Objectives : The present study was conducted to examine whether Cynanchi wilfordii radix (CWR) with or without fermentation has an ameliorative effect on hyperlipidemia in rats.Methods : We analyzed the contents of Conduritol F on Cynanchi wilfordii radix. The experimental animals were divided into six groups; normal diet fed group (N), high cholesterol fed control group (Con), Lovastatin 20 mg/kg (L), CWR-W 300 mg/kg (CWR-W), and CWR-F 300 mg/kg on hyperlipidemia model induced by feeding 1.25% cholesterol. Rats were administrated orally every day for 8 weeks. And lipid profile of serum and weight change were observed.Results : The vehicle displayed a markedly increased body weight and significantly increased liver and epididymal fat weight, however, the administration of CWR improved the body, liver, and epididymal fat weights. All drug treatment reduced significantly the serum level of total cholesterol and LDL-cholesterol elevated by intake of high cholesterol diet. TG displayed a reducing tendency all drug treatment, however, CWR-W decreased significantly. Atherogenic index and cardiac risk factor increased high cholesterol diet fed control group, while the administration of CWR-W and CWR-F decreased significantly. The major index of liver injury such as AST and ALT improved in all drug treatment.Conclusions : These results suggest that CWR extended the effect of lipid enhanced. Therefore CWR with or without fermentation may be useful for therapeutic treatment of clinical conditions associated with hyperlipidemia. Finally, these require more investigations about the action mechanism of CWR in the future.

Purpose: Postharvest handling of onions (harvesting, cleaning, grading, cooling, storing, and transport) should be performed continually to reduce costs and improve quality. The purpose of this study is to a) determine the design parameters and operating conditions of anion auto-dumping that constitutes a key component of the postharvest bulk handling machinery system, and b) to perform a performance test with the auto-dump prototype system. Methods: Kinematic analyses and computer simulations of the auto-dump mechanism were applied to analyze the operating conditions and design parameters. Results: The optimum working condition for the auto-dump was determined from kinetic analyses. In addition, the interaction between the velocity of the hydraulic cylinder and the angular velocity of the auto-dump were analyzed in order to control the bulk handling machinery system. The acting forces and optimum operating conditions of the hydraulic cylinder were determined by analyzing the forces related to the mass of inertia of the auto-dump assembly during rotation. The method of controlling the feeding rate of onions in terms of the uniformity of the stacking pattern and the control of the entire system was better than the two-stage method of controlling the rotational speed of the auto-dump. Based on the performance test with the prototype for the auto-dump, the stacking pattern and rigidity of the system were analyzed. Conclusions: These results would be of great importance in the postharvest bulk handling machinery system for onions.

Ticks are the arthropod vector capable of transmitting diverse pathogens, which include bacteria, viruses, protozoan and fungi. Ticks are able to survive under stressful environmental conditions. One of evolutionary outcomes of these obligatory hematophagous arthropods is the survival for extended periods of time without a blood meal during off-host periods. Water conservation biology and heat tolerance have allowed ticks to thrive even under high temperatures and low relative humidity, thus they have become highly successful arthropods as they are distributed globally. Tick osmoregulatory physiology is a complex mechanism, which involves multiple osmoregulatory organs (salivary glands, Malpighian tubules, hindgut and synganglion) for the acquisition and excretion of water and ions. Blood feeding and water vapor uptake have been early reported as the primary passages for ixodid tick to acquire water. Recently, we have learned that ticks can actively drink environmental water allowing hydration. The acquired water can be traced to the salivary glands (type I acini) and the midgut diverticula. This opens new avenues for tick management through the delivery of toxic agents into their drinking water, in addition to an alternative strategy for the study of tick physiology. Here we address the osmoregulatory physiology in the ixodid ticks as a potential target physiological mechanism for tick control. We discuss the implications of water drinking behavior for tick control through the delivery of toxic agents and discuss the dermal excretion physiology as an additional pathway to induce tick dehydration and tick death.
Antioxidative function of uncooked powdered food (Sangsik) was evaluated in rats consuming nutritionally unbalanced diet including 1% cholesterol, high proportion of animal lipids (lard : soybean oil : 8 . 2) , sub-optimal levels of vitamin and mineral mixture along with 0.5% ethanol in drinking water. The uncooked powdered food tested in the present study was a mixture composed of 42 kinds of plant foods (cereals, legumes, seaweeds, vegetables, and fruits) supplemented with vitamins and minerals, and dietary fiber. Control rats were fed the semi-purified diet based on the AIN-93G composition, and nutritionally unbalanced rats were divided into 3 groups, and fed one of the following diets with 0.5% ethanol in drinking water for 5 weeks : unbalanced control diet (UC) ,20% Sangsik powder supplemented diet (S20), and 40% Sangsik powder supplemented diet (S40) . Food efficiency ratio was significantly higher in rats fed S40 compared to the value for rats fed UC (p < 0.05). Hepatic level of thiobarbituric acid reactive substances (TBARS) was significantly lower in rats fed UC compared to that for control rats (p < 0.05) , and was not influenced by dietary supplementation of the Sangsik powder. Hepatic superoxide dismutase (SOD) activity was significantly higher in rats fed UC compared to that for control rats (p < 0.05) , and significantly reduced in rats fed S20 or S40 compared to the value for unbalanced control rats. Feeding unbalanced control diet significantly reduced the ratio of hepatic GSH-Px + catalase/SOD activities compared to the value for control rats, and this decrease in the ratio of antioxidant enzyme activities was reversed by adding the Sangsik powder to the diet at 20% (p <0.05) . Based on the results of antioxidant enzyme activities, feeding uncooked powdered diet appears to provide a favorable environment for body's antioxidative defense mechanism. Serum levels of Fe and Cu were significantly lower in rats fed the Sangsik powder supplemented diets compared to the value for unbalanced control rats (p < 0.05) , and levels of Se, Mn, and Zn were also tended to be decreased by dietary supplementation of the Sangsik powder. These results postulate the possibility that ingredients used in the uncooked powdered food may decrease the bioavailability of trace elements in rats.

This study was performed to investigate the anti-diabetic mechanism of crude polysaccharides isolated from the fruiting bodies of Grifola frondosa. We treated 3T3-L1 adipocyte cells to observe whether the crude polysaccharides isolated from Grifola frondosa would stimulate insulin sensitivity. Significant insulin sensitizing activity was observed in the 3T3-L1 adipocytes, and giving the crude polysaccharide of Grifola frondosa with 1 nM of insulin caused glucose uptake to increase to a similar level as giving 50 nM of insulin alone. To confirm the mechanism for the anti-diabetic effect of the crude polysaccharides, we performed further examinations within $KK-A^{y}$ mice, an animal model of type 2 diabetes. The crude polysaccharides reduced blood glucose levels in the $KK-A^{y}$ mice for 2 weeks after feeding, and also significantly lowered plasma insulin levels. These results suggest that the anti-diabetic mechanism of the crude polysaccharide of Grifola frondosa is related to the enhancement of insulin sensitivity.
Objective: Feed consumption contributes a large percentage for total production costs in the poultry industry. Detecting genes associated with feeding traits will be of benefit to improve our understanding of the molecular determinants for feed efficiency. The objective of this study was to identify candidate genes associated with feed conversion ratio (FCR) via genomewide association study (GWAS) using sequence data imputed from single nucleotide polymorphism (SNP) panel in a Chinese indigenous chicken population. Methods: A total of 435 Chinese indigenous chickens were phenotyped for FCR and were genotyped using a 600K SNP genotyping array. Twenty-four birds were selected for sequencing, and the 600K SNP panel data were imputed to whole sequence data with the 24 birds as the reference. The GWAS were performed with GEMMA software. Results: After quality control, 8,626,020 SNPs were used for sequence based GWAS, in which ten significant genomic regions were detected to be associated with FCR. Ten candidate genes, ubiquitin specific peptidase 44, leukotriene A4 hydrolase, ETS transcription factor, R-spondin 2, inhibitor of apoptosis protein 3, sosondowah ankyrin repeat domain family member D, calmodulin regulated spectrin associated protein family member 2, zinc finger and BTB domain containing 41, potassium sodium-activated channel subfamily T member 2, and member of RAS oncogene family were annotated. Several of them were within or near the reported FCR quantitative trait loci, and others were newly reported. Conclusion: Results from this study provide valuable prior information on chicken genomic breeding programs, and potentially improve our understanding of the molecular mechanism for feeding traits.

Objective: In this study, we investigated the adverse effects of dietary zearalenone (ZEA) (0.5 to 1.5 mg/kg diet) on the localization and expression of the growth hormone receptor (GHR) in the uteri of post-weaning gilts and explored alternative mechanism of the reproductive toxicity of ZEA on piglets. Methods: A total of forty healthy piglets (Duroc${\times}$Landrace${\times}$Large White) aged 28 d were selected for study. Piglets were transferred to single cages after 10 days' adaptation on an obstetric table. The animals were allocated to one of four treatments: a normal basal diet supplemented with 0 (Control), 0.5 (ZEA0.5), 1.0 (ZEA1.0), or 1.5 (ZEA1.5) mg/kg purified ZEA, and fed for 35 d after the 10-d adaptation. Analyzed ZEA concentrations in the diets were 0, $0.52{\pm}0.07$, $1.04{\pm}0.03$, and $1.51{\pm}0.13mg/kg$, respectively. At the end of the feeding trial, piglets were euthanized after being fasted for 12 h. Two samples of uterine tissue from each pig were rapidly collected, one of which was stored at $-80^{\circ}C$ for analysis of the relative mRNA and protein expression of GHR, and the second was promptly fixed in Bouin's solution for immunohistochemical analysis. Results: The relative weight of the uteri and thickness of the myometrium and endometrium increased linearly (p<0.001) and quadratically (p<0.001) with an increasing level of ZEA. The results of immunohistochemical analysis indicated that GHR immunoreactive substance was mainly localizated in the cytoplasm of uterine smooth muscle, glandular epithelial, luminal epithelial, stromal, and vascular endothelial cells. In contrast, nuclear staining was rarely observed. The immunoreactive integrated optic density of GHR in the myometrium, luminal epithelium, glandular epithelium, and whole uteri of weaning gilts increased linearly (p<0.001) and quadratically (p<0.05) with an increasing level of ZEA. The mRNA and protein expression of GHR in the uteri of weaning gilts increased linearly (p<0.001) and quadratically (p<0.05) with an increasing level of ZEA. Conclusion: In conclusion, ZEA at a concentration of 0.5 mg/kg was sufficient to significantly thicken the myometrium and endometrium, and at a concentration of 1.0 mg/kg induced a high level of GHR expression to promote growth and development of the uteri. This revealed an alternative molecular mechanism whereby ZEA induces growth and development of the uteri and provides a theoretical basis for the revision of Chinese feed hygiene standards.
Production of green pepper has been increased due to customer's preference and a projected ten-year boom in the industry in Korea. This study was carried out to develop an automatic grading and sorting system for green pepper using machine vision. The system consisted of a feeding mechanism, segregation section, an image inspection chamber, image processing section, system control section, grading section, and discharging section. Green peppers were separated and transported using a bowl feeder with a vibrator and a belt conveyor, respectively. Images were taken using color CCD cameras and a color frame grabber. An on-line grading algorithm was developed using Visual C/C++. The green peppers could be graded into four classes by activating air nozzles located at the discharging section. Length and curvature of each green pepper were measured while removing a stem of it. The first derivative of thickness profile was used to remove a stem area of segmented image of the pepper. While pepper is moving at 0.45 m/s, the accuracy of grading sorting for large, medium and small pepper are 86.0%, 81.3% and 90.6% respectively. Sorting performance was 121 kg/hour, and about five times better than manual sorting. The developed system was also economically feasible to grade and sort green peppers showing the cost about 40% lower than that of manual operations.
Soybean is a rich source of isoflavones such as genistein and daidzein. Soy isoflavones have both weak estrogenic and anti-estrogenic effects and are structurally similar to tamoxifen, an agent that has an effect similar to that of estrogen in terms of reducing postmenopausal bone loss. The purpose of this study was to determine the effects of differences in protein source (casein vs soy) and isoflavone levels (reduced vs higher levels) on selected bone markers and hormones in growing male rats. Thirty weanling Sprague-Dawley young rats were divided into 3 groups: The control group was fed a casein-based diet, the soy concentrate group was fed soy protein with totally reduced isoflavones content (isoflavones 0.07 mg/g protein), and the soy isolate group was fed soy protein with a higher than normal isoflavones content (isoflavones 3.4 mg/g protein). The degree of bone formation was estimated by measuring serum osteocalcin and alkaline phosphoatase (ALP). By determining collagen cross-linkage by immunoassay and correcting with creatinine values, the bone resorption rate was compared. Serum osteocalcin, growth hormone, estrogen and calcitonin were analyzed using radio immunoassay kits. The bone formation marker and ALP activity were differentiated by protein source, showing higher values than casein in feeding either soy isolate or soy concentrate. In this study using growing rats, the differences in isoflavone contents were not a significant factor in either bone formation or bone reaborption markers. Moreover, the soy isolate group had significantly higher levels of growth hormone than the casein group. The findings of this study suggest that growth hormone is partially responsible for its bone-formation effects in young growing rats. Soy protein and the isoflavones in soy protein are beneficial for bone-formation in growing male rats. Therefore, exposure to soy protein and isoflavones early in life may have long-term health benefits in preventing bone diseases such as osteoporosis. Neuropeptides produced in neurosecretory cells are the largest group of insect hormones. They regulate various physiological functions, such as fat body homeostasis, feeding, digestion, excretion, circulation, reproduction, metamorphosis, and behavior throughout all life stages. The PRXamide peptide family (X, a variable amino acid) is a well-characterized neuropeptide component with a common amino acid sequence, PRXamide (NH2), at the C-terminal end conserved across Insecta. The PRXamide peptides are classified into three subfamilies, each having diverse biological roles in insects: (1) pyrokinin (PK) includes the pheromone biosynthesis activating neuropeptide (PBAN) and the diapause hormone (DH), (2) the capability (CAPA) peptides, and (3) the ecdysis-triggering hormone (ETH). PBAN as a member of PK subfamily was first identified to stimulate pheromone biosynthesis in moths three decades ago. Since then, PBAN peptides have been extensively studied by various research groups from a broad spectrum of arthropods. In this paper, we briefly review insect PBAN molecules with emphasis on gene structure and expression, signal transduction, physiological mechanism in sex pheromone biosynthesis, and application for pest management.
