Carboxymethyl Chitosan Promotes Migration and Inhibits Lipopolysaccharide-Induced Inflammatory Response in Canine Bone Marrow-Derived Mesenchymal Stem Cells

  • The study was conducted to evaluate the effects of carboxymethyl chitosan (CMC) on proliferation, migration, and lipopolysaccharide (LPS)-induced inflammatory response in canine bone marrow-derived mesenchymal stem cells (BMSCs). The proliferation and migration of BMSCs were examined after treatment with CMC. The effect of CMC on the mRNA expression of inflammatory cytokines, such as interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α, IL-10, and transforming growth factor (TGF)-β, was also evaluated by reverse transcription polymerase chain reaction (RT-PCR). In the proliferation assay, no significant changes were found at all CMC concentrations compared with controls. The migration assay showed that CMC dose-dependently stimulated the migration of BMSCs in normal and LPS-treated conditions. RT-PCR showed that TNF-α and IL-10 expressions were suppressed in the BMSCs after CMC treatment. However, other genes were not affected. Taken together, CMC promoted BMSC migration and inhibited TNF-α and IL-10. Therefore, CMC may be possible to regulate wound healing when mesenchymal stem cells are applied in inflammatory diseases.

쥐에서 Sodium Carboxymethylcellulose 및 Dextran 70의 유착방지 효과 (Effects of Sodium Carboxymethylcellulose and Dextran 70 on Prevention of Adhesion in Rats)

  • The 2% Sodium carboxymethylcellulose(CMC) and 10% Dextran 70 solutions were evaluated for the prevention of postoperative adhesion formation in rats. The artificial injuries(2cm longitudinal incision and suture) were performed on the colon and uterine horns in the rats. 2% CMC and 10% dextran solutions were applied to the injured surfaces and peritoneal cavity in the rats. Ten days later, there were significant difference(p<0.01) in the prevention of adhesion formation between control and treated groups(2% CMC and 10% dextran 70), but the,$\boxUl$ was no significant difference between 2% CMC and 10% dextran 70 treated groups. The increment rate of body weight in the 10% dextran 70 treated group was lower than 2% CMC and control groups(p<0.05.). A positive correlation was found between the severity of adhesion formation and the plasma fibrinogen concentration in blood.

알로인이 함유된 창상치료제용 PVA/CMC 하이드로젤의 개발 (Development of Aloin Loaded PVA/CMC Hydrogel for Wound Healing)

  • 알로인이 첨가된 하이드로젤을 제조하여 창상치유의 효과를 관찰하였다. Poly(vinyl alcohol)(PVA)과 carboxyl methyl cellulose(CMC)를 동결-융해 방법으로 결합시킨 하이드로젤을 제조하였다. 젤의 특성과 약물의 방출, in vivo 창상치유 실험 및 병리학적 평가를 진행하였다. 쥐 등에 만든 상처의 치유 실험은 알로인이 첨가된 PVA/CMC(5:5) 하이드로젤 그룹과 대조군 그룹의 알로인이 함유된 카보머934 젤 혹은 순수 카보머934 젤로 처치한 그룹보다 CMC의 수분과 알로인의 피토케미칼 활성 때문에 빠른 치유력을 확인할 수 있었다. 결론적으로 알로인이 첨가된 창상용 드레싱으로 5% PVA, 5% CMC 그리고 0.125% 알로인의 조성은 상처치료에 효과적이며 창상용 드레싱으로 활용을 기대할 수 있다.

  • 이 연구는 글리시딜 숙시네이트 양이온 제미니 계면활성제의 합성에 관한 것이다. 제미니 계면활성제는 2-메틸 디소디움 숙시네이트와 에피클로로히드린, N, N-디메틸 도데실 아민을 사용하여 합성하였다. FT-IR과 $^1H$-NMR을 통해 물성을 확인하였다. 표면장력과 cmc 측정, 유화력 및 기포력을 각각 주어진 조건을 이용하여 측정하였다. 제미니 계면활성제의 표면장력은 33~34 dyne/cm이며 cmc 값은 $10^{-4}{\sim}10^{-3}mol/L$ 였다. 제미니 계면활성제의 유화력은 유기용매에서 더 우수하였고 기포력이 안정한 것을 확인하였다.

급식 대상 유형과 위탁급식전문업체 규모별 고객 만족도에 영향을 미치는 서비스 품질 속성의 규명 (Identifying the Effect of Service Quality Attributes on an Overall Customer Satisfaction by the Foodservice Type and the Contract Management Company(CMC) Scale)

  • The purposes of this study were to a) measure the service quality attributes of foodservice type such as school foodservice, hospital foodservice and business & industry(B&I) foodservice, managed by contract management company(CMC), b) compare with service quality attributes by CMC scale, c) analyze overall customer satisfaction(CS) by the foodservice type and the CMC scale, and d) identify the effect of service quality attributes on an overall CS by the foodservice type and the CMC scale. The questionnaires were handed out to 6,620 customers of 207 school, 38 hospital, and 86 B&I foodservices in 108 CMCs. The statistical data analysis was completed using SPSS Win(ver 12.0) for descriptive analysis, t-test, reliability analysis, and multiple linear regression analysis. From an analysis on service quality attributes, 'proper arrangement of table and chair at hall distribution(3.53)', 'operation of nutrition education(3.50)' were highly perceived to student, 'correctable serving(4.08)', 'serve at fixed distribution time(4.08)', 'kindness of serving employee(4.04)' were highly perceived to patient, 'employee's kindness(3.84)' were highly perceived to customer of B&I. In comparison of service quality attributes by CMC scale, most scores of large enterprise(LE) were significantly higher than small and medium sized enterprise(SME) in school foodservice, hospital foodservice and B&I foodservice. Overall CS levels were 3.53 out of a maximum 5 on B&I, 3.46 on school, and 3.44 on hospital and were evaluated differently CS score by CMC scale. Finally, regression results for the effects of service quality attributes on overall CS by each of foodservice type were identified significantly different service quality attributes by foodservice type such as school, hospital, B&I(p<.001) and by CMC scale. For considering the goal of enterprise on profit-making through CS and the needs of customer on CS at moment of truth(MOT), the findings should be applied to the CMC and the foodservice industry.

발효빵에 첨가한 Carboxymethyl Chitosan이 품질에 미치는 영향 (Effect of Carboxymethyl Chitosan on Quality of Fermented Pan Bread)

  • 식빵을 직접반죽법에 따라 제조하면서 원료의 혼합공정에 carboxymethyl chitosan (CMC)를 처리하여 제품의 품질수명을 연장시키는데 미치는 영향요인 중 노화도, 항균성 및 관능적 특성, 수분활성도를 조사한 결과는 다음과 같았다. CMC로 처리한 처리구 C-I과 C-II는 대조구 C-0에 비하여 노화가 지연되었으며, 특히 빵의 저장 중 부패의 원인이 되는 Aspergillus, Penicillium 등과 같은 곰팡이의 생육을 억제시켰다. 또한 평점법에 따라 관능검사를 실시한 결과 향, 이취, 전체적인 기호도는 저장기간동안 유의적인 차이를 나타내지 않았으나, 조직감은 C-0와 C-II 처리구 간에는 5%수준에서 유의적인 차이를 보였다.

  • The effect of pasting agent on the defibrillation of Tencel fabric was investigated. It was evaluated by the weigth loss of fabric when the fabric was treated with cellulase containing various kinds of pasting agents. The surface appearance of Tencel fabric was checked by SEM. Under the treatment condition without pasting agent, the weight loss of fabric was high at pH 5.0 and $60^\circ{C}$. This means that the cellulase activity was high at this condition. By increasing the concentration of carboxymethyl cellulose(CMC), the weight loss of fabric was decreased monotonously. This tendency was not appeared in other pasting agents. CMC is synthesized by the reaction of chloroacetic acid and cellulose. The glucose units may be remained after the reaction. So, it was considered that the degradation of glucose unit in Tencel was decreased, because cellulase had to attack both Tencel and CMC.

쥐에서 Carboxymethylcellulose 및 Ibuprofen을 이용한 유착형성 방지에 관한 연구 (Prevention of Adhesion Formation by Use of Carboxymethylcellulose and Ibuprofen in Rats)

  • With the use of a rat surgical model, the ability of carboxymethylcellulose and ibuprofen in the reduction of abdominal adhesion was examined. Seventy seven female rats were randomly divided into 7 groups : (1) control, (2) 2% CMC, (3) 3% CMC, (4) ibuprofen 25mg, (5) ibuprofen 50mg, (6) combination of ibuprofen 25mg and 2% CMC and(7) combination of ibuprofen 50mg and 3%, CMC. Following induction of abrasion injuries on ileum, colon and both uterine horns with a surgical blads, the rats in groups (2), (6) were infused with 2% CMC solution singly or in combined Infection of 25 mg/kg of ibuprofen for three consecutive days, the rats In groups (3), (7) were infused with 3% CMC solution singly or In combined Injection of 50mg/kg of ibuprofen for three consecutive days. The rats in groups (4), (5) were injected only with 25 mg or 50 mg/kg of ibuprofen for three consecutive days. After 10 days the abdominal cavities were opened and the appearance of formed adhesion were graded. The changes of body weight, CBC and blood chemicals were also evaluated at 3, 6 and 10 days after operation. In ileum, the rats in the groups (2), (6) and (7) showed less adhesion formation. In colon, there were significant differences(p<0.05) in adhesion formation in all treated groups as compared to control. In both uterine horns, there were significant decrease(p<0.05) of adhesion formation in groups(2), (6) and (7) in comparison with other groups. The increasing rate of body weight was evident in group (3) and fibrinogen concentrations at 6 and 10 days revealed significant decrease (p<0.01) in group (7), whereas there was no consistent change in CBC and blood chemicals. Therefore, it can be sugested that the infusion of 2% CMC solution with or without the injection of 25 mg/kg of ibuprofen and 3% CMC solution with the injection of 50 mg/kg of ibuprofen are effective and safe following abdominal surgery,

랫드에서 고농도의 Carboxymethyl Chitosan이 복벽 유착 예방에 미치는 영향 (Effect of high concentrations of carboxymethyl chitosan on preventing formation of peritoneal adhesion in rats)

  • The objective of this study was to determine the effective concentration of carboxymethyl chitoaan(CMC) solution on preventing intraperitoneal adhesions. In this study, 60 rats were divided into four groups : an untreated control group and three experimental groups that were treated with 3 ml of 3, 5, and 7% CMC solution. Adhesions were induced by suturing both the ileal serosa and peritoneum scraped until petechial bleeding occurred. CMC solutions were instilled intraperitoneally immediately before closure of the peritoneomuscular layer. The tensile strength of formed adhesions was measured by using a computerized tensiometer. Histopathological changes of adhesions were evaluated by hematoxylin-eosin for light microscopic examination and Massons's trichrom staining for collagen fibers. All of CMC solutions reduced significantly the tensile strength of the adhesions. CMC inhibited the increase of inflammatory cell infiltration and damage of adhesion spot, and accelerated the collagen synthesis and neovascuiarization. No significant differences were observed among all of CMC concentration at inhibiting adhesion formation. Therefore, we could conclude that 3% CMC, the lowest concentration adopted in this study, was appropriate to prevent the formation of peritoneal adhesion.

흰쥐에서의 카르복시메칠셀룰로오스나트륨과 겔란검 혼합겔의 유착방지 효과 (Prevention of Postsurgical Adhesions with a Mixed Gel of Sodium Carboxymethyl Cellulose and Gellan Gum in the Rat)

  • The aim of the present study was to investigate the efficacy of a mixed gel formulation composed of sodium carboxymethyl cellulose and gellan gum (Na-CMC gel) for the prevention of adhesions after laminectomy. The anti-adhesive effect of the Na-CMC gel was tested in a controlled randomized study using an animal model of lumbar laminectomy. The animals (60 female Sprague-Dawley rats) were randomly allocated into two treatment groups to receive the Na-CMC gel on the injured area or no gel (control). The incidence of adhesions and their grade were blindly evaluated at 4, 8 and 12 weeks after surgery. The amount of scar tissue and tenacity were grossly reduced by the Na-CMC gel at postoperative 4, 8, and 12 weeks. The mean adhesion scores were 0.75, 1.25, and 1.38 at 4, 8, and 12 weeks in the gel-treated group, respectively. No significant inflammatory reaction was observed and the healing of wound was not affected by the Na-CMC gel. The Na-CMC gel reduced the amount of scar formation and tenacity in rat laminectomy model without affecting the healing of operation wound and other complications. Therefore, the Na-CMC gel may be the potential to prevent postsurgical adhesions in clinical state.
