Lysosomal acid phosphatase mediates dedifferentiation in the regenerating salamander limb

  • In this study, monoclonal antibodies against lysosomal acid phosphatase (LAP) of a salamander, Hynobius leechii, were used to determine the spatial and temporal expression of the LAP in the regenerating limbs. The Western blot and immunohistochemical analysis in the limb regeneration revealed that LAP was highly expressed at the dedifferentiation stage, especially in the wound epidermis and dedifferentiating limb tissues such as muscle and cartilage. With RA treatment, the LAP expression became upregulated in terms of both level and duration in the wound epidermis, blastemal cell and dedifferentiating limb tissues. In addition, in situ activity staining of LAP showed a similar result to that of immunohistochemistry. Thus, the activity profile of LAP activity coincides well with the expression profile of LAP during the dedifferentiation period. Furthermore, to examine the effects of lysosomal enzymes including LAP on salamander limb regeneration, lysosome extract was microinjected into limb regenerates. Interestingly, when the lysosome extract was microinjected into limb regenerates with a low dose of RA($50\;{\mu}g/g$ body wt.), skeletal pattern duplication occurred frequently in the proximodistal and transverse axes. Therefore, lysosomal enzymes might cause the regenerative environment and RA plays dual roles in the modification of positional value as well as evocation of extensive dedifferentiation for pattern duplication. In conclusion, these results support the hypothesis that dedifferentiation is a crucial event in the process of limb regeneration and RA-evoked pattern duplication, and lysosomal enzymes may play important role(s) in this process.

Gene Discovery Analysis from Mouse Embryonic Stem Cells Based on Time Course Microarray Data

  • An embryonic stem cell is a powerful tool for investigation of early development in vitro. The study of embryonic stem cell mediated neuronal differentiation allows for improved understanding of the mechanisms involved in embryonic neuronal development. We investigated expression profile changes using time course cDNA microarray to identify clues for the signaling network of neuronal differentiation. For the short time course microarray data, pattern analysis based on the quadratic regression method is an effective approach for identification and classification of a variety of expressed genes that have biological relevance. We studied the expression patterns, at each of 5 stages, after neuronal induction at the mRNA level of embryonic stem cells using the quadratic regression method for pattern analysis. As a result, a total of 316 genes (3.1%) including 166 (1.7%) informative genes in 8 possible expression patterns were identified by pattern analysis. Among the selected genes associated with neurological system, all three genes showing linearly increasing pattern over time, and one gene showing decreasing pattern over time, were verified by RT-PCR. Therefore, an increase in gene expression over time, in a linear pattern, may be associated with embryonic development. The genes: Tcfap2c, Ttr, Wnt3a, Btg2 and Foxk1 detected by pattern analysis, and verified by RT-PCR simultaneously, may be candidate markers associated with the development of the nervous system. Our study shows that pattern analysis, using the quadratic regression method, is very useful for investigation of time course cDNA microarray data. The pattern analysis used in this study has biological significance for the study of embryonic stem cells.

A Study on Changing Process of Nonlinear Expression Methods appeared in Frank Gehry's works according to Digital Archhitecture's introduction (디지털 건축의 도입에 따른 프랭크 게리의 작품에서 나타나는 비선형적 표현 기법의 변화 과정에 관한 연구)

  • Conceptual and technical design designated as digital architecture has been discussed with disorderly meaning as a conception only. Thus, this study aims at verification of development possibility for the digital architecture by analyzing how works of Frank Gehry have effect on nonlinearity of the digital architecture. Nonlinear characteristics appeared on works of Frank Gehry analyzed on the basis of expression techniques and modeling principle are as follows: 1) Initial works show a trend to gradually change and develop from horizontal and vertical linear structure to nonlinear pattern. 2) From a nonlinear modeling principle, initial works appear the primary deformation elements only, while afterwards, they are gradually developing to a pattern to mix the primary and secondary deformation elements as introduction of digital architecture. 3) Through specific cases, Frank Gehry has conducted attempts with various method for nonlinear pattern expression. 4) From the initial works to the latest works, continuity becomes higher and changed to nonlinear pattern. This study is significant from a viewpoint that it has verified development possibility of digital architecture by analyzing nonlinear trend of digital architecture for Frank Gehry and it is required to conduct multilateral researches related to the digital architecture.

Robust Facial Expression Recognition Based on Signed Local Directional Pattern (Signed Local Directional Pattern을 이용한 강력한 얼굴 표정인식)

  • In this paper, we proposed a new local micro pattern, Signed Local Directional Pattern(SLDP). SLDP uses information of edges to represent the face's texture. This can produce a more discriminating and efficient code than other state-of-the-art methods. Each micro pattern of SLDP is encoded by sign and its major directions in which maximum edge responses exist-which allows it to distinguish among similar edge patterns that have different intensity transitions. In this paper, we divide the face image into several regions, each of which is used to calculate the distributions of the SLDP codes. Each distribution represents features of the region and these features are concatenated into a feature vector. We carried out facial expression recognition with feature vectors and SVM(Support Vector Machine) on Cohn-Kanade and JAFFE databases. SLDP shows better classification accuracy than other existing methods.

A study on the Contemporary Art Inspiration in Contemporary Fashion Design (현대패션에 나타난 Contemporary Art Inspiration 연구)

  • The purpose of this study is to suggest more specific method to express art connected to the factor of fashion design by classifying fashion case combined with contemporary art, which is expressed through visual factor of fashion, analyzing exchange phenomenon, trend and change aspect of art and fashion and researching its expression type under the background of art preference phenomenon of contemporary art. From the result of measuring the frequency of contemporary art type, it was found that various types and artists' works were applied to fashion industry. Therefore, the scope of contemporary art, which is used for fashion, is being enlarged and new type is appearing every year continuously. Especially, its frequency was highest in 2008 S/S. In addition, it was found that it was more frequent in S/S season rather than in F/W season. From the result of analyzing expression method of contemporary art inspiration shown in fashion by classifying it into structure aspect, print aspect and 3D decoration aspect, in most cases, the contemporary art was used as a print or pattern for dress or accessory. The print aspect could be divided into geometric abstraction pattern, expressional abstract pattern, trompe-l'oeil pattern, graffiti pattern, picture image pattern and cartoon pattern in detail.

Expression of Chimeric Chicken-Yeast-Chicken H2B Histone Gene

  • A chicken H2B histone gene was cloned and expressed in Rat 3 cell line. Its messenger RNA level was about 10 times higher during S phase than during $G_1$ phase. A chimeric chicken-yeast-chicken H2B histone gene was made to change some of wobble sequences of chicken H2B gene. When the chimeric H2B gene was transfected into the Rat 3 cell line, it showed a pattern of expression similar to that of the original chicken H2B gene. At least in this gene, it was concluded that the wobble sequences were not required for the cell-cycle regulated pattern of expression.

Homeobox Gene (OSH1) Expression in Embryonic Mutants of Rice (Oryza sativa L.)

  • Recent identification and characterization of plant homeobox genes suggest that they play important roles in morphogenetic events. OSH1, one of the rice homeobox genes, is thought to be related to organ development since the changes of OSH1 gene expression cause morphological abnormalities of leaves by the ectopic expression and is expressed during early embryogenesis. In this experiment, the expression pattern of OSH1 was analyzedinmutants by in situ hybridization, and OSH1's potential as a molecular marker was explored. Region-specific expression of OSH1 during early embryogenesis shows that OSH1 could be used as a molecular marker for characterizing embryo mutants. Although several organless and shootless mutants showed normal expression of OSM1, some mutants exhibited abnormal expression patterns. In a minute organless cle1-1 embryo whose epidermis resembled morphologically the epithelium of scutellum, OSH1 expression was limited to a small basal region. This expression pattern suggests the gross deletion of the basal part. In a radicleless mutant, odm115, OSH1 expression was detected in a basal region instead of subcentral region of the ventral side. Together with other characteristics (short embryo and normal adventitious roots), odm115 was estimated to be derived from the deletion of basal region. Among five shootless mutants, three showed normal expression of OSH1. In the shl2 embryo, no expression of OSH1 was observed. In the shl1 embryo, however, OSH1 expression was extended to a dorsal side, indicating that SHL2 might be related to dorsoventral patterning. The above results of in situ hybrydization clearly indicate that OSH1 can be utilized as a marker for characterizing gene functions of embryo mutants.

  • This study investigated expression pattern of PHGPx gene in male rat reproductive organs exposed to 17$\beta$-estradiol. First, in view of quantitative change, the exposure to 17$\beta$-estradiol for 1 week increased PHGPx mRNA level in testis and prostate. PHGPx mRNA level in epididymis decreased weakly as compared to control group.(omitted)

  • This paper proposes the method of facial expression recognition based on decision tree structure. In the image of facial expression, ASM(Active Shape Model) and LBP(Local Binary Pattern) make the local features of a facial expressions extracted. The discriminant features gotten from local features make the two facial expressions of all combination classified. Through the sum of true related to classification, the combination of facial expression and local region are decided. The integration of branch classifications generates decision tree. The facial expression recognition based on decision tree shows better recognition performance than the method which doesn't use that.

  • The purpose of this study is to contribute to the role of lines in creative design development by analyzing the expression types and aesthetic characteristics of modern fashion using geometric formativeness of symmography. A literature study was conducted of works since 2009 to examine the general consideration of lines together with analysis of the concept and characteristics of symmography in the formative arts field, and to analyze the expression types and aesthetic characteristics of modern fashion design using the formativeness of symmography. The infinite sense of formativeness and original expression of symmography are used in formative arts such as space design, installation art, and industrial design. Expression types of modern fashion design using geometric formativeness of symmography can be classified into the following three types: two-dimensional graphic pattern, relief surface, and three-dimensional spatial. First, the two-dimensional graphic pattern type forms an optical pattern, providing individuality and visual interest to the textile design. Second, the relief surface type expresses the plane in various ways, so that the thickness changes according to how lines overlap. Third, the three-dimensional spatial type expands the boundaries of clothing and creates a fantastic spatial beauty. Next, the aesthetic formativeness of fashion design using symmography can be classified into repetitive rhythmicity, geometric self-similarity, and optical spatiality. Symmography enables a myriad of geometric patterns to be developed depending on material, color, and the designer's imagination, and helps inspire a variety of designs in fashion that sculpt a three-dimensional human body.
