all groups at 35 days. Adrenal gland weight of ovariectomized groups was smaller than control groups at 49 days and untreated group in ovariectomized groups was considerably smaller than the other 3 groups at 63 days after treatment. 5. In control rats, the ovary weight treated group with estradiol benzoate was larger than the untreated group at 35 days after treatment, but was smaller than the untreated group after 49 days. 6. The weight of reproductive tract in ovariectomized groups were markedly smaller than control groups till 35 days after treatment. The growth of reproductive tract untreated in ovariectomized groups was almost completely depressed.
