측백나무 열매 추출물의 항균활성 (Antimicrobial Activities of Organic Extracts from Fruit of Thuja orientalis L.)

  • This study was carried out to investigate the antimicrobial activities of organic extracts obtained from the fruit of Thuja orientalis L. The native fruits in Korea were collected and extracted by 80% ethanol, and the extract was sequentially fractionated with n-hexane, chloroform, ethylacetate, and butanol. The fraction yields of n-hexane, chloroform, ethylacetate, butanol and water of ethanol extract were 10.15%, 10.05%, 1.45%, 45.35% and 27.55%, respectively. n-Hexane-soluble fraction showed the highest antibacterial activity against gram positive bacteria, while the chloroform, ethylacetate, butanol and aqueous fractions did not show any antibacterial activity. Minimum inhibitory concentration (MICs) on Staphyloycoccus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction were $100\;{\mu}g$, $500\;{\mu}g$ and $50\;{\mu}g$/disc, respectively. The antibacterial activity was not destroyed by heating at 80, 100, $120^{\circ}C$ for 30 min and was not affected by pH. In the inhibitory test against the Staphylococcus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction showed potent growth inhibition at the concentration of 0.1 and $0.5\;{\mu}g/mL$ for 12~24 hours and n-hexane-soluble fraction did not show any mutagenic activity.

Anthraquinones with Immunostimulating Activity from Cassia tora L.

  • Many of plants had been reported having immunostimulating activity. This study reports the immunostimulating activity of Cassia tora L. (Leguminosae) seed, by means of solvent extraction method. Ethanol extract and solvent fractions, n-hexane, chloroform, ethylacetate, n-butanol and aqueous layer of Cassia tora L. seed were tested for immunostimulating activity in vitro. The ethylacetate-soluble fraction caused significant inhibition on the production of nitric oxide by murine macrophages (RAW 264.7), and mouse splenocytes were also stimulated at the concentration of 10 pgfmL. Three anthraquinones, chrysophanol (1), isochrysophanol (2) and aloe-emodin (3) with immunostimulating activity were isolated from the ethylacetate-soluble fraction of Cassia tora L. seed through activity-monitored fractionation and isolation method. These results permit Cassia tora L. to be useful as one the of natural immunostimulating crops.

유용식물로부터 Human Low Density Lipoprotein(LDL)에 대한 항산화제의 탐색 (Screening of Antioxidative Compounds toward Human Low Density Lipoprotein (LDL) from Useful Plants)

  • 본 연구는 유용식물로부터 DPPH free radical scavenger및 사람의 low density lipoprotein(LDL)의 산화에 대한 항산화 효과를 조사하기 위하여 25종의 식물로부터 메탄올로 추출하여 조사하였다. 각종 유용식물 중 항산화 활성은 우수한 달맞이꽃이 가장 높았으며 다시 메타놀로 추출하여 조사한 결과 $\alpha$-tocopherol과 항산화능이 거의 비슷하였다. 메타놀 추출물을 ethylacetate로 추출한 획분이 항산화 활성이 높았으며 ethylacetate 층 중 acidic soluble 획분을 다시 분리한 후 Sepadex LH-20 column chromatography로 추출한 결과 Fraction F-2가 전자공유능이 가장 높았다. 따라서 항산화 활성이 높은Fraction, F-2의 구조를 동정한 결과 3,4-dihydroxybenzoic acid와 3-hyoxycinnamic acid로 확인되었다.

경상도 전통마른오징어 식해의 향기성분 및 기능성 (Functional and Volatile Flavor Compounds in Traditional Kyungsando Squid sikhe)

  • 경상도 전통마른오징어 식해를 제조하여 $5^{\circ}C$에서 보관하면서 GC와 GC-MS를 이용하여 향기성분의 원인 물질을 분석, 동정하였다. 식해에서 동정된 물질 중에서 ${\alpha}-zingibirene$이 19.73 mg/kg으로 전체상대 농도에서 가장 높았으며 (Z)-Di-2-propenyl disulfide, ${\alpha}-curcumene$, methyl alkyl disulfide, $(E-E)-{\alpha}-farnesene$, pentanol, z-citral, $3-ethyl-1,2-dithi-5-ene-{\beta}-elemene$, acetic acid 및 ${\beta}-phelland$ rene의 함량 순으로 동정되어 이러한 성분들이 식해의 주요성분으로 나타났다. 식해의 향기성분은 hydrocarbone류가 49종, 알데히드류가 15종, 알콜류가 33 종, 케톤류 및 에스테류 11 종을 포함하여 총 162 종을 동정하였다. 식해의 메탄올 추출물을 용매 분획하여 얻은 각 분획물의 전자공여능을 측정한 결과 $200\;{\mu}m$ DPPH radical을 50% 환원시키는데 필요한 $SC_{50}$ 값이 헥산과 물층에서는 효과가 나타나지 않았으며, ethylacetate 층은 $310.64\;{\mu}g/mL$, butanol 층은 $1096.49\;{\mu}g/mL$으로 나타났다. 혈압상승 억제효과를 살펴본 결과 헥산과 물 분획물에서는 전혀 효과가 없었으며 ethylacetate층에서는 $IC_{50}$이 1.623 mg/mL, butanol 층에서는 1.303 mg/mL의 저해효과를 나타내었으며, xanthine oxidase에 대한 $IC_{50}$은 ethyl acetate 층은 3.591 mg/mL, butanol 층은 2.083 mg/mL로 나타났다.

수용성 Silica에 대한 항균성물질의 흡착 및 내열성 (Adsorption and Thermostability of Antimicrobial compounds on Water-soluble Silica)

  • 항균성 물질이 함유된 투명한 신기능 포장필름의 제조를 위하여 흡착제로서 수용성 silica를 선택하여 항균성 물질의 흡착 및 내열성을 조사하였다. Methanol 자화 방선균 (MO-16, MO-17)이 생산하는 천연 항균성 물질을 ethylacetate로 추출, 농축하여 silica에 흡착시킨 후 열처리한 결과 15$0^{\circ}C$에서 5분까지 항균효과가 유지되었다. 따라서 투명필름의 제조시에 흡착제로서 silica의 첨가가 효과적이라고 판단되었다. 항균제인 benzoic acid(BA) 는 세균 및 곰팡이에 강한 항균효과를 나타내었으며, 특히 곰팡이에 대한 항균력이 우수하였고, 10$0^{\circ}C$, 5분간 열처리시에도 항균력을 유지하였다. 또한 항균성 물질과 공동 첨가시 약간의 상승효과를 나타내어 필름 제조시 공동 첨가하므로서 첨가량의 감소에 따른 비용의 절감 및 항진균 효과가 보완된 필름제조가 가능하다고 사료되었다.

소나무수피 프로안토시아니딘(Proanthocyanidin)의 분리 및 구조분석 (Isolation and Structure Elucidation of Proanthocyanidin in Bark of Pinus densiflora)

  • To elucidate the structure of procyanidin in Korean red pine (Pinus densiflora S. et Z.), bark, the extractives were extracted with acetone-water mixture(7:3, v/v) from inner bark of Korean red pine. The extracts separated three fractions which were extracted by liquid-liquid extraction. The extracting solvents were chloroform and ethyl acetate and water. The part of ethylacetate soluble was chromatographed by liquid chromatography. The ethylacetate soluble portion yielded four natural procyanidin dimers, two known epicatechin-($4{\beta}{\rightarrow}6$)-catechin, catechin-($4{\alpha}{\rightarrow}8$)-catechin and two unknown catechin-($4{\beta}{\rightarrow}6$)-catechin and conformational isomer of epicatechin-($4{\alpha}{\rightarrow}6$)-catechin. The additional catechins was also isolated. The structures of these procyanidins were elucidated by their $^{13}C$-NMR spectra.

벼자나무 잎의 추출성분 (Extractives from the Needles of Torreya)

  • 비자나무 잎을 아세톤-물의 혼합용액으로 추출하고 n-hexane, chloroform, ethylacetate 및 물 분획으로 분류하여 동결건조하고 분말상 화합물로 조제하였다. 그 중에서 ethylacetate 용성 추출물을 메탄올-물 및 에탄올-헥산(n-hexane)의 혼합용액을 사용하여 Sephadex LH-20 칼럼상에서 크로마토그래피를 수행하였다. 비자나무 잎의 추출성분은 대부분 (+)-catechin, (-)-epicatechin, (+)-gallocatechin 및 (-)-epigallocatechin과 같은 flavan 화합물과 3'-O-methyl-(+)-catechin 및 3'-O-methyl-(-)-epicatechin과 같은 flavan의 methyl ether 화합물로 구성되어 있었으며 그 이외에 benzoic acid계 화합물인 protocatechuic acid도 단리 되었다. 단리된 화합물은 $^1H$- 및 $^{13}C$-NMR, NOE와 CI-MS 분석에 의해 화학구조를 구명하였다.

오동나무 잎의 페놀성 화합물 (Phenolic compounds from the leaves of Paulownia Coreana Uyeki)

  • 강원대학교 구내림에서 오동나무 잎을 채취하여 아세톤-물(7:3, v/v)로 추출하고 n-hexane, methylene chloride, ethylacetate 및 $H_2O$ 등 네 개의 분획으로 분리한 후 에틸아세테트용성 분획물을 Sephadex LH-20으로 칼럼 크로마토그래피를 수행하였으며 용리용매는 메탄올 수용액과 에탄올-헥산 혼합액을 사용하였다. 그 결과 flavonoid류 화합물인 (+)-catechin, (-)-epicatechin, apigenin, luteolin, taxifolin과 phenolic acid류 화합물인 $\rho$-hydroxybenzoic acid, vanillic acid 및 $\rho$-coumaric acid를 단리 하였다. 단리된 화합물은 NMR 및 MS 스펙트럼을 이용하여 구조를 결정하였다.

Phenolic Compounds from Japanese Anise (Illicium anisatum L.) Leaves

  • Japanese anise (Illicium anisatum L.) leaves were collected and ground after drying, then immersed with 50 % aqueous acetone for 3 days. After filtration, the extracts were fractionated with n-hexane, chloroform ($CHCl_3$), ethylacetate (EtOAc) and $H_2O$, and then freeze dried after concentration. A portion of EtOAc (3.12 g) and $H_2O$ (6.08 g) soluble fractions were chromatographed on a Sephadex LH-20 column with various aqueous MeOH solution to isolate the compounds. Compound 1 ((+)-catechin) was isolated from EtOAc soluble fraction. Compounds 2 (quercetin), 3 (quercitrin) and 4 (2''-O-rhamnosylvitexin) were isolated from $H_2O$ soluble fraction. For the first time, quercitrin (3) and 2''-O-rhamnosylvitexin (4) of the isolated compounds were obtained from the extracts of japanese anise leaves.

Extraction and Bioassay of Allelochemicals in Jerusalem Artichoke

  • Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.
