Enhancement of storage stability of red beet pigment using broccoli extracts (브로콜리 추출물을 이용한 레드 비트 색소의 저장 안정성 향상 연구)

  • The effect of broccoli extracts on the storage stability of red beet extracts under various conditions (temperature, light condition, and pH) was analyzed. Regardless of the pH and light conditions, the absorbance of the red beet pigment at 537 nm was relatively stable (less than 10% reduction) at 4℃ for up to 5 days. The absorbance of the control extracts was rapidly reduced during storage at an ambient temperature in the dark, but the degree of reduction was relatively low at pH 3.8. As a positive control, the addition of vitamin C to the control extracts slightly inhibited the reduction (40% reduction), but the degree of the inhibition was much higher (15% reduction) with the addition of broccoli extracts (in 70% ethanol). Among all the samples, the addition of the broccoli floret extracts using 70% ethanol was the most effective method for the enhancement of the storage stability of red beet pigment at an ambient temperature in the dark.

  • Recently, the advance of accurate dynamic displacement measurement devices, such as GPS, computer vision, and optic laser sensor, has enhanced the structural monitoring technology. In this study, the dynamic displacement data was used to verify the applicability of the structural physical parameter estimation method through subspace system identification. The subspace system identification theory for estimating state-space model from measured data and physics-based interpretation for deriving the physical parameter of the estimated system are presented. Three-degree-freedom steel structures were fabricated for the experimental verification of the theory in this study. Laser displacement sensor and accelerometer were used to measure the displacement data of each floor and the acceleration data of the shaking table. Discrete state-space model generated from measured data was verified for precision. The discrete state-space model generated from the measured data extracted the floor stiffness of the building after accuracy verification. In addition, based on the story stiffness extracted from the state space model, five column stiffening and damage samples were set up to extract the change rate of story stiffness for each sample. As a result, in case of reinforcement and damage under the same condition, the stiffness change showed a high matching rate.

  • Sufficient understanding of oversea construction market status is crucial to get profitability in the international construction project. Plenty of researchers have been considering the news article as a fine data source for figuring out the market condition, since the data includes market information such as political, economic, and social issue. Since the text data exists in unstructured format with huge size, various text-mining techniques were studied to reduce the unnecessary manpower, time, and cost to summarize the data. However, there are some limitations to extract the needed information from the news article because of the existence of various topics in the data. This research is aimed to overcome the problems and contribute to summarization of market status by performing topic modeling with Latent Dirichlet Allocation. With assuming that 10 topics existed in the corpus, the topics included projects for user convenience (topic-2), private supports to solve poverty problems in Africa (topic-4), and so on. By grouping the topics in the news articles, the results could improve extracting useful information and summarizing the market status.

The Sensory Characteristics of Brown Sauce by Adding Different Ratio of Chicken Bone (닭뼈 혼합비에 따른 Brown Sauce의 관능적 성질)

  • The purpose of this study was to investigate sensory characteristics of brown sauce made with different ratio of chicken bone. Brown sauces made from chicken bone and beef bone respectively were evaluated by pH, viscosity, turbidity, Hunter's color and sensory evaluation. With chicken bone increased, pH of brown sauce changed from 5.11 to 5.72 (p<0.05) that was lower than optimal bacterial growth condition between 6.8 and 7.0. Viscosity increased with higher chicken bone ratio (p<0.05) and desired viscosity can be obtained by controlling chicken bone content. Turbidity increased with chicken bone (p<0.05), which might be caused by the thick chicken bone extract. Lower Hunter "L" value and higher "a" and "b" value were shown with increase of chicken bone (p<0.05). The sensory evaluation was performed with scoring tests for color, flavor, viscosity and taste by 20 professional panels who were professional cooks working in I hotel. Better scores of color, flavor, taste and overall evaluation except viscosity were observed with increase of cattle bone (p<0.05). There was not a significant difference between chicken bone and beef bone 0.0 : 10.0 and 2.5 : 7.5 (p>0.05). Brown sauce made with 2.5 : 7.5 can be prepared without reducing sensory quality.

Studies on the Formation of L-Threonine by auxotrophic mutants of Brevibacterium flavum (Brevibacterium Flavum의 Auxotrophic Mutants에 의한 L-Threonine 생성(生成)에 관한 연구(硏究))

  • This study was attempted to increase the production of L-Threonine by Brevibacterium Flavum ATCC 14067, To select the strain which produce the highest threonine, mutants ere induced by N-methyl-N'-nitro-N-nitrosoguanidine treatment. The composition of media and cultural condition for its overproduction of threonine were also studied. In a threonine producer, strain B-13(Met-) was the strain producing the highest amount of threonige among methionine, lysine and isoleucine auxotrophs. The following results were obtained. 1. The wild strain and B-13(Met-) produced threonine 1.4mg/ml and 4.86mg/ml , respectively. 2. The optimum composition of medium for producing threonine by Brevibacterium Flavum B-13 was glucose 10%, ammonium sulfate 4%, potassium phosphate monobasic 0.2%, magnesium sulfate 0.05%, biotin $200{\mu}l$, thiamine $300{\mu}l$. Addition of nicotinic acid also led to increase L-threonine production. 3. In addition of organic nutrients to the fermentation medium, peptone n'ere effective and addition of methionine $100{\mu}g/ml$ produced the highest amount of L-Threonine. Aspartic acid and homoserine were also effective when these amino acid were added to the fermentstion medium. 4. Cultural conditon on threonine production by B-16 were investigated. The optimum pH was 7.0-8.0. The highest amount of threnine was produced after 4 days of cultural period.

Effect of Temperature, Solvent Concentration, and pH on the β-Glucan Extraction (β-Glucan 추출에 미치는 온도, 용매 농도 및 pH의 영향)

  • This study investigated the effects of temperature, solvent concentration, and pH on the ${\beta}$-glucan extraction. Oat bran ${\beta}$-glucan was extracted with different extraction conditions, using various combinations of experiment factors, such as temperature (40, 45, 50, 55, and $60^{\circ}C$), ethanol concentration (0, 5, 10, 15, and 20%), and pH (5, 6, 7, 8, and 9). Under the various extraction conditions, ${\beta}$-glucan extraction rate and overall mass transfer coefficient of oat bran ${\beta}$-glucan, and viscosity of oat bran extracts were investigated. As increasing the extraction time, the extraction rate of ${\beta}$-glucan increased. The overall mass transfer coefficient of ${\beta}$-glucan ranged from $3.36{\times}10^{-6}$ to $8.55{\times}10^{-6}cm/min$, indicating the lowest at the extraction condition of $45^{\circ}C$, 15% and pH 8, and the highest at $50^{\circ}C$, 0% and pH 7. It was significantly greater with increasing extraction temperature and decreasing ethanol concentrations of extraction solvent, except for solvent pH. There were positive correlations among the overall mass transfer coefficient, the extraction rate of ${\beta}$-glucan, and the viscosity of extract.

Mechanism of Apoptosis & Tumor Growth Inhibition of Agrimonia pilosa Ledebour(APL) in vitro and in vivo (선학초(짚신나물)에 의한 in vitro와 in vivo에서의 암세포사멸 기전 탐색)

  • Objectives : The aim of this study was to experiment the antitumor activity of Agrimonia pilosa Ledebour (APL) in human stomach cancer (AGS) cell lines (in vitro) and male C57BL/6J mouse (in vivo). Methods : The effects of the ethanol extract from the plant on several transplantable rodent tumors were investigated in vitro by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxy phenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt (MTS) assay. DNA content analysis and Western blot analysis. Agrimonia pilosa Ledebour (APL) was given to rats with Lewis Lung Carcinoma (LLC) cells. The experimental rats were divided into 3 groups in vivo. Saline was injected into the abdominal cavity in the first group, 50 mg/kg APL was injected into the abdominal cavity in the second group and 100 mg/kg was injected into the abdominal cavity in the third group. After that, we checked their tumor volume periodically. Results : At first, human gastric cancer (AGS) cell lines (in vitro) showed decreased cell viability, and increased $sub-G_1$ contents. When we experimented rat intestinal epithelial (RIE)l as same condition, this result didn't show. With this, compared to normal cells, Agrimonia pilosa Ledebour (APL) led selectively to the extinction of cells only in human gastric cancer. Moreover, we showed that the traditional herbal medicine APL induced caspase-dependent apoptosis in AGS cells. Next, APL inhibited the growth of LLC-bearing mouse tumor. However, we could not verify APL induced caspase-dependent apoptosis in LLC-bearing mouse tumor. Conclusions : The roots of Agrimonia pilosa Ledebour (APL) contain some antitumor constituents.

Effects of Siegesbckiae Herba on the Brain Ischemia (희렴 이 뇌허혈에 미치는 효과)

  • This experimental study was designed to investigate the effects of SIEGESBECKIAE HERBA extract (SHE) on the change of cerebral hemodynamics 〔regional cerebral blood flow (rCBF) and mean arterial blood pressure(MABP)〕 in normal condition and cerebral ischemic rats, and to determine the mechanism of action of SHE. This study was designed to investigate whether or not SHE inhibit lactate dehydrogenase (LDH) activity in neuronal cells and cytokines production in serum of cerebral ischemic rats. The results were as follows SHE increased rCBF significantly in a dose-dependent manner, but MABP was not changed by SHE in normal rats. The SHE-induced increase in rCBF was significantly inhibited by pretreatment with indomethacin (IDN), an inhibitor of cyclooxygenase but was increased by methylene blue (MTB), an inhibitor of guanylate cyclase. SHE inhibited lactate dehydrogenase (LDH) activity significantly in neuronal cells. rCBF was increased significantly and stably by SHE(10㎎/㎏, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group in ischemic rats. In serum by drawing from femoral arterial blood after middle cerebral arterial occlusion(MCAO) for 1hr and reperfusion for 1hr, the sample group was decreased IL-1β production significantly compared to that of the control group. In serum by drawing from femoral arterial blood after MCAO 1hr and reperfusion 1hr, sample group decreased TNF-α production significantly compared to that of the control grolilp. In serum by drawing from femoral arterial blood after reperfusion 1hr, sample group increased TGF-β production significantly compared to that of the control group. In serum by drawing from femoral arterial blood after MCAO for 1hr and reperfusion for 1hr, IL-10 production of the sample group was similar to that of control group. These results suggested that SHE had inhibitive effect on the brain damage by inhibited LDH activity, IL-1β and TNF-α production, but accelerated TGF-β production.

Influence of Environmental Factors on Conidial Germination of Alternaria dauci (당근 검은잎마름병원균 Alternaria dauci의 포자 발아에 미치는 환경인자의 영향)

  • Effects of temperatures, relative humidity, pH, and dry periods on conidial germination of Alternaria dauci were evaluated under in vitro conditions. Conidial germination was accelerated at over 95% relative humidity in $15^{\circ}C$ to $25^{\circ}C$ condition. Conidial germination was rapidly reduced at 5 regardless of relative humidity conditions. More than 50% of the conidial germination were initiated within 2 h at $25^{\circ}C$ through pH 5 to 7. The highest conidial germination of A. dauci was on 0.2% of carrot leaf extract. Conidia could survive longer than 12 h, even though its germination decreased. After a 12 h dry period, around 10% of conidia revived and germinated when conidia were hydrated again. These results could be used as the useful information on conidial germination of A. dauci and ecology of Alternaria leaf blight.

Lignin Removal from Barley Straw by Ethanosolv Pretreatment (Ethanosolv 전처리에 의한 보릿짚의 리그닌 제거)

  • Lignocellulose represents a key sustainable source of biomass for transformation into biofuels and bio-based products. Unfortunately, lignocellulosic biomass is highly recalcitrant to biotransformation, both microbial and enzymatic, which limits its use and prevents. As a result, effective pretreatment strategies are necessary. The vast majority of pretreatment strategies have focused on achieving a reduction of lignin content. In this work, an ethanosolv pretreatment has been evaluated for extracting lignin from barley straw. 75% ethanol was used as a pretreatment solvent to extract lignin from barley straw. The influence on delignification of three independent variables are temperature, time, catalyst (1 M $H_2SO_4$) dose. The best pretreatment condition observed was $180^{\circ}C$, 120 min, 0.2% $H_2SO_4$ and delignification was 38%. A combined roasting and ethanosolv, 2-step pretreatment, was developed in order to improve the delignification. Roasting didn't increase the delignification but reduced the pretreatment time. X-ray diffraction results indicated that these physical changes enhance the enzymatic digestibility in the ethanosolv treated barley straw. The cellulose in the pretreated barley straw becomes more crystalline without undergoing ethanosolv.
