Ultrasound-assisted Extraction for Development of Skin Whitening and Anti-wrinkling Cosmetic Materials from Spirulina platensis (스피루리나(Spirulina platensis)로부터 미백과 주름개선 생리활성 물질 분리를 위한 초음파 추출공정 개발)

  • Ultrasound-assisted extraction (UAE) conditions, including extraction time, extraction temperature, and ethanol concentration, were optimized to increase the total flavonoid content (TFC), tyrosinase inhibitory activity (TIA), and collagenase inhibitory activity (CIA) of Spirulina platensis through central composite design (CCD). For the optimization of the three dependent variables, a quadratic regression model was derived from 17 experimental sets for the simultaneous maximization of TFC, TIA, and CIA. The predicted optimal UAE conditions were 44.0 min of extraction time, 82.8℃ of extraction temperature, and 96.0% of ethanol concentration. Under these conditions, 0.93 mg QE/g DM of TFC, 81.9% of CIA, and 92.1% of TIA were predicted. The CCD-based UAE optimization enabled an increase in TFC, CIA, and TIA, thereby confirming that the S. platensis extract can be used in the development of a cosmetic material with skin whitening and anti-wrinkle effects.

Determination of the Conditions for Anthocyanin Extraction from Purple-Flashed Sweet Potato (자색고구마 Anthocyanin 색소의 추출조건 결정)

  • To establish the optimum conditions for the extraction of anthocyanin pigment from purple-fleshed sweet potato, a suitable extraction solvent with the optimum citric acid concentration for acidification of the solvent, and the optimum extraction time and temperature were determined. Twenty percent ethanol solution acidified with citric acid was found to be a good solvent for the extraction of the pigment from purple-fleshed sweet potato. About 10 hour extraction at room temperature was appropriate for the extraction. pH of the extract was below 3 when more than 0.7% citric acid was added. The higher the concentration of citric acid added was, the higher the total optical density (TOD) of the extract was. However, the increase in TOD of the extract was insignificant when more than 1% of citric acid was added. Therefore, addition of 1% citric acid was determined for acidification of the extracting solvent. Though the initial rate of the pigment extraction increased as the extracting temperature increased, extraction at higher temperatures of 60 or 8$0^{\circ}C$ for an extended time caused a decrease in the extraction yield due to degradation of the pigment. The optimum extraction temperature for the anthocyanin pigment from purple-fleshed sweet potato with the solvent used was determined as 4$0^{\circ}C$.

Extraction Conditions for Phenolic Compounds with Antioxidant Activities from White Rose Petals

  • The extract of white rose petals has an antioxidant effect and can be used to treat allergic disease. The purpose of this study was to identify optimal conditions for extracting antioxidative compounds from white rose petals with 2,2-diphenyl-1-picrylhydrazyl scavenging activities. A response surface methodology based on a central composite design was used to investigate the effects of three independent variables: ethanol concentration ($X_1$), extraction temperature ($X_2$), and extraction time ($X_3$). The estimated optimal conditions for obtaining phenolic compounds with antioxidant activities were as follows: ethanol concentration of 42% ($X_1$), extraction time of 80 min ($X_3$), and extraction temperature of $75^{\circ}C$ ($X_2$). The estimated optimal conditions for obtaining flavonoid compounds with antioxidant effects were an ethanol concentration of 41% ($X_1$), extraction time of 119 min ($X_3$), and an extraction temperature of $75^{\circ}C$ ($X_2$). Under these conditions, predicted response values for the phenolic and flavonoid contents were 243.5 mg gallic acid equivalent/g dry mass and 19.93 mg catechin equivalent (CE)/g dry mass, respectively.

Monitoring for optimum antioxidant extraction condition of Gugija (Lycium chinensis Mill) extract (구기자 추출물의 최적 항산화 추출조건 모니터링)

  • This study optimized the extraction of antioxidants from Gugija (Lycium chinensis Mill). To determine operational parameters, including ethanol concentration ($X_1$, 0~80%) and extraction time ($X_2$, 1~5 hr), response surface methodology was applied to monitor yield, anthocyanins, flavonoids and DPPH radical scavenging activity. Coefficients of determinations ($R^2$) of the models were range of 0.8645~0.9859 (p<0.01~0.1) in dependant parameters. Yield of Gugija extracts was maximized 23.12% in extraction conditions of 4.22 h at 8.25% ethanol. Anthocyanins was maximized 1.43 (OD in 530 nm) in extraction conditions of 3.06 h at 79.98% ethanol. Flavonoids was maximized $3,100{\mu}g/100g$ in extraction conditions of 3.37 h at 67.02% ethanol. DPPH radical scavenging activity was maximized 96.93% in extraction conditions of 1.67 h at 69.81% ethanol. Optimum extraction conditions (2.5 h extraction at 70% ethanol) were obtained by superimposing the contour maps with regard to anthocyanins, flavonoids and DPPH radical scavenging activity of Gugija. Maximum values of anthocyanins, flavonoids and DPPH radical scavenging activity in optimum extraction condition were 1.0080 (OD in 530 nm), $3,145{\mu}g/100g$, 96.96%, respectively. But values of anthocyanins, flavonoids and DPPH radical scavenging activity in water extraction condition (1 h at water) were 0.4652 (OD in 530 nm), $1,633{\mu}g/100g$, 86.98%, respectively.

Monitoring of Extraction Yields and Functional Properties of Ginger (Zingiber officinale) Extracts Using Response Surface Methodology (반응표면분석을 이용한 생강의 추출효율 및 기능성에 대한 모니터링)

  • Extraction characteristics of ginger (Zingiber officinale) and the functional properties of the corresponding extracts were monitored by the response surface methodology (RSM). The maximum extraction yield of 38.83% was obtained under the specific extraction conditions, such as microwave power of 126.68 W, ethanol concentration of 53.39% and extraction time of 8.15 min. The maximum electron donating ability (EDA) was found as 77.92% at the conditions of 58.25 W microwave power, 91.87% ethanol concentration and 5.54 min extraction time. The maximum inhibitory effect on tyrosinase was 79.39% at 106.08 W, 2.06% and 5.61 min. The maximum polyphenol content was 105.59% under the extraction conditions of 81.82 W, 99.52% and 4.06 min. Based on superimposition of four dimensional RSM with respect to extraction yield and electron donating ability and polyphenol content obtained under the various extraction conditions, the optimum ranges of extraction conditions were found to be microwave power of $0{\sim}80\;W$, ethanol concentration of $0{\sim}40%$ and extraction time of $3{\sim}7$ min.

Optimization of Extraction Conditions of Sarcodon aspratus by Response Surface Methodology (반응표면분석에 의한 능이버섯 추출물의 추출조건 최적화)

  • Response surface methodology (RSM) was used to monitor extraction characteristics of extracts from Sarcodon aspratus. Based on a central composite design, independent variables were microwave power (30~150 W), ethanol concentration (0~100%), and extraction time (1~9 min). Dependent variables were yield, electron-donating ability, total phenol contents, and SOD-like activity. Coefficients of determination ($R^2$) for dependent variables ranged from 0.80 at 0.97. The maximum extraction yield was 50.28% under conditions of 125.1 W microwave power, 18.67% ethanolic concentration, and 7.06 min extraction time. The maximum extraction electron-donating ability was 22.14% under conditions of 31.09 W, 45.76%, and 4.32 min. The maximum extraction total polyphenol content was 30.54 mg tannic acid equivalent/g at 122.54 W, 48.05%, and 8.36 min. The maximum extraction SOD-like activity was 33.44% at 121.17 W, 47.42%, and 8.41 min. Based on superimposition of four dimensional RSM with respect to extraction yield, electron-donating ability, total polyphenol content, and SOD-like activity obtained under various extraction conditions, optimum ranges of extraction conditions were found to be microwave power of 78~88 W, ethanol concentration of 39~57%, and extraction time of 3.5~9 min.

Optimization of Crude Papain Extraction from Papaya Latex Using Response Surface Methodology (반응표면 분석법에 의한 crude papain 추출 조건의 최적화)

  • Crude papain was extracted from papaya latex under various conditions. Extraction conditions of crude papain were optimized by the response surface methodology (RSM). Extraction yield of papain was affected by concentration of $NaHSO_3$, extraction time and pH. Three independent factors were chosen to determine their effects on the various responses and the function was expressed in terms of a quadratic polynomial equation, which measures the linear, quadratic and interaction effects. According to the results of RSM, the stationary point for quantitatively dependent variable was found to be the maximum point for extraction yield. Optimum conditions of papain extraction were 4% of $NaHSO_3$, 120 min of extraction time and pH 7.6. Under these conditions, 793.16 mg of papain per g latex was extracted from papaya latex.

Prediction of Optimal Microwave-Assisted Extraction Conditions for Functional Properties from Fluid Cheonggukjang Extracts (액상청국장 추출물의 기능성에 대한 마이크로웨이브 최적 추출조건 예측)

  • Response surface methodology (RSM) was employed to optimize extraction conditions in order to find the maximal functional properties of fluid Cheonggukjang. Based on central composite design, a study plan was established with variations of microwave power, ethanol concentration, and extraction time. Regression analysis was applied to obtain a mathematical model. The maximum inhibitory of tyrosinase activity was found as 26.75% at the conditions of 30.56W microwave power, 2.40 g/mL of ratio of solvent to sample content and 10.00 min extraction time, respectively. The maximum superoxide dismutase (SOD)-like activity was 53.23% under the extraction conditions of 108.42 W, 4.38 g/mL and 7.84 min. Based on superimposition of three dimensional RSM with respect to extraction yield, inhibitory of tyrosinase activity and SOD-like activity obtained under the various extraction conditions, the optimum ranges of extraction conditions were found to be microwave power of $55{\sim}75$ W, ratio of solvent to sample content of $2{\sim}5$ g/mL and extraction time of $3.5{\sim}15$ min, respectively.

Optimization of Ethanol Extraction Conditions for Artemisis capillaris Effective Components Using Response Surface Methodology (반응표면분석법을 이용한 인진쑥 유효성분의 에탄올 추출조건 최적화)

  • This study was conducted to monitor the quality characteristics of Artemisis capillaris ethanolic extract by response surface methodology. The independent variables were extraction temperature ($X_1$; 60, 70, 80, 90, and $100^{\circ}C$), extraction time ($X_2$; 1, 2, 3, 4, and 5 hr), and ethanol concentration ($X_3$; 0, 20, 40, 60, and 80%). Soluble solid content ($Y_1$), chlorogenic acid content ($Y_2$), and coumaric acid content ($Y_3$), etc. were analyzed as the dependent variables. Estimated optimal conditions for soluble solids were an extraction temperature of $87.65^{\circ}C$, extraction time of 3.19 hr, and ethanol concentration of 42.40%. The optimal extraction conditions for chlorogenic acid were $84.30^{\circ}C$, 3.14 hr, and 47.85%, respectively. Further, those for coumaric acid were $83.45^{\circ}C$, 3.40 hr, and 45.39%, respectively. Extraction conditions for effective components of Artemisis capillaris were superimposed by response surface plots on optimization extraction condition of each dependent variable, including soluble solid, chlorogenic acid, and coumaric acid contents. As a result, superimposed extraction conditions were $80{\sim}90^{\circ}C$, 3~4 hr, and 40~50%, respectively. Under these conditions, soluble solid, chlorogenic acid, and coumaric acid contents were 1.09%, 25.66 mg%, and 20.25 mg%, respectively.

Improved cosmetic activity by optimizing the Citrus madurensis (Citrofortunella microcarpa) INEFU extraction process. (둥근금감 (깔라몬딘 오렌지)의 이네푸 추출공정의 최적화를 통한 화장품 기능성 활성 증가 연구)

  • This study was expand the use of Citrus madurensis (Citrofortunella microcarpa), which is a good source of skin whitening and antioxidants activation cosmetics. The goal was to provide cosmeceutical activity data about the extraction yield and total phenol of this calamondin orange by optimizing the focused high ultrasound(INEFU) and ultrasound extraction(UE) conditions. Under optimal extraction conditions, which consisted of 1800W for 45min and 500W for 45 min, 21.55 and 13.27 % (w/w) of the highest extractions yield and polyphenol contents were obtained. For the skin whitening activity, tyrosinase inhibitory activity was observed at 69.24 % in the INEFU extracts, which was ca. 18 % higher than that of the UE extracts (58.82 %). To reduce melanin production in Clone M-3 cells, 86.9 % melanin production was observed following the addition of control, WE(water extraction) relative to the control without a sample, and 81.2 and 78.9 % were found in the UE and INEFU conditions. The highest total phenol secretion was conclusively obtained under the optimal conditions and resulted in a significant improvement of the cosmetic activities of Citrus madurensis(Citrofortunella microcarpa) extracts.
