A Study on Gamma ray effects on Stress Response and Cellular Toxicity using Bacterial Cells

  • 본 연구는 5가지의 발광성 미생물을 이용하여 유해 방사선으로 알려져 있는 ${\gamma}-rays$가 여러가지 cellular stresses 중, 특히 유전자 손상과 생물막 손상을 유발하였는데, 이들의 손상 정도가 총 방사선량과 상관관계가 있음을 발생하는 bioluminescence 로써 확인하였다. 뿐만 아니라, 선량률의 변화를 통하여 방사선으로 인한 유전자 손상 및 일반적인 독성 효과가 큰 영향을 받는 것을 확인하였는데, 선량률 증가에 따라 이들 손상정도가 증가하는 것으로 보아 선량률이 genetic 및 radioprotecion에 심각한 영향을 미치는 것을 확인하였다.

Production of Recombinant Humanized Anti-HBsAg Fab Fragment from Pichia pastoris by Fermentation

  • In this report, we describe the high-yield secretory expression of the recombinant human anti-HBsAg Fab fragment from Pichia pastoris that was achieved by co-integration of the genes encoding the heavy and light chains (both under the control of alcohol oxidase promoter) into the genome of the yeast cells. The fed-batch fermentations were carried out in a 5 L scale. Both chains of the Fab were successfully expressed upon methanol induction. The absorbance ($OD_{600}$) of the broth can reach 350~500 at the end of fed-batch phase. After the induction, the expression level of the recombinant Fab (soluble) reached 420~458 mg/L. The recombinant Fab fragment was purified from the crude culture supernatant by ion exchange chromatography and the purity of the recombinant Fab fragment was over 95%. The affinity activities of the crude fermentation supernatant and the purified Fab were analyzed by indirect ELISA, which showed that the purified recombinant Fab fragment had high affinity activity with hepatitis B surface antigen.

A Whole Cell Bioluminescent Biosensor for the Detection of Membrane-Damaging Toxicity

  • The recombinant bacteria strain DPD2540, containing a fabA::luxCDABE fusion, was used to detect the toxicity of various chemicals in this study. Membrane damaging agents such as phenol, ethanol, and cerulenin induced a rapid bioluminescent response from this strain. Other toxic agents, such as DNA-damaging or oxidative-damaging chemicals, showed a delayed bioluminescent response in which the maximum peak appeared over 150 min after induction. This strain was also tested for measurement of toxicity in field samples such as wastewater and river water effluents.

Streptomyces sp. 50634 균주가 생산하는 tipA Promotor 활성화 물질, Sulfomycin Ia

  • In the course of screening for the tipA promoter-inducing substances, we isolated an active compound, sulfomycin Ia, from the mycelium of a microorganism designated 50634. The producing organism was identified as Streptomyces sp. on the basis of taxonomic studies. Sulfomycin Ia was purified from mycelial extract by silica gel column chromatography, LH-20 column chromatography, silica gel TLC, and preparative HPLC. The molecular weight of sulfomycin Ia was determined to be m/z 1129 (M+Na)$^{+}$ by FAB mass measurement and $^{1}$H NMR spectroscopic analysis. The structure was assigned as a derivative of sulfomycin I with thiazole, methyloxazole, oxazole, and pyridine rings by $^{1}$H NMR spectral data.

재조합 발광 박테리아를 이용한 환경독성탐지 및 휴대용 바이오 센서의 개발

  • Bioluminescent bacteria fusing the stress promoter and lux gene have been developed as a toxicity biosensor. The light emitting bioluminescent bacteria have been used to measure the toxicity of many different chemicals. In this study, specially, DPD2540 (fabA::luxCDABE) was used to detect and classify phenolic toxicity to the cells membrane fatty acids, and then the relationship between phenolic toxicity and the distribution of various phenols in the cell was determined, with a model and equations provided. In addition, to show the possibility of detecting and classifying the toxicity of a chemical mixture, which may be present in wastewater, various bioluminescent bacteria having different stress promoters were used and their distinct response to the sample mixture was measured. To extend the applicable area of these bioluminescent bacteria to field, the portable biosensor using freeze-drying methods was developed and confirmed successfully.
