Hesperidin Lowers Activities of 3-Hydroxy-3-methylglutaryl-CoA Reductase and Acyl-CoA:Cholesterol Acyltransferase in Rats Fed High -Cholesterol Diet

  • In this study, a potential mechanism through which the hesperidin might work on the effect was examined in vivo. Male rats were fed a high cholesterol synthetic diet (1%, wt/wt) with hesperidin (0.1%, wt/wt) for 42 days. Activity of hepatic HMG-CoA reductase was significantly lowered by the hesperidin supplement compared to the control. Hesperidin did not significantly alter plasma or hepatic lipids, but tended to lower those lipid levels. Hesperidin also subsequently reduced the fecal neutral sterols compared to the control(253.3mg/d vs.521.9 mg/d). The inhibition of HMG-CoA reductase resulting from the hesperidin supplementation could count for the reduction in fecal neutral sterols that appears to compensate for the decreased cholesterol biosynthesis. The dose of hesperidin in a high choles-terol diet should apparently be more than 0.1% to exhibit the hypocholesterolemic response in these rats. It remains to be determined whether the observed alterations in cholesterol metabolism are specific to the rat or also could be applied to the humans.

마(Dioscorea)의 콜레스테롤 저하작용 및 그 작용기전 (Effects of Yam on Lowering Cholesterol Level and Its Mechanism)

  • The effects of yam(Dioscorea) on lowering cholesterol level and its mechanism were investigated. The concentrations of plasma and liver lipids, and the excretions of fecal neutral sterol and bile acid were measured in male Sprague-Dawley rats. Five Groups of 8 rats were fed hypercholesterolemic diet(1% cholesterol, 10% lard ; control), hypercholesterolemic diet plus 15% or 30% dried yam powder prepared by either hot-air(15HY, 30HY) or freeze dry(15FY, 30FY) for 4wk. Plasma total lipid, total cholesterol and cholesterol level was also significantly lower(28%, p<0.05), buy HMG-CoA reductase activity was higher in 30FY(230%, p<0.05) than in control. Although no significant differences in fecal neutral sterols were observed among groups, the yam-fed rats apparently had less bacterial degradation of cholesterol as indicated by a significantly greater of fecal cholesterol to coprostanol than in controls. Total fecal bile acids were significantly greater in rats fed yam(15HY : 5 folds, 15FY ; 12,30HY ; 12, 20FY ; 22) than in controls. The ratio of secondary to primary bile acids was almost 8 times lower in 30FY than in control. These data indicate that yam lowers cholesterol both in plasma and in liver through increasing fecal bile acid excretion as well as HMG-CoA reductase activity. Freeze-dried yam, which possesses viscosity, was more effective in cholesterol-lowering action than hot-air dried one.

Xylooligo당이 고콜레스테롤 식이 횐쥐 간조직의 UDP-Glucuronyl Transferase 활성과 분변중 Sterol류 조성에 미치는 영향 (Effects of Dietary Xylooligosaccharide on Hepatic UDP-Glucuronyl Transferase Activity and Compositions of Fecal Sterols in Rat Fed High Cholesterol Diets)

  • 고콜레스테롤 식이 흰쥐에서 xylooligo당이 간조직중의 UDP-glucronyl transferase 활성과 분변을 통한 지질, 담즙산 및 중성 sterol류의 배설 및 장기능개선 효과를 관찰하였다. 실험동물은 100$\pm$10g의 Sprague-Dawley 횐쥐를 이용하여 정상 식이군과 1% 고콜레스테롤 실험군으로 나누고 다시 실험군은 1%의 고콜레스테롤 식이에 xylooligo당을 0%, 5%, 10% 및 15% 농도 첨가군(C, C5XO, C10XO 및 C15XO군)로 나누어 각군마다 10마리씩 4주간 자유섭식 시켰다. 분변중의 총지질 및 TG함량은 C군에 비해 xylooligo당 공급군에서 현저하게 증가되었다. 간 microsome의 UDP-glucuronyl transferase의 활성은 정상군에 비해 C군에서 35% 증가되었고 또 C5XO, C10XO 및 X15XO군은 C군에 비해 15%, 41% 및 21%씩 각각 증가되었다. 분변중의 bile acid 함량은 C군에 비해 C5XO, X10XO 및 C15XO군에서 3.1배, 3.6배, 2.8배씩 각각 증가되었다. 분변중 중성 스테롤류인 cholesterol, coprostanol, coprostanone 함량은 C군에 비해 xylooligo당 공급군에서 현저하게 증가되었다. 이상의 결과에서 xylooligo당은 고콜레스테롤 식이 횐쥐에서 간조직의 UDP-GTase활성을 증가시키고 분변으로 총지질 및 중성지방 그리고 스테롤류의 배설을 증가시키는 효과가 있었고 또 분변량도 증가시키는 장기능 개선 효과도 뚜렷하였다.

Chitin.Chitosan이 흰쥐의 콜레스테롤 대사에 미치는 영향 (Effects of Chitin and Chitosan on Lipid Metabolism in Rats)

  • The effects of chitin and chitosan on the contents of plasma and liver lipids and the fecal excretion of neutral and acidic steroids were investigated in rats fed with atherogenic diet. Dietary chitin did not affect plasma total cholesterol, LDL cholesterol and HDL cholesterol, while chitosan diet de creased total cholesterol and LDL cholesterol. Plasma content of triglyceride was reduced signif icantly by chitin diet but not by chitosan. Increases in fecal weight were observed in rats fed with either chitin or chitosan, and the increase was greater with chitosan than with chitin. Dietary chitosan resulted in an increase of fecal excretion of neutral steroids without any changes in the excretion of acidic sterols. Putting the results together, dietary chitosan reduces plasma cholesterol probably due to the reduction of cholesterol absorption. The decrease in plasma triglyceride level by dietary chitin requires further research to understand the mechinism.

감귤박, 다시마, 손바닥 선이장 분말을 함유한 식이의 급여가 고콜레스테롤혈증 흰쥐의 체내 지질수준과 장내 콜레스테롤 흡수, 혈소판 응집성 및 간 조직에 미치는 영향 (Hypocholesterolemic Effect of Tangerine Pulp, Sea Tangle or Prickly Pear Cactus on Lipid Level, Intestinal Cholesterol Absorption, Platelet Aggregation and Liver Tissue in Hypercholesterolemic Rats)

  • This study done to investigate the hypocholesterolemic effect of ow dietary fiber in a high cholesterol diet in aspect of cholesterol absorption and excretion using Sprague Dawley rats. After feeding diets containing 0.5% cholesterol and 5% tangerine pulp, sea tangle or prickly pear cactus for four weeks, we measured the levels of plasma and liver cholesterol and triglyceride, fecal neutral sterols and fecal radioactivity after ingestion 14(sup)C-cholesterol. We also examined platelet aggregation and histological change in liver tissues in association of hypercholesterolemia. The liver to body weight ratio was significantly(p<0.01) lower in rats fed prickly pear cactus than in other groups. The levels of plasma total cholesterol, LDL-cholesterol and triglyceride were decreased significantly(p<0.01) in the prickly pear cactus group compared with the control, while there was no difference in the liver levels of total cholesterol and triglyceride among groups. Fecal corprostanol and cholesterol were significantly(p<0.01) higher groups of control and prickly pear cactus compared to the other two groups. Radioisotope excretion after ingestion of 14(sup)C-cholesterol was higher in the control group than in tangerine pulp group or sea tangle group, with the highest in prickly pear group. Radioisotope excretion was the highest during the 2nd and 3rd days in all groups. Hematocrit and platelet aggregation were decreased in all fiber groups compared to the control, but not statistically different. Microscopic examination showed that cholesterol diet cause a fat accumulation in the liver and prickly pear cactus decreased the fat accumulation. Result indicates that prickly pear cactus has hypocholesterolemic effect by decreasing absorption and increasing excretion of cholesterol, thereby protective effect on fatty liver. Control group fed diet containing high cholesterol and low fiber seems to have a self control system in cholesterol absorption and excretion preventing hypercholesterolemia. 식이 섬유소 섭취상태와 변 답즙산 조성과의 관계 (The Relationship to Dietary Fiber Intake and Fecal Bile Acid Profiles)

  • Bacterial transformation of bile acids is possibly involved in colorectal carcinogenesis. n several epidemiological studies, the fecal bile acid dietary fiber are related to the indicence of colonic cancer. This study investigated the influence of age, dietary fiber intake on fecal bile acid profiles in healthy subject. The dietary fiber were assessed by mean of 24 hour dietary recall method, the subjects consist of 238 members aged 20 to 64 years old and feces are collected from the subjects. Fecal bile acids and neutral sterols were analyzed by gas chromatography. Mean dily crude fiber intake level was 7.7$\pm$1.4g(dietary fiber : 16.7$\pm$3.5g), dietary fiber intake range being 6.5-36.8g. The dietary fiber intake in elederly subject was significantly lower than in the other younger groups. Dietary fiber intakes was negatively correlated with the total bil acid concentation in feces. Probably, a decrease in dietary fiber intake results in higher fecal bile acid concentrations. The secondary bile acid concentration is related to the colon cancer, deoxycholic acid and lithocholic acid were significantly higher in elderly subjects. Concentration of fecal total bile acid, deoxycholic acid, coprostanol, coprostanone were higher in low dietary fiber intake group. These results suggest that the risk factor for colon cancer might be reduced, when dietary fibers are consummed more.

  • This study is conducted to evalute the effect of ginseng fraction component (ginseng extract solution, GES; ginseng protein, GP; ginseng saponin, GSA; ginseng residue, GR) upon hyperlipidemia and fatty liver induced by high fat administration. In doing so, the serum, liver and epididymal adpoid tissue have been examined for lipid components level and lipoprotein fraction. Feces bile acid and neutral sterol excretion have been also measured. 1'he results obtained from this study are as follows. 1. Serum, liver, epididymal lipid components of GP and GSA group were significantly lower than the controlgroup. 2. During the feeding experiment, VLDL and LDL increase while HDL decrease in all group. However the degree of VLDL and LDL increase and HDL decrease were signficantly small in GP and GSA group compared with control group. 3. In the excretion of bile acid and neutral sterol, all experiment group showed increased excretion in the comparison of control group.

식이 Cholesterol의 수준에 따라 우유가 흰쥐의 Cholesterol 대사에 미치는 영향 (Effect of Milk on Cholesterol Metabolism of Rats with Different Levels of Dietary Cholesterol)

  • The effect of milk in low and high cholesterol diet was invesigated on serum cholesterol metabolism and lipid contents of serum, aorta, liver of rats. Weanling male Sprague-Dawley rats were divided into low(0.01% w/w) and high(1.01% w/w) cholesterol-diets groups. Low cholesterol groups subdivided into four groups ; control group was given water and three milk groups were given low heat milk(LM), ultra-high heat milk(HM), and powder milk(PM), respectivily, instead of water. High cholesterol groups were consisted of three groups ; control, LM, and HM groups. After feeding these experimental diets for six weeks, lipid levels were measured in serum and tissure and dried feces were analyzed for neutral and acidic sterols. Results obtained from this study are as follows : 1) Nutrient intakes, body weight gains and aorta weights did not differ among groups, but liver weights were higher in high cholesterol fed rats than low cholesterol fed rats. 2) Serum protein contents were increased independently by intakes of high cholesterol and milk. 3) Serum total cholesterol and triglyceride levels were increased but phospholipid levels and HDL-cholesterol/total-cholesterol ratios were decreased by high cholesterol in diet. And milk supplementation decreased serum total cholesterol and triglyceride levels but increased phospholipid levels and HDL-cholesterol/total-cholesterol ratios. 4) Contents of cholesterol and triglyceride in aorta and liver were elevated by dietary high cholesterol and lowered by consumption of all three types of milk. 5) Levels of cholesterol and triglyceride among serum, aorta and liver were highly correlated (r=0.7-0.9, p<0.001). 6) Fecal excretion of total sterols was three times high in high cholesterol group, compared with low cholesterol groups and were increased about 20% by milk consumption. 7) The effects of milk were more pronouncely shown in low cholesterol groups and mostly confined to LM and HM groups, rarely shown in PM group. It is concluded from the present study that milk had the hypolipidemic as well as hypocholes terolemic effect, which appears to be mediated through increased fecal bile acid excretion. But the effect is likely to be shaded by excess consumption of dietary cholesterol and was almost absent in powder milk.

실험적으로 동맥경화증을 유발시킨 랫드에서 ${\beta}$-Glucan의 동맥경화 방어기전에 대한 연구 (Preventive Mechanisma of ${\beta}$-Glucan on the Experimental Atherosclerosis in Rats)

  • Mechanisms for the hypocholesterolemic effetcs of $\beta$-glucan remain unclear. Rats were divided into 3 groups; normal control group, atherogenic group(oral administration of cholesterol 40 mg/kg/day and vitamin $D_2$320,000 IU/kg/day),${\beta}$-glucan treatment group(oral administration of atherogenic treatment and ${\beta}$-glucan 0.135 g/kg/day). The ${\beta}$-glucan treatment group showed moderate increases of serum lipids concentration compared with atherogenic group. In histopathological examination, aortas showed no critical lesions. The total fecal neutral sterols and bile acids excreted for 6 days was increased compared with both normal and atherogenic group. These result\ulcorner suggest that mechanisms for the hypocholesterolemic effect of ${\beta}$-glucan on rats were due to the inhibition of cholesterol absorption in the intestinal lumen and acceleration of cholesterol catabolism in the liver.

  • The study was designed to observe the effect of dietary calcium and fats on plasma cholesterol level, hepatic microsomal fluidity and HMG-CoA reductase activity as well as the excretion of fecal bile acids and neutral sterols in 1, 2-dimethylhydrazine(DMH)-treated rats. Male Sprague Dawley rats, at 7 weeks of age, were divided into 2 groups, 0.3% and 1.0% Ca levels and each group again subdivided into 2 groups of corn oil and perilla oil. Each rat was intramuscularly infused with DMH for 6 weeks to give total dose of 180mg/kg body weight and also fed experimental diet containing 15%(w/w) different fit and Ca(0.3% or 1.0%) for 20 weeks. High dietary calcium(1.0%) did not significantly influence on plasma cholesterol as well as hepatic microsomal fluidity and HMG CoA reductase activity, but significantly reduced the excretion of total bile acid per gram of faces and increased the excretion of total neutral sterol. However, high dietary Ca reduced the excretion of secondary bile acid(deoxycholic and lithocholic acids) which was known as promoter for colon cancer. Perilla oil rich in n-3 ${\alpha}$-linolenic acid significantly decreased plasma cholesterol by increasing hepatic microsomal fluidity compared with corn oil, but did not influence on HMG CoA reductase activity. Perilla oil did not influence on fecal excretion of total and primary bile acids, but reduced the excretion of secondary bile acids. Therefore, it could be recommended to consume more fish product and food rich in calcium and use more perilla oil in meal preparation to prevent from coronary hear disease and colon cancer especially when high fit diet has been practiced.
