Physicochemical Properties and Plant Coverage of Wood-based Growing Media on Slopes

  • The use of wood waste as substrate for plant growth exemplifies a strategy for turning waste into resources. The overall objective of this research was to evaluate the effects of wood-based growing media on plant cover in a slope area. Moreover, we tried to find out what physicochemical properties affect plant cover on a slope. For treatments, we tested natural soil, soil mixed with wood-based growing medium (1:1, w/w), and wood-based growing medium by itself. Physical and chemical characteristics were evaluated after four months from the date of treatment application to the experimental slope site. Soil coverage with seedlings of Lespedeza cyrtobotrya was measured for plant growth evaluation. Physicochemical properties were altered by mixing the natural soil with wood-based growing medium. Particularly, soil moisture and organic matter contents were significantly changed in soils treated with wood-based growing medium compared to soil alone. We confirmed that plant coverage rate was high when wood-based growing medium was mixed with the natural soil. There was a significant linear relationship between moisture content and CEC (Cation Exchange Capacity) of all growth media tested and plant coverage. This result was expected, as moisture content tends to increase with organic matter content, such as in wood-based growing medium. In conclusion, the high moisture content of the wood-based growing medium was considered effective for plant growth in the experimental slope site, and this wood-based growing medium provides a means to improve the harmony between the slope and the surrounding environment.

Effect of cryopreservation of ginseng (Panax ginseng C.A. Meyer) seeds on redox ratio of ascorbate and glutathione (인삼종자 초저온보존 후 Ascorbate 및 Glutathione의 산화환원 변화)

  • Ginseng seeds are one of short-lived seeds species which loose their viability easily in the condition of conventional storage. Cryopreservation using liquid nitrogen (LN) has been recommended as a alternative storage for this kind of germplasm short lived or dessiccation-sensitive. This study was performed to find out whether cryopreservation could affect physiological change such as enzyme activity induced by reactive oxygen species. In this work, the redox ratio of ascorbate and glutathione were examined onto ginseng seedlings before and after LN storage of seeds for 1 day using spectrophotometer method. Reduced ascorbate (ASA) was increased while oxidized ascorbate (DHA) was decreased slightly for both after 1d-LN storage. And for glutathione also, reduced form (GSH) was increased while oxidized form (GSSG) was decreased slightly for both after 1d-LN storage. Consequently total phenol compound and ion leakage after LN storage showed no significant differences. Additionally root growth from the seeds after LN storage was not affected by ultra low temperature. From the above results, we may suggest that cryopreservation could be recommended for storage tool of ginseng seeds even with low viability also and expected to make slower seed aging process during preservation period through further study.

Evaluating Solar Light Collectors for Use in Closed Plant Production Systems (폐쇄형 식물생산 시스템에서 태양광 채광시스템 연구)

  • In this study, a solar light collector that collects and transmits solar light required for crop production in a closed plant production system was developed. The solar light collector consisted of a Fresnel lens for collecting solar light, and a tracking actuator for tracking solar light from sunrise to sunset to increase the light collection efficiency. The optical fiber that transmitted solar light was made of Glass Optical Fiber (GOF), and it had an excellent optical transmission rate. After collecting the solar light, the amount of light was measured at 5, 10, 15, 20, 25, and 30 cm distances from the GOF through the darkroom by using a light sensor logger connected to a quantum and pyranometer sensor. Compared with solar light, the light intensity of pyranometer sensor measured at 5 cm was 114% higher than solar light, and 61% at 10 cm. In addition, it was observed that it is possible to transmit the necessary amount of light for growing crops up to about 15 cm (as over 22%) through GOF. Therefore, adding diffusers to the solar light collector should be expected to replace artificial light in plant factories or plug seedlings nurseries for leafy vegetables. More studies on the solar light collection devices and the light transmission devices that have high light collection efficiency should be conducted.

Expression Patterns of SQS in Different Tissues in Amaranth Grains (Amaranthus cruentus L.)

  • To date, there have been no reports on the cloning and characterization of a gene encoding SQS from Amaranthus, although there have been some reports on methods of extracting and purifying squalene from Amaranthus seeds. In this study, we monitored the expression pattern of the amaranth SQS gene in seeds at different developmental stages and in different tissues. The transcript expression pattern of the SQS gene was investigated using total RNA isolated from seeds at different stages of development. There were low levels of SQS transcripts at the early stage of seed development, and the levels remained low until the middle developmental stage. The expression of SQS increased rapidly to reach a peak at the mid-late developmental stage, and then declined dramatically. This pattern of expression was consistent with the results of RT-PCR analyses. All RNA samples generated a fragment of the expected size (183-bp). The amaranth SQS was expressed at low levels during the initial to middle stages of seed development, and its expression level increased at the mid-late development stage. Also The tissue-specific expression of amaranth SQS was determined by quantifying its mRNA in total RNA isolated from the leaves, petioles, stems, and roots of seedlings at the four- and six-leaf stages. Using qRT-PCR and RT-PCR analysis, we detected amaranth SQS transcripts in some of the tissues at the six-leaf stage, but in none of the tissues from plants at the four-leaf stage. SQS transcripts accumulated in almost equal amounts in stems and roots, while a lower level accumulated in leaves and petioles during seedling development at the four- to six-leaf stages. This study provides useful information about the molecular characterization of the SQS clone isolated from grain amaranth. A basic understanding of these characteristics will contribute to further studies on the amaranth SQS.

Evaluation of Drought Tolerance for Biomass Production of Salix gracilistyla Miq.

  • Salix gracilistyla is widely distributed along riversides in Korea and very good for biomass production by SRC because of its excellent germination ability, but it is necessary to measure drying tolerance for cultivation. The drought tolerance of S. gracilistyla was tested using cuttings, and growth and physiological analysis were performed after irrigation was stopped. The growth inhibition of S. gracilistyla was observed from the day irrigation was stopped, and the soil moisture content decreased to less than 10% on the 25th day after irrigation was stopped. Over 50% of the seedlings turned brown 25 days after watering was stopped. The chlorophyll content of S. gracilistyla decreased dramatically after 25 days of stopping of irrigation. RWC values were unchanged until day 12 after irrigation was stopped but decreased rapidly until day 21, but there was a slightly decreasing trend after that. RWL levels increased slightly during irrigation stops. The proline content of plants subjected to drought stress was 0.91-2.63 mg/0.05 g, 2.75 times higher than that of the control treatment. The sugar content of the drought stress treatment group was 29.77 to 350.66 mg/0.05 g, which increased 12.24 times that of the control treatment. As a result of this study, S. gracilistyla was found to have a drought tolerance almost comparable to that of evergreen broad-leaved trees growing on the land. This study is expected to contribute to the resource utilization S. gracilistyla, a native willow tree of Korea, and the mass production of biomass by SRC.

Distributional Characteristics, Population Structures and Fruition Dynamics of Korean Endemic plant, Prunus choreiana H. T. Im (한국특산 복사앵도나무(Prunus choreiana H. T. Im)의 분포특성, 개체군구조 및 결실동태)

  • Following the adoption of the global plant conservation strategies at the Conference of the Parties for Biodiversity Conservation, diligent actions to achieve each targets are actively carried out. In particular, the need for ecological conservation research to achieve targets 2 and 7 of GSPC-2020 has increased. The priority taxa to accomplish the objectives of GSPC-2020 are rare and endemic plants. In particular, endemic plants with limited distribution in specific regions are evaluated to face a high risk of extinction. To address the necessity to preserve endemic plants, we investigated the distribution of Prunus choreiana H. T. Im, a Korean endemic plant. After that, we examined the vegetational environment of the habitat of P. choreiana and evaluated its population structure. The productivity of its fruits and the effects of pollinators on fruit production were evaluated as well. The fruiting ratio was calculated based on the number of flowers produced. Lastly, we observed the annual growth characteristics of P. choreiana. The habitats of P. choreiana did not show a specific type of vegetation. All of them were located in a limestone area of Gangwon-do in the central Korean Peninsula and occupied a site where the coverage of the tree layer and the sub-tree layer was not high or did not exist. The population structure of P. choreiana contained a high proportion of mature plants capable of producing fruits and a low proportion of seedlings and Juvenile plants. We found that the production of fruits required pollinators and was affected by the performance of each plant. Although P. choreiana produces many flowers, only a maximum of 20% and only 2-6% on average bear fruits. These flowering characteristics may be due to pollinators' low abundance and activity during the flowering season (between mid-March and early April), suggesting that many flowers are needed to attract more pollinators. We rarely observed the re-establishment of seedlings in the population of P. choreiana. Despite that, we predict the population to persist owing to its long lifespan and periodic production of numerous fruits. However, if the tree layer and sub-tree layer in competing status with P. choreiana increase their crown density, they are expected to inhibit the growth of P. choreiana and affect the risk of its extinction. Therefore, the current changes in the vegetational environment of the habitats are expected to decrease the number and extent of P. choreiana in the long term. The results of this study may serve as primary and important data necessary for the achievement of GSPC-2020 objectives.

Growth Response of Pinus rigida × P. taeda to Mycorrhizal Inoculation and Efficiency of Pisolithus tinctorius at Different Soil Texture and Fertility with Organic Amendment (리기테다 소나무의 균근(菌根) 접종(接種) 반응(反應)과 토양비옥도(土壤肥沃度)에 따른 모래밭 버섯의 효과(効果) 및 그 생태학적(生態學的) 의미(意味))

  • Potted, germinating Pinus rigida ${\times}$ P. taeda seedlings were inoculated with Pisolithus tinctorius (Pt) ectomycorrhizal fungus to test the effectiveness of Pt in relation to organic amendment and changes in soil fertility and soil texture. Pt was cultured as mycelia in vermiculite-peat moss mixture with nutrients and added to sterilized pot soils with or without organic amendment (fully fermented compost) at three soil texture levels (sand, loamy sand, and sandy loam) in a factorial design. Plants were grown in a greenhouse for 4 months and harvested to compare their growth with non-mycorrhizal plants and plants infected by natural fungi. Regardless of sod texture, soil fertility, or organic amendment, seedlings inoculated with Pt were better in dry weight and height than non-mycorrhizal plants or those infected by natural fungi. An exception was observed in the most fertile soil (0.075% N and 1.32% organic matter content in sandy loam with organic amendment), where non-mycorrhizal plants were slightly bigger (8%) and heavier (18%) than Pt-inoculated plants. In over-all average, Pt-inoculated seedlings were 30% taller and 107% heavier than those infected by natural fungi and 31 % taller and 60% heavier than non-mycorrhizal plants. Growth stimulation of seedlings by Pt was more pronounced in less fertile sand soil when organic was not amended. Mycorrhizal frequency of Pt (% of mycorrhizal root tips) was reduced to about half (from 84 to 33% in sandy loam and from 77 to 40% in loamy sand) by organic amendment, while that of natural fungi was not significantly affected. Severe nitrogen deficiency was observed in the needles of non-mycorrhizal plants (1.38% N), while both Pt-inoculated plants (1.68% N) and those infected by natural fungi (1.89% N) did not develop symptom, suggesting an active role of mycorrhizae in absorption of soil nitrogen. Top to root ratio increased with organic amendment to non-mycorrhizal plants, but was not significantly affected by fungal treatment. It was concluded from this study that relative effectiveness of Pt was determined by soil fertility. Organic amendment to less fertile sand soil increased effectiveness of Pt, while the same amendment to more fertile loamy sand and sandy loam decreased effectiveness of Pt. Benefits of Pt mycorrhizae would be expected most either when organic was not added to the soil, or when soil nutrients were not abundant.

Distribution and Stand Dynamics of Subalpine Conifer Species (Abies nephrolepis, A. koreana, and Picea jezoensis) in Baekdudaegan Protected Area (백두대간 보호지역 내 아고산 침엽수종(분비나무, 구상나무, 가문비나무)의 분포 현황과 임분 변화 특성)

  • Data from an investigation of vulnerable conifer species in the subalpine zone in Korea obtained by the Korea Forest Service in 2017-2018 and monitoring research conducted by the National Institute of Forest Science since 2014 were used to analyze the status of distribution and growing condition of three major conifer species (Abies nephrolepis, Abies koreana, and Picea jezoensis) in the subalpine zone in the Baekdudaegan protected area. The distribution area of the studied species in the Baekdudaegan protected area was ca. 74% (8,035 ha) of the total distribution area in Korea, indicating that Baekdudaegan is a core area for conservation and restoration of subalpine conifer species. From decline index [A. nephrolepis in Mt. Taebaeksan and Mt. Deogyusan increased by 77.3% and 29.6%, respectively; A. koreana in Mt. Jirisan (Chunwangbong Peak) increased by 45.2% in four years; and P. jezoensis in Mt. Jirisan (Chunwangbong Peak) increased by 47.8% in two years] and seedling frequency (lower frequency of newly recruited seedlings than dead seedlings) results, the studied species are expected to face difficulties in sustainability. In contrast, at Mt. Seseoksan and Chunwangbong Peak in Mt. Jirisan, the health of trees and seedling frequency showed a partial tendency to recover and increase. In addition, we identified the relationship between the decline index and seedling frequency. These results will support the implementation of conservation strategies for vulnerable conifer species in the subalpine zone.

Effects of the Mixing Ratio of Substrates and Cuttings on the Growth of Virus-free Sweetpotato Seedlings in Hydroponic Bag Culture (고구마 무병묘 자루재배시 배지의 혼합 비율 및 삽수의 종류가 생육에 미치는 영향)

  • This experiment was carried out to investigate the growth characteristics of virus-free sweetpotato in hydroponic bag culture. The rooted one-node and upper-shoot cuttings of 'Yeonhwangmi' and 'Mannamiwere' transplanted into the plastic bag (0.5 mm of thickness, $W300{\times}L1,200{\times}H120mm$) culture system filled with commercial horticultural media (TKS-2, Flora Gard Co.) and cocopeat + perlite (3 : 7, 5 : 5, 7 : 3, (v/v). Nutrient solution of National Horticultural Research Station in Japan was separately irrigated $3times{\times}30minutes$ per day (10:00, 14:00 and 20:00 with timer control) by the drip irrigation. The growth of two varieties was not significantly different among four substrates in 15 days, but it was excellent in commercial horticultural media (TKS-2) and cocopeat 3 : perlite 7 (v/v) in 20 days. The growth in 'Yeonhwangmiwas' promoted than that in 'Mannami', and the upper-shoot cuttings of 'Yeonhwangmi' and 'Mannami' showed a significant vine elongation (over 10 and 2 cm, respectively) compared to the one-node cuttings. Fresh weight and dry weight in cocopeat 3 : perlite 7 (v/v) were significant increased than that in cocopeat 7 : perlite 3 (v/v). Therefore, the bag culture system filed with cocopeat 3 : perlite 7 (v/v) was successfully applied on propagation of virus-free sweetpotato seedling, and the result expected that the bag culture system could be commercialized without high financial costs for farmers.

Analysis of Research for the Actual State and Management of Automated Horticultural Facilities (경북지역 현대화 원예시설의 관리실태 조사분석)

  • This study was carried out in order to understand the plan, design, constructing and actual condition of management of modernized horticultural facilities in Kyungpook Province which had been constructed from 1992 to 1995 funded by Government support. The aim of this study is to provide reference data for success of the forth project. It was performed by making up a question about driving of project and management condition of equipment after constructing. The results obtained from this study are as follows: 1. 73.5% of facilities horticulture farmhouse recognized that the prospect of greenhouse is bright, but 92.5% of the farmhouse also recognised that they need technical consultation on protected horticulture farming. Therefore, technical educations would must be enhanced about foundation of greenhouse and cultivation technique. 2. The holding times of explanatory meetings, cause of understanding to farmhouse, were one or two times in greenhouse construction, and 62.5% of the farmhouse expressed the insufficiency at the explanation and educational data. For this reason, it was judged that the construction contract had been delayed more than 5 months in 49.3% of the farmhouse after the decision of project budget. 3. In constructing after a contract, the rates of construction delay is 53.4% and defect occurrence is 41.1%. The biggest reasons of construction delay was insufficiency of worker and materials supply. Each percentage is 29.1%. And the reason of defect occurrence is badness of machinery equipment(62.9% ). 4. In management of greenhouse, a pipe-constructed plastic film greenhouse changes plastic film every one and three years because of sticking dust on plastic film. It was needed to about in cleaning technique of coverings. Because that used 3-5 years only half of the expected life span. 5. The order of broken rating in the subsidiary equipment is like this lollop ventilator (42.8%), a general control system(33.3%) especially, in the case of a general control system, the rate of all family can control is 52.7%. so, it is time to develop easy control equipment which every one could use as soon as possible. 6. When choose heat generator as decide capacity, the most priority is the mount of heat generator the percent is 45.5% heat generator and as decide model, the private purchase's percent is 77.3%. It is higher than a public bidding heat generator the percent is 22.7% heat generator when it compare with a public bidding. In the case of $CO_2$ generator, using rate is only 19.0%. The using rate is very low, so it needs education how to use depends on the way of the subsidiary equipment. 7. In the case of seedlings, it is asked to use factory-processed seedling effectively. because it's difficult to get security of labors(58.8%), hoped crops (55.9%) access same crops(29.4%) much more and changing of crops depends on market situation. that is the main reason the lack of knowhow.
