Monitoring of Chicken RNA Integrity as a Function of Prolonged Postmortem Duration

  • Gene expression profiling has offered new insights into postmortem molecular changes associated with meat quality. To acquire reliable transcript quantification, high quality RNA is required. The objective of this study was to analyze integrity of RNA isolated from chicken skeletal muscle (pectoralis major) and its capability of serving as the template in quantitative real-time polymerase chain reaction (qPCR) as a function of postmortem intervals representing the end-points of evisceration, carcass chilling and aging stages in chicken abattoirs. Chicken breast muscle was dissected from the carcasses (n = 6) immediately after evisceration, and one-third of each sample was instantly snap-frozen and labeled as 20 min postmortem. The remaining muscle was stored on ice until the next rounds of sample collection (1.5 h and 6 h postmortem). The delayed postmortem duration did not significantly affect $A_{260}/A_{280}$ and $A_{260}/A_{230}$ ($p{\geq}0.05$), suggesting no altered purity of total RNA. Apart from a slight decrease in the 28s:18s ribosomal RNA ratio in 1.5 h samples (p<0.05), the value was not statistically different between 20 min and 6 h samples ($p{\geq}0.05$), indicating intact total RNA up to 6 h. Abundance of reference genes encoding beta-actin (ACTB), glyceraldehyde 3-phosphate dehydrogenase (GAPDH), hypoxanthine-guanine phosphoribosyltransferase (HPRT), peptidylprolylisomerase A (PPIA) and TATA box-binding protein (TBP) as well as meat-quality associated genes (insulin-like growth factor 1 (IGF1), pyruvate dehydrogenase kinase isozyme 4 (PDK4), and peroxisome proliferator-activated receptor delta (PPARD) were investigated using qPCR. Transcript abundances of ACTB, GAPDH, HPRT, and PPIA were significantly different among all postmortem time points (p<0.05). Transcript levels of PDK4 and PPARD were significantly reduced in the 6 h samples (p<0.05). The findings suggest an adverse effect of a prolonged postmortem duration on reliability of transcript quantification in chicken skeletal muscle. For the best RNA quality, chicken skeletal muscle should be immediately collected after evisceration or within 20 min postmortem, and rapidly preserved by deep freezing.

A Study on Content Characteristics, Consumer Behavior and Economic Value According to the Degree of Consideration of Graphic Content (그래픽 콘텐츠 고려 정도에 따른 콘텐츠 특성, 소비자 행동, 경제적 가치에 관한 연구)

  • This study verified what differences in screen golf content characteristics, intention to reuse, customer satisfaction and economic value experienced by consumers according to the image feeling, expression method, and image color provided by screen golf graphic content. In addition, the purpose of this study was to analyze what kind of influence the content characteristics of screen golf have on the economic value and what kind of influence the intention to reuse and customer satisfaction have in this process. From September 1, 2021 to September 30, 2021, a survey of 225 copies of consumers using the screen golf course was conducted. For data processing, frequency analysis, factor analysis, reliability analysis, cluster analysis, chi-square analysis and 3-step mediated regression analysis were performed. The research results are as follows. First, the preferred image feeling showed a high level of clean and sophisticated feeling and the preferred expression method showed a high realistic image. In addition, the preferred image color showed a high level of green color. Second, there were differences in competitiveness, ease of use, sense of solidarity and realism according to the degree of consideration of graphic content and differences in consumer's intention to reuse, customer satisfaction, and economic value. Third, in the relationship between screen golf content characteristics and economic value, customer satisfaction and re-use intention had a mediating effect. Through this study, by providing basic data to derive the graphic design model of screen golf, the operating entity suggested a way to improve economic benefits and tried to contribute to the growth of the screen golf industry.

Generating Rank-Comparison Decision Rules with Variable Number of Genes for Cancer Classification (순위 비교를 기반으로 하는 다양한 유전자 개수로 이루어진 암 분류 결정 규칙의 생성)

  • Microarray technology is extensively being used in experimental molecular biology field. Microarray experiments generate quantitative expression measurements for thousands of genes simultaneously, which is useful for the phenotype classification of many diseases. One of the two major problems in microarray data classification is that the number of genes exceeds the number of tissue samples. The other problem is that current methods generate classifiers that are accurate but difficult to interpret. Our paper addresses these two problems. We performed a direct integration of individual microarrays with same biological objectives by transforming an expression value into a rank value within a sample and generated rank-comparison decision rules with variable number of genes for cancer classification. Our classifier is an ensemble method which has k top scoring decision rules. Each rule contains a number of genes, a relationship among involved genes, and a class label. Current classifiers which are also ensemble methods consist of k top scoring decision rules. However these classifiers fix the number of genes in each rule as a pair or a triple. In this paper we generalized the number of genes involved in each rule. The number of genes in each rule is in the range of 2 to N respectively. Generalizing the number of genes increases the robustness and the reliability of the classifier for the class prediction of an independent sample. Also our classifier is readily interpretable, accurate with small number of genes, and shed a possibility of the use in a clinical setting.

Effects of Teaching Behavior Type of Expression Activity Class on Flow and Competence in Elementary School (초등학교 표현활동수업에서의 교수행동유형이 몰입과 유능감에 미치는 영향)

  • The purpose of this study was to know the effects of teaching behavior type of expression activity class on flow and competence in elementary school. The subject for this research consisted of elementary students located in Busan were selected as a population. The sampling method was convenience sampling out of non-probability sampling method. A total of 300 questionnaires were distributed and 280 copies were collected back. Out of those returned questionnaires, insincerely replied or double-replied questionnaires were excluded and finally 216 questionnaires were analyzed for this study. Collected data were put to frequency analysis, exploratory factor analysis reliability analysis, correlation analysis, and multiple regression analysis using the SPSS Win Ver. 22.0 program. The results of these analyses are as follows. First, the commanding type, the autocratic type, the democratic type and the reward type which are the subordinate variables of the teaching behavior type, were all found to have significant effects on the flow. Second, the democratic type and the reward type which are the subordinate variables of the teaching behavior type, were found to have significant effects on the competence. But, the commanding type and the autocratic type which are the subordinate variables of the teaching behavior type, were not found to have significant effects on the competence. Third, the flow were found to have significant effects on the competence.

Development And Applying Detailed Competencies For Elementary School Students' Data Collection, Analysis, and Representation (초등학생의 데이터 수집, 분석, 표현 수업을 위한 세부역량 개발 및 적용)

  • From 2019, software education has become a required subject for all elementary school students. However, many teachers are still unfamiliar with how the classes should be instructed. So this paper presented the meaning, detailed competencies and achievement standard in order to help in the collection, analysis and representation of data among the computational thinking that are key to software education. And it also suggested the applicability of the classes. The full course of the paper is summarized as follows. First, existing studies have summarized the meaning, detail and achievement standard of data related competencies. Based on this, a preliminary investigation was instructed. Pilot study carried out both FGI and closed questions at the same time. This was done in response to the survey's questionnaire reflecting the opinions of experts. Second, the results of the questionnaire generated as a result of the above were verified for validity, stability, and reliability among the PhD, PhD courses, software education teachers, and software education workers. Third, I developed and applied the five lessons as a class objective as 'Choosing collection method-Select the collection method according to the problem situation.', 'Searching for meaning of data-Understand what the analyzed data mean..', 'Using various expression methods-Use a variety of expression tools.' using the backward design model to integrate education, class, and assessment. As a result, the detailed competencies of data collection, analysis, and representation and achievement standard were presented. This may help in setting specific and specific criteria for what direction classes are recommended when planning data-related classes in elementary schools.

The influencing factors on procalcitonin values in newborns with noninfectious conditions during the first week of life

  • Purpose: Although procalcitonin (PCT) level is useful for the diagnosis of neonatal sepsis, PCT reliability is inconsistent because of the varied conditions encountered in neonatal intensive care units. This study aimed to investigate PCT levels and factors influencing increased PCT levelin newborns without bacterial infection during the first week of life. Methods: In newborns hospitalized between March 2013 and October 2015, PCT levels were measured on the first, third, and seventh days after birth. Newborns with proven bacterial (blood culture positive for bacteria) or suspicious infection (presence of C-reactive protein expression or leukocytosis/leukopenia) were excluded. Various neonatal conditions were analyzed to identify the factors influencing increased PCT level. Results: Among 292 newborns with a gestational age of $35.2{\pm}3.0$ weeks and a birth weight of $2,428{\pm}643g$, preterm newborns (n=212) had higher PCT levels than term newborns (n=80). Of the newborns, 7.9% had increased PCT level (23 of 292) on the firstday; 28.3% (81 of 286), on the third day; and 3.3% (7 of 121), on the seventh day after birth. The increased PCT level was significantly associated with prenatal disuse of antibiotics (P=0.004) and surfactant administration (P<0.001) on the first day after birth, postnatal use of antibiotics (P=0.001) and ventilator application (P=0.001) on the third day after birth, and very low birth weight (P=0.042) on the seventh day after birth. Conclusion: In newborns without bacterial infection, increased PCT level was significantly associated with lower gestational age and respiratory difficulty during the first week of life. Further studies are needed for clinical applications.

A Study on Development of Fashion Orientation Scale for the On-line Image Consulting of Career Men (직장남성의 이미지 컨설팅을 위한 측정도구 개발에 관한 연구 -패션성향을 중심으로-)

  • This study was aimed at the development of scales measuring appearance management behaviors of career men for on-line image consulting. The purposes of this study were to develop a scale which can measure fashion orientation of career men and classify them according to fashion orientation. The data were collected from 380 career men for the first survey and 372 career men for the second survey in Seoul, Korea and were analyzed by factor analysis, reliability test, cluster analysis, one-way ANOVA and Duncan test. The results from this study were as follows: 1) Ten factors of fashion orientation were identified: clothing interest, satisfaction in clothing-selecting ability, individuality and self expression, interest in appearance management, appearance improvement and mood enhancement, clothing conformity, putting value on attractive appearance, maintenance of neat appearance, weight control and pursuit of fashion. The total variance explained by the ten factors was $65.70\%$ and Cronbach's $\alpha$ of 10 factors ranged from $.60\~.92.$ 2) Three groups were classified by the representative items of fashion orientation factors : passive appearance management group who were conscious of other people, positive appearance management group who were highly fashion-oriented and indifferent group to appearance management. The fashion orientation characteristics of groups classified in the second survey were nearly similar to those in the first.

A Study on In-line Skate Clothing Interest of In-line Skaters (인라인스케이터의 인라인스케이트 의복흥미 연구(Ⅱ))

  • As the life styles have been changed in modern society and sports gave been emphasized for spending spare time and improving health condition, sports are regarded to lave a leading role to fashion as one of the prominent factors. This study is to analyze the gender-identity and how different levels of involvement in In-line skating affects clothing interests. 291 In-line skaters ranged in age from middle twenties to middle thirties were surveyed and measured for this study. MANOVA(multivariate analysis of variance), ANOVA(univariate analysis of variance), Cronbach ${\alpha}$ reliability, t-test, and correlation analysis were performed with SPSS 10.0 program on the selected data. The results of this study are as follow. There were no obvious differences in In-line skating clothing interests by gender of In-line skaters. There were, however, differences in the reasons for In-line skating, the hours to skate per week. Those whose purpose for skating was diet had more interest in style focused on expression. In case of skating for health, they showed interest in activity-functionality style and anti-deviation style. The group of In-line skaters who skated 15 to 18 per week gave more interest in activity-functionality style out of the clothing interest-factors. Those who had strong masculinity in gender-identity of In-line skaters were more involved in In-line skating. There were some differences in clothing interests according to gender-identity of In-line skaters. In regard of In-line skating clothing interest by involvement in In-line skating, those who had higher involvement in In-line skating had higher In-line skating clothing interest.

  • In the large scale recent reclaiming works performed within the wide spatial boundary, evaluation of long-term consolidation settlement and residual settlement of the whole construction area is sometimes made with the results of the limited ground investigation and measurement. Then the reliability of evaluation has limitations due to the spatial uncertainty. Additionally, in case of large scale deep excavation works such as urban subway construction, there are a lot of hazardous elements to threaten the safety of underground pipes or adjacent structures. Therefore it is necessary to introduce a damage prediction system of adjacent structures and others. For the more accurate analysis of monitoring information in the wide spatial boundary works and large scale urban deep excavations, it is necessary to perform statistical and spatial analysis considering the geographical spatial effect of ground and monitoring information in stead of using diagrammatization method based on a time-series data expression that is traditionally used. And also it is necessary that enormous ground information and measurement data, digital maps are accumulated in a database, and they are controlled in a integrating system. On the abovementioned point of view, we developed Geomonitor 2.0, an Internet based real time monitoring program with a new concept by adding GIS and geo-statistical analysis method to the existing real time integrated measurement system that is already developed and under useful use. The new program enables the spatial analysis and database of monitoring data and ground information, and helps the construction- related persons make a quick and accurate decision for the economical and safe construction.

Proteomic Analysis of Bovine Pregnancy-specific Serum Proteins by 2D Fluorescence Difference Gel Electrophoresis

  • Two dimensional-fluorescence difference gel electrophoresis (2D DIGE) is an emerging technique for comparative proteomics, which improves the reproducibility and reliability of differential protein expression analysis between samples. The purpose of this study was to investigate bovine pregnancy-specific proteins in the proteome between bovine pregnant and non-pregnant serum using DIGE technique. Serums of 2 pregnant Holstein dairy cattle at day 21 after artificial insemination and those of 2 non-pregnant were used in this study. The pre-electrophoretic labeling of pregnant and non-pregnant serum proteins were mixed with Cy3 and Cy5 fluorescent dyes, respectively, and an internal standard was labeled with Cy2. Labeled proteins with Cy2, Cy3, and Cy5 were separated together in a single gel, and then were detected by fluorescence image analyzer. The 2D DIGE method using fluorescence CyDye DIGE flour had higher sensitivity than conventional 2D gel electrophoresis, and showed reproducible results. Approximately 1,500 protein spots were detected by 2D DIGE. Several proteins showed a more than 1.5-fold up and down regulation between non-pregnant and pregnant serum proteins. The differentially expressed proteins were identified by MALDI-TOF mass spectrometer. A total 16 protein spots were detected to regulate differentially in the pregnant serum, among which 7 spots were up-regulated proteins such as conglutinin precursor, modified bovine fibrinogen and IgG1, and 6 spots were down-regulated proteins such as hemoglobin, complement component 3, bovine fibrinogen and IgG2a three spots were not identified. The identified proteins demonstrate that early pregnant bovine serum may have several pregnancy-specific proteins, and these could be a valuable information for the development of pregnancy-diagnostic markers in early pregnancy bovine serum.
