The herbal components should be transformed to bioactive compounds by the intestinal bacteria and then expressed the pharmacological action of herbal medicines. Human fecal enzyme activities related to the metabolism of herbal components were measured. The metabolic activities of puerarin, poncirin, glycyrrhizin, ginsenoside Rb1 and ginsenoside Rb2 to their bioactive compounds were $3.5{\pm}1.18,{\;}333.1{\pm}183.64,{\;}95.7{\pm}107.1,{\;}20.8{\pm}10.32{\;}and{\;}20.8{\pm}13.3{\;}{\mu}mo1/h/g$, respectively. The profile of these metabolic activities of glycyrrhizin and ginsenosides were not changed even if herbal extracts, water extract of Glycyrrhizae Radix and Ginseng Radix, instead of the isolated compounds were used . All the enzyme activities tested were not different between male and female, and between ages. However, the difference of these enzyme activities in individuals was significant. These results suggest that the metabolic activity of herbal components to bioactive compounds may be a factor of constitutional classification, and could be available for constitutional classifications, if the constitutional herbal medicines were used .

라오스의 Houay Pano 유역은 상업적 조림으로 인해 2011년부터 2013년까지 급속한 토지이용 변화를 겪어왔다. 본 연구는 이러한 토지이용변화가 박테리아 거동에 어떠한 영향을 주는지 이해하기 위해 Soil and Water Assessment Tool (SWAT) 모형을 활용한 박테리아 거동 모델링을 수행하였다. SWAT 모형은 수치 표고 모델, 토양 특성, 토지 이용 등의 정보를 종합하여, 유역 내수량 및 수질의 변화를 모의할 수 있는 모형으로, 본 연구는 대표적인 분원성 지표 세균 (Fecal Indicator Bacteria)인 대장균 (Escheichia coli, E. coli)을 대상으로 모델링을 수행하였다. SWAT 모형은 지표면 위 박테리아를 1)식물 위, 2)토양 용액상, 3)토양 입자상으로 구분하여 모의한다. 각 상태로 분할된 박테리아는 소멸 (die-off), 씻김 (wash-off), 침투, 표면 유출을 통한 수계로의 이동 등의 단계를 통하여 유역 내에서 거동한다. 본 연구는 서로 다른 기후의 영향을 배제하기 위해 각 토지이용 시나리오를 (2011, 2012, 2013) 실제 기후 조건과 동일 기후(2011-2013 평균) 조건으로 분류하여 분석하였다. 실제 기후 조건에서 SWAT 모형은 표면 유출, 토사 유출, E. coli 거동에 대해 2011년부터 2012년까지 감소, 2012년부터 2013년까지 증가로 모두 동일한 양상을 모의하였다. 이는 강수량의 양상과 동일한 것으로, 강수량이 표면 유출의 양을 결정하고, 달라진 표면 유출에 따라 토사 유출과 E. coli 거동이 결정되기 때문이다. 하지만 동일 기후 조건에서는, E. coli 거동 동인인 표면 유출과 토사 유출이 비교적 일정해짐에 따라, 각 상태로 분할된 박테리아의 초기 부하량값이 E. coli 거동을 결정하는 주된 요인임을 확인 할 수 있었다. 따라서 초기 부하량 분할에 활용되는 엽면적 지수 (Leaf Area Index)와 분배계수 (BACTKDDB)의 정확도가 요구된다. 추가적으로 본 연구는 박테리아의 유입원인 비료 모델링과, LAI를 활용한 박테리아 초기 부하량 산정, 토양 특성 변수와 토지 이용 변수의 분리, 지하수를 통한 박테리아 거동 등을 중심으로 SWAT 모형의 한계점과 개선점을 제시하였으며, 본 연구 결과는 토지이용변화가 박테리아 거동에 주는 영향을 모형적으로 이해하고, 또한 추후 박테리아 모델링 개발에 도움을 줄 것으로 예상된다.
Campylobacter species are known to the high optimum growth temperature ($42^{\circ}C$) and the cause of enteritis in people. Erythromycin has a curative effect for enteritis caused by the bacteria. However, the rate of erythromycin-resistant bacteria was not well known until recently in Korea. Swine are one of sources of the infection with a Campylobacter species which cause the symptom of a high temperature. In this study, we cultured rectum fecal specimens of 100 pigs in an area of Buan-gun, Jeonbuk Province during July 2009. As a result, the detection rate of C. jejuni and C. coli and the rate of erythromycin-resistant bacteria for the separated Campylobacter species on the condition of high temperature were investigated. The possession or not of hipO and glyA gene and ciprofloxacin-resistant gene gyrA was also reviewed with biochemical characteristics and PCR.

This study aims to assess the efficacies of exogenous enzymes, derived from invertebrate gut-associated microbes, as feed additives, in reducing volatile organic compound (VOC) emissions using an in vitro pig intestine continuous fermentation system. An in vitro continuous fermentation model was used to simulate a comparable bionic digestion system by co-reacting feed, enzymatic additives (arazyme, mannanase, and xylanase, derived from the gut bacteria of Nephila clavata, Eisenia fetida, and Moechotypa diphysis, respectively), and gastrointestinal microbes, followed by an analysis of their correlations. A significant correlation was observed between exogenous enzyme supplementation and reduced VOC emissions in the fecal phase of continuous fermentation (p < 0.05). The concentration of VOCs decreased by 3.75 and 2.75 ppm in the treatment group following arazyme and multi-enzyme supplementation, respectively, compared to that in the control group (7.83 ppm). In addition, supplementation with arazyme and multiple enzymes significantly affected the microbial composition of each fermentation phase (p < 0.05). In particular, Lactiplantibacillus pentosus and Pediococcus pentosaceus, which changed in abundance according to arazyme or multi-enzyme supplementation, exhibited a positive relationship with VOC emissions. These results suggest that exogenous enzymes derived from invertebrate gut-associated bacteria can be efficiently applied as feed additives, leading to a reduction in VOC emissions.
This experiment was carried out to evaluate the sanitary quality of commercially frozen sea foods. One hundred and sixteen samples in six different items from several refrigeration plant in Busan city were examined from March to December in 1974. In addition, the changes in bacterial density through the process from thawing, round or semifilleted frozen alaska pollack to the finishing as frozen fillet blocks were observed. To evaluate the sanitary quality, sanitary indicative bacteria such as total coliform, fecal coliform, fecal streptococci and enterococci as well as plate counts were determined. From the results, the median value of fecal coliform MPN was 20 per 100 grams of the samples and that of enterococci was 790. The median value of plate counts was $2.2\times10^4$ per gram. The plate counts were not correlated with the number of sanitary indicative bacteria. The results suggest that enterococci could be used advantageously in preference to coliform organisms as indicative bacteria for the evaluation of sanitary quality of frozen sea foods. The plate counts at $20^{\circ}C$ of the samples were 14 times higher than that at $35^{\circ}C$. Geometric mean of total coliform MPN was 310 and that of enterococci was 143. Bacterial density was reduced by fleering. Morethan 50 percent for total coliform MPN and $35^{\circ}C$ plate counts, and about 35 percent for enterococci MPN and $20^{\circ}C$ plate counts were reduced under the contact freezing unit which was generally operated at $-40^{\circ}C$. About fifty-five percent of the samples were negative in fecal coliform test and 10 percent of those were exceeded $1.0\times10^5$ per gram in $35^{\circ}C$ plate counts.
This study was conducted to determine the effect of probiotics as an alternative for antibiotics on growth performance, nutrient digestibility, noxious gas emission and fecal microbial population in growing piglets. A total of 96 piglets ($22.5{\pm}1.3$kg average body weight) were allotted to 3 different treatment groups and replicated 4 times with 8 piglets per replicate in randomized complete block design. Treatments were T1) (Control, basal diet+0.2% antibiotics), T2) 0.2% probiotics complex and T3) 0.3% Bacillus probiotics. During the whole experiment period, there were no differences (p>0.05) in average daily gain (ADG), average daily feed intake (ADFI) and feed efficiency. However, digestibility of dry matter, crude protein, ether extract, nitrogen free extract and crude ash were showed higher in probiotics groups (T2 and T3) than those of control. In noxious gas emission, ammonia, amine, hydrogen sulfide and mercaptan were significantly (p<0.05) reduced in T2 and T3 treatments compared to those in control. Moisture content of feces was not significantly different among treatments. The colony forming units (CFU) of total bacteria, E. coli and thermoduric bacteria in feces were significantly different among treatments. The CFU of total bacteria, E. coli and thermoduric bacteria in T3 treatment were reduced by feeding probiotics B. From this study, we suggest that probiotics A and B are likely able to improve the growth performance and nutrients digestibility, reduce noxious gas emission and change the fecal microbial composition in growing piglets.
Careful cleaning and disinfection of pigpens is essential to prevent disease spread and avoid the resultant economic losses. Hygiene in pigpens is generally evaluated by visual monitoring supplemented with bacteriological monitoring, which includes counting the total aerobic bacteria (TAB) and/or fecal indicator bacteria (FIB). However, these methods present drawbacks such as time and labor requirements. As adenosine triphosphate (ATP) is ubiquitous in all living organisms including microorganisms, this study aimed to directly compare the results of microbial assessment and ATP quantification, and to suggest possible detailed application methods of the ATP test for hygiene evaluation in pigpens of a farrowing unit. Before and after standard cleaning procedures, samples were collected from the floor corner, floor center, and feeding trough of four pigpens at different time points. No FIB were detected and both the TAB and ATP levels were significantly decreased in the floor center area after cleaning. FIB were continuously detected after cleaning and disinfection of the floor corners, and there was no significant ATP level reduction. The feeding trough did not show any significant difference in these values before and after cleaning, indicating insufficient cleaning of this area. The levels of TAB and ATP after cleaning were significantly correlated and the average ATP value was significantly lower in the absence of FIB than in their presence. In the absence of standard references, a more thorough hygiene management could be achieved evenly by supplementing cleaning or disinfection based on the lowest ATP results obtained at the cleanest test site, which in the present study was the floor center. Overall, these results indicate that the on-farm ATP test can be used to determine the cleanliness status, in addition to visual inspection, as an alternative to laboratory culture-based testing for the presence of microorganisms.
The aim of this study was to evaluated the efficacy of mixture of Lactic Acid Bacteria (LAB) and bifidobacteria supplement, which are contained with Lactobacillus acidophilus, Bifidobacterium longum SPM1205, and Pediococcus pentosaceus for the management of constipation in animal model and clinical trials. 5 ICR mice and 4 female constipation subjects were orally taken mixture of LAB and bifidobacteria for 2 weeks. We investigated the number of fecal LAB and harmful enzymes activities before and after mixture of LAB and bifidobacteria application. As a result, fecal LAB count was increased and harmful enzymes activities of intestinal microflora were generally decreased after mixture of LAB and bifidobacteria application. Also, 61 female subjects were randomly assigned to receive either mixture of LAB and bifidobacteria or lactose and were taken three times a day for 2 weeks. Then, we analyzed mixture of LAB and bifidobacteria effect through the questionnaires. Daily consumption of this mixture of LAB and bifidobacteria improved the constipation in constipation group (56.3%) compared with lactose application group (26.7%). Furthermore, after mixture of LAB and bifidobacteria treatment, frequency of hard stool decreased from 0.22 to 0.03. These results indicated that mixture of LAB and bifidobacteria application is effective to improve the constipation.

This study was conducted to investigate the effects of probiotic mixture on fecal ammonia, caecal microorganism, immune response, egg quality and production in layer under heat stress (HS).A total of four hundred 50 week olds Hy-Line Brown layers were randomly divided into four groups of 100 heads each: C (control, room temperature $25^{\circ}C$), HS (heat stress $33^{\circ}C$), PM (HS plus probiotic mixture 500, 750 mg/kg of diets). Egg production, egg quality, spleen weight, blood IgG and lymphocyte concentrations were increased in the PM group compared to the HS group, while mortality, the heterophil:lymphocyte (H:L) ratio, and corticosterone levelswere significantly decreased. Lactobacillus was increased in the PM group compared to the HS group, but E. coli, coliform bacteria and aerobic bacteria were significantly reduced. Fecal ammonia production was significantly increased in the HS group compared to the PM group. In conclusion, the results of this study that these mixed probiotics can reduce the heat damage of the summer laying hens and can be an effective nutritional strategy to reduce odor generation from feces, and to improve egg quality and laying production through immune response and caecal microbial balance.
Hand washing is an important component of hygiene program for food handlers. Hands can be a source of direct or indirect contamination of foods with pathogenic microorganisms. In this study, the effectiveness of hand washing methods and the use of 70% alcohol solution against transient skin bacteria was tested in an university foodservice facility. 70% alcohol solution is sprayed for 5 seconds automatically when hands are placed in the dispenser. Samples were taken using swab technique in meat cutting area, vegetable trimming area, and vegetable cutting area: before and after washing hands according to planned methods, and after being sprayed with 70% alcohol solution after washing hands. The bacteriological analysis of total plate counts, coliform, fecal coliform of food handlers' hands was done. Statistical data analysis was completed with Mann-Whitney U test and Kruskal-Wallis model using the SPSS program. The levels of initial contamination of workers' hand were significantly different by the work areas($x^2$=9.156, p<0.01). Workers in the vegetable trimming area had more heavily soiled hands than in the other work areas. Mean of TPC counts and coliform was 8.97×$10^5$ CFU/12.4$cm^2$, 2.93×$10^2$ MPN/12.4$cm^2$ respectively, but fecal coliform was not detected. Transient bacteria were removed from hands after washing and using 70% alcohol solution but were not removed completely. Mean reduction percentage in TPC varied among work areas and ranged from 93.19% to 94.99%, and in coliforms from 97.31% to 100%. A significant difference in TPC was found between before and after hand disinfection (Z=-2.714, p<.01) and between standardized hand washing procedures and un-standardized hand washing procedures(z=-2.301, p<.01). Subjects using the hand sanitizer showed a great elimination of TPC(99.45% reduction), but this effect was valid only after following proper washing procedures. Based on the results, the most effective hand washing method was recommended as the combination of the standardized hand washing procedure with warm-water and use of the 70% alcohol solution. The results can be used to develop hand hygiene programs and training strategies for enhancing hand hygiene practices for food handlers in foodservice operations.
