환경수계 레지오넬라균 오염 지표의 메타분석 (Meta-Analysis of Risk Factors for Contamination of Environmental Waters by Legionella)

  • 레지오넬라균에 의한 자연환경의 수체 및 각종 시설 용수의 오염도를 조사하는데 적용할 수 있는 오염도 지표를 통상적으로 측정되는 수질항목 중에서 선정하려는 목적으로, 다양한 수체에 적용된 수질항목들이 레지오넬라균 오염도와 어느 정도 유의한 상관관계를 맺는 지에 관한 메타분석을 실시하였다. 환경부의 수질관리 지침에서 레지오넬라균 오염도의 위험지수로 사용되고 있는 대장균의 경우, 총대장균군 항목과 레지오넬라균 검출여부의 교차비(odds ratio; OR)로 분석하였는데, 교차비는 1.05(0.36-3.12, 95% CI)로, 오즈의 차이가 없을 확률이 0.92에 달하였다. 또한, 총대장균군 자료는 유의한 수준의 자료간 이질성을 보였으며(P=0.008), 무작위적인 자료간 이질성이 전체 교차비 변량의 63%를 차지하였다. 종속영양세균수(HPC)의 교차비가 2.72(2.04-3.63)로 1 보다 매우 유의하게 높았고(P<0.0001), 자료간 이질성이 없었다($Q_{df=8}$=12.7, P=0.12). 탁도와 염소농도, 철과 구리 이온의 농도 등이 레지오넬라균 검출여부와 함께 조사되는 통상적 수질항목이었으나, 자료수의 부족과 정량 자료의 통일성 부족으로 적절한 메타분석이 수행 될 수 없었다. 결론적으로, 분변균의 오염도 보다, 수중 레지오넬라균 서식지(아메바와 생물막)의 양을 표현하는 종속영양세균수가 레지오넬라균의 오염도 지표로 활용될 수 있을 것으로 사료되며, 실험 방법의 표준화와 경계값을 판단하는 후속 연구가 필요하다.

Microbial Community of Healthy Thai Vegetarians and Non-Vegetarians, Their Core Gut Microbiota, and Pathogen Risk

  • Pyrosequencing analysis of intestinal microflora from healthy Thai vegetarians and non-vegetarians exhibited 893 OTUs covering 189 species. The strong species indicators of vegetarians and non-vegetarians were Prevotella copri and Bacteroides vulgatus as well as bacteria close to Escherichia hermanii with % relative abundance of 16.9 and 4.5-4.7, respectively. Core gut microbiota of the vegetarian and non-vegetarian groups consisted of 11 and 20 different bacterial species, respectively, belonging to Actinobacteria, Firmicutes, and Proteobacteria commonly found in both groups. Two species, Faecalibacterium prausnitzii and Gemmiger formicilis, had a prevalence of 100% in both groups. Three species, Clostridium nexile, Eubacterium eligens, and P. copri, showed up in most vegetarians, whereas more diversity of Collinsella aerofaciens, Ruminococcus torques, various species of Bacteroides, Parabacteroides, Escherichia, and different species of Clostridium and Eubacterium were found in most non-vegetarians. Considering the correlation of personal characters, consumption behavior, and microbial groups, the age of non-vegetarians showed a strong positive correlation coefficient of 0.54 (p = 0.001) to Bacteroides uniformis but exhibited a moderate one to Alistipes finegoldii and B. vulgatus. Only a positive moderate correlation of body mass index and Parabacteroides distasonis appeared. Based on the significant abundance of potential pathogens, the microbiota of the non-vegetarian group showed an abundance of potential pathogen varieties of Bilophila wadsworthia, Escherichia coli, and E. hermannii, whereas that of the vegetarian group served for only Klebsiella pneumoniae. These results implied that the microbiota of vegetarians with high abundance of P. copri and low potential pathogen variety would be a way to maintain good health in Thais.

Comparative Efficacy of Plant and Animal Protein Sources on the Growth Performance, Nutrient Digestibility, Morphology and Caecal Microbiology of Early-weaned Pigs

  • The present study was conducted to evaluate and compare the effects of various animal and plant protein sources on piglet' performance, digestibility of amino acids and gut morphology in weaned pigs until 28 days after weaning. The plant protein sources used were soybean meal (SBM), fermented soy protein (FSP), rice protein concentrate (RPC); and animal protein sources tested were, whey protein concentrate (WPC) and fishmeal (FM). Iso-proteinous (21%) diets were formulated and lysine (1.55%) content was similar in all the diets. The level of each protein source added was 6% by replacing SBM to the same extent from the control diet containing 15% SBM. The ADG was higher (p<0.05) in the groups fed animal proteins as compared with plant proteins at all the levels of measurement, except during 15-28 days. The highest ADG was noted in WPC and FM fed diets and lowest in SBM fed diet. The feed intake was higher in animal protein fed groups than plant proteins at all phases, but the feed:gain ratio was not affected by protein sources except during overall (0 to 14 day) measurement which was improved (p<0.05) in animal protein fed diets compared to plant protein sources. The digestibilities of gross energy, dry matter and crude protein were higher in animal protein fed groups than for plant protein fed sources. The apparent ileal digestibilities of essential amino acids like Leu, Thr, and Met were significantly (p<0.05) higher in animal proteins fed animals as compared with plant protein fed animals. But the apparent fecal digestibilities of essential amino acids like Arg and Ile were significantly higher (p<0.05) in plant protein diets than animal protein sources. The villous structure studied by scanning electron microscope were prominent, straight finger-like, although shortened and densely located in FM fed group as compared with others. The lactic acid bacteria and C. perfringens counts were higher in caecal contents of pigs fed plant proteins than the animal proteins. Overall, it could be concluded that animal protein sources in the present study showed better effects on growth performance, nutrient digestibility and gut morphology than plant protein sources.

The Effects of Cellulose, Pectin and Starch on Standardized Ileal and Apparent Total Tract Amino Acid Digestibilities and Bacterial Contribution of Amino Acids in Feces of Growing Pigs

  • Eight ileally cannulated pigs (BW $35.9{\pm}0.9kg$) were randomly allotted according to a $4{\times}3$ Latin square design to determine the effects of cellulose, pectin and starch on standardized ileal digestibility (SID) and apparent total tract digestibility (ATTD) of crude protein (CP) and amino acids (AA) as well as on the bacterial AA contribution in feces. The pigs were fed the control diet (20.2% CP, % dry matter (DM)) or one of the three experimental diets in which 25% of the control diet was substituted by cellulose, starch or pectin. Due to this substitution, dietary CP levels were lower in the cellulose (15.5% CP, % DM), pectin (15.4% CP, % DM) and starch diet (15.2% CP, % DM). Following a 15-d adaptation period, feces were collected for 5 d and ileal digesta for a total of 24 h. Starch increased SID of CP, while cellulose and pectin had no significant effect on the digestibility of CP. Overall, starch supplementation resulted in higher (p<0.05) SID values of histidine, isoleucine, threonine, alanine, aspartic acid, cysteine, glycine and serine compared with cellulose, while pectin decreased (p<0.05) SID of valine and proline compared with the starch and control diet. Both cellulose and pectin reduced (p<0.05) the ATTD of CP and AA, while starch decreased (p<0.05) ATTD of phenylalanine, alanine, proline and serine compared with the control. With regard to bacterial AA composition of the fecal mixed bacterial mass (MBM), cellulose supplementation increased (p<0.05) its content of N and almost all AA, except for valine, while pectin caused higher contents of arginine, histidine and proline compared with the control (p<0.05). The bacterial contribution of arginine in feces was higher (p<0.05) in the cellulose treatment, while pectin reduced (p<0.05) the bacterial contribution of leucine, alanine, glutamic acid and proline in feces compared with the control. In conclusion, the effects of cellulose, starch and pectin on SID were rather small. Bacterial activity in the large intestine can only explain the reduced ATTD values for arginine in the cellulose treatment, but not for the other AA in the cellulose and pectin treatments, suggesting higher endogenous losses of these AA in the large intestine.

수영만의 수질환경과 대장균 2. 동계의 수질환경과 대장균에 관하여 (The Environmental Factors and Coliform Group in Suyeong Bay 2. The environmental factors and coliform group during winter in Suyeong bay)

  • 부산인구의 약 23.5%인 110만명 이상이 거주하는 수영천으로 인하여 도시하수, 가정폐수 등의 오염의 영향을 직접받는 수영만에서 동계동안(1990년 11월, 12월, 1991년 1월, 2월) 대장균군의 분포를 조사하고 여기에 관련된 환경요인을 분석한 결과를 아래와 같이 요약하였다. 염분은 16.84~34.93%의 범위였으며, 용존산소는 동계임에도 불구하고 수영만 하구부근에서는 $1m{\ell}/{\ell}$ 이하의 분포를 보였으며, COD는 0.48~5.06ppm, 영양염류중 아질산염은 $0.42{\sim}18.82{\mu}g-at/{\ell}$, 질산염 $0.5{\sim}53.23{\mu}g-at/{\ell}$, 암모니아는 $3.73{\sim}56.16{\mu}g-at/{\ell}$, 규산염은 5.18~147.29범위였다. 조사기간 대장균군수의 변화는 $1.1{\times}10^2{\sim}2.4{\times}10^5/100m{\ell}$, 분변성 대장균군은 $1.7{\times}10{\sim}3.5{\times}10^4/100m{\ell}$였으며, TC/FC비는 1.73~58.49로 크게 나타났다. 분리된 117개 균주중 E. coli 가 68.4% 로 가장 우점하였으며, 그 중 E. coli I type이 97.47%로 가장 많은 부분을 차지하였다.

말 분변을 이용한 주스박의 in vitro 발효 특성 (In vitro Fermentation Characteristics of Juice Pomaces Using Equine Fecal Inoculum)

  • 본 연구는 바실러스, 효모 그리고 유산균으로 발효한 다양한 주스박들과 말 분변을 첨가한 in vitro 발효 시험을 통해 pH, 건물 분해율, 암모니아성 질소, 가스 발생량, 휘발성 지방산의 변화를 조사하였다. 주스박 첨가에 따른 발효액의 pH의 범위는 6.4~7.1로 미생물 발효에 의한 소화가 정상적으로 나타났다. 48시간 후 발효액에서 $10^9CFU/m{\ell}$ 정도 미생물이 증식함에 따라 사용한 주스박들이 장내 미생물에 의한 소화작용에 유용하게 이용될 것으로 판단되었다. 건물 분해율은 배양 12시간 이후부터 증가하여 48시간째에 사과박 39.19%, 포도박 38.22%, 당근박 37.02%, 감귤박 36.2% 그리고 혼합박 34.35%로 나타났다. 암모니아성 질소의 농도는 배양 12시간까지는 전 처리구에서 $1.5mg/100m{\ell}$의 수준이었으며 배양 24시간 이후부터 급격히 증가하였다. 가스 발생량은 혼합박이 가장 낮았으며, 배양 12시간 이후부터 급격히 증가하였다. 총 VFA는 배양 24시간부터 증가하여 48시간에 가장 높았다. 본 연구결과로부터 발효 주스박은 말 in vitro 소화과정에서 적정 pH를 유지하면서 건물 분해가 진행되었고 말의 맹장과 대장에서 섬유소 분해 미생물의 작용이 활발히 이루어질 수 있음을 제시하였다.

어린잎채소 생산 농장의 위생지표세균과 병원성미생물 오염도 조사 (Investigation of Microbial Contamination Level during Production of Baby Leafy Vegetables)

  • 본 연구는 어린잎 채소와 생산환경에서 미생물학적 오염도를 조사하고자 수행하였다. 이를 위하여 11종의 어린잎채소와 종자, 관개용수, 상토, 작업도구 및 작업자 장갑 등 총 126개의 시료를 채취하여 위생지표세균(대장균군, E. coli)과 병원성미생물(Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus)을 조사하였다. 그 결과, 종자와 관개용수를 제외한 대부분의 시료에서 대장균군이 검출되었다. E. coli는 분석시료 중 10.3% (13/126)에서 검출되었으며 검출된 시료로는 관개용수, 칼, 작업자 장갑, 및 다채, 적무, 청경채였다. 또한 B. cereus는 상토, 작업도구 및 청경채, 비트, 적근대 등 총 38% (48/126)의 시료에서 검출되었다. 한편 E. coli O157:H7, Salmonella spp., L. monocytogenes는 검출되지 않았다. 본연구의 결과로 미루어 볼 때 어린잎채소는 미생물 안전성면에서는 크게 우려할 수준은 아니지만 어린잎채소의 미생물오염을 사전 예방하는 차원에서 농업현장에서 쉽게 실천할 수 있는 위생관리 기술 개발과 보급이 필요하다고 판단된다.

두 돼지 종의 다양한 성장단계에 따른 장내미생물 비교분석 (Comparison Analysis of Swine Gut Microbiota between Landrace and Yorkshire at Various Growth Stages)

  • 연구에서는 차세대염기서열분석법(Next Generation Sequencing)을 이용하여 상업적으로 농가에 가장 많이 보급되어 있는 요크셔와 랜드레이스를 포함한 두 종의 장내미생물생태 분석을 실시하였다. 박테리아의 16S rRNA 유전자는 분변샘플로부터 추출한 DNA에서 V4 지역을 증폭할 수 있도록 디자인된 유니버설 프라이머 세트를 이용하여 증폭되었다. 두 종에 대한 장내미생물생태 비교분석은 성장단계에 따라 차이를 보이는 반면, 종에 따른 차이는 거의 없다는 것을 확인하였다. 하지만, 두 종간의 장내미생물생태 내에서 특정 미생물의 수가 차이가 있다는 것을 확인하였다. 요크셔는 특히 섬유질 소화를 통해 에너지 생산률을 높여준다고 보고된 바가 있는 Xylanibacter 속(Genus)의 미생물을 많이 포함하는 것으로 나타났다. 또한, 랜드레이스는 숙주 내에서 면역에 중요한 역할을 하는 것으로 알려진 Clostridium_IV 종을 상당히 많이 포함하는 것으로 나타났으며, 반면 요크셔는 기회감염미생물들을 많이 포함하는 것으로 나타났다. 본 연구는 요크셔와 랜드레이스를 포함한 두 종간의 장내미 생물생태 비교분석을 통해 그 차이점이 종에 의한 차이보다는 성장단계에 따라 큰 차이가 있다는 것을 확인하였다. 하지만 두 종 사이에서 성장에 영향을 미칠 가능성이 있는 몇 장내미생물의 수가 차이가 있다는 것을 확인하였다.

곡물 및 사료오염 데옥시니발레놀 및 대사체에 의한 인축질환 연계 생체지표 및 바이오모니터링 (Human and Animal Disease Biomarkers and Biomonitoring of Deoxynivalenol and Related Fungal Metabolites as Cereal and Feed Contaminants)

  • Deoxynivalenol (DON) and related trichothecene mycotoxins are extensively distributed in the cereal-based food and feed stuffs worldwide. Recent climate changes and global grain trade increased chance of exposure to more DON and related toxic metabolites in poorly managed production systems. Monitoring the biological and environmental exposures to the toxins are crucial in protecting human and animals from toxicities of the hazardous contaminants in food or feeds. Exposure biomarkers including urine DON itself are prone to shift to less harmful metabolites by intestinal microbiota and liver metabolic enzymes. De-epoxyfication of DON by gut microbes such as Eubacterium strain BBSH 797 and Eubacterium sp. DSM 11798 leads to more fecal secretion of DOM-1. By contrast, most of plant-derived DON-glucoside is also easily catabolized to free DON by gut microbes, which produces more burden to body. Phase 2 hepatic metabolism also contributes to the glucuronidation of DON, which can be useful urine biomarkers. However, chemical modification could be very typical depending on the anthropologic or genetic background, luminal bacteria, and hepatic metabolic enzyme susceptibility to the toxins in the diet. After toxin exposure, effect biomarkers are also important in estimating the linkage and mechanisms of foodborne diseases in human and animal population. Most prominent adverse effects are demonstrated in the DON-induced immunological and behavioral disorders. For instance, acutely elevated interleukin-8 from insulted gut exposed to dietaty DON is a dominant clinical biomarker in human and animals. Moreover, subchronic exposure to the toxins is associated with high levels of serum IgA, a biological mediator of IgA nephritis. In particular, anorexia monitoring using mouse models are recently developed to monitor the biological activities of DON-induced feed refusal. It is also mechanistically linked to alteration of serotoin and peptide YY, which are promising biomarkers of neurological disorders by the toxins. As animal-alternative biomonitoring, huamn enterocyte-based assay has been developed and more realistic gut mimetic models would be useful in monitoring the effect biomarkers in resposne to toxic contaminants in the future investigations.

2019년 충남지역 고등학교에서 발생한 다병원체에 의한 집단식중독의 역학적 분석 (Epidemiological Analysis of a Food Poisoning Outbreak Caused by Multiple Pathogens in a High School in Chungnam Korea, 2019)

  • Objectives: This study was performed in order to report the epidemiological features of a food poisoning outbreak caused by multiple pathogens in a high school in Chungcheongnam-do Province, Korea in April 2019 and to suggest measures to prevent a similar incidence. Methods: A total of 39 patients with diarrhea were examined. Environmental samples were obtained from 6 food handlers, 4 food utensils, 72 preserved foods served during the food poisoning outbreak, 9 door handles, 10 drinking water samples from water dispensers, and 6 ground water samples from water taps. These analyzed to detect viruses and bacteria. Results: Among the 39 patients, 21 cases (53.8%) of enteroaggregative E. coli (EAEC), 7 cases of Staphylococcus aureus (17.9%), and 17 cases of norovirus (43.6%) were positive, and in 16 of the cases a co-infection with at least one other pathogen were observed. EAEC was assumed to be transmitted from contaminated drinking water because it was also detected in the water sample from a water dispenser in the dormitory. Staphylococcus aureus was isolated only in the fecal samples of patients, meaning it was not possible to trace its origin. The genotype of norovirus detected in the drinking water and ground water was consistent with that isolated from patients, and it was determined that the norovirus infection originated from the school's water environment. Conclusions: These findings indicate that a lack of environmental hygiene management related to school meals caused the food poisoning incident. In particular, a lack of management of drinking water, water supply, and personal hygiene should be pointed out. This should be urgently addressed and continuous monitoring should be carried out in the future. In addition, students and staff should be educated and trained to improve their personal hygiene.
