Reproductive ecology of the sharp toothed eel in the southern Korean waters (한국 남해에 서식하는 갯장어, Muraenesox cinereus (Forsskal)의 생식생태)

  • Reproductive ecology of the sharp toothed eel, Muraenesox cinereus was investigated based on the samples captured in southern Korean waters from January 2010 to December 2011. Gonadosomatic index began to increase in April, and reached maximum between July to August. After spawning it began to decrease from October. Reproductive season was estimated to April-September, with peak in July. Fecundity was proportional to the size of the female, with the clutch size varying from 56,000 eggs in the smallest female (anal length, 27.0cm) to 1,400,000 eggs in the largest (anal length, 49.5cm). Size at 50% sexual maturity ($AL_{50}$), determined from mature females, were 21.9cm. Annual reproductive cycles of this species could be divided into four successive stages; immature stage (October-February), maturing stage (March-May), mature stage (June-August) and spent stage (August-October).

Induction of Sexual Maturation in Female Eels Anguilla japonica by Recombinant Eel Gonadotropin Produced in Silkworm (누에에서 생산된 뱀장어 재조합 생식소자극호르몬이 암컷 뱀장어(Anguilla japonica)의 성성숙 유도에 미치는 영향)

  • This study investigated the effects of recombinant eel gonadotropin hormone (rJeGTH) produced in silkworms, with and without a carboxyl-terminal peptide from equine chorionic gonadotropin (eCG), on the induction of sexual maturation in female eels Anguilla japonica. Experiments were conducted both in vivo and in vitro. In in vitro trials, germinal vesicle breakdown (GVBD) induction did not significantly differ between rJeFSH and $rJeFSH{\cdot}eCG$ treatments and the control group. However, previous studies did find that rJeLH and $rJeLH{\cdot}eCG$ treatments induced GVBD in female eels. Our in vitro exploration of $estradiol-17{\beta}$ ($E_2$) levels in immature ovarian tissues revealed significantly higher $E_2$ levels in the group treated with $rJeFSH{\cdot}eCG$ $1{\mu}g/mL$ than in the control group. In contrast, the in vivo experiments showed no effect of recombinant hormones on the sexual maturation of feminized eels. Previous studies and our own in vitro results have clearly shown that rJeGTH and $rJeGTH{\cdot}eCG$ have a positive effect on the sexual maturation of feminized eels. To develop the activity of rJeGTH in vivo, further studies should confirm circulation time and activity of these hormones in eels' bloodstream, modify the structure of the recombinant gene, and implement additional glycosylation.

Studies on a Nematode, Anguillicola crassa Parasitic in the Air Bladder of the Eel (뱀장어 부레에 기생(寄生)하는 선충(線虫), Anguillicola crassa에 관(關)한 연구(硏究))

  • In Korea, studies on a Nematode, Anguillicola crassa parasitic in the air bladder of eel are not yet reported. This reason led the author to study the parasitic species, state and life history of the A. crassa parasitized in the air bladder of eel in order to take effective control measures against its damage. The size of fully developed eggs was 80 to $92(86.7){\times}62$ to $71(67.4)\;{\mu}m$, larva was 210 to $240(225){\times}18$ to $23(20.6)\;{\mu}m$. The intermediate host of A. crassa was Thermocyclops hyalinus, it was capable for parasitizing the eel after 4 days of invasion and then the size of larva was 360 to $420(390){\times}28$ to $35(31)\;{\mu}m$. Fifty days after eel had ingested the Thermocyclops hyalinus infected with larva of A. crassa, the larvae matured into adult worms in the air bladder of eel. The size of detected adult worms was 7.3 to $31.0(16.5){\times}0.5$ to 2.2(1.2) mm, 4.9 to $13.3(8.3){\times}0.3$ to 0.9(0.4) mm. Investigating the morphology of the worms, they were identified as A. crassa. Monthly the parasitic rate of the worms in the eel was high in June, September and December, but low in January to March. After the investigation on the significance between non-parasitic fish and parasitic fish, it was not significant, therefore it can be considered that there is no effect of infection in the growth of eel. Any abnormality of eels air bladder tissue was not seen by the infection of A. crassa. At 25.0 to $26.7^{\circ}C$ of water temperature the death time of Thermocyclops hyalinus by masoten treatment was 14 hours in 0.5 ppm, 20 hours in 0.4 ppm, 22 hours in 0.3 ppm, 30 hours in 0.2 ppm and 42 hours in 0.1 ppm.

Profile of Plasma Estradiol-17β According to Ovarian Development of the Pike Eel, Muraenesox cinereus (갯장어 Muraenesox cinereus의 난소 발달에 따른 혈중 Estradiol-17β 변동)

  • This study correlated changes in the plasma levels of estradiol-$17{\beta}$ (E2) with changes in the gonadosomatic index (GSI) and ovarian development during the annual reproductive cycle of the pike eel Muraenesox cinereus, collected at the Tongyung coast region. Ovarian maturity was classified based on histological observations; the perinucleolus stage (November to February), the oil droplet stage (March to April), the early vitellogenic stage (April to May) and the late vitellogenic stage (June to October). Seasonal changes in the GSI were correlated with water temperature and reflected the degree of ovarian maturity. Plasma E2 levels were correlated with changes in the GSI, which increased from April to a peak in July, and the levels remained comparatively high until October. These data indicated that changes in the GSI and plasma E2 levels are correlated with the annual ovarian activity of the pike eel. In this study, however, female pike eels were not collected during the spawning stage. Therefore, spawning of this species seemed to be closely related to its migration toward the deep sea of offshore.

Effects of Estradiol-17β on the Feminization of Japanase Eel, Anguilla japonica (극동산 뱀장어, Anguilla japonica 자성화에 미치는 estradiol-17β 유도 효과)

  • The effects of oral administration of estradiol-$17{\beta}$ (E2) on glass eels (Body weight: $0.16{\pm}0.05g$, Total length: $6.2{\pm}0.9cm$) and young eels (Body weight: $2.6{\pm}0.6g$, Total length: $13.2{\pm}0.6cm$) on gonadal sex and growth were examined, respectively. Glass eels were fed a diet containing E2 at a dose of 10 mg/kg or 25 mg/kg, respectively, for five months. The female ratio significantly increased in all E2-treated groups (10 mg/kg diet group: 70%; 25 mg/kg diet group: 90%) when compared to the control group (10%). Young eels were fed a diet containing E2 at a dose of 25 mg/kg for four months. The female ratio also significantly increased in the E2-treated groups (60%) compared to the control group (20%). The highest female ratio was observed in the stage of glass eels rather than young eels. In all experiments, however, the growth of eels treated with E2 was similar to that of controls to the end of the experiment. Thus, oral administration of E2 could be a good approach to controlling sex differentiation.

Correlationship between Artificial Maturation Season and Reproduction Coefficient in the Cultured Eel Anguilla japonica (양식산 뱀장어 Anguilla japonica에 있어서 인공성성숙시기와 번식률과의 상관관계)

  • This study investigated the correlationship between artificial maturation season and reproduction coefficient of cultured eel Anguilla japonica from May (spring) to next January (winter). The brood stock, female eels ($400{\sim}600\;g$) were artificially matured by weekly intramuscular injections of salmon pituitary extracts (SPE, 20 mg/fish) to induce a completion of vitellogenesis. After completion of vitellogenesis, final oocyte maturation and ovulation was induced by injection of $17{\alpha}$, $20{\beta}-dihydroxyprogesterone$ (DHP) at about $2\;{\mu}g/g$ body weight. Most fish ovulated $15{\sim}18\;h$ following the DHP injection. The ovulated fish were induced to natural spawning or artificial fertilization by the dry method. Males ($200{\sim}350\;g$) were received weekly intramuscular injections of human chorionic gonadotropin (HCG) at a dosage of 1 IU/g body weight to induce testicular maturation and spermiation. Seasonal reproduction coefficient which includes the rate of ovulation, buoyancy, fertilization and hatching of eggs in the artificially matured eel during spring to summer ($May{\sim}July$) were significantly higher than the other season, while there were no significant difference among spring and summer (P<0.05). Furthermore, the number of eggs spawned and larvae hatched in the artificially maturated eel during spring to summer ($May{\sim}July$) were significantly higher than the other season, while there were no significant difference in spring and summer (P<0.05). These results indicate that artificial maturation by hormone treatment of A. japonica was successful only during spring to summer, which is the maturation period in the wild stock in nature. Consequently, it is possible to determine the period of artificially induced sexual maturity by the reproduction coefficient which includes the rate of ovulation, buoyancy, fertilization and hatching of eggs in the cultured eel A. japonica.

Pathogenicity of Lactobacillus pentosus PL11 isolated from eel (Anguilla japonica) intestine and single oral toxicity of its culture broth in rats (랫트에서 뱀장어로부터 분리된 Lactobacillus pentosus PL11의 병원성 및 배양액에 대한 단회 경구독성 시험)

  • The pathogenicity and acute toxicity of Lactobacillus (L.) pentosus PL11 from eel (Anguilla japonica) were investigated using male and female Sprague-Dawley rats. The pathogenicity of L. pentosus PL11 was examined after treating the rats with $10^{11}$ CFU/mL, $10^9$ CFU/mL or $10^7$ CFU/mL doses of L. pentosus PL11 culture or 0.85% NaCl (Control) intragastrically. For acute toxicity studies, rats were treated with dried culture broth of L. pentosus PL11 at doses of 5,000 mg/mL, 2,500 mg/mL, 1,250 mg/mL or 625 mg/mL or Lactobacilli MRS broth (Control), and clinical signs or mortalities were monitored for two weeks. The results of the present investigation revealed no mortalities or obvious clinical signs in rats administered with the live bacterial cultures or dried culture broth at any investigated dose level. Also, no significant differences were observed in net body weight gain, gross pathological findings, feed and water consumption and body temperature among the different treatment groups and between the treated and control rats. It can be concluded from the above findings that L. pentosus PL11 is a safe probiotic strain with potential as feed additive to increase the feed efficiency or health of fish.

  • The sand-eel, Ammedytes Personatus is a commercially important fish abounantly found in west and east coast of Korea. Samples were collected from the Baegryeong Island (Yellow Sea), Geoje Island(southern coast) and Jumunjin (East Sea) from May 1973 to December 1974. In this paper the author dealt with some biological point of the fish, especially the relationship between total length ang body weight, and the major spawning season and sex ratio. 1. The major spawning season was confined to the end of November and the end of December at Jumunjin. 2. The sex ratio (male/female) is 0.75 before the main spawning season and increases up to 1.36 after spawning at Jumunjin in 1974. 3. The relationship between the total length (L) and body weight (In were represented as follows: $W=0.0001906L^{3.1998319}$ for the sample from Baegryeong Is. $W=0.0003419L^{3.0213438}$ for the sample from Geoje Is. $W=0.0002655L^{3.1408629}$ for the sample from Jumunjin.

  • In order to study the reproductive physiology of Sand ell(Hypoptichus dybowskii) catched at kangwon-do, Wooamjin Bay, spawning and egg development were investigated in April and May, 1996. The observed spawning period was from the middle of April to the end of May with the water temperature above $8^{\circ}$C. Body size and colouration were distinguished between male and female. The type of testicular structure is lobular, and the ovary is a pair of sac-sahaped covering with a fibromuscula capsule and comprising a mature egg of average 86. The unfertilzed egg are demersal and adhesive, and their diameter were 1.65-1.95 mm. the colour of yolk is light yellow and contained a lot of tiny oil globules. The average spawned egg was 62 in sargasso weeds. Hatching was observed in 12 days after fertilization with the water temperature of 14 ${\pm}1^{\circ}$C. The average size of larva after hatching was 6.85${\pm}$0.23mm in total length, and larva were containing yolk including globule.
