Effect of Water Extract from Hordeum vulgare L. with Medicinal Herb on Plasma Lipid Status and Glucose in Rats Fed High Fat Diet (맥아와 한약재추출물이 고지방 식이를 섭취시킨 흰쥐의 혈장지질 및 혈당농도에 미치는 영향)

  • Objectives : This study was designed to investigate the effect of water extract from Paeonia Lactiflora Pasll. with medicinal herb on plasma lipid and glucose in rats fed high fat diet. Methods : Experimental groups were divided into control diet group(C), high fat diet group(HF), high fat diet and 5% water extracts from Paeonia Lactiflora Pasll. with medicinal herb group(HFE I), high fat diet and 10% water extract from Paeonia Lactiflora Pasll. with medicinal herb group(HFE II). Weight gain, food intake, food efficiency, plasma lipid concentration and glucose were determined for 10 weeks during high fat diet. Results : In this experiment, body weight and food efficiency were significantly increase in high fat diet. High fat diet group showed elevated levels of total cholesterol, LDL-cholesterol and free fatty acid. Consumption of 10% water extract Paeonia Lactiflora Pasll. with medicinal herb lowers level of total cholesterol, LDL-cholesterol and free fatty acid. But levels of plasma triglyceride, HDL-cholesterol and glucose did not affect by high fat diet and water extract Paeonia Lactiflora Pasll. with medicinal herb Conclusions : In these results, water extract Paeonia Lactiflora Pasll. with medicinal herb in high fat diet group decreased plasma total cholesterol, LDL-cholesterol and free fatty acid. Thus the Paeonia Lactiflora Pasll. with medicinal herb might be useful in the treatment of hyperlipemia.

Anti-obesity Effects of SBY-III in High Fat Diet-Fed Obese Rats Continued by High Fat Diet and Regulated by Normal Diet (SBY-III이 비만 및 비만 후 식이조절 흰쥐에 미치는 영향)

  • Objectives : This experimental study was designed to investigate the effect of SBY-III extract on the weight, cell size of epididymal fat-pad, fat accumulation area in liver, serum lipid level and UCP1 mRNA in brown adipose tissue of high fat diet-fed obese rats continued by high fat diet and regulated by normal Diet. Methods : The body weight gain, weight of the internal organs(epididymis, liver, brown adipose tissue), insulin, triglyceride, total cholesterol, total lopod, free fatty acid, expression of UCP1 mRNA were measured in high fat diet-fed obese rats continued by high fat diet and regulated by normal diet. The experimental study are divided into exp-I and exp-II. Each study was administered normal diet, high fat diet and SBY-III according to each situation. Normal group is normal diet for 8 weeks. Exp-I are divided into control group(high fat diet for 8 weeks) and sample group(high fat diet for 8 weeks and SBY-III for last 2 weeks). Exp-II are divided into control group(high fat diet for 6 weeks and normal diet for 2 weeks) and sample group(high fat diet for 6 weeks and normal diet with SBY-III for 2 weeks). These were then compared mutually. Results : 1. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease of weight gain than control group and diet control-fed sample group with SBY-III showed the more effective decrease of weight loss including weight gain than control group. 2. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease cell size of epididymal fat-pad, fat accumulation area in liver than control group. 3. Non diet control-fed sample group taken SBY-III showed the more effective decrease of serum triglyceride, total lipid, free fatty acid than control group and diet control-fed sample group taken SBY-III showed the decrease of serum triglyceride, free fatty acid than control group. 4. Only diet control-fed sample group taken SBY-III showed the decrease of UCP1 volume. Conclusions : These results shows that SBY-III has effects on anti-obesity, especially keeping pace with diet control.

The Effects of Wolbiga-banha-tang on Obese Rats Fed a High-fat Diet (월비가반하탕이 고지방식이로 유도된 비만 쥐에 미치는 영향)

  • This study investigated the effects of Wolbiga-banha-tang (WBBT) on high fat diet-induced rats. We divided the rats into 4 groups: normal (fed a general-fat diet), control (fed a high-fat diet and no treatment), $WBBT{\times}1$ (fed a high-fat diet and 250 mg/kg of Wolbiga-banha-tang extracts), and $WBBT{\times}2$ (fed a high-fat diet and 500 mg/kg of Wolbiga-banha-tang extracts). We measured the body weight, total cholesterol, HDL cholesterol, LDL cholesterol, triglycerides, free fatty acids, total lipids, AST, ALT, and CBC in the serum of the obese rats for 7 weeks. There was a significant decrease in the control group, $WBBT{\times}1$, and $WBBT{\times}2$ in triglycerides and free fatty acids. The results suggest that WBBT is effective for lowering triglycerides and free fatty acids. Therefore, WBBT may be applied to the treatment of lipid metabolism in obesity and further clinical studies are needed.

A study of metabolic effect in high and low fat diet on Albino Rat (지방 함량에 따르는 흰쥐의 체내 대사 연구)

  • The present study was undertaken to determine the metabolic effect of various levels of fat in the diet. Forty males and the same number of females weighing $35{\pm}29$ were divided into three experimental groups and one control group, 10 rats each in both sexes. The dietary lipid contents were included in three levels, 2% as low, 30% as high and fat free diet in order to reflect the lipid consumption of present Korean diet. 20% sugar casein diet were employed as standard for control animals. This study was carried for 16 weeks. After these period animals were sacrificed to collect the internal organs and blood samples by heart puncture. In the result of this study, high fat diet group is lower than low fat diet group in the body weight gain, food efficiency ratio, protein efficiency ratio, urinary nitrogen and organ nitrogen contents were same trend but there is no significant difference in these respects. It was noteworty that high, and free fat diet group revealed more glucose total protein, albumin and globulin contents in the serum than other compared groups. It can be concluded that fat content whether low or high are free in the diet did not show significant effect on body nitrogen metabolism. But 30% high fat diet increased the total lipid and total cholesterol contents in the liver and the serum. This fact can be interpreted that 2% low fat diet and fat free diet do not need to increase the amount of fat content in the diet as high as 30% fat diet. This result might indicate the one possible reason to decide the recommanded dietary fat levels in Korean diet.

Effect of administration with fat-free diet and dietary iron on polyunsaturated fatty acid, vitamin E and their derivatives in male rats (Rat에 있어서 탈지사료 및 철분투여가 다불포화 지방산, vitamin E 및 그 유도체에 미치는 영향)

  • This study was designed to elucidate the effects of administration with dietary iron and fat-free diet on the contents of unsaturated fatty acid in phospholipid molecules, vitamin E contents and malondialdehyde contents in liver, kidney, muscle and testis of the male rats. The rats were divided into 3 experimental groups, namely, control, iron injection and fat-free diet administration groups. The control group was fed with normal diet, iron injection group injected intraperitoneally 20mg of ferric hydroxide/100g of body weight 20 times every 3 days and fat-free diet group administered lipid extraction diet with hexane in normal diet. All experimental groups were maintained for 60 days with feeding on the respective ration. The results obtained were summarized as follows: 1. In the mean contents of unsaturated fatty acid in phospholipid of liver, kidney, muscle and testis among groups, control group was 21.31mg/g, 19.38mg/g, 1.67mg/g, 13.68mg/g, iron injection group was 13.83mg/g, 16.53mg/g, 0.71mg/g, 10.11mg/g and fat-free diet group was 21.07mg/g, 19.38mg/g, 1.49mg/g and 13.40mg/g, respectively. 2. In the mean contents of vitamin E in liver, kidney, muscle and testis among groups, control group was 6.77mg/g, 1.93mg/g, 0.12mg/g, 0.17mg/g, iron injection group was 3.16mg/g, 0.86mg/g, 0.07mg/g, 0.09mg/g and fat-free diet group was 7.41mg/g, 1.50mg/g, 0.11mg/g and 0.16mg/g, respectively. 3. In the mean contents of malondialdehyde in liver, kidney, muscle, testis and serum among groups, control group was 11.29nM/0.1g, 23.25nM/0.1g, 42.47nM/0.1g, 7.01nM/0.1g, 4.33nM/ml, iron injection group was 34.98nM/0.1g, 40.55nM/0.1g, 72.21nM/0.1g, 12.26nM/0.1g, 11.27nM/ml and fat-free diet group was 8.07nM/0.1g, 20.63nM/0.1g, 39.92nM/0.1g, 6.95nM/0.1g and 4.27nM/ml, respectively.

Functional Studies of Acyl-CoA Synthetase 4 in the Rat Liver (흰쥐 간장에 있어서 아실-CoA 합성효소4의 기능연구)

  • Acyl-CoA synthetase 4 (ACS4) is an arachidonate-preferring enzyme abundant in steroidogenic tissues. We examined ACS4 in rat liver, which contains a variety of pathways that use acyl-CoAs, in order to determine subcellular locations. We demonstrate that ACS4 protein was present most abundantly in the mitochondria and to a much lesser extent in the peroxisomes and microsomes. To determined the dietary effects on the level of ACS4 mRNA, northern blotting was carried out using total RNA from the livers of adult male rats fed various diets. Fasting, high fat diet, and fat-free high sucrose diet increased the hepatic level of ACS4 mRNA approximately 2-fold. Furthermore, the levels of ACS4 mRNA were induced by DEHP[Di- (2-ethylhexyl) phthalate]. These data suggest that ACS4 expression in the liver is regulated with a variety of pathways, including $\beta$-oxidation, hormone, and insulin.

Changes of body composition in obese women with short-term low calory diets (비만 여성을 대상으로 한 단기간의 저열량 식사요법에서 체구성 성분의 변화)

  • Background: A reasonable weight reduction is to reduce fat-body mass while preserving free-fat mass. Recently, many people in Korea have been trying reckless diet therapy for weight reduction by low calory and protein diets, which gave rise to many side effects consequently. For reasonable weight reduction, this study was undertaken to investigate the influential diet factors that change body composition during short-term low calory diet. Methods: 26 healthy obese women aged 23-27 years whose BMI was greater than 24volunteered for the 6 week diet therapy from January to April in 2003. All subjects were instructed to eat approximately 1200kca1/day and write dietary diary. In the beginning of the study, the 3rd week and the 6th week, subjects' body weight and fat body weight were measured by BlA. In the 6th week, the analysis of dietary diary was conducted. Results : There was significant positive correlation between protein intake per ideal bodyweight in the first 3 weeks and free fat mass increase in the later 3 weeks (P<0.05). At the same time, fat intake in the first 3 weeks had positive correlation with free fat mass in the second 3 weeks, too(P<0.05). But, we could not find any significant values that had effect on free fat mass increase in multiple regression analysis. Conclusion: The protein intake in the first 3 weeks helps preserve fat free mass in the later 3 weeks, and has positive effects.

Influence of fat levels and types of dietary fiber on lipid metabolism of rats (식이중 지방 수준과 Fiber 종류가 흰쥐의 지방대사에 미치는 영향)

  • The influence of fat level and dietary fiber on serum and liver lipid concentration of the rat was investigated. Groups of rats were fed respectively 3%, 5%, 20% fat containing and free fiber diet until 5th week. From 5th week to 9th week high fat diet groups of rates were devided high fat diet group, high fat pectin supplemented diet group and high fat celulose supplemented diet group. Pectin and cellulose at 10% was supplemented to high fat diets. After rats were fed for 9 weeks, all rats were sacrificed to collect the liver and blood samples by heart puncture. Serum and liver lipids were determined on all rats and compared the histochemical lipid staining method with the biochemical analysis of serum and liver lipids. A high fat diet caused increased level of the total lipids, total cholesterol and free cholesterol contents in the liver and the serum. Rats fed pectin reduced body weight, werum lipid and liver lipid but cellulose had no effect to reduce serum and liver lipid concentration. These results indicate that dietary fat level led to changes in the lipid metabolism of rats and that pectin was more effective in lowering serum and liver lipid than cellulose.

A Study of the Free Amino Acids in the Plasma and Erythrocytes in the Male Adult Rats Fed with the Low Protein Diets (저단백영양(低蛋白營養)에 있어서 성숙(成熟)흰쥐의 Plasma 및 Erythrocytes Free Amino Acid에 대하여)

  • An analysis of the free amino acid contained in the plasma and erythrocytes of the six groups of Wistar Strain male adult rats(body weight 200-300g) having fasted for sixteen hours was made by means of the HITACHI Amino Acid Autoanalyzer and the result of which was corrected with RC-24 B TOMY Micro Hematocrit Centrifuge. There was a depression of the plasma and erythrocytes free amino acid level on the noprotein diet with ad libitum feeding. But on the 20% casein diet there was an elevation in the levels of free amino acid and consequently alanine, glysine, lysine, serine and arginine level in the erythrocytes and threonine glutamic acid and taurine level in the plasma increased on the high protein diet. There was more plasma and erythrocytes free amino acid level on the 5% casein-30% fat diet than on the 5% casein-no fat diet with pair-feeding. In comparison, on the low calorie diet more free amino acids were found in plasma than in erythrocytes, but on the higher calorie diet more free amino acids were found in the erythrocytes than in the plasma. On the 20% casein-30% fat diet with pair-feeding the erythrocytes free amino acids level increased but in plasma free amino acids level decreased. Such as an opposite result was given in plasma and erythrocytes free amino acids level. In the pair-fed four groups, erythrocytes per plasma generally increased in the rate of less than 10.0 as the calorie increased. The essential amino acid per non essential amino acid generally increased in the ratio as protein level and calorie increased, and that ratio range was from 0.2 to 0.7. And essential amino acid per non essential amino acid of plasma was higher than that of erythrocytes.
