The base sequences representing human- and cow-specific 168 rRNA gene markers identified in a T-RFLP analysis were recovered from clone libraries. The human- and cow-specific primers were designed from these sequences and their specificities were analyzed with fecal DNAs from human, cow, and pig. The AllBac primer set showed positive results for all human, cow, and pig samples, whereas the human-specific primer set showed positive result only for the human sample but not for the cow or pig samples. Likewise, the cow-specific primer set showed positive results only for the cow sample but not for the human or pig samples. Real-time PCR assay with these primers was developed for the identification and quantification of fecal pollution in the river water. The human- and cow-specific markers were detected in the order of 9 $\log_{10}$ copies per gram wet feces, which were two orders of magnitude lower than those of total Bacteroidales. For the river water samples, the human-specific marker was detected in $1.7-6.2\;\log_{10}$ copies/100 ml water, which was 2.4-4.9 orders of magnitude lower than those of total Bacteroidales. There was no significant correlation between total Bacteroidales and conventional fecal indicators, but there was a high correlation between Bacteroidales and the human-specific marker. This assay could reliably identify and quantify the fecal pollution sources, enabling effective measures in the watersheds and facilitating water quality management.
In 2005, the 5.84-Km length of Cheonggyecheon stream, previously covered with concrete road, was uncovered in the middle of Seoul, Korea. We investigated microbial water quality in various sites in Cheonggyecheon stream. We took water samples on three different days. The sampling sites included inflow water from upper stream (Mojeongyo), midstream (Ogansugyo), and downstream (Muhakgyo). Fecal pollution indicator microorganisms were measured by both IDEXX $Colilert^{(R)}$ and $Enterolert^{(R)}$. Microbial community from these sampling sites was also characterized based on 16S rRNA gene sequences. The average concentrations of total coliform are 5 CFU/100 mL, 1474 CFU/100 mL, and 1776 CFU/100 mL at Mojeongyo, Ogansugyo, and Muhakgyo, respectively. The average concentrations of fecal coliform were 28 CFU/100 mL, 47 CFU/100 mL in Ogansugyo, and Muhakgyo, respectively. The concentrations of other fecal indicator microorganisms including E. coli and Enterococcus sp. increased in downstream. When we characterized the microbial community, unique microbial community were discovered at different sampling sites. This study suggests that Cheonggyechoen stream is likely affected by non-point fecal sources and has unique microbial environment as the river flows downstream.

Colorectal cancer is a major cause of mortality worldwide. Fecal occult blood testing has proven a very effective screening tool for early detection and mortality reduction. The aim of this study was to determine predictors factors related to fecal occult blood testing using the Health Belief Model method among residents of Bushehr, Iran. A cross sectional study was performed on a sample of 600 men and women more than 50 years of age. The sample was selected by a convenience method from patients referred to public and private laboratories throughout the city. Each subject filled out a questionnaire which was designed and developed based on Health Belief Model constructs. Statistical analysis was conducted using ANOVA, T-test, chi-square test, and logistic regression. Fecal occult blood tests were performed on 179 (29.8%) out of 600 subjects, of which 95 patients (58.1%) did a periodic examination test and 84 patients (46.9%) had a doctor's advice for testing. According to the logistic regression model, the perceived barriers (P=0.0, Exp(B)= 0.3), perceived benefits (P<0.01, Exp(B)= 1.9) and self-efficacy (P<0.01, Exp(B)= 1.6) were predictive factors related to occult blood testing among subjects. The results showed that reducing people's perception of barriers to testing, increasing perceived benefits of screening, and reinforcing self efficacy can have major effect in increasing the rate of fecal occult blood screening for colorectal cancer prevention.
The influence of dietary supplementation with blended essential oil on growth performance, nutrient digestibility, blood profiles and fecal characteristics was evaluated in 125 crossed ((Duroc ${\times}$Yorkshire) ${\times}$Landrace) pigs (6.21${\pm}$0.20 kg initial body weight and 21 d average age). The pigs were allotted to the following treatments: i) NC (antibiotic free diet), ii) PC (NC diet+44 ppm tylosin), iii) T1 (NC diet+0.1% essential oil), iv) T2 (NC diet+0.1% essential oil (with 0.3% Benzoic acid)) and v) T3 (NC+22 ppm tylosin and 0.05% essential oil). Average daily gain (ADG) was improved in the T2 group on d 14 (p<0.05). In addition, nutrient digestibility was partially affected (both positively and negatively) by the treatments. Furthermore, the immune system was stimulated and the fecal pH and fecal noxious gases were improved in pigs that received the diets supplemented with essential oil (p<0.05). The appearance and score of diarrhea also tended to be lower in pigs that were subjected to the essential oil treatments. Collectively, the results of this study indicate that supplementation of the diet with blended essential oils could replace treatment with antibiotics to improve growth performance and fecal characteristics.
Objective: To investigate the effect of host genetics on gut microbial diversity, we performed a structural survey of the fecal microbiota of four purebred boar pig lines: Duroc, Landrace, Hampshire, and Yorkshire. Methods: The V3-V4 regions of the 16S rRNA genes were amplified and sequenced. Results: A total of 783 operational taxonomic units were shared by all breeds, whereas others were breed-specific. Firmicutes and Bacteroidetes dominated the majority of the fecal microbiota; Clostridia, Bacilli, and Bacteroidia were the major classes. Nine predominant genera were observed in all breeds and eight of them can produce short-chain fatty acids. Some bacteria can secrete cellulase to aid fiber digestion by the host. Butyric, isobutyric, valeric, and isovaleric acid levels were highest in Landrace pigs, whereas acetic and propionic acid were highest in the Hampshire breed. Heatmap was used to revealed breed-specific bacteria. Principal coordinate analysis of fecal bacteria revealed that the Landrace and Yorkshire breeds had high similarity and were clearly separated from the Duroc and Hampshire breeds. Conclusion: Overall, this study is the first time to compare the fecal microbiomes of four breeds of boar pig by high-throughput sequencing and to use Spearman's rank correlation to analyze competition and cooperation among the core bacteria.
Canine parvovirus(CPV) is a very highly contagious virus causing hemorrhagic enteritis and myocarditis mainly in young dogs. The diseases were first recognized in 1978, and then spread throughout the world by 1980. The main source of the infection seems to be the feces of infected dogs, at the same time feces are suitable materials for detection of virus in the enteric form exactly for the same reasons. Recently, a new technique of in vitro DNA amplification, Known as the polymerase chain reaction (PCR), has been widely applied to clinical viral diagnosis because of its sensitivity, specificity and rapidity. In this research, we attemped to set up the PCR for the detection of CPV in fecal samples and conformed the canine parvpviral enteritis by PCR. To increase the sensitivity and specificity of a PCR, the nested PCR (two-step PCR) was performed. We also surveyed the contamination status of CPV in the research using fecal specimen was highly sensitive and specific. Of the 100 fecal specimens suspected canine parvoviral enteritis, 45 fecal specimens were positive in HA test, 64 fecal specimens were positive in the first PCR, and 87 fecal specimens were positive in the second PCR. CPV contamination status of animal clinics and breeding centers was serious, wo hygienic management of environment in which dogs are reared is required. The nested PCR described here seems to be a rapid, sensitive and specific for the detection of canine parvovirus.
Fecal coliform bacteria are indicators of fecal contamination and of the potential presence of pathogens associated with animal excreta, wastewater, sludge. This study examined the distribution of fecal coliform in sand of the playground at educational facilities and parks in Seoul. Fecal coliform were detected from 61 (22.1%) places out of 276 playgrounds. Escherichia (E) coli (75.4%) was found most frequently, followed by Cronobacter (Cro) sakazakii (8.7%), Klebsiella (K) pneumoniae (7.2%) and Actinobacter baumannii (4.3%). We also tested the bacteria for their susceptibility to 16 antibiotics using disk diffusion method. All the isolates were highly susceptible to cefepime, gentamicin, tetracycline, ciprofloxacin, enrofloxacin, nalidixic acid and sulfamethoxazole/ trimethoprim, but resistant to apramycin and cephalothin. E. coli and Cro. sakazakii were highly resistant to apramycin (100% and 83.3%), followed by cephalothin (51.9% and 50%) and ampicillin (19.2% and 16.7%). K. pneumoniae was highly resistant to ampicillin and apramycin (100%, respectively) but the remaining antibiotics were susceptible.
Effects of Dasom Valley and Bora Valley on fecal microflora, fecal moisture, and fecal pH of twelve healthy human volunteers were investigated. Numbers of Bifidobacterium, Clostridium perfringens, Escherichia coli, and Lactobacillus of control group were $9.24{\pm}0.63$, $4.44{\pm}1.21$, $7.75{\pm}0.38$, and $6.98{\pm}0.81$ (Log CFU/g wet feces), respectively. During administration of Dasom Valley, numbers of Bifidobacterium and Lactobacillus were $10.70{\pm}0.44$ and $8.84{\pm}0.77$, whereas those of C. perfringens and E. coli were $2.96{\pm}1.50$ and $6.69{\pm}0.29$, respectively. Administration of Dasom Valley significantly increased growth responses of beneficial bacteria, Bifidobacterium and Lactobacillus, whereas those of harmful bacteria, C. perfringens and E. coli, significantly decreased. Moisture content of feces increased and fecal pH decreased with intake of Dasom Valley. Intake of Bora Valley slightly increased numbers of Bifidbacterium and Lactobacillus and slightly decreased those of C. perfringens and E. coli. Results indicate Dasom Valley has greater intestinal-modulating effect than Bora Valley and Atlantic. Daily intake of Dasom Valley may normalize disturbed physiological functions, resulting in improvement of growth and composition of microbial community within intestinal tract.
A study was investigated the effects of the probiotic mixture (PM) contained Lactobacillus plantarum (L. plantarum) SY-99 $2.4{\times}10^9CFU/kg$ and Bacillus subtilis (B. subtilis) SJ-61 $2.2{\times}10^9CFU/kg$ on growth performance and fecal microflora counts of broiler chickens. Referred to the previous studies, a total of 160 one-day-old male broiler chicken (Ross 308) were randomly assigned to four experimental groups, which were a basal diet alone (control) and supplemented with PM 0.1 g/kg feed (T1), PM 0.2 g/kg feed (T2), and PM 0.4 g/kg feed (T3). After 28 days of administration, the average daily feed intake of T1 and T3 was significantly different compared to that of control (p<0.05). In addition, the feed conversion ratio of T2 and T3 was significantly improved compared to that of control (p<0.05). On the 28th day after administration, all treatment groups were significantly increased fecal lactic acid bacteria (LAB) counts and decreased fecal Enterobacteriaceae (ENT) counts compared to T1 (p<0.05). The results of this study indicated that the combination of L. plantarum and B. subtilis strains in the range of 0.2-0.4 g/kg feed could be used for the improvement of growth performance and fecal microflora in broiler chickens.
This study aimed to determine the effect of feeding Hermetia illucens (H.illucens) pellets with canine diets on fecal properties and digestibility. The canine breeds used in this experiment were nine dogs without discriminating between genders: 3 Maltese (average weight 2.0 kg), 3 Chihuahua (average weight 1.0 kg), and 3 Poodles (average weight 2.5 kg). As canine diets, 70 g of regular diets were mixed with 30 g of H.illucens pellets, and 100 g was weighed on a scale. Dog were fed the experimental diets once a day. No significant changes were observed in the fecal properties in the three types of canines fed a mixture of pellets and canine diets at Days 0 and 7 (p>0.05). At Day 0, the fecal properties of all canines appeared in a less solid state, with scores ranging from 2.00 to 2.22; however, at Day 7, the fecal properties improved to normal (3.00 to 3.33). In addition, the results for nutrient digestibility showed that dry matter and crude fat digestibility were significant (p<0.05); however, crude protein and crude ash digestibility were not affected (p>0.05). In conclusion, these results suggest that feeding a mixture of H.illucens pellets and canine diets to companion canines has beneficial effects on fecal properties and digestibility without adverse effect.
