  • During embryo implantation in pigs, the uterine endometrium undergoes dramatic morphological and functional changes accompanied with dynamic gene expression. Since the greatest amount of embryonic losses occur during this period, it is essential to understand the expression and function of genes in the uterine endometrium. Although many reports have studied gene expression in the uterine endometrium during the estrous cycle and pregnancy, the pattern of global gene expression in the uterine endometrium in response to the presence of a conceptus (embryo/fetus and associated extraembryonic membranes) has not been completely determined. To better understand the expression of pregnancy-specific genes in the endometrium during the implantation period, we analyzed global gene expression in the endometrium on day (D) 12 and D15 of pregnancy and the estrous cycle using a microarray technique in order to identify differentially expressed endometrial genes between D12 of pregnancy and D12 of the estrous cycle and between D15 of pregnancy and D15 of the estrous cycle. Results showed that the global pattern of gene expression varied with pregnancy status. Among 23,937 genes analyzed, 99 and 213 up-regulated genes and 92 and 231 down-regulated genes were identified as differentially expressed genes (DEGs) in the uterine endometrium on D12 and D15 of pregnancy compared to D12 and D15 of the estrous cycle, respectively. Functional annotation clustering analysis showed that those DEGs included genes involved in immunity, steroidogenesis, cell-to-cell interaction, and tissue remodeling. These findings suggest that the implantation process regulates differential endometrial gene expression to support the establishment of pregnancy in pigs. Further analysis of the genes identified in this study will provide insight into the cellular and molecular bases of the implantation process in pigs.

  • Objective: An experiment was conducted to determine the relationship between the KAP11.1 and the regulation wool fineness. Methods: In previous work, we constructed a skin cDNA library and isolated a full-length cDNA clone termed KAP11.1. On this basis, we conducted a series of bioinformatics analysis. Tissue distribution of KAP11.1 mRNA was performed using semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) analysis. The expression of KAP11.1 mRNA in primary and secondary hair follicles was performed using real-time PCR (real-time polymerase chain reaction) analysis. The expression location of KAP11.1 mRNA in primary and secondary hair follicles was performed using in situ hybridization. Results: Bioinformatics analysis showed that KAP11.1 gene encodes a putative 158 amino acid protein that exhibited a high content of cysteine, serine, threonine, and valine and has a pubertal mammary gland) structural domain. Secondary structure prediction revealed a high proportion of random coils (76.73%). Semi-quantitative RT-PCR showed that KAP11.1 gene was expressed in heart, skin, and liver, but not expressed in spleen, lung and kidney. Real time PCR results showed that the expression of KAP11.1 has a higher expression in catagen than in anagen in the primary hair follicles. However, in the secondary hair follicles, KAP11.1 has a significantly higher expression in anagen than in catagen. Moreover, KAP11.1 gene has a strong expression in inner root sheath, hair matrix, and a lower expression in hair bulb. Conclusion: We conclude that KAP11.1 gene may play an important role in regulating the fiber diameter.

  • The objective of this study was to determine the mRNA expression patterns of several putative imprinted genes in in vivo and in vitro fertilized, parthenogenetic, and cloned porcine preimplantation embryos. Both maternally (Dlk1, IGF2, Peg1/Mest and Ndn) and paternally (IGF2r, H19 and Xist) imprinted genes were selected. We have used reverse transcription polymerase chain reaction (RT-PCR) to investigate gene expression patterns in the porcine embryos. IGF2 transcripts were detected in the most of embryos. In nuclear transfer (NT), Peg1/MEST transcripts showed fluctuating pattern. Dlk1 was only expressed partially from the morula and blastocyst stage of NT embryos. Ndn gene expression was started somewhat early for in vivo embryos. However, the expressions of maternally imprinted genes were similar in all types of blastocysts (NT, in vivo and in vitro fertilized, and parthenogenetic embryos). The IGF2R gene expression level was somewhat irregular and varied among samples. However, for the majority samples of all types of embryos, IGF2R expression was diminished after one- to two-cell stages and reappeared at the morulae or blastocyst stage embryos. H19 gene was only expressed early in parthenogenetic and in vivo embryos. For NT embryos, H19 was only expressed in blastocysts. Xist expression was detected in all blastocysts with the earliest being in vivo 8-cell stage embryos and the last one being NT blastocysts. These putative imprinted genes appeared to have stage specific expression patterns with a fluctuating pattern for some genes (Peg/Mest, IGF2r, H19). These results suggest that stage specific presence of imprinted genes can affect the embryo implantation and fetal development.

  • To clarify the role of stem cells in hepatocarcinogenesis, glypican-3 (GPC-3) and E-cadherin expression was investigated in embryonic cell lineages. Mouse embryonic stem cells (ESCs), hepatic progenitor cells (HPCs) and hepatocyte like cells (HCs), representing 0, 22 and 40 days of differentiation, respectively, were treated in vitro with diethylnitrosamine (DEN) at four doses (0, 1, 5 and 15 mM; G1, G2, G3 and G4, respectively) for 24 h and GPC-3 and E-cadherin expression was examined by relative quantitative real-time PCR and immunocytochemistry. GPC-3 mRNA expression was significantly different for G4 at day 0 (p<0.001) and for G4 at day 22 (p<0.01) compared with the control (G1). E-cadherin mRNA expression was significantly different for G3 and G4 at day 0 (p<0.05 and p<0.001, respectively), for G2 and G4 (p<0.05 and p<0.001, respectively) at day 22 and for G2 and G4 (p<0.01 and p<0.001, respectively) at day 40 compared with G1. Immunofluorescence staining for GPC-3 showed a membranous and/or granular expression in cytoplasm of ESCs and HPCs and granular and/or diffuse expression in cytoplasm of HCs, which were also stained by E-cadherin. DEN treatment increased GPC-3 expression in ESCs, HPCs and HCs, with increase of E-cadherin expression. Taken together, the expression of GPC-3 was altered by DEN treatment. However, its expression pattern was different at the stage of embryo stem cells and its derived hepatic lineage cells. This suggests that GPC-3 expression may be modulated in the progeny of stem cells during their differentiation toward hepatocytes, associated with E-cadherin expression.

  • The purpose of this study was to research the characteristics of the pattern design and the weave construction of 67 fashion fabrics in women's Hanbok. All the fabrics were made by 100% silk. Arrangement, size, and the type of patterns were investigated for research of the pattern characteristics. Pattern weave and background weave were analyzed, then the weight and the thickness were measured for studying the fabric construction. The results were as follows. First, irregular arrangement was more commonly used than regular arrangement, and then the complex arrangement and the combination arrangement were appeared considerably because of the existence of the various patterns in a fabric. Third, the types of pattern weave construction were much more than those of background weave construction. This is caused by the expression of dimensional effect for patterns. This survey can be practically used for development of pattern design for Hanbok industry and furthermore it will contribute to revitalizing of Hanbok market to overcome design limit in the future.

  • We consider the irrational part of the basic pattern drafting of the Geo-Ko-ri so that we want to design it more scientically. So there are problems which is found after we considered with comparison about the existingly basic pattern drafting of Geo-go-ri. (1) there is a difference between the practical and Calculation. (2) The part of the curved line was mad indistinctly There is a difference betweence the calculating expression and calculating value. (4) There are some parts which lack objectivity about the reference size. (5) The calculating method of Collar-width and Neck-Nidth are not formative and rational. These are items that we must consider and study further. Especially if we can make up of the scientific form in consideration of the formativeness focused on "git" I think it will make a great contribution to not only the standardization of the basic pattern of Geo-go-ri and the use of the use of education but also the manufacture of individual clothing the production by Auto CAD. • In machining with cutting tool inserts having complex chip groove shape, the flow, curl and breaking patterns of the chip are different than in flat-face type inserts. In the present work, an effort is made to understand the three basic phenomena occurring in a chip since its formation in machining with groove type and pattern type inserts. These are the initial chip flow, the subsequent development of up and side curl and the final chip breaking due to the development of torsional and banding stresses. In this paper, chip flow angle in a groove type and pattern type inserts. The expression for chip flow angle in groove type and pattern type insets is also verified experimentally using high speed filming techniques.

  • Silhouette extraction is widely used in many computer graphics applications. In this paper, we proposed a method for extracting 3D silhouette and internal pattern from 3D vector data. To do this, we first make an edge-list, secondly define the silhouette, and finally remove hidden lines. After getting the silhouette, we extract internal pattern using adjacent edge's dihedral. The proposed method not only effectively improves the performance of extracting 3D silhouette and internal pattern from 3D vector data but also reduces the computational complexity.

  • This paper presents the temporal classification method based on data mining techniques for discovering knowledge from measured load patterns of distribution transformers. Since the power load patterns have time-varying characteristics and very different patterns according to the hour, time, day and week and so on, it gives rise to the uninformative results if only traditional data mining is used. Therefore, we propose a temporal classification rule for analyzing and forecasting transformer load patterns. The main tasks include the load pattern mining framework and the calendar-based expression using temporal association rule and 3-dimensional cube mining to discover load patterns in multiple time granularities.
