The Effect of External Divalent Cations on Intestinal Pacemaking Activity

  • Electrical rhythmicity in the gastrointestinal (GI) muscles is generated by pacemaker cells, known as interstitial cells of Cajal (ICC). In the present study, we investigated the effect of external divalent cations on pacemaking activity in cultured ICC from murine small intestine by using whole-cell patch clamp techniques. ICC generated pacemaker currents under a voltage clamp or electrical pacemaker potentials under a current clamp, and showed a mean amplitude of $-500{\pm}50$ pA or $30{\pm}1$ mV and the frequency of $18{\pm}2$ cycles/min. Treatments of the cells with external 0 mM $Ca^{2+}$ stopped pacemaking activity of ICC. In the presence of 2 mM $Ca^{2+}$, 0 mM external $Mg^{2+}$ depolarized the resting membrane potential, and there was no change in the frequency of pacemaking activity. However, 10 mM external $Mg^{2+}$ decreased the frequency of pacemaking activity ($6.75{\pm}1$ cycles/min, n=5). We replaced external 2 mM $Ca^{2+}$ with equimolar $Ba^{2+}$, $Mn^{2+}$ and $Sr^{2+}$, and they all developed inward current in the sequence of $Ba^{2+}$>$Mn^{2+}$>$Sr^{2+}$. Also the frequency of the pacemaking activity was stopped or irregulated. We investigated the effect of 10 mM $Ba^{2+}$, $Mn^{2+}$ and $Sr^{2+}$ on pacemaking activity of ICC in the presence of external 0 mM $Mg^{2+}$, and found that 10 mM $Ba^{2+}$ and $Mn^{2+}$ induced large inward current and stopped the pacemaking activity of ICC (n=5). Interestingly, 10 mM $Sr^{2+}$ induced small inward current and potentiated the amplitude of pacemaking activity of ICC (n=5). These results indicate that extracellular $Ca^{2+}$ and $Mg^{2+}$ are requisite for the pacemaking activity of ICC.

A Study on Switching Characteristics of Active Clamp Type Flyback Converter with Synchronous Rectifier Driving Signals Controlling Auxiliary Switch (보조스위치가 동기정류기 구동 신호로 제어되는 능동 클램프형 플라이백 컨버터의 스위칭 특성에 관한 연구)

  • In this paper, the switching characteristics of the active clamp type flyback converter, which is deemed suitable for the miniaturization of the external power supply for home appliance, were analyzed and the process of reducing the switching loss was explained. The active clamp type flyback converter operating in the DCM has confirmed that the surge voltage of the main switch does not occur and the turn-off / on loss of the switch do not occur in principle. Also, in the case of the switch for synchronous rectifier, it was showed that the switch current showed half-wave rectified sinusoidal characteristic, and the switching loss was reduced. The switching characteristics of the experimental results gathered from 120 W class prototype were compared with the theoretical waveform in the steady-state and it was confirmed that the power conversion efficiency of the active clamp type flyback converter was maintained high due to the reduction of the switching loss.

A Study on the Effect of Preloading in Clamp (클램프에서 예하중의 영향에 관한 연구)

  • The clamp, as the structure which is used for supporting the pipe in the atomic power plant, is produced with a certain degree of anticlastic curvature in the current manufacturing process. In this study, the structural analysis of the clamp and the pipe was performed using ABAQUS. And the finite element modelling for the analysis was made by an HyperMesh. The contact forces which are transferred between the clamp and the pipe for the external force are changed according to the binding force of bolts and keeps the clamp tightly and protects the slipping between the clamp and the pipe. The clamps with the anticlastic curvature and with the flat curvature are considered in order to invest the anticlastic effect. In this study, another case is suggested. The present case does not have the stiffness ring on the end of the clamp but the suggested case has the ring. For the present case, the results showed that the equivalent stress is higher in the anticlastic curvature case than in the flat curvature case and the equivalent stresses on the pipe are almost the same as the binding force increses. For the suggested case, the result showed that the equivalent stress in the anticlastic curvature case decreases until some binding force and increases as the binding forces increase and is lower in some range than in the flat curvature case. From this study, the clamp with the anticlastic curvature in the suggested method is better than the clamp with the flat curvature and the optimal binding force are given.

Evaluation on Damage Weak Part of Rail Fastening System for Concrete Tracks (콘크리트 궤도용 탄성레일체결장치 손상취약부 분석)

  • The purpose of this study is to derive the damage weak part of the elastic rail fastening system for concrete tracks (System 300-1). In the concrete tracks, the elastic rail fastening system sticks the rail and the sleeper and reacts all the time when the train is running. Among the components of the rail fastening system, the resilience pad and tension clamp were fatigue members and were constantly deformed in response to compressive and uplift forces. In this study, the residual deformation characteristics of the tension clamp according to the period of use were analyzed using by specimens taken on site in the same section for 6, 11, and 16 years on the serviced urban transit. In addition, the damage mechanisms for each component were derived based on finite element analysis. As a result of the numerical analysis, the stress (strain) of each part of the tension clamp according to the external force from the applied clamping force was analyzed to derive the damaged weak part of the tension clamp.

Expression and Characterization of G Protein-activated Inward Rectifier $K^+$ Channels in Xenopus Oocytes

  • The G protein-activated inwardly rectifying $K^+$ channel (GIRK1) was coex-pressed in Xenopus oocytes along with the $5-HT_{1A}$ receptor, a 7-helix receptor known to be coupled to $K^+$ channels in many neural tissues. Thus, the activation of the $5-HT_{1A}$ receptor by its agonist leads to the opening of GIRK1. The GIRK1 current was measured using the two electrode voltage clamp technique with bath application of 5-HT in the presence of various external potassium concentrations $[K^+]_0$. GIRK1 showed a strong inward rectification since only hyperpolarizing voltages evoked inward currents. $K^{+}$ was the major ion carrier as evidenced by about 44㎷ voltage shift corresponding to a 10-fold external 〔$K^+$〕 change. 5-HT induced a concentration-dependent inward $K^+$ current ($EC_{50}{\equation omitted}10.7nM$) which was blocked by $Ba^{2+}$. Pertussis toxin (PTX) pre-treatment reduced the $K^+$ current by as much as about 70%, suggesting that PTX-sensitive G protein ($G_i or G_o$ type) are involved in the $5-HT_{1A}$ receptor-GIRK1 coupling in Xenopus oocytes.

Effects of Samchulkunbi-tang in Cultured Interstitial Cells of Cajal of Murine Small Intestine

  • We studied the modulation of pacemaker activities by Samchulkunbi-tang (SCKB) in cultured interstitial cells of Cajal (ICC) from murine small intestine with the whole-cell patch-clamp technique. Externally applied SCKB produced membrane depolarization in the current-clamp mode. The pretreatment with $Ca^{2+}$-free solution and thapsigargin, a $Ca^{2+}$-ATPase inhibitor in endoplasmic reticulum, abolished the generation of pacemaker potentials and suppressed the SCKB-induced action. The application of flufenamic acid (a nonselective cation channel blocker) abolished the generation of pacemaker potentials by SCKB. However, the application of niflumic acid (a chloride channel blocker) did not inhibit the generation of pacemaker potentials by SCKB. In addition, the membrane depolarizations were inhibited by not only GDP-${\beta}$-S, which permanently binds G-binding proteins, but also U-73122, an active phospholipase C inhibitor. These results suggest that SCKB modulates the pacemaker activities by nonselective cation channels and external $Ca^{2+}$ influx and internal $Ca^{2+}$ release via G-protein and phospholipase C-dependent mechanism. Therefore, the ICC are targets for SCKB and their interaction can affect intestinal motility.

Experimental Analysis on Yield Strength of Pipe Connectors and Joints for Pipe Framed Greenhouses (파이프 골조 온실의 조립 연결구 내력 시험)

  • Experiments on the yield strength of pipe connectors made of metal wire, joint pins, pole pipes, multi span insertion joints, and T-clamp joints used in pipe houses were conducted. The strength of connections of a pipe connector made of metal wire was adequate but it had a big difference according to loading direction. Therefore as it is installed, its direction should be taken into consideration. The collapse load of pipes connected with a joint pin was lower than that of single pipes. In the part of frame member at which the great bending moment occurs, the use of joint pin should be avoided. Also experimental results showed that pole pipes for use in a part of frame buried under the ground were safe, and the strength of multi span insertion joints should be increased. The resistant moment of T-clamp was about 13.7% of a single pipe. In case that the external forces acting on left and right rafter are different. a unsymmetrical rotational force is produced at the multi span joint. If it is expected that the actual bending moment on the multi span joint is larger than resistant moment of T-clamp, a reinforcement to safely resist the rotational force is required.

Loosening behavior of Internal and External Connection Dental Implants under Cyclic Loads Considering Pre-fastening Force (체결력을 고려한 내부 및 외부연결형 임플란트의 반복 하중에 대한 풀림 연구)

  • This paper presents the loosening behavior that occurs after the application of an external load to internal and external connection types of dental implants using the finite element method. We use fastening force between an abutment and a fixture to clamp the dental implant system; however, loosening and breakage may occur owing to cyclic external loads. In this study, we considered the initial fastening condition in the pre-load analysis and then investigated the change in stress and contact surface status when applying external loads. After the application of the initial fastening load, we verified that the internal connection-type model exhibited a relatively lower stress distribution than that of the external connection-type one. Moreover, we found that the former model showed a lower stress concentration after the application of the external load. In addition, after the application of this load, we found that the higher the shear load acting on the implant system, the higher the possibility of loosening. The study results showed the change in stress distribution and contact surface according to the connection type of the dental implants and the phenomenon of loosening by cyclic loads. We expect that the results of this study will be useful for the study of reliability and design of dental implant systems.

Inwardly Rectifying $K^+$ Currents in Gastric Myocytes of Guinea-pig

  • To identify the presence of inwardly rectifying $K^+$ channels and its characteristics, membrane currents were measured using a whole-cell patch clamp from isolated gastric myocytes of guinea-pig. Change of external $K^+$ concentration from 5 to 90 mM induced an inward current at a holding potential of -80 mV. The high $K^+-induced$ inward current was blocked by $Ba^{2+}$ and $Cs^+,$ but not by glibenclamide. With 90 mM $K^+$ in bath, the $Ba^{2+}-$ and $Cs^+-sensitive$ currents showed strong inward rectification. Ten mM TEA weakly blocked the inward current only at potentials more negative than -50 mV. With 90 mM $K^+$ in bath, hyperpolarizing step pulses from -10 mV induced inward currents, which were inactivated at potentials more negative than -70 mV. Reduction of external $K^+$ to 60 mM decreased the amplitudes of the currents and shifted the reversal potential to more negative potential. The inactivation of inward $K^+$ current at negative clamp voltage was not affected by removing external $Na^+.$ These results suggest that the inwardly rectifying $K^+$ channels may exist in gastric smooth muscle.

External pH Effects on Delayed Rectifier $K^+$ Currents of Small Dorsal Root Ganglion Neuron of Rat

  • Under certain pathophysiological conditions, such as inflammation and ischemia, the concentration of H^+$ ion in the tissue surrounding neurons is changed. Variations in H^+$ concentration are known to alter the conduction and/of the gating properties of several types of ion channels. Several types of K^+$ channels are modulated by pH. In this study, the whole cell configuration of the patch clamp technique has been applied to the recording of the responses of change of external pH on the delayed rectifier K^+$ current of cultured DRG neurons of rat. Outward K^+$ currents were examined in DRG cells, and the Charybdotoxin and Mn^{2+}$ could eliminate Ca^{2+}-dependent$ K^+$ currents from outward K^+$ currents. This outward K^+$ current was activated around -60 mV by step depolarizing pulses from holding potential -70 mV. Outward K^+$ currents were decreased by low external pH. Activation and steady-state inactivation curve were shifted to the right by acidification, while there was small change by alkalization. These results suggest that H^+$ could be alter the sensory modality by changing and modifying voltage-dependent K^+$ currents, which participated in repolarization.
